Characteristics of Biological Nitrogen Removal for Low C/N Ratio Municipal Wastewater Using Methanol as an External Carbon Source in $A_2O$ Fluidized Media Process (유동여재 $A_2O$공정에서 외부탄소원으로 메탄올을 이용한 낮은 C/N비 하수의 생물학적 질소제거 특성)

  • This experiment was performed to evaluate the characteristics of BNR system performance, behavior of pollutants as organic and nitrogen at each basin and the effects of C/N ratio on biological nitrogen removal with methanol as an external carbon source for a low C/N ratio municipal wastewater. A lab-scale $A_2O$ system by employing the aerobic basin with the fluidized polyurethane media, which was $10{\sim}20$ mm rube type like a sponge, was used. The aerobic basin was hybrid type to be suspended and fixed biomass. The obtained results from this study were as follows; When no methanol was added, suspended biomass was 3 times more than that of the fluidized media in this system(total biomass 80 g). Biomass growed by an external carbon was firstly attached on media, and then suspended. $COD_{Cr}$ concentration for the effluent was a range of 13 to 29 mg/L regardless of pouring an external carbon. For nitrogen, the effluent concentration was $20.0{\sim}35.9mg/L$(removal efficiency; 18%) in case of no addition of an external carbon, but was $2.5{\sim}9.0mg/L$(removal efficiency ; $71{\sim}83%$) with addition of methanol. For the characteristics of pollutants removal, most of $COD_{Cr}$ were removed at the anaerobic basin when no external organic carbon was added, and were removed at the anoxic basin in case of adding external organic carbon but at the aerobic basin in case of adding excess external organic carbon. On the other hand, most TIN(total inorganic nitrogen) were removed at the anaerobic basin when no external organic carbon was added, but when an external organic carbon was added, they were removed at the anaerobic basin under unstable condition and at the anoxic basin under stable condition.

Physical and Chemical Characteristics of Cotton Waste Substrate According to Fermentation Conditions for Oyster Mushroom Bed Cultivation (느타리버섯 폐면배지의 발효조건별 이화학적 특성)

  • We have surveyed the variation of physical and chemical characteristics of aerobic and anaerobic outdoor fermentation of cotton wastes using for oyster mushroom cultivation. The inner temperature of cotton wastes fermented aerobically covered with thin cloth and setting pallet at bottom was higher than that of anaerobic fermented cotton wastes covered with P.E vinyl and the maximum temperature was $75^{\circ}C$ at 5th day after fermentation. pH of cotton wastes fermented aerobically was increased up to 8.9 after fermentation of $9{\sim}12$ days, but that of anaerobically fermented was decreased up to 5.0. Total carbon content was decreased but total nitrogen content was increased when fermentation was in progress. Oxygen concentration of cotton wastes fermented aerobically was decreased until 6 days after fermentation but increased after 9 days of fermentation. Ammonia concentration of cotton wastes fermented aerobically and anaerobically was below 10 ppm and $20{\sim}85\;ppm$ respectively. In anaerobic condition the cotton wastes was contaminated with mold ($15{\sim}50%$), where no contamination was found in aerobic condition during spawn running stage. Yields of mushroom grown on cotton wastes aerobically fermented for $6{\sim}9$ days was $23.0{\sim}23.6\;kg$ per $3.3\;m^2$ area.

Characterization of ATPase Activity of Free and Immobilized Chromatophore Membrane Vesicles of Rhodobacter sphaeroides

  • The intracytoplasmic membrane of Rhodobacter sphaeroides readily vesiculates when cells are lysed. The resulting chromatophore membrane vesicle (CMV) contains the photosynthetic machineries to synthesize ATP by ATPase. The light-dependent ATPase activity of CMV was lowered in the presence of $O_2$, but the activity increased to the level observed under anaerobic condition when the reaction mixture was supplemented with ascorbic acid (${\geq}0.5mM$). Cell lysis in the presence of biotinyl cap phospholipid (bcp) resulted in the incorporation of bcp into the membrane to form biotinylated CMV (bCMV), which binds to streptavidin resin at a ratio of approximately $24{\mu}g$ bacteriochlorophyll a/ml resin. The ATPase activity of CMV was not affected by biotinylation, but approximately 30% of the activity was lost by immobilization to resin. Interestingly, the remaining 70% of ATPase activity stayed constant during 7-day storage at $4^{\circ}C$. On the contrary, the ATPase activity of bCMV without immobilization gradually decreased to approximately 40% of the initial level in the same comparison. Thus, the ATPase activity of CMV is sustainable after immobilization, and the immobilized bCMV can be used repeatedly as an ATP generator.

  • In this study, we carried out torrefaction experiment using PKS(Palm Kernel Shell), and Bagasse as a raw material of oversee of herbaceous biomass and using waste wood and logging residue as a raw material of domestic of woody biomass. And then, by analyzing the physical & chemical properties, we investigated the characteristics as a fuel. By using the result of thermo gravimetric analysis, the biomass residue was torrefied for 30 minutes at a temperature range of $250-350^{\circ}C$ in anaerobic condition. As a result, torrefied materials of moisture content are lower than raw, but of fixed carbon, calorific value and ash are higher than raw.

  • Purpose : The study objective was to assess the antibacterial activity of essential oil of basil against S. mutans and P. gingivalis and to explore its potential to prevent dental caries and peridontal disease. Method : Essential oil of basil, extracted using steam distillation, was diluted with triple distilled water and Tween 20 to generate samples at various concentration, that is 30%, 50%, and 70% (v/v). Strains of S. mutans and P. gingivalis were incubated in the medium under anaerobic condition. Broth microdilution susceptibility testing and plate incubation diffusion were utilized to determine the minimum inhibitory concentration (MIC) and to measure antibacterial activity, respectively. Result : An upsurge in antibacterial activity was seen to correlate with and increase in the concentration used for both bacterial strains, but was more significant with S. mutans. A statistically significant growth inhibition effect and reduction in the number of colonies was also observed with both strains dependent on the concentration used following 24 hours of incubation. Conclusion : Thus, the current study finding was that essential oil of basil was effective against dental caries and periodontal disease and could be used in dentifrice to help prevent oral disease.

Solubilization of Sewage Sludge by Inoculation of Lactic Acid Bacteria (유산균 접종에 의한 하수 슬러지의 가용화)

  • A new approach to the solubilization of excess activated sludge by the inoculation of lactic acid bacteria was studied to reduce the amount of sludge produced in the activated sludge treatment process. Aerobic microorganism in sludge was lysed in anaerobic condition and the cytoplasmic substance eluted was utilized as a carbon source by lactic acid bacteria. On the basis of sludge solubilization efficiency, Lactobacillus brevis and Leuconostoc mesenteroides subsp mesenteroides were selected the best candidates among five kinds of Lactobacillus sp. and seven kinds of Leuconostoc sp. The sludge solubilization efficiency by heterofermentative lactic acid bacteria was more efficient than that of homofermentative bacteria. Initial value of soluble COD (sCOD) was 1050 mg/L at the initial inoculation time increased to 3070 mg/L (192% solubilization) at 96 h of the incubation time. The inoculation of lactobacillus brevis to the sludge resulted in 2824% increase in sCOD value after 96 h of incubation than the control experiment. Leuconostoc mesenteroides subsp mesenteroides showed 152% increase of solubilization and 30% increase of S-COD/T-COD on 96 h of incubation time. Considering the increase of S-COD by the inoculation of Leuconostoc sp. on 24 h, 10% inoculation of lactic acid bacteria to the sludge was most effective.

Growth-Inhibiting Effects of Various Traditional Drinks of Plant Origin on Human Intestinal Bacteria

  • A total of 28 traditional drinks derived from 23 plant species in 19 families were tested for their in vitro growth-inhibiting effects against Bifidobacterium adolescents, Bifidobacterium bifidum, Lactobacillus acidophilus, Lactobacillus casei, Clostridium perfringens, and Escherichia coil using a paper disc agar diffusion method under anaerobic condition. The responses varied with bacterial strain, plant species and tissue sampled. In a test with C. perfringens at 5 and 10 mg/disc, potent growth inhibition was produced from the extracts of Eucommia ulmoides stems, Pinus densiflora leaves and shoots, Thea sinensis leaves (green and oolong teas) and Zingiber officinale roots. All materials tested did not adversely affect the growth of bifidobacteria, lactobacilli, and E. coli. These results may be an indication of at least one of the pharmacological activities of these plant-derived drinks.

  • With the population growth and industrialization, the characteristics of discharged waste water and sewage have become more diverse. The removal of phosphorus (P) in the wastewater is essential for the prevention of eutrophication in the river and stream. This study was performed in order to estimate the field application of the Bacillus sp. 3434 BRRJ. Bacillus sp. 3434 BRRJ was cultured in the raw wastewater and synthetic medium at the 5 L reactor. The best optimum conditions for P removal by Bacillus sp. 3434BRRJ in the synthetic medium at the 5 L reactor were as follows: temperature, $30^{\circ}C$; P concentration, 20 mg/L; carbon sources, glucose + acetate (1:1); oxygen concentration, alternatively anaerobic and aerobic conditions. P removal efficiency under the optimum condition was 89.4%. In case of wastewater, P removal efficiency was 95.5% under controlled at $30^{\circ}C$. Through this study we confirmed that P removal by Bacillus sp. 3434BRRJ in case of wastewater was as effective as the synthetic medium. It is considered that Bacillus sp. 3434 BRRJ can be applied to the treatment of wastewater in order to biologically remove P from the wastewater on a large scale.

Study on the 2,4,6-Trinitrotoluene reduction rate by mediation of extracellular material of mixed culture (혼합배양 미생물의 세포외 물질이 2,4,6-Trinitrotoluene 변환에 미치는 영향에 관한 연구)

  • 2,4,6-Trinitrotoluene(TNT) was reduced into intermediate products by mixed culture incubated in anaerobic condition. To test the effects of extracellular material to electron transfer between sulfide and TNT, filtered medium of mixed culture was loaded in the test tubes with TNT and sulfide. The transformation rate was measured under four different conditions. The rate under microbial activity was the fastest among under different conditions. With sulfide or filtrate alone and TNT, the reactions were measured as the slowest reactions or no reactions occured, respectively. The reaction rate coefficient were calculated by linear regression and the first order kinetic was fitted best. Also, the plot of rate coefficients (K$_{f}$) showed linear relationships when at time zero TNT and sulfide concentration were 20 mg/1 and 6.0 mM, respectively. By extrapolation, reaction rate coefficient of 100% filtrate could be calculated as 0.0054/minute. However, reaction rate was affected by different concentration of sulfide, so it is a dependent of sulfide concentration. The results of this test showed TNT reduction rate can be limited more by microbial reaction than by mediation of filtrate or sulfide and filtrate alone.

Effect of Minerals surface characteristics On Reduction Dehalogenation of chlorination solvents in water-FeS/FeS$_2$ system

  • FeS/FeS$_2$ minerals have been known to be potentially useful reductant to the removal of common organic contaminants in groundwater and soil. This research is aimed at improving our understanding of factors affecting the pathways and rates of reductive transformation of Hexachloroethane by catalytical iron minerals in natural system. Hexachloroethane is reduced by FeS/FeS$_2$ minerals under anaerobic condition to tetrachloroethylene and trichloroethylene with pentachloroethyl radical as the intermediate products. The kinetics of reductive transformations of the Hexachloroethane have been investigated in aqueous solution containing FeS, FeS$_2$. The proposed reduction mechanism for the adsorbed nitrobenzene involves the electron donor-acceptor complex as a precursor to electron transfer. The adsorbed Hexachloroethane undergo a series of electron transfer, proton transfer and dehydration to achieve complete reduction. It can be concluded that the reductive transformation reaction takes place at surface of iron-bearing minerals and is dependent on surface area and pH. Nitrobenzene reduction kinetics is affected by reductant type, surface area, pH, the surface site density, and the surface charge. FeS/FeS$_2$-mediated reductive dechlorination may be an important transformation pathway in natural systems.
