  • Two landfills of this study containing municipal wastes without any bottom liner and leachate treatment system have different landfill age, waste volume and most importantly different hydrogeologic settings. The one (Cheonan) is situated in an open flat area while the other (Wonju) is located in a valley. In the interior of the landfills, typical anaerobic conditions revealed by low DO and ${NO_3}^-$ concentrations, negative ORP values, high $NH_3$, alkalinity and $Cl^-$ concentrations were observed. Generally higher levels of contaminants were detected in the dry season while those were greatly lowered in the wet season. Significantly large decrease of Cl concentration in the wet season indicates that the dilution or mixing is one of dominant attenuation mechanisms of leachate. But detailed variation behaviors in the two landfills are largely different and they were most dependent on permeability of surface and subsurface layers. The intermediately permeable surface of 1.he landfills receives part of direct rainfall infiltration but most rainwater is lost to fast runoff. The practically impermeable surface of clayey silt (paddy field) at immediately adjacent to the Cheonan landfill boundary prevented direct rainwater infiltration and hence redox condition of the groundwaters were largely affected by that of the upper landfill and the less permeable materials beneath the paddy fields prohibited dispersion of the landfill leachate into downgradient area. In the Wonju landfill, there exist three different permeability divisions, the landfill region, the sandy open field and the paddy field. Roles of the landfill and paddy regions are very similar to those at the Cheonan. The very permeable sandy field receiving a large amount of rainwater infiltration plays a key role in controlling redox condition of the downgradient area and contaminant migration.

  • An electrolytic oxidation process was applied to remove odorous compounds from non-point odor sources including wastewater pipelines and manholes. In this study, a distance between the anode and the cathode of the electrolytic process was varied as a system operating parameters, and its effects on odor removal efficiencies and reaction characteristics were investigated. Odor precursors such as sediment organic matters and reduced sulfur/nitrogen compounds were effectively oxidized in the electrolytic process, and a change in oxidation-reduction potential (ORP) indicated that an stringent anaerobic condition shifted to a mild anoxic condition rapidly. At an electrode distance of 1 cm and an applied voltage of 30 V, a system current was maintained at 1 A, and the current density was 23.1 $mA/cm^{2}$. Under the condition, the removal efficiency of hydrogen sulfide in gas phase was found to be 100%, and 93% of ammonium ion was removed from the liquid phase during the 120 minute operating period. Moreover, the sulfate ion (${SO_4}^{2-}$) concentration increased about three times from its initial value due to the active oxidation. As the specific power consumption (i.e., the energy input normalized by the effective volume) increased, the oxidation progressed rapidly, however, the oxidation rate was varied depending on target compounds. Consequently, a threshold power consumption for each odorous compound needs to be experimentally determined for an effective application of the electrolytic oxidation.

  • This study was conducted to elucidate degradation pattern of two synthetic pyrethroid insecticides, bifenthrin having 2-methylbiphenyl group and cyhalothrin having ${\alpha}$-cyano benzyl ester group in theirs alcohol moiety, in two soils and aqueous media under laboratory conditions. The half-life of bifenthrin was 85.1 days and 12,4 days in Chilgok and Bokhyen soil of aerobic upland condition, respectively, and that of cyhalothrin was 54.6 days and 32.2 days. Bifenthrin and cyhalothrin were degraded very slowly under anaerobic flooded condition and sterilized. Their degradation seemed to be mainly mediated by aerobic microorganisms in soil. Bifenthrin and cyhalothrin were degraded more rapidly in Bokhyen soil with rich organic matter than Chilgok soil. Cyhalothrin was degraded 30 days faster than bifenthrin under aerobic upland condition of two soils. Cyhalothrin was degraded more than bifenthrin in alkaline solution of pH 10, but cyhalothrin and bifenthrin were degraded very slowly in acidic solution of pH 2 and 6.

When Chong Ju and Chung Ju soils possessing different physicochemical properties were treated with 500 ppm of TOK and incubated in flooded anaerobic condition for 2, 4, and 6 months, respectively, they produced 4-Chloro-4'-amino diphenyl ether, 2,4-Dichloro-4'-amino diphenyl ether(amin-TOK), N-[4'-(4-Chlorophenoxy)] phenyl acetamide, and N-[4'-(4-Chlorophenoxy)] phenyl formamide as the metabolities. This result indicates that TOK undergose the reduction of its $NO_2\;to\;NH_2$ group, dechlorination, acetylation, and formylation under this condition. The cleavage of ether linkage does not occur. In addition, TOK degrades more readily in Chung Ju soil which is characterized by pH 6.43 and higher contents of $Ca^{++}$ and C.E.C. than in Chong Ju soil which is lower in pH, $Ca^{++}$, and C.E.C. 2. In the aerobic incubation of TOK of 25ppm in Chung Ju soil suspension for 21 days, the ratio of the resulting metabolites, TOK : amino-TOK : 4-Chloro-4'-amino diphenyl ether was 100 : 130 : 76. Meanwhile, in the 42 day incubation, the ratio was 100 : 19 : 5, which indicates that TOK in aerobic condition dose not necessrily degrade as a function of the incubation period. 3. The citrate buffer extract of Chung Ju soil has the capability of degrading TOK, which was verified to be due to the action of the microorganisms involved. 4. Twelye strains of soil bacteria were isolated from the TOK-treated soils. In the incubation of TOK in pure cultures of the respective isolates, the strain T-1-1 isolated from Chong Ju soil had almost no degradability whereas the strain T-2-3 was the most potent. The degradation of TOK by the isolates constituted mostly the reduction of the nitro group to amino group. 5. In a test for the degradability of TOK by some selected microorganisms, Pseudomonas species were more potent than fungi. Yet, Isolate B which had been isolated from Chung Ju soil suspension was the most potent.

Preparation and Storage Conditions of Oleoresin from Root Portion of Peeled Garlic (박피마늘 뿌리부분을 이용한 Oleoresin의 제조 및 저장조건)

  • Extraction and storage conditions of oleoresin were studied from root portion of peeled garlic, an waste of garlic industry. Extraction with ethanol and methanol showed an high solid yields of $27{\sim}37%$ at the temperature range of $30{\sim}50^{\circ}C$. Two hours of extraction were found to be economic because of no significant increase in furthur extraction. Storage of the oleoresin under anaerobic condition such as vaccum or nitrogen resulted in a less changes in pH, total acidity, color and thiosulfinate content, than those changes under aerobic condition. Most of the thiosulfinate was decreased for all conditions after 14 days of storage at $25^{\circ}C$. Glucose and sucrose were increased and most of organic acids were decreased during storage under nitrogen condition. Addition of ascorbic acid and cysteine into oleoresin retained the garlic flavor, effectively.

Stability of Matrine and Oxymatrine from the Biopesticide from Sophora flavescens under Aquatic and Soil Environment (고삼 추출물을 주성분으로 하는 유기농자재의 alkaloid계 살충성분 2종의 토양 및 수계 노출 안정성)

  • BACKGROUND: The stabilities of the two alkaloidal insecticides of S. flavescens including matrine and oxymatrine are important factor to establish expiry date and usage manual for crop protection. However, the environmental stability of the compounds had not been studied with the extract and its commercial biopesticide. METHODS AND RESULTS: The environmental stabilities of the two alkaloids were performed with extract of S. flavescens, and its two commercial biopesticides both in controlled aquatic and soil conditions. The half-lives of the total matrines for the extract and its two commercial biopesticides were estimated over 200 days both under aerobic and anaerobic water condition. Under dry soil condition, the initial decay rates of the matrines were calculated 0.0804-0.1275 ($t_{1/2}$ 5.4-8.6 days), and the half-lives under wet soil condition were calculated 33.0-231 days. Total soil bacteria on the wet soil ranged 6.0-8.0 log CFU/g-soil during the experiments period. CONCLUSION: The aquatic mixture of the extract showed excellent stability both with the extract and its biopesticides, however, the stability of soil mixture were shorter than the aquatic mixture, suggesting that soil metal consider as a catalyst for the degradation of the two alkaloids.

Study on the Effects of Supplemented Factors on the Production of Vitamin $B_{12}$ by Propionibacterium shermanii (Propionibacterium shermanii에 의한 비타민 $B_{12}$의 생성에 영향을 미치는 배지첨가물들에 대한 연구)

  • Following the study on the fermentation conditions influencing the production of vitamin $B_{12}$ by Propionibacterium shermanii(Korean J Biotechnol. Bioeng. 7,126-131, 1992), the effects of some factors supplemented in the medium on the production of vitamin $B_{12}$ were studied. Maximum production of vitamin $B_{12}$ was observed when $Co^{+2}$ was supplemented at the concentration of 2-4 ppm in the fermentation medium. Increase of the supplemented $Co^{+2}$ to 12 ppm did not inhibit the growth of the organism, but it accelerated the lysis of the organism. In the literature, peptone was reported to activate the biosynthesis of vitamin $B_{12}$. Examination of the effect of peptone on the growth and the production of vitamin $B_{12}$ showed that at early stage more vitamin$B_{12}$ was observed in the supplemented medium, but no difference was observed in the later stage of fermentation. Examination of the time for addition and the amount of 5,6-dimethylbenzimidazole, a precursor known to influence the production of vitamin $B_{12}$, showed that a maximum yield of vitamin $B_{12}$ was observed when 15 mg/L was added to the fermentation medium after 2 days' incubation. The effect was comparable with the increase of the production of vitamin $B_{12}$ when the fermentation condition was changed to aerobic condition after 2 days' culture under anaerobic condition.

Study of Biodegradable Ability of Biodegradable Plastic in Anaerobic Digestion (혐기성소화에 의한 생분해성 플라스틱의 생분해능 검토)

  • This study is to estimate that food waste bags with biodegradable plastic are really decomposed by microorganism in composting with food waste and to examinate how biodegradable plastic affects composting. 6 kinds of 30%, 4 kinds of 100% and 2 kinds of none biodegradable plastics were used in d1is study. In 30% biodegradable plastics the highest Degradation rare is 6% in meso-condition and 10% in thermal-condition. Srain at auto break decreased to 150% in meso-condition and 120% in thermal-condition. Stress at max load were also reduced to $180kgf/cm^2$ in mesocondition and $200kgf/cm^2$ in thermal-condition. Usually, LLDPE decreased larger than HDPE in physical characreristics but HDPE is higher in degradation rate. 1n stain at auto break and stress ar max load 100% biodegradable plastic declined to 230% and to $380kgf/cm^2$ in meso-condition and to 440% and to $400/cm^2$ in thermal-condition respectively. 100% biodegradable plastics showed higher biodegradation and decomposition then 30%. They appeared clearly through SEM observation. As a result, it was not appropriate to use 30% biodegradable plastics as food waste bag because they were not decomposed perfectly. It is possible to use 100% biodegradable plastic as it but cost is too high. So development of technique is needed.

Analysis of the Factors Affecting Anaerobic Thermophilic Digestibility of Food Wastes (음식물쓰레기의 고온 혐기성 소화도에 미치는 요소에 대한 분석)

  • Serial basic tests were conducted for the determination of fundamental kinetics and for the actual application of kinetic parameter to food waste digestion with precise measurement of methane production under a thermophilic condition. The effects of food particle size, sodium ion concentration, and volatile solid (VS) loading rate on the anaerobic thermophilic food waste digestion process were investigated. Results of serial test for the determination of fundamental kinetic coefficients showed the value of k (maximum substrate utilization rate coefficient) and KS (half-saturation coefficient) as $0.24hr^{-1}$ and $700mg/{\ell}$, respectively, for non-inhibiting organic loading range. No inhibition effect was shown until $5g/{\ell}$ of sodium ion concentration was applied to a serum bottle reactor. However, the volume of methane gas was decreased gradually when the concentrations of more than $5g/{\ell}$ of sodium ion applied. All sizes of food waste particle showed the same constants (A : 0.45) but the maximum substrate utilization rate constant ($k_{HA}$) was inversely proportional to particle size. As an average particle size increased from 1.02 mm to 2.14 mm, $k_{HA}$ decreased from $0.0033hr^{-1}$ to $0.0015hr^{-1}$. The result reveals that particle size is one of the most important factors in anaerobic food waste digestion. There was no inhibition effect of sodium ion when VS loading rate was $30g/{\ell}$. And maximum injection concentration of VS loading rate was determined about $40g/{\ell}$.

Optimization of Culture Conditions for 1,3-propanediol Production from Glycerol Using Klebsiella pneumoniae (글리세롤로부터 1,3-propanediol 생산을 위한 Klebsiella pneumoniae 배양 조건 최적화)

  • To improve the productivity of 1,3-propanediol(1,3-PD) with K. pneumoniae DSM4799 using pure glycerol and crude glycerol derived from the biodiesel process, optimizing fermentation conditions was performed by changing environmental factors such as anaerobic/aerobic condition, temperature, glycerol concentration, and pH. When anaerobic conditions were maintained, there was an improved 1,3-PD production compared with that from aerobic/anaerobic 2-stage fermentation. From the results with temperature $26{\sim}37^{\circ}C$, the higher 1,3-PD production yield was observed at $30{\sim}33^{\circ}C$. For an initial glycerol concentration higher than 60 g/L, cell growth and 1,3-PD production were inhibited. When crude glycerol was used, the initial 1,3-PD production appeared to be inhibited. After 48 hr of incubation, however, 1,3-PD production with crude glycerol was even higher than that with pure glycerol, demonstrating the feasibility of 1,3-PD production using crude glycerol as a substrate. Fed-batch fermentation was applied for the high concentration of 1,3-PD without substrate inhibition. By regulating pH at 7 during the fed-batch with glycerol lower than 40 g/L, the yield of 1,3-PD was 25% higher than that without pH regulation(0.56 g/g vs. 0.45 g/g). In conclusion, based on our results, anaerobic conditions, temperature at $30^{\circ}C$, pure or crude glycerol lower than 40 g/L, and pH regulation at 7 were the optimized conditions for 1,3-PD production using K. pneumoniae DSM4799, making it more feasible to produce 1,3-PD at higher concentration and a lower price.
