  • Rhodopseudomonas sp. KCTC 1437은 글루타민산이 질소원으로 존재할 때, 포도당으로부터 질소고 정효소 (nitrogenase)에 의해 효율적으로 수소를 생성하였다. in vivo에서 질소고정 효소의 활성도를 조사해 본 결과, 암모니아 이온의 제한 농도에서 키운 균체가 다른 질소원에서 키운 균체보다 더 큰 정도로 수소를 글루타민산 존재하에서 포도당으로부터 생성하였다. 이 균주는 또한 수소 생성의 억제물질 및 조건으로 알려져 있는 암모니아 이온의 높은 농도와 암조건에서 높은 정도로 수소를 생성했는데 이것은 수소효소 (hydrogenase)의 일종인 포르믹산 수소분해효소 (formic hydrogenlyase)에 의한 것이라 고려된다.

Biochemical characterization of cotton stalks biochar suggests its role in soil as amendment and decontamination

  • Cotton is the major fiber crop in Pakistan that accounts for 2% of total national gross domestic product (GDP). After picking of cotton, the dry stalks are major organic waste that has no fate except burning to cook food in villages. Present research focuses use of cotton stalks as feedstock for biochar production, its characterization and effects on soil characteristics. Dry cotton stalks collected from agricultural field of Bahauddin Zakariya University, Multan, Pakistan were combusted under anaerobic conditions at $450^{\circ}C$. The physicochemical analysis of biochar and cotton stalks show higher values of % total carbon, phosphorus and potassium concentrations in biochar as compared to cotton stalks. The concentration of nitrogen was decreased in biochar. Similarly biochar had greater values of fixed carbon that suggest its role for carbon sequestration and as a soil amendment. The fourier transformation infrared spectroscopic spectra (FTIR) of cotton stalks and biochar exposed more acidic groups in biochar as compared to cotton stalks. The newly developed functional groups in biochar have vital role in increasing surface properties, cation exchange capacity, and water holding capacity, and are responsible for heavy metal remediation in contaminated soil. In a further test, results show increase in the water holding capacity and nutrient retention by a sandy soil amended with biochar. It is concluded that cotton stalks can be effectively used to prepare biochar.

건각탕(健脚湯) 섭취가 엘리트 장거리 달리기 선수의 유산소성 능력 향상에 미치는 영향 (Development of Herbal Drink to Improve Aerobic Capacity in Elite Endurance Runners)

  • The purpose of this study was to identify the effects of oriental herbal drink to improve aerobic capacity in elite endurance runners. 14 male elite college runners were participated and divided into two group; i ) oriental herbal drink group (OG), ii ) placebo group (PG). All subjects were completed treadmill exercise protocol using GXT at before (B) and after (A) experimental treatment of one week. The V02max, anaerobic threshold (AT) were measured by gas analysis and heart rate (HR) were measure by polar system at pre, max, post, post 30 min and post 60 min. Blood samples were collected to analyze blood components. The V02max were significantly increased in OGA compared to OGB whereas the V02max and AT in OGA were significantly higher than PGA. The blood lactate concentration were shown higher decrease rate in OGA compared to Doth OGB and PGA during recovery whereas LDH and Na were significantly increased in OGA compared to both OGB and PGA. The blood concentrations of CI and K were significantly increased in OGA compared to PGA. There were no significant differences were founded in WBC, RBC, Hct, Hb and other components associated with energy sources(glucose, TG, TC, HDL, LDL, creatinine, CPK). These results suggested that this oriental herbal drink can be administrated to improve aerobic capacity in long distance runners.

The Influences of Addition of Sugar with or without L. buchneri on Fermentation and Aerobic Stability of Whole Crop Maize Silage Ensiled under Anaerobic Silos

  • The whole plant of crop maize was chopped and ensiled in airtight 1-L capacity glass jars to determine the influence of residual sugar on anaerobic yeast growth and on the fermentation of lactic acid by L. buchneri in whole crop maize silage. There were a total of six treatments used in this experiment as follow: added 25 g de-mineralised water per kg chopped maize serving as control (con), 37.5 g glucose solution containing 12.5 g glucose ($g_1$), 75 g glucose solution containing 25 g glucose ($g_2$), 25 g L. buchneri suspension intended for $10^6$ cfu $g^{-1}$ (L.b.), $g_1+L.b.$ and $g_2+L.b.$ All silos were stored in the dark at $20^{\circ}C$ until end of experiment. Jars were opened on duplicates at day 2, 7, 14, 28, 56 or triplicates at day 91 after ensiling for measuring the pH, microbiological enumeration and fermentative products. Results indicated that acidification rates for all silages were very fast, no difference occurred among treatments before day 28. After day 28 the pH values for silages inoculated by L. buchneri. with or without sugar tended to increase especially for treated only with L. buchneri, resulting in higher (p<0.01) finial pH than uninoculated silages. Compared with control silage, the added sugar significantly (p<0.01) increased dry matter (DM) loss, L. buchneri enhanced (p<0.01) DM loss further at different sugar existence. Silages inoculated by L. buchneri only or in combination with sugar addition contained less (p<0.01) lactic acid than the correspondent silages without inoculation with L. buchneri. In comparison with control, ethanol production is about 3 or 6 fold higher due to addition 12.5 or 25 g glucose per kg chopped maize at ensiling. The added sugar resulted in less acetic acid concentration (p<0.01) than control, but inoculation with L. buchneri increased (p<0.01) acetic acid than correspondent uninoculated silages at different sugar levels. No butyric acid and propionic acid were found in uninoculted silages, silages inoculated with L. buchneri. produced more propionic acid, 1-propanol and butyric acid. Lactobacilli counts were not influenced by added sugar, but increased (p<0.01) with inoculation of L. buchneri. The added sugar increased significantly (p<0.01) the yeast count, whereas L. buchneri showed the contrary effect. No differences were found in the aerobic stability among all treatments. In conclusions, 1) the added sugars encourage the growth of yeast and yeasts convert extra sugar into ethanol in maize silages. 2) The added sugars and L. buchneri do not influence the aerobic stability of silages stored in anaerobic silos.

음식물쓰레기의 특성이 메탄생성량에 미치는 영향분석 (Effect of food waste properties on methane production)

  • 음식물 쓰레기는 산완충능(buffer capacity)이 충분하지 않아 포집과 운송기간 동안 유기산이 축적될 수 있고 몰리브덴이나 코발트와 같은 미량원소가 부족할 수 있기 때문에 가축분뇨는 음식물 쓰레기와 혼합소화 시 부족한 메탄생성량을 보충할 수 있는 혼합기질로서 적합한 것으로 판단된다. 본 논문에서는 성공적인 혐기공정 설계 및 안정적 운영을 위한 우리 나라 유래 대상기질의 실제성상 특징과 메탄가스 발생사이의 상관성을 알아보고자 하였다. 서울시의 8개 기초자치단체(강남, 강동, 송파, 영등포, 관악, 구로, 동작, 용산)의 음식물 쓰레기 중간 집하장이나 처리시설에서 음식물 쓰레기를 채취하였다. 음식물 쓰레기의 고형물 함량은 평균 16%를 보였고 잠재 메탄발생량은 평균 $446.6STP-m{\ell}/g-VS$로서 $334.8{\sim}567.5STP-m{\ell}/g-VS$의 범위를 가진다. 우분의 고형물 함량과 잠재메탄 발생량은 각각 평균 26%과 $280.6STP-m{\ell}/g-VS$로 타나났다. 잠재 메탄발생량은 고위발열량, 지방 함량, 탄소함량, 수소함량과 양의 상관관계를 가지고 탄수화물 함량과 음의 상관관계를 가지는 것으로 나타났다($r^2&gt;0.8$). 따라서 이러한 기질특성 분석결과를 통해 잠재 메탄발생량을 비교적 정확하게 예측하였다. 본 연구에서는 단일 기질과 잠재 메탄발생량을 분석하였으나 향후는 최대 메탄발생량을 위하여 혼합기질 (음식물쓰레기, 가축분뇨, 수분조절제 등)의 최적조합을 결정하는 연구가 수행하여야 할 것으로 여겨진다.

혐기성 박테리아, 효모 및 곰팡이로 제조된 synbiotics 첨가 축우용 완전혼합사료의 성분 변화 및 발효 특성에 미치는 영향 (Effects of Supplemental Synbiotics Composed of Anaerobic Bacteria, Yeast and Mold on the Change of Chemical Composition and Fermentation Characteristics of Total Mixed Ration for Cattle)

  • 본 시험은 혐기성 박테리아, 효모 및 곰팡이로 제조한 synbiotics를 TMR에 첨가하여 7일 동안 발효시킨 후, 성분함량의 변화 및 개봉하여 공기에 노출시킨 기간에 따른 발효특성의 변화를 알아보기 위하여 수행되었다. 처리구는 무처리구인 US구, 혐기성 박테리아와 prebiotics로 구성된 BS구, 혐기성 효모와 prebiotics로 구성된 YS, 혐기성 곰팡이와 prebiotics로 구성된 MS구, 혐기성 박테리아와 혐기성 곰팡이 및 prebiotics를 조합한 BMS구, 혐기성 효모와 혐기성 곰팡이 그리고 prebiotics를 조합한 YMS구, 혐기성 박테리아와 혐기성 효모 그리고 prebiotics를 조합한 BYS구, 혐기성 박테리아, 효모 및 곰팡이 복합물과 prebiotics를 조합한 BMYS구로서 총 8 처리구로 나누었다. 개봉 후 노출기간(1, 3, 5, 7, 14, 및 21일)별 3반복으로 총 144개의 bag을 공시사료로 제조하였다. 혐기 발효 TMR의 개봉 후 성분함량과 공기노출에 따른 발효특성에 대한 결과를 요약하면 다음과 같다. 혐기 발효시킨 TMR의 수분함량은 약 41${\sim}$45% 범위로서원 사료와 비슷한 수준이었다. 조단백질 함량은 기초 사료에 비해 무처리 대조구에서는 11.7${\sim}$14.8% 줄어들었으나, BMYS 처리구에서는 약 11%가 증가되었다. BMYS 처리구에서는 조섬유 함량이 기초사료에 비해 약 32% 감소되었고, NDF 및 ADF도 각각 15.5% 및 26.1%가 감소되었다. 공시사료의 내부 온도는 개봉 7일째에 전 처리구에서 높게 나타났다. 발효 TMR의 pH는 개봉 5일까지는 처리 간에 차이가 없었으나, 개봉 7일 이후부터는 높아졌고, BMS구에서 개봉 14일째에 가장 높았다(P<0.05). 산에 대한 완충능력은 개봉 후, 시간이 경과함에 따라 산의 첨가량이 많아져 완충능력이 높아지는 경향으로서 대체로 개봉 7일 이후부터 시작하여 14일째에 peak를 이루었다. 발효 TMR 즙액의 $NH_3-N$ 농도는 개봉 후 5일째에 peak를 이루었으며, 휘발성지방산 함량은 매우 낮은 수준이었다. 이상의 결과로 볼 때, BMYS 처리구에서 단백질 함량은 높아지고, 섬유소함량은 낮아졌지만, 공기 중 노출기간별 발효특성에서는 혐기성 synbiotics의 첨가에 따른 영향은 없는 것으로 나타났다.

두부제조폐기물과 하수슬러지의 화학/초음파 전처리에 의한 가용화 및 혐기발효 수소생산 (Fermentative Hydrogen Production from the Pretreated Food-Processing Waste and Sewage Sludge using Chemical/Ultra-Sonication)

  • Acid and alkali pretreatments were applied to tofu processing waste (TPW) to increase the solubility of ingredients in TPW. Pretreatment at 1.0% of HCl and 2.5% of NaOH condition resulted in the increase of SCOD concentration from 3.2 g COD/L to 27 g COD/L and 33 g COD/L, respectively. The acid and alkali-pretreated TPW was studied for its fermentative $H_2$ production capacity in batch mode using a thermophillic mixed culture. Alkali pretreatment on presence of 2.5% NaOH exhibited more soluble portion released compared to acid pretreatment using HCl, however the $H_2$ production from acid pretreated TPW was better than alkali-pretreated TPW probably due to the sodium inhibition on microbial activity. In addition, sewage sludge was externally added to the acid-pretreated (1.0% HCl) TPW by 20% (on volume basis). Average H2 production rate was increased from 31 to 78 ml/L-broth/hr, and it was attributed to the high buffer capacity and abundant nutrients especially divalent cation in sewage sludge.

영양상담을 통한 식사행동의 변화가 운동선수의 운동능력 향상에 미치는 효과 (A Behavior-Modification Approaches to Improved Exercise Performance for Athletes Through the Multipe Nutritional Counseling)

  • Management of nutrient intakes through behavior-mordification can be important for improving exercise performance in athletes. The purpose of this study was to determine the effectiveness of nutritional counseling for improving exercise performance capability in athletes. The subjects were seven golfers and fourteen ju-do athletes from Hoseo university, and they have nor received professional hel[ from dieticians. Prior to nutritional counseling, their dietary intakes, lifestyles and dietary habits were evaluated. Nutritional counseling sessions were conducted by a trained dietician every 2-3 weeks for 3-4 month. After 5 counseling sessions, nutrient intakes, lifestyle and dietary habits were re-evaluated. Exercise performance capability was measured by maximal exercise stress test using treadmills. Individual data were assessed as quality index represents the better nutritional status. The common dietary and lifestyle problems were overeating binging, overeating, snacking and drinking alcohol prior to the nutritional counseling. After counseling sessions, the quality index of dietary habits seemed to decrease, especially in ju-do athletes, representing nutritional problems were resolved. Body fat and body mass index decreased in ju-do athletes, but not in golfers. Both athletes have shown to increase oxygen uptake at anaerobic threshold and maximal oxygen uptake status, which represents that the athletic performance capacity was improved after counseling sessions. Their respiratory quotients were decreased. In conclusion, nutritional counseling sessions over five times are an effective and efficient approach to change dietary habits to improve exercise performance capacity. Furthermore, clients can have good dietary habits and learn how to manago muscle strength by behavior modification through multiple nutritional counseling sessions. pH조절이 음식폐기물의 유기산발효에 미치는 영향 (Effect of pH on the $VFA_s$ fermentation in the anaerobic treatment of food waste)

  • The purpose of this study was to investigate the created liquid post-acid fermentation of usability of denitrification as exterior carbon sources by pH control. The time of acid fermentation of food waste, the slower loading capacity of organic matter, the faster decomposition rate, but the density of generation Volatile Fermentation Acids(VFAs), was weak and, $SBOD_{5}$ : ST-N rate and $SBOD_{5}/SCOD_{Cr}$ rate was low. Between TS and VS, VS was decreased to 4.5th day fast, and then was decreased slowly. 1.5 days after stating the experiments, $SCOD_{Cr}$, $SBOD_{5}$, STOC and $VFA_{s}$ was decreased of increased slowly, and then increased fast. And after showing the highest density, it was tended to decreased fast. At the time of $SBOD_{5}$ with the highest density, at $SBOD_{5}$ : ST-N ratio, $R_1$ was 303:1, $R_2$ was 319:1, $R_3$was the highest. After studying $SBOD_{5}$ : ST-N ratio and $SBOD_{5}/SCOD_{Cr}$ ratio, as a carbon source of biological denitrification it was profitable composition ratio.

Effect of Initial Glucose Concentrations on Carbon and Energy Balances in Hydrogen-Producing Clostridium tyrobutyricum JM1

  • The carbon metabolism of newly isolated Clostridium tyrobutyricum JM1 was investigated at varying initial glucose concentrations (27.8-333.6mM). Because an understanding of metabolic regulations was required to provide guidance for further effective metabolic design or optimization, in this case, maximizing hydrogen production, carbon and energy balances by C. tyrobutyricum JM1 were determined and applied in anaerobic glucose metabolism. The overall carbon distribution suggested that initial glucose concentrations had strong influence on the stoichiometric coefficients of products and the molar production of ATP on the formation of biomass. C. tyrobutyricum JM1 had a high capacity for hydrogen production at the initial glucose concentration of 222.4 mM with high concentrations of acetate and butyrate.
