전분자원의 효율적 이용을 위한 방법의 일환으로 분리동정된 H. anomala var. anomala FRI YO-32와 S. cerevisiae와의 세포융합가능성을 검토하기 위하여 원형질체형성을 위한 기본적인 제반조건에 대하여 실험하였다. 세포벽분해효소로서 달팽이 (Helix pomatia) 추출 효소를 사용하여 원형질체의 형성시수율에 영향을 미치는 중요한 인자로서, 함유황화합물에 의한 전처리유무 및 이러한 화합물의 처리농도 및 처리방법, 세포벽분해효소의 농도 및 처리시간, 공시효모의 성장시기 및 효모세포의 수와 삼투압안정제 (KCI)의 농도 등이 고려되었으며, 원형질체형성을 위한 이러한 인자들의 최적처리조건이 검토되었다.
녹말로부터 에탄올을 직접 생산하는 균주를 개발하고자 amylase 활성이 높은 Aspergillus oryzae와 알콜발효능이 있는 Saccharomyces cerevisiae의 원형질체를 융합하였다. 이들의 융합을 유도하기 위해서 fusogen으로 35% PEG 4000을 사용하였으며, 융합체의 선별을 위한 유전적 지표로는 아미노산 요구성을 사용하였다. 융합률은 $4.6\times 10^{-6}$이었다. 선별된 융합체들은 효모의 형태였으며 amylaseghkf성을 나타내었고, 에탄올을 생사하였다. 그리고 이들 성질 중에서 형태는 후세대에도 안정하게 유지되었으나 녹말로부터의 알콜 생성능은 현저히 감소하였다. 비록 포도당 배지에서의 융합체들의 알콜 생산은 S. cerevisiae의 0.5배에 지나지 않았으나, Prototroph인 S. cerevisiae와는 달리 이들은 녹말로부터 직접 에탄올을 생산할 수 있었다.
$\beta$-Amylase를 생산하여 전분 분해능을 갖는 산업용 효모를 개발하기 위해 산업용 Saccharomyces cerevisiae에서 Achlya bisexualis $\beta$-amylase (BAMY)유전자를 ADC1 promoter에 연결하여 구성적으로 발현시켰다. 효모의 형질전환은 $\delta$-서열을 재조합 부위로 하는integration 시스템을 이용하였다. Integration 시스템의 세균 유전자 부분은 제거되고 BAMY 유전자와 $\delta$-서열을 갖고 있는 짧은integrative cassette를 제조하였다. BAMY 유전자를 발현하는 재조합 S. cerevisiae 형질전환체는 세포외 배지로 45 kDa의 $\beta$-amylase를 분비하였고, $\beta$-amylase 활성은 A. bisexualis에 비해 약 18.5배 높았다. 형질전환체에 다중도입된 BAMY 유전자는 비선택배지에서 100세대 생장 후에도 안정되게 유지되었다. 각종전분을 기질로 했을 매 $\beta$-amylase의 활성은soluble starch를 기질로 했을 경우와 유사하게 높았고, 가수분해산물 분석 결과 maltose가 주 분해산물이었다.
짚에서 60균주를 분리하여 생균제제로서의 기능이 있는 청국장 제조용 starter로 사용가능성을 조사하였다. 분리한 60균주 중 단백 분해력, 전분 분해력, 대두 분해력을 나타낸 균주는 B-59 등 8균주 이었으며 모두 그람 양성 간균으로 포자를 형성하였다. 선별된 8균주의 Staphylococcus aureus, Listeria monocytogenes, Shigella sonni, Pseudomonas fluorescens, Vibrio parahaemolyticus에 대해 항균활성을 나타낸 균주는 B-59이었다. 항균활성을 나타낸 분리균주 B-59에 의해 L. monocytogenes, S. aureus, V. parahaemolyticus, P. fluorescens의 성장은 억제되었으며. API 50CHB kit를 이용하여 동정한 결과 99.0%의 유사율로 Bacillus licheniformis로 간이 동정되었다. 분리균주인 B-59는 pH 2.5로 조정된 인공 위액에 초기 생균수 8.33 CFU/mL에서 $37^{circ}C$에서 3시간 배양 후의 6.91 CFU/mL로 높은 생존율을 나타내었으며, 인공담즙산에서 초기 균수 6.90 CFU/mL에서, 배양 24시간 후 8.24 CFU/mL로 증가하여 내성을 나타내었다. 분리균주인 B-59는 NaCl 0, 2, 4% 농도에서는 각각 7.63, 7.70, 7.67 CFU/mL로 성장에 영향을 미치지 않았으나, 32% 농도에서도 생존하여 NaCl에 대한 내성을 나타내었으며, 알코올농도 32%에서도 4.22 CFU/mL를 나타내어 에탄올에 대한 내성을 나타내었다. DPPH radical 소거능으로 측정한 B-59 배양액의 항산화 활성이 균주의 성장에 따라 증가하였다.
Objective: This study investigated the association of enzyme-producing microbes and their enzymes with starch and hemicellulose degradation during fermentation of total mixed ration (TMR) silage. Methods: The TMRs were prepared with soybean curd residue, alfalfa hay (ATMR) or Leymus chinensis hay (LTMR), corn meal, soybean meal, vitamin-mineral supplements, and salt at a ratio of 25:40:30:4:0.5:0.5 on a dry matter basis. Laboratory-scale bag silos were randomly opened after 1, 3, 7, 14, 28, and 56 days of ensiling and subjected to analyses of fermentation quality, carbohydrates loss, microbial amylase and hemicellulase activities, succession of dominant amylolytic or hemicellulolytic microbes, and their microbial and enzymatic properties. Results: Both ATMR and LTMR silages were well preserved, with low pH and high lactic acid concentrations. In addition to the substantial loss of water soluble carbohydrates, loss of starch and hemicellulose was also observed in both TMR silages with prolonged ensiling. The microbial amylase activity remained detectable throughout the ensiling in both TMR silages, whereas the microbial hemicellulase activity progressively decreased until it was inactive at day 14 post-ensiling in both TMR silages. During the early stage of fermentation, the main amylase-producing microbes were Bacillus amyloliquefaciens (B. amyloliquefaciens), B. cereus, B. licheniformis, and B. subtilis in ATMR silage and B. flexus, B. licheniformis, and Paenibacillus xylanexedens (P. xylanexedens) in LTMR silage, whereas Enterococcus faecium was closely associated with starch hydrolysis at the later stage of fermentation in both TMR silages. B. amyloliquefaciens, B. licheniformis, and B. subtilis and B. licheniformis, B. pumilus, and P. xylanexedens were the main source of microbial hemicellulase during the early stage of fermentation in ATMR and LTMR silages, respectively. Conclusion: The microbial amylase contributes to starch hydrolysis during the ensiling process in both TMR silages, whereas the microbial hemicellulase participates in the hemicellulose degradation only at the early stage of ensiling.
A $\alpha$-l, 4-D-glucan maltohydrolase $(\beta$-amylase), secreted by the mesophilic aerobic bacterium Bacillus polymyxa No.26, was purified and characterized. The enzyme production was increased after a logarithmic phase of bacterial growth and paralleled with the onset of bacterial sporulation. By applying anion exchange chromatography and gel filtration the enzyme was purified 16.7-fold and had a specific activity of 285.7 units/mg. Two enzyme activities were eluted on a column of DEAE-Sephadex chromatography, and they were designated as E-I for a major enzyme peak and E-II for a minor peak. Of them, E-I enzyme peak was further purified by using gel chromatography. The molecular mass of this enzyme was determined to be 64, 000 daltons and consisted of a single subunit, showing an isoelectric point of 8.9. The enzyme was able to attack specifically the $\alpha$-l, 4-glycosidic linkages in soluble starch and caused its complete hydrolysis to maltose and $\beta$-limited dextrin. This amylolytic enzyme displayed a temperature optimum at $45^\circ{C}$ and a pH optimum at 7.0. The amino acid composition of the purified enzyme was quite similar to the other bacterial $\beta$-amylases reported. Surprisingly, the purified enzyme from this aerobe only exhibited hydrolytic activity on soluble starch, not on starch granules. The degradation of from starch by $\beta$-amylase was greatly stimulated by pullulanase addition. These results differentiated from other $\beta$-amylases reported. Based on a previous result that showed the enzyme system involves in effective degradation of raw starch granules, this result strongly suggested that the purified enzyme (E-I) can be a synergistic part of starch granule-digestion and E-II plays a crucial role in digestion of starch granules.

Amylases and cellulases are important enzymes that can be utilized for various biological activities. Ten different wild Nigerian mushrooms (Agaricus blazei, Agaricus sp., Corilopsis occidentalis, Coriolus versicolor, Termitomyces clypeatus, Termitomyces globulus, Pleurotus tuber-regium, Podoscypha bolleana, Pogonomyces hydnoides, and Nothopanus hygrophanus) were assayed for production of these secondary metabolites. The results revealed that most of the tested wild fungi demonstrated very good amylase and cellulase activities. With the incorporation of carboxymethyl-cellulose (a carbon source) into the culture medium, Agaricus blazei had the highest amylolytic activity of 0.60 unit/mL (at $25^{\circ}C$, pH 6.8). This was followed in order by P. tuber-regium and Agaricus sp. with 0.42 and 0.39 unit/mL, respectively ($p {\leq} 0.05$). Maltose and sucrose supplementation into the submerged liquid medium made N. hygrophanus and P. hydnoides to exhibit very low amylase activities of 0.09 and 0.11 unit/mL, respectively. Introducing peptone (an organic nitrogen source) into the basal medium enhanced the ability of C. versicolor to produce a cellulase value of 0.74 unit/mL. Other organic nitrogen sources that supported good cellulase activities were yeast extract and urea. Sodium nitrate (inorganic nitrogen source) generally inhibited cellulase production in all mushrooms. The best carbon source was carboxymethyl-cellulose, which promoted very high cellulase activity of 0.67 unit/mL in C. versicolor, which was followed in order by P. tuber-regium, T. chypeatus, and C. occidentalis ($p {\leq} 0.05$). Sucrose was the poorest carbon compound, supporting the lowest values of 0.01, 0.01, and 0.14 unit/mL in P. hydnoides, A. blazei, and Agaricus sp., respectively.
In 2004, bacterial spot-causing xanthomonads (BSX) were reclassified into 4 species-Xanthomonas euvesicatoria, X. vesicatoria, X. perforans, and X. gardneri. Bacterial spot disease on pepper plant in Korea is known to be caused by both X. axonopodis pv. vesicatoria and X. vesicatoria. Here, we reidentified the pathogen causing bacterial spots on pepper plant based on the new classification. Accordingly, 72 pathogenic isolates were obtained from the lesions on pepper plants at 42 different locations. All isolates were negative for pectolytic activity. Five isolates were positive for amylolytic activity. All of the Korean pepper isolates had a 32 kDa-protein unique to X. euvesicatoria and had the same band pattern of the rpoB gene as that of X. euvesicatoria and X. perforans as indicated by PCR-restriction fragment length polymorphism analysis. A phylogenetic tree of 16S rDNA sequences showed that all of the Korean pepper plant isolates fit into the same group as did all the reference strains of X. euvesicatoria and X. perforans. A phylogenetic tree of the nucleotide sequences of 3 housekeeping genes-gapA, gyrB, and lepA showed that all of the Korean pepper plant isolates fit into the same group as did all of the references strains of X. euvesicatoria. Based on the phenotypic and genotypic characteristics, we identified the pathogen as X. euvesicatoria. Neither X. vesicatoria, the known pathogen of pepper bacterial spot, nor X. perforans, the known pathogen of tomato plant, was isolated. Thus, we suggest that the pathogen causing bacterial spot disease of pepper plants in Korea is X. euvesicatoria.
메품종의 거대배아미인 화청거대배아미, 남풍거대배아미, 그리고 찰품종인 화청찰거대배아미, 신선찰거대배아미 등 4품종의 거대배아미 품종의 볍씨를 각각 $27^{\circ}C$에서 3일간 발아시켜, 발아에 따른 전분 가수분해 효소의 활성 및 전분입자의 이화학적 특성을 각각 비교하였다. 3일간 발아시킨 벼의 ${\alpha}-amylase$의 활성은 맥아에 비해서 활성이 높게 나타났으며, 특히 화청거대배아미 및 신선찰거대배아미는 맥아보다 약 2배 정도의 활성을 나타내고 있었다. 이에 비해서 ${\beta}-amylase$의 경우는 일반품종보다는 거대배아미품종의 활성이 높기는 하지만 비교군인 맥아에 비해서 상당히 낮은 활성을 나타내고 있었다. 전분분자 중 아밀로오스 분자 유래의 긴 포도당 사슬의 양은 발아와 더불어 메품종 거대배아미에서는 줄어들고 있었으며, 찰품종에서는 증가하고 있었다. 아밀로펙틴 분자 유래의 포도당 사슬길이 분포는 찰벼와 메벼 품종에 관계없이 발아와 더불어 중합도가 14부터 60가지 비율은 증가하고 있었으며, 중합도 130이상 또는 13이하의 비율은 감소하고 있었다. Glucoamylase에 의한 가수분해도는 거대배아미메품종의 경우에는 발아와 더불어 현저히 낮아지고 있었고, 찰품종의 경우에는 오히려 증가하고 있었다. 그리고 호화개시온도, 호화종료 및 호화엔탈피는 감소하고 있었다.
Aamylase 활성이 높은 Aspergillus oryzae와 알콜발효능이 있는 Saecharomyccs cerevisiae의 원형질체융합을 위한 기초연구로서, amylase 활성이 있는 A. oryzae의 종내원형질체를 융합시켜 이들융합체의 특성을 조사하였다. 영양요구성 돌연변이 균주의 mycellia로부터 원형질체를 생성하기 위해서는 lytic enzyme으로 Novozyme 234가 효과적이였고 완충용액의 pH는 5.5에서 6.0사이가 최적이었다. F usogen으로 30% PEG4,000를 사용하였을 때 효과적으로 원형질체의 융합이 이루어졌으며 이들 융합체의 대부부은 heterokaryons이었다. 원형질체의 형태와 PEG처리후 융합되는 과정을 광학현미경으로 관찰하였다. 원형질체의 재생율은 재생배지와 균주에 따라 1.46~14%이었고, A. oryzae 종내융합율은 0.12-0.16이었다. 융합체의 DNA함량을 조사한 결과 모균주보다 약 1.5배정도 증가됨을 보였고 융합체들의 amylase 활성은 융합체에 따라 다소 차이를 냐타내었다. 가장 높은 amylase 활성을 나타낸 융합체들의 amylase 활성은 융합체에 따라 다소 차이를 나타내었다. 가장 높은 amylase 활성을 나타낸 융합체 F2-2에 있어서는 야생균주 ATCC 22788의 그것보다 amylase 활성이 약 1.5배 가량 높았다.
