Studies on L-Glutamic Acid-Producing Bacteria(II) (L-Glutamic acid 생성균에 관한 연구 2)

  • Searches for the nutrition requirements of three strains of Brevibacterium ammoniagenes reported in the previous paper were carried out with an aim of achieving the striking accumulation of L-glutamic acid and the large multipication of cells. It was recognized that all three strains required both biotin and thiamine, together with amino acids such as histidine or cysteine, for their good growth and extracellular L-glutamic acid accumulation. The quantity of biotin required for remarkable growth of these microorganisms was quite different from that for the maximum production of L-glutamic acid. This result, however, did not apply in the case of thiamine. It was also confirmed that, of 18 amino acids, histidine and cysteine were the msot effective organic nitrogen sources, while the most available inorganic ammonium salt resulting in a large amount of L-glutamic acid-production and considerable cell gorwth was found to be only urea. Maximum accumulation of extracellular L-glutamic acid, more than 50%(w/w) of the initial sugar content, could be obtained from fermentation in the medium containing wheat-bran extract(Brev. ammoniagenes T-1 and Brev.ammoniagenes Y-2) or rice-bran extract(Brev. ammoniagenes YR-2), which confirmed us a possibility that these bacteria might be employed for industrial fermentation of L-glutamic acid.

  • Various disorders cause hyperammonemia during childhood. Amongthem are those caused by inherited defects in urea synthesis and related metabolic pathways. These disorders can be grouped into two types: disorders of the enzymes that comprise the urea cycle, and disorders of the transporters or metabolites of theamino acids related to the urea cycle. Principal clinical features of these disorders are caused by elevated levels of blood ammonium. Additional disease-specific symptoms are related to the particular metabolic defect. These specific clinical manifestations are often due to an excess or lack of specific amino acids. Treatment of urea cycle disorders and related metabolic diseases consists of nutritional restriction of proteins, administration of specific amino acids, and use of alternative pathways for discarding excess nitrogen. Although combinations of these treatments are extensively employed, the prognosis of severe cases remains unsatisfactory. Liver transplantation is one alternative for which a better prognosis is reported.

Extraction Equilibria and Solvent Sublation for Determination of Ultra Trace Bi(Ⅲ), In(Ⅲ) and TI(Ⅲ) in Water Samples by Ion-Pairs of Metal-2-Naphthoate Complexes and Tetrabutylammonium Ion

  • The solvent sublation using ion pairs of metal-2-naphthoate(2-HNph) and tetrabutyl ammonium ($TBA^+$) ion has been studied for the concentration and determination of ultra trace Bi(III), In(III) and Tl(Ⅲ) ions in water samples. The partition coefficients ($K_p$) and the extraction percentages of 2-HNph and the ion pairs to methyl isobutyl ketone (MIBK) were obtained as basic data. After the ion pair $TBA^+$·M$(Nph)_4^-$ was formed in water samples, the analytes were concentrated by the solvent sublation and the elements were determined by GF-AAS. The pH of the sample solution, the amount of the ligand and counter ion added and stirring time were optimized for the efficient formation of the ion pair. The type and amount of optimum surfactant, bubbling time with nitrogen and the type of solvent were investigated for the solvent sublation as well. 10.0 mL of 0.1 M 2-HNph and 2.0 mL of 0.1 M $TBA^+$ were added to a 1.0 L sample solution at pH 5.0. After 2.0 mL of 0.2%(w/v) Triton X-100 was added, the ion pairs were extracted into 20.0 mL MIBK in a flotation cell by bubbling. The analytes were determined by a calibration curve method with measured absorbances in MIBK, and the recovery was 80-120%.

Modified Activated Carbons from Olive Stones for the Removal of Heavy Metals

  • The activated carbon "C" was obtained by carbonization followed by activation with steam at 40% of burn-off. Oxidized carbons C-N, C-P and C-H were obtained by oxidizing the activated carbon C with concentrated nitric acid, ammonium peroxysulfate and hydrogen peroxide, respectively. The textural properties of the carbons were determined from nitrogen adsorption at 77 K. The acidic surface functional groups were determined by pH titration, base neutralization capacity and electrophoretic mobility measurements. The cation exchange capacities of un-oxidized and oxidized carbons were determined by the removal of Cu(II) and Ni(II) from their aqueous solutions. The surface area and the total pore volume decreased but the pore radius increased by the treatment of activated carbon with oxidizing agents. These changes were more pronounced in case of oxidation with $HNO_3$. The surface pH of un-oxidized carbon was basic whereas those of the oxidized derivative were acidic. The removal of Cu(II) and Ni(II) was pH dependent and the maximum removal of the both ions was obtained at pH of 5-6. Cu(II) was more adsorbed, a phenomenon which was ascribed to its particular electronic configuration.

  • The titled dye of DBPI gives amplified spontaneous emission (ASE) with maximum at 580 nm upon pumping by nitrogen laser (${\lambda}_{ex}\;=\;337.1\;nm$). The ground state absorption cross section (${\sigma}_A$) and emission cross section (${\sigma}_E$) as well as effective emission cross section(${\sigma}^*_E$) have been determined. The electronic absorption spectra of DBPI were measured in ethanol and tetrahydrofuran at room and low temperature. DBPI displays molecular aggregation in water. The photochemical reactivity of DBPI was also studied in carbon tetrachloride upon irradiation with 525 nm light. The electrochemical investigation of DBPI dye has been carried out using cyclic voltammetry and convolution deconvolution voltammetry combined with digital simulation technique at a platinum electrode in 0.1 mol/L tetrabutyl ammonium perchlorate (TBAP) in two different solvents acetonitrile ($CH_3CN$) and dimethylformamide (DMF). The species were reduced via consumption of two sequential electrons to form radical anion and dianion (EE mechanism). In switching the potential to positive direction, the compound was oxidized by loss of two sequential electrons, which were followed by a fast dimerization and/or aggregation process i.e $EC_{dim1}EC_{dim2}$ mechanism. The electrode reaction pathway and the chemical and electrochemical parameters of the investigated compound were determined using cyclic and convolutive voltammetry. The extracted electrochemical parameters were verified and confirmed via digital simulation method.

The Optimal Producing Conditions of Bacteriocin Produced by Lactobacillus sp. FF-3 Isolated from Korean Dongchimi (동치미에서 분리한 Lactobacillus sp. FF-3가 생산하는 bacteriocin의 최적 생산조건)

  • The optimal culture conditions on bacteriocin producing of Lactobacillus sp. FF-3 isolated from Korean Dongchimi, were studied for enhancing its production with regard to environmental and nutritional factors. The optimal cultivation time, initial pH and temperature were 21 hours, pH 7.0 and 30∼37$^{\circ}C$ respectively. Optimal compositions of culture medium for bacteriocin production were glucose 3% as carbon source, tryptone 4% as nitrogen source, and manganese sulfate 0.005% as inorganic salt with other basal components. The maximum antimicrobial activity was 484 BU/mL under the optimal culture condition.

Evaluation of Agrotain Efficiency for Suppression of Ammonium Volatilization Under Chinese Cabbage Cultivation Fields

  • Chinese cabbage cultivation and ammonia volatilization experiments were done to evaluate the efficiency of Agrotain coated urea (GSP 80% + Agro; GSP 100% + Agro) against conventional urea (GSP 80%; GSP 100%). Fresh weight of Chinese cabbage were 17.2% and 7.3% higher in the treatments that received GSP 80% + Agro and GSP 100% + Agro, respectively, of those from the treatments that received urea alone. Likewise, the nitrogen use efficiency of Chinese cabbage in the treatments that received Agrotain coated urea were significantly higher at the rate of 3.5% (GSP 80% + Agro) and 1.9% (GSP 100% + Agro) compared to urea alone treatments. Ammonia emission was substantially higher at the rate of $107.6N\;mg\;chamber^{-1}$ with the application of only GSP 100%. However, nearly 28.3% of ammonia emission was considerably reduced with the use of Agrotain coated urea. Hence, we recommend the use of Agrotain coated urea in conventional farming for increased crop yield as well as simultaneous reduction of nitrogenous fertilizer use.

Purification of A Novel Protein Induced by Salt Treatment in Rice Seedling (NaCl 처리로 벼 유묘에서 유도된 단백질의 분리와 정제)

  • The monoclonal antibody against a novel protein, which was induced in the roots of rice seedlings treated with NaCl, was produced. Oryzea sativa L. cv. Annapruna grains were sown on clean sands and grown for 10days and then the seedlings were soaked in 50mM NaCl aqueous solution. In 48hrs of the NaCl treatment, the roots were collected and homogenated with liquid nitrogen and extraction buffer. The homogenate was centrifuged and to the supernatant 75% ammonium sulfate was added to pre-cipitate proteins. From these proteins a novel protein was purified through DEAE-ion chromatography and FPLC(Phenyl column). This protein appeared as a single band in the native electrophoresis. Using this protein as antigen, monoclonal antibody was produced. Five cell lines that secreted antibodies specifically bound to this protein were constructed.

  • This study was performed to investigate the disinfection efficiency and the formation characteristics of disinfection by-products(DBPs) by chlorination in the sewage effluent. The effluent was sampled from the sewage treatment plants operated in the activated sludge process and the advanced sewage process. The type of DBPs investigated were Trihalomethanes(THMs), Dichloroacetonitrile(DCAN), Chloral hydrate(CH), Dichloroacetic acid(DCAA), Trichloroacetic acid(TCAA). Major findings are as follows. First, the optimum injection concentration for chlorination in sewage effluent were found to be in the range $0.5{\sim}1.0mg\;cl_2/L$. Also, It was found that the chlorine dosage in the effluent of activated sludge process was higher than in the effluent of advanced sewage process. Second, the maximum formation concentration of THMs were $12.7{\mu}g/L$. The THMs formation reaction was finished in a short time of several seconds and chloroform was mainly formed. Also, it was found that the concentration of ammonium nitrogen is higher, the concentration of THMs is lower. Third, it was found that DCAA and TCAA were mainly formed as DBPs by disinfection.

Removing nitrogenous compounds from landfill leachate using electrochemical techniques

  • In this research, applicability of electrochemical technology in removing nitrogenous compounds from solid waste landfill leachate was examined. Novel cathode material was developed at laboratory by introducing a Cu layer on Al substrate (Cu/Al). Al and mild steel (MS) anodes were investigated for the efficiency in removing nitrogenous compounds from actual leachate samples collected from two open dump sites. Al anode showed better performances due to the effect of better electrocoagulation at Al surface compared to that at MS anode surface. Efficiency studies were carried out at a current density of $20mA/cm^2$ and at reaction duration of 6 h. Efficiency of removing nitrate-N using Al anode and developed Cu/Al cathode was around 90%. However, for raw leachate, total nitrogen (TN) removal efficiency was only around 30%. This is due to low ammonium-N removal as a result of low oxidation ability of Al. In addition to the removal of nitrogenous compounds, reactor showed about 30% removal of total organic carbon. Subsequently, raw leachate was diluted four times, to simulate pre-treated leachate. The diluted leachate was treated and around 88% removal of TN was achieved. Therefore, it can be said that the reactor would be good as a secondary or tertiary treatment step in a leachate treatment plant.
