Effect of Ammonium Concentration on the Emission of $N_2O$ Under Oxygen-Limited Autotrophic Wastewater Nitrification

  • A significant amount of nitrous oxide ($N_2O$), which is one of the serious greenhouse gases, is emitted from nitrification and denitrification of wastewater. Batch wastewater nitrifications with enriched nitrifiers were carried out under oxygen-limited condition with synthetic (without organic carbon) and real wastewater (with organic carbon) in order to find out the effect of ammonium concentration on $N_2O$ emission. Cumulated $N_2O$-N emission reached 3.0, 5.7, 6.2, and 13.5 mg from 0.4 l of the synthetic wastewater with 50, 100, 200, and 500 mg/l ${NH_4}^+$-N, respectively, and 1.0 mg from the real wastewater with 125 mg/l ${NH_4}^+$-N. The results indicate that $N_2O$ emission increased with ammonium concentration and the load. The ammonium removal rate and nitrite concentration also increased $N_2O$ emission. Comparative analysis of $N_2O$ emission from synthetic and real wastewaters revealed that wastewater nitrification under oxygen-limited condition emitted more $N_2O$ than that of heterotrophic denitrification. Summarizing the results, it can be concluded that denitrification by autotrophic nitrifiers contributes significantly to the $N_2O$ emission from wastewater nitrification.

  • 본 연구는 생물학적 질산화 공정에서 암모늄 이온과 활성을 가진 질산화균의 초기 농도가 질산화에 미치는 영향을 확인하고, 이에 따른 동역학식을 제시하고자 하였다. 먼저 실험에 이용된 슬러지의 질산화균 농도는 미생물 호흡률 실험으로 측정하였는데, 배양된 슬러지 중 42.8%가 활성을 가진 질산화균으로 나타났다. 암모늄 이온과 질산화균의 초기 농도를 달리하여 $N_0/X_0$비가 0.025~0.493의 조건에서 잘산화 실험을 실시하였으며, 이를 통해 암모늄 이온과 질산화균의 농도가 상이하더라도 $N_0/X_0$ 비가 동일할 경우 암모늄 산화율이 동일함을 확인하였다. 또한 $N_0/X_0$ 비와 비기질이용율의 관계는 Contois 형태의 관계식으로 표현되었으며, 최대 비암모늄산화율($q_{Nmax}$)은 4.32 gN/gVSS/day, 반 포화속도 상수($K_N{^{\prime}}$)는 0,013 gN/gVSS인 것으로 확인되었다.

  • The crystal and molecular structure of 2-methyl-3-(N-trimethyl ammonium)phenol iodide, $C_{10}H_{16}NOI, was determined by X-ray diffraction method. The compound crystallizes in the orthorhombic space group $P_{na2}_{1}$ with a=13.327(3), b=12.496(3), C=7.227(2)A and Z=4. A total of 489 independent observed reflections were collected by the automated Four-circle diffractometer and was solved by heavy atom method and refined by anisotropic block-diagonal least-squares method to the R value of 0.04. The benzene ring is slightly distorted from regular hexagon. The I atom and 2-methyl-3-(N-trimethyl ammonium)phenol group is held together by van der Waals forces in the crystal. Intermolecular hydrogen bond is of the type O-H....I with the length 3.35.angs.. Apart from the hydrogen bonding system the molecules are held together by van der Waals forces in the crystal.

An Optical-Density-Based Feedback Feeding Method for Ammonium Concentration Control in Spirulina platensis Cultivation

  • Cultivation of Spirulina platensis using ammonium salts or wastewater containing ammonium as alternative nitrogen sources is considered as a commercial way to reduce the production cost. In this research, by analyzing the relationship between biomass production and ammonium-N consumption in the fed-batch culture of Spirulina platensis using ammonium bicarbonate as a nitrogen nutrient source, an online adaptive control strategy based on optical density (OD) measurements for controlling ammonium feeding was presented. The ammonium concentration was successfully controlled between the cell growth inhibitory and limiting concentrations using this OD-based feedback feeding method. As a result, the maximum biomass concentration (2.98 g/l), productivity (0.237 g/l d), nitrogen-to-cell conversion factor (7.32 gX/gN), and contents of protein (64.1%) and chlorophyll (13.4mg/g) obtained by using the OD-based feedback feeding method were higher than those using the constant and variable feeding methods. The OD-based feedback feeding method could be recognized as an applicable way to control ammonium feeding and a benefit for Spirulina platensis cultivations.

  • Various kinds of water soluble N,N,N-trimethylchitosan ammonium chloride(TMC) with different molecular weights were synthesized to examine the antimicrobial activity against Methicillin Sensitive Staphylococcus aureus(MSSA) and Methicillin Resistant Staphylococcus aureus(MRSA), which causes serious hospital infection, and to apply them to antimicrobial finishing agents for textiles. Chitosan samples were highly deacetylated with sodium hydroxide solution and degraded with hydrogen peroxide to control the molecular weight. TMC has the antimicrobial activities against MRSA and MSSA. TMC showed an excellent antimicrobial activity below the molecular weight of 70,000, especially at 40,000. The minimum inhibitory concentration (MIC) of TMC with optimum molecular weight against MRSA and MSSA was 250ppm. Because MRSA did not resist TMC in the subculture test of bouillon medium, it was expected that the successive use of TMC against MRSA was possible.

상수리나무림 임상에 공급한 무기질소가 토양호흡에 미치는 영향 (Effects of Nitrogen Addition on Soil Respiration)

  • In order to gain a better understanding of how forests participate in the cycling of carbon, effects of nitrogen addition on soil respiration were investigated on the oak forest in Kongju, Korea. Study site was divided into control, treatment f and treatment 2 plots, with 5 replication in each plot. In each replicate of treatment 1 and treatment 2 were fertilized with ammonium nitrate (NH$_4$NO$_3$), 30 g/$m^2$ and 60 g/$m^2$, respectively. Soil respiration, soil temperature, ammonium-N and nitrate-N were measured during the experimental period. Ammonium-N and nitrate-N in Ta were higher than those in control and T$_1$. Ammonium-N and nitrate-N in top-soil and sub-soil decreased sharply in August after bi9 rainfall in July in T$_1$ and T$_2$, however, those in control plot increased. Soil respiration in T$_2$ Plot showed consistently higher than those in control and T$_1$ until the end of July. However, soil respiration was similar among the control, T$_1$ and T$_2$ in mid-August and September The amount of Co$_2$ released from soil respiration in control, T$_1$ and T$_2$ in mid-August was 8.0$\pm$0.4, 9.3$\pm$0.6 and 10.2$\pm$0.5 $\mu$mol$^{-1}$ ㆍm$^{-2}$ ㆍs$^{-1}$ , respectively. However, those in control, T$_1$ and T$_2$in mid-August was 13.0$\pm$0.4, 13.5$\pm$0.5, 13.3$\pm$0.6 $\mu$mol$^{-1}$ ㆍm$^{-2}$$^{-1}$ , respectively. The results suggest that nitrogen addition in this oak forest has a positive effect on soil respiration.

Atmosphere-forest Exchange of Ammoniacal Nitrogen in a Subalpine Deciduous Forest in Central Japan during a Summer Week

  • The present study aimed to investigate the diurnal variations in air concentrations and exchange fluxes of ammoniacal nitrogen ($NH_x$: ammonia ($NH_3$) and particulate ammonium) in a subalpine deciduous forest in central Japan during a week in summer. The $NH_3$ concentrations ($0.50\;{\mu}g\;N\;m^{-3}$ on average) showed a clear circadian variation, i.e., high and low in the daytime and nighttime, respectively. The concentration of particulate ammonium in the coarse fractions was extremely low, whereas that for the PM2.5 fraction was relatively high $0.55\;{\mu}g\;N\;m^{-3}$ on average). The main inorganic ion components of PM2.5 at the study site were ammonium and sulfate. The exchange fluxes of $NH_x$ were bidirectional. Both the maximum and minimum values occurred in the daytime, i.e., $0.39\;mg\;N\;m^{-2}\;hr^{-1}$ of downward flux and $0.11\;mg\;N\;m^{-2}\;hr^{-1}$ of upward flux for $NH_3$ and $0.25\;mg\;N\;m^{-2}\;hr^{-1}$ of downward flux and $0.13\;mg\;N\;m^{-2}\;hr^{-1}$ of upward flux for PM2.5 ammonium. The exchange fluxes of $NH_x$ at night could be considered as zero. The mean deposition velocity during the research period was almost zero for both $NH_3$ and PM2.5 ammonium. The atmosphere-forest exchange of $NH_x$ in the forest during the study period was balanced. The remarkably large deposition of $NH_x$ was attributable to meteorological events such as showers the night before that thoroughly washed the forest canopy and subsequent clear skies in the morning, which enhanced convection. The cleaning effect of rainfall and the rapid change in convection in the early morning should be monitored to evaluate and generalize the gas and particle exchange in a forest.

  • P. oleovorans의 유가식 배양에서 탄소원으로 octanoic acid, 질소원으로 $NH_4NO_3$를 이용한 혼합기질을 배양액의 pH 변화에 따라 공급하는 pH-stat 기질공급전략을 개발하였다. 공급기질의 탄소원/질소원 비 (C/N 비)를 변화시킴으로써 최종 균체농도, PHA 농도, PHA 함량 등을 변화시킬 수 있었으며, 최대 균체농도는 C/N 비가 10 (g octanoic acid/g $NH_4NO_3$)일 때 65 g/L, 최대 PHA 농도는 C/N 비가 20일 때 41 g/L, 최대 PHA 함량은 C/N 비가 20일 때 75%였으며 최대 PHA 생산성은 C/N 비가 10일 때 1.03 g/L/h였다.

Characterization of Bacterial Structures in a Two-Stage Moving-Bed Biofilm Reactor (MBBR) During Nitrification of the Landfill Leachate

  • Differences in DNA banding patterns, obtained by ribosomal intergenic spacer analysis (RISA), and nitrification were followed in a moving-bed biofilm reactor (MBBR) receiving municipal landfill leachate. Complete nitrification (>99%) to nitrate was obtained in the two-stage MBBR system with an ammonium load of 1.09 g N-$NH_4/m^2{\cdot}d$. Increasing the ammonium load to 2.03 g N-$NH_4/m^2{\cdot}d$or more caused a decline in process efficiency to 70-86%. Moreover, at the highest ammonium load (3.76 g N-$NH_4/m^2{\cdot}d$), nitrite was the predominant product of nitrification. Community succession was evident in both compartments in response to changes in ammonium load. Nonmetric multidimensional scaling (NMDS) supported by similarity analysis (ANOSIM) showed that microbial biofilm communities differed between compartments. The microbial biofilm was composed mainly of ammonia-oxidizing bacteria (AOB), with Nitrosomonas europeae and N. eutropha being most abundant. These results suggest that high ammonium concentrations suit particular AOB strains.

득량만 퇴적물로부터 영양염 용출 평가 (Estimation of Nutrients Released from Sediments of Deukryang Bay)

  • The purpose of the present study is to estimate the release of dissolved Inorganic nitrogen and photophorous from sediments of Deukryang Bay. One method used In this study is to calculate nutrients released from a concentration gradient between sediment porewaters and the overlying water based on periments. The calculated and measured ammonium released from the sediments were 8.93mg-atN/$m^2$ hr and 60.4mg/atN/$m^2$ hr, respectively In July. 8.57mg-atN/$m^2$. hr and 32.9mg/atN/$m^2$ hr, respectively in October. The ammonium was released more highly in truly than in October, and the measured ammonium flux was higher than the calculated one. The calculated nitrate plus nitrite released from the sediments were 0.31mg/atN/$m^2$. hr in July and 0.84mg/atN/$m^2$. hr in October. The measured nitrate plus nitrite released from the sediments was 282mg/atN/$m^2$. hr in October. The calcuated was lower than the measured because the content of the nutrients in the sediments was always much more than in the overlying waters, and it has shown a differently seasonal pattern compared to the ammonium flux. The calculated phosphorous released from the sediments were 0.97mP/atN/$m^2$ hr and measured negative fluxed -6.50mP/atN/$m^3$ hr In truly, and alculated 0.18mg/atP/$m^2$. hr and measured 24.6mg/atP/$m^2$. hr, respectively in October.
