16S rRNA 유전자 분석방법을 이용한 동해 울릉분지 심해 퇴적물 내 고세균 군집 구조 및 다양성의 수직분포 특성연구 (Community Structure, Diversity, and Vertical Distribution of Archaea Revealed by 16S rRNA Gene Analysis in the Deep Sea Sediment of the Ulleung Basin, East Sea)

  • To assess community structure and diversity of archaea, a clone sequencing analysis based on an archaeal 16S rRNA gene was conducted at three sediment depths of the continental slope and Ulleung Basin in the East Sea. A total of 311 and 342 clones were sequenced at the slope and basin sites, respectively. Marine Group I, which is known as the ammonia oxidizers, appeared to predominate in the surface sediment of both sites (97.3% at slope, 88.5% at basin). In the anoxic subsurface sediment of the slope and basin, the predominant archaeal group differed noticeably. Marine Benthic Group B dominated in the subsurface sediment of the slope. Marine Benthic Group D and Miscellaneous Crenarchaeotal Group were the second largest archaeal group at 8-9 cm and 18-19 cm depth, respectively. Marine Benthic Group C of Crenarchaeota occupied the highest proportion by accounting for more than 60% of total clones in the subsurface sediments of the basin site. While archaeal groups that use metal oxide as an electron acceptor were relatively more abundant at the basin sites with manganese (Mn) oxide-enriched surface sediment, archaeal groups related to the sulfur cycle were more abundant in the sulfidogenic sediments of the slope. Overall results indicate that archaeal communities in the Ulleung Basin show clear spatial variation with depth and sites according to geochemical properties the sediment. Archaeal communities also seem to play a significant role in the biogeochemical carbon (C), nitrogen (N), sulfur (S), and metal cycles at each site.

Microbial Community Diversity in Anaerobic Reactors Digesting Turkey, Chicken, and Swine Wastes

  • The microbial community structures of two continuous stirred tank reactors digesting turkey manure with pine wood shavings as well as chicken and swine manure were investigated. The reactor fed with chicken/swine wastes displayed the highest organic acids concentration (up to 15.2 g/l) and ammonia concentration (up to 3.7 g/l ammonium nitrogen) and generated a higher biogas yield (up to $366ml/g_{VS}$) compared with the reactor supplied with turkey wastes (1.5-1.8 g/l of organic acids and 1.6-1.7 g/l of ammonium levels; biogas yield was up to $195ml/g_{VS}$). The microbial community diversity was assessed using both sequencing and profiling terminal restriction fragment length polymorphisms of 16S rRNA genes. Additionally, methanogens were analyzed using methyl coenzyme M reductase alpha subunit (mcrA) genes. The bacterial community was dominated by members of unclassified Clostridiales with the prevalence of specific clostridial phylotypes in each reactor, indicating the effect of the substrate type on the community structure. Of the methanogenic archaea, methanogens of the genus Methanosarcina were found in high proportions in both reactors with specific methanosarcinas in each reactor, whereas the strict hydrogenotrophic methanogens of Methanoculleus sp. were found at significant levels only in the reactor fed with chicken/swine manure (based on the analyses of 16S rRNA gene). This suggests that among methanogenic archaea, Methanosarcina species which have different metabolic capabilities, including aceticlastic and hydrogenotrophic methanogenesis, were mainly involved in anaerobic digestion of turkey wastes.

TREATMENT OF HIGH-CONCENTRATION SWINE WASTEWATER BY ANAEROBIC DIGESTION AND AN AQUATIC PLANT SYSTEM

  • The treatment of high-strength swine wastewater by anaerobic digestion combined with an aquatic plant system was investigated. Anaerobic digestion of swine wastewater gave volatile solids (VS) removal efficiencies of 43.3%, 52.1% and 54.5% for hydraulic retention times (HRTs) of 20, 30, 40 days, respectively. The removal efficiencies of VS, total chemical oxygen demand (TCOD) and soluble chemical oxygen demand (SCOD) decreased with increasing VS volumetric loading rate (VLR). Higher organic removal efficiency was observed at longer HRTs for the same VS volumetric loading rate. As VS volumetric loading rate increased, biogas production increased and the methane content of the biogas decreased. Experiments using duckweed (Lemna species) as an aquatic macrophyte gave the following results. In the case of nitrogen, removal efficiency was above 60% and effluent concentration was below 10.0 mg/L when the influent ammonia-N loading was about $1.0\;g/m^2/day$. In the case of phosphorus, removal efficiency was above 55% and effluent concentration was below 2.0 mg/L when the influent $PO_4$-P loading was about $0.15\;g/m^2/day$. In addition, crude protein and phosphorus content of duckweed biomass increased from 15.6% to 41.6% and from 0.8% to 1.6%, respectively, as the influent nutrient concentration increased. The treatment of high-strength swine wastewater by anaerobic digestion combined with an aquatic plant system offers good performance in terms of organics and nutrient removal for relatively low operation and maintenance costs. The results indicate that under appropriate operational conditions, the effluent quality is within the limits set by Korean discharge criteria.

콩 발효 식품(청국장, 물두시, 낫또)의 특성, In Vitro 항돌연변이 및 항암 효과 (Comparisons of Properties, In Vitro Anti-Mutagenicity, and Anti-Cancer Effects of Short-Term Fermented Soybean Foods (Chungkukjang, Shuidouchi, and Natto))

  • 본 연구에서는 콩에서 유래한 단기 발효식품인 청국장, 물두시, 낫또 제품 중 대표적인 제품을 각각 하나씩 선택하여 이들의 일반적 특성과 항돌연변이 효능 및 항암 효능을 비교 연구하였다. 청국장은 아미노태 질소 및 암모니아태 질소의 함량이 높았으나, 낫또는 그 함량이 낮게 나타났다. 물두시의 경우 여타 발효식품에 비해 pH가 낮고 산도가 높았다. Ames test 및 HT-29 인체 대장암세포를 이용한 실험에서는 청국장이 우월한 항돌연변이 및 항암 효능을 나타냈다. 이상의 결과로 볼 때 청국장은 전반적으로 기능성이 우수한 것으로 나타났으며, 물두시는 낫또에 비해 기능성이 더 나은 것으로 나타났다. 이는 발효 기간의 차이, 제조방식, 원재료의 품질 등에 의한 것으로 여겨지며, 향후 각국의 식문화 및 조리 방식 등의 차이에 기초를 둔 추가적인 연구가 필요할 것으로 여겨진다.

  • 주형합성법을 이용하여 메조기공(mesoporous pore)을 지닌 나노실리카 물질들과 나노카본볼의 대량생산법이 개발되었다. 암모니아, 트리메틸아민, 아세트알데히드 그리고 메틸메르캅탄과 같은 악취 물질들이 마크로기공 코어-메조기공 쉘(macro-porous core/mesoporous shell) 구조체인 나노카본볼에 흡착되는 현상이 상업용 탈취제인 활성탄과 비교 조사되었다. 나노카본볼에서의 악취의 흡착 및 분해는 활성탄에 비해 우수하게 관측되었고 촉매가 나노카본볼 내부에 첨착되면 더욱 악취 분해 특성이 증가되었다. 나노카본볼의 우수한 흡착 및 분해 특성을 이해하기 위해 물리화학적 특성인 균일한 기공, 넓은 표면적, 큰 기공 부피에 관한 기공 특성과 악취의 분해 현상이 연구되었다. 이러한 나노카본볼은 탈취제 분야에서 많은 응용성이 있을 것으로 전망된다.

  • 양송이 합성배지(合成培地)의 야외발효과정(野外醱酵過程)을 성력기계화(省力機械化)하기 위한 방법(方法)으로서 분쇄(粉碎)볏짚에 영양원(營養源)을 첨가(添加)하여 단기간(短期間) 발효(醱酵)시킨 다음 후발효(後醱酵)하는 시험(試驗)을 실시(實施)한 결과는 마음과 같다. 1. 분쇄(粉碎)볏짚의 단기발효배지(短期醱酵培地)는 Ammonia 태질소(態窒素)의 잔류량(殘留量)은 낮으나 전실소함량(全室素含量)이 관행배지보다 낮았다. 2. 미생물(微生物)은 분쇄볏짚이 관행배지보다 밀도(密度)가 높아서 발효기간(醱酵期間)의 단축(短縮)이 가능함을 보였다. 3. 양송이 자실체수량(字實體收量)은 관행배지보다 낮은 경향이었으나 영양원(營養源)의 첨가(添加)를 개선(改善)하므로서 향상(向上)시킬수 있는 가능성이 보였다. 4. 단기발효배지(短期醱酵培地)의 제조시(製造時) 수분(水分) 및 영양원(營養源)의 조절(調節)이 곤란하였고 이를 위해서 최소(最少) 5일간(日間)의 퇴적(堆積) 및 뒤집기작업(作業)이 필요하였다. 5. 배지재료(培地材料)로서는 일본형품종(日本型品種)의 볏짚이나 통일계품종(統一系品種) 모두 사용(使用)할수있었고 영양원(營養源)은 들깻묵과 미강을 혼합(混合) 첨가(添加)하는 것이 가장 좋았다.

질소와 암모니아 존재하에서 1 keV 에너지의 알곤과 수소 이온 조사에 의한 PTFE(polytetrafluoroethylene)의 표면형상 변화연구 (Surface Modification of Polytetrafluoroethylene by 1 keV Argon and Hydrogen Irradiated in Nitrogen and Ammonia Gas Environment)

  • Polytetrafluoroethylene (PTFE) surface was modified for improving hydrophilicity by ion irradiation in environmental gas of $N_2$ and $NH_3$, respectively. The water contact angle onto the PTFE surface increased from $104{\circ}$ to over $140{\circ}$ by Ar ion irradiation in $N_2$ gas. In the case of $NH_3$ as environmental gas, there were a slight increase of contact angle from ion dose of $1{\times}10^{15}\;to\;5{\times}10^{15}\;ions/cm^2$, and its dramatic decrease to the value of 35o at the conditions of ion dose higher than $1{\times}10^{16}\;ions/cm^2$. It was found from SEM results that the surface morphology of PTFE was changed into one with filament structure after Ar ion irradiation in $N_2$ gas environments. On the contrary, Ar ion irradiation in $NH_3$ gas condition induced the PTFE surface with network structure. Hydrogen ion irradiation resulted in a little change of PTFE surface morphology, comparing with the case of Ar ion irradiation. The water contact angle of hydrogen ion irradiated PTFE surface in reactive gas decreased with increment of ion dose. Hydrogen ion irradiation could improve hydrophilicity with little change of surface morphology. It might be considered from FT-IR results that the improvement in wettability of PTFE surface by ion irradiation in $N_2$ and $NH_3$ gases could be due to the hydrophilic groups of NHx bonds.

정수처리장 사여과지의 이단이중여과재 시스템으로의 개량 (Improvement of Rapid Sand Filtration to Two Stage Dual Media Filtration System in Water Treatment Plant)

  • This study aimed for evaluating the applicability of the two stage dual media filtration system in field water treatment plant. The field plant of two stage and dual media filtration system was operated for 2 months. Average iron concentrations of the settled water, existing filtered water and second stage filtered water was 0.041 mg/L, 0.007 mg/L and 0.005 mg/L, respectively. Removal efficiency of iron concentration in the second stage is appropriately 35% more than in existing filtered water. Also removal efficiency of residual chlorine in the dual media filtration system is relatively 42.3% more than in existing filtered water due to adsorption of activated carbon, but the removal of ammonia nitrogen by adsorption is insufficient. Average concentrations of THM and chloroform in the settled water are 0.033 mg/L, 0.026 mg/L, respectively and in existing filtered water are 0.023 mg/L and 0.023 mg/L. Average concentrations of THM and chloroform in the dual media filtration system are 0.008 mg/L and 0.013 mg/L. Therefore removal efficiency of THM concentration in second stage is more than 66.4% in existing filtrated water. Also removal efficiency of chloroform in the dual media filtration system is more than 50.0% in existing filtered water because of the adsorption of activated carbon. In this case backwashing period in dual stage system is 4~5 days, but in existing filtration system is 1~2 days.

  • 인산감수성(燐酸感受性)이 다른 9개대두재배종(個大豆栽培種) Lee, Hill, Harosoy, Clark-63, Chippewa 및 R56-49을 서로 다른 질소원(窒素源)으로 수경재배(水耕栽培)하여 엽중유리당(葉中遊離糖)과 유기산(有機酸)을 gas chronatograph로 조사(調査)하여 미동정(未同定)의 X, Y peak와 Sucrose peak가 주(主) peak로 나타났다. X 화합물(化合物)은 NO3-N 급여(給與)경우 흔적으로 나타난데 반하여 NH4-N나 urea 급여(給與)경우에 다량(多量)나타났다. Y 화합물(化合物)을 Urea 급여(給與)경우 감소하는 경향이었다. Sucrose는 NH4-N 급여(給與)에서 흔적으로 나타나고 Urea에서 NO3-N에서보다 높았다. X 화합물(化合物)은 erythrose에서 유래(由來)한 4 탄소(炭素) 산성당(酸性糖)이거나 Purine이나 Pyrimidine에서 유래(由來)한 환상화합물(環狀化合物)로 Y 화합물(化合物)은 glycolysis path에서 유래(由來)한 6 탄당(炭糖)으로 추정(推定)되었다. Y/X 치(値)는 인산감수성(燐酸感受性)의 좋은 지표(指標)가 되므로(역관계(逆關係)) 이 두 화합물(化合物)은 ammonia 독작용(毒作用) 및 인산감수성(燐酸感受性) 기작(機作)에 관여(關與)하는 요인화합물(要因化合物)로 보였다.

매립지 침출수 현장 처리를 위한 폴리우레탄과 개질토의 특성 분석 실험에 관한 연구 (Characterization of Polyurethane and Soil Layers for In-situ Treatment of Landfill Leachate)

  • A chemical and biological permeable barrier with economic feasibility is suggested to treat landfill leachate in this study. The proposed composite layers consist of bentonite, and polyurethane (PU) foam that is mixed with powdered activated carbon (PAC) and inoculated with microorganisms from local wastewater treatment plant. Each layer is mixed with local sand, and yellow brown soil. Batch tests were conducted to investigate the sorptions of nitrate on the PU foam and PAC, and nitrification/denitrification rate of each layer material. Nitrification occurred in 30 minutes with initial ammonia concentration of 100 mg/L, and the concentration of nitrate attached in the PU foam increased after 270 minutes. Results of denitrification batch tests showed 76.6%, 87.3% and 88% of nitrate removal efficiency at 10%, 20% and 30% of the volume ratio of PU foam, respectively. The pH increased from 7 to 9.42, and alkalinity increased from 980 mg/L to 1720 mg/L during the denitrification batch tests. In the column experiments using the proposed composite layers with 20% of the volume ratio of the PU foam, about 96% of BOD, 63% of COD, 58.1~79.5% of total nitrogen were removed.
