The retinae and the lens bodies of the frogs were hydrolyzed with 20% hydrochloric acid, and their amino acids were separated by paper chromatography. (2) The free amino acids in the retinae were extracted with 80% ethyl alcohol and then separated by paper chromatography. Though their separation was not so definite , serine, gultaminc acid, and glycine were always separated regardless of the amount of the sample. When the amount of the smaple was enough , $\beta$-alanine , ${\gamma}$-amino butyric acid and methionine +valine were also separated. (2) The free amino acids in the retinae were extracted with 80% ethyl alcohol and then separated by paper chromatography. DL-1-aminobutylphosphonic acid was synthesized from the pentanoic acid which was prepared from the butyl alcohol, by the modified Curtius reaction. DL-2-amino-2-carboxyethylphosphonic acid was also synthesized from the pyruvic acid was also synthesized from the pyruvic acid. Four previously unreported N-acylated derivatives were prepared according to the modified Schotten-Baumann method. They are as follows; N-acetyl-DL-1-aminobutylphosphonic acid, N-benzoyl-DL-1-aminobutylphosphonic acid, N-benzoyl-DL-2-amino-2-carboxyethylphosphonic acid, N-p-chlorobenzoyl-DL-2-amino-2-carboxyethylphosphonic acid. The products were identified by the methods of elemental analysis, infrared spectra, ninhydrin test and neutralization equivalent.
The antibacterial actions of two amino acid oxidases, a D-amino acid oxidase from hog kidney and a L-amino acid oxidase from the venom of Agkistrodon halys, were investigated, demonstrating that both enzymes were able to inhibit the growth of both Gram-positive and Gram-negative bacteria, and that hydrogen peroxide, a product of their enzymatic reactions, was the antibacterial factor. However, hydrogen peroxide generated in the enzymatic reactions was not sufficient to explain the degree to which bacterial growth was inhibited. A fluorescence labeling assay showed that both of these two enzymes could bind to the surfaces of bacteria. To the best of our knowledge, this is the first report regarding the antibacterial activity of the D-amino acid oxidases.
We have isolated and characterized a new insect chicken type (c-type) lysozyme gene from the common cutworm, Spodoptera litura. The full-length cDNA of Spodoptera lysozyme is cloned by rapid amplification of cDNA ends PCR (RACE-PCR). The isolated cDNA consists of 1039 bp including the coding region for a 142-amino acid residue polypeptide, which included a signal peptide of 21-amino acid residue and a mature protein of 121-amino acid residue. The predicted molecular weight of mature lysozyme and its theoretical isoelectric point from amino acid composition is 13964.8 Da and 9.05, respectively. The deduced amino acid sequence of Spodoptera lysozyme gene shows the highest similarity (96.7%) to Spodoptera exigua lysozyme among other lepidopteran species. Amino acid sequence comparison with other the c-type lysozymes, Spodoptera lysozyme has the completely conserved $Glu^{32}$ and $Asp^{50}$ of the active site and eight Cys residues are completely conserved in the same position as that of other lepidopteran lysozymes.
Simple and rapid screening methods were developed to screen mutant strains of Trigonopsis variabilis ATCC10679 (TW). D-amino acid oxidase (D-AAO) from a mutant strain, T26, showed about 30% higher specific activity against cephalosporin C than from its wild type, TW. D-AAO genes from both TW and T26 strains were cloned and sequenced. There was one nucleotide changed from T to C at 811 position, resulting in an amino acid codon changed from Phe-258 to Ser-258.
This experiment was undertaken the purpose of investigating the difference of the content of free amino acids in the diseased and healthy leaves of the resistance and susceptible rice varieties, to blast by means of paper chromatography. The results obtained are summarized as follows. (1) In the healthy and diseased leaves, the former was founded nine to tell sorts of free amino acids and later was observed eleven. In the both of leaves, alanine and valine were contained in larger amount, and aspartic acid, glycine and arginine were found relatively less in general. (2) In the diseased leaves, the amino acids were increased in quantity, especially glutamine and aspartic acid were remarkable, and cystine were found only in the diseased ones. (3) It seems to that the health leaves in all varieties were observed the amino acid contents, in notably increase and decrease, but in the diseased ones were not changed in amount. Tatacho, susceptible variety was more contained the amount of glutamine and aspartic acid than Zenith, resistance one.

Quantification of endogenous amino acid loss at the terminal ileum is an essential means for calculation of the true amino acid digestibility of a feedstuff. Since nitrogen appeared in the determined diet or not could shift the results very much, also, none of digestibility markers could be recovered with 100% rate at the terminal ileum, the objectives of the present study were: (1) to determine endogenous amino acid losses when fed either a casein diet or a protein-free diet and (2) to examine the reliability of chromic oxide or acid insoluble ash in the protein-free diet. Six ileal-cannulated pigs ($65{\pm}1.85 kg$ BW) with a simple T-cannula in the terminal ileum were used in a replicated $3{\times}3$ Latin square designed trial, after allowed a 14 d recuperation period. Each test period ran for 12 days comprised of a 10 d adjustment period and a 2 d collection period. The endogenous AA losses of His, Ile, Lys, Cys, Thr, Val, Trp, Asp, Glu, and Ser from pigs fed the casein diet were significantly higher than those of the protein-free diet (p<0.05). No significant difference was found in the amount of endogenous amino acid loss when determined with the different markers in the protein-free diet (p>0.05). These data suggest that endogenous amino acid loss could be underestimated when a protein-free diet is used. A direct effect of dietary peptides on the endogenous amino acid loss was found when the casein diet was fed. Our results also indicate that acid insoluble ash can be used as an inert marker as an alternative to chromic oxide when measuring endogenous amino acid loss.
Takju (Korean native Sake) was mashed with two different materials. One of the material was polished rice and Nuruk(mold wheat), the other one was corn and Nuruk. The amino acids in those fermenting mashes were identified by paper partition chromatography at regular intervals. The results were summarized as follows; a) Following 14 kinds of amino acids were identified in the mash of rice material; lysine, valine, proline, leucine, serine, glycine, aspartic acid, alanine, cystine, tyrosine, histidine, glutamic acid, tryptophan and phenylalanine b) Following 12 kinds of amino acids were identified in the mash of corn material; lysine, valine, proline, leucine, serine, glycine, aspartic acid, alanine, cystine, histidine, argrinine and tryptophan. c) The main amino acids in the Takju mash according to the color density of the each amino acid spot on the paper chromatograms were checked as lysine, valine, leucine, serine, proline and glycine.
Four fibrous feedstuffs from alfalfa meal (AFM), cassava leaf meal (CLM), rubber seed meal (RSM) and leucaena meal (LM) were included in semi-purified diets for growing pig (45 kg body wt.) at 20%, to investigate the effects of these fiber sources and fractions on amino acid digestibility. Cellulose (C), a purified fiber source was included in another diet at 5% level for comparison. The barrows fitted with ileal T-cannula were used in the digestion trials with latin square design. The digestibilities of amino acids were measured at both terminal ileum and fecal level. NDF and hemicellulose content were the highest in AFM-diet whereas LM-diet had the highest ADF and lignin content. RSM-diet contained the highest crude fiber and cellulose content. The digestibilities of amino acids at ileal level were found the highest with CLM-diet, while LM-diet was the least. At fecal level, control diet and CLM-diet were the highest in amino acid digestibility while AFM-diet was the least. The digestibility of amino acids was higher at ileal than fecal level. The digestibility of arginine was not affected with fiber fractions but was found to be the most disestible across all diets. The most depressed amino acid was methionine at both levels; praline and glycine, in the dispensable amino acid group, were depressed at ileal and fecal level, respectively. Lignin did not depress amino acid digestibility in general but specifically depressed methionine, histidine, isoleucine and threonine digestibility. Cellulose content did not affect amino acid digestibility but undesirable factors might be responsible.
Alpha-linolenic acid is an important polyunsaturated fatty acid that exhibits anticancer, anti-inflammatory, and antioxidative effects. In this study, we investigated the protective effects of alpha-linolenic acid on the cell proliferation and differentiation of C2C12 cells under essential amino acid-deficient conditions. Different concentrations of alpha-linolenic acid and essential amino acids were added to the growth and differentiation media. The concentrations of 10 µM of alpha-linolenic acid and 2% essential amino acid were chosen for subsequent experiments. Supplementation with alpha-linolenic acid and essential amino acids improved the proliferation and differentiation of C2C12 cells and significantly increased the mRNA levels of catalase, superoxide dismutase, B-cell lymphoma-2, and beclin-1 as well as the protein levels of PPARγ coactivator-1α compared to those in the controls. Moreover, supplementation with alpha-linolenic acid and essential amino acids reduced the levels of phosphorylated H2A.X variant histone, Bcl-2-associated X, p53, and light chain 3 during C2C12 cell proliferation, and increased the expression levels of myogenic factors 4 (myogenin) and 5 during C2C12 cell differentiation. Overall, we determined that alpha-linolenic acid and essential amino acids maintained the cell proliferation and differentiation of C2C12 cells via their anti-oxidative, anti-apoptotic, and anti-autophagic effects.
