The Effects of Enzyme Complex on Performance, Intestinal Health and Nutrient Digestibility of Weaned Pigs

  • Two experiments were conducted to evaluate the effect of supplementing a corn-soybean meal-based diet with an enzyme complex containing amylase, protease and xylanase on the performance, intestinal health, apparent ileal digestibility of amino acids and nutrient digestibility of weaned pigs. In Exp. 1, 108 piglets weaned at 28 d of age were fed one of three diets containing 0 (control), 100, or 150 ppm enzyme complex for 4 wks, based on a two-phase feeding program namely 1 to 7 d (phase 1) and 8 to 28 d (phase 2). At the end of the experiment, six pigs from the control group and the group supplemented with 150 ppm enzyme complex were chosen to collect digesta samples from intestine to measure viscosity and pH in the stomach, ileum, and cecum, as well as volatile fatty acid concentrations and composition of the microflora in the cecum and colon. There were linear increases (p<0.01) in weight gain, gain: feed ratio and digestibility of gross energy with the increasing dose rate of enzyme supplementation during the whole experiment. Supplementation with enzyme complex increased the digesta viscosity in the stomach (p<0.05) and significantly increased (p<0.01) the concentrations of acetic, propionic and butyric acid in the cecum and colon. Enzyme supplementation also significantly increased the population of Lactobacilli (p<0.01) in the cecum and decreased the population of E. coli (p<0.05) in the colon. In Exp. 2, six crossbred barrows (initial body weight: $18.26{\pm}1.21$ kg), fitted with a simple T-cannula at the distal ileum, were assigned to three dietary treatments according to a replicated $3{\times}3$ Latin Square design. The experimental diets were the same as the diets used in phase 2 in Exp. 1. Apparent ileal digestibility of isoleucine (p<0.01), valine (p<0.05) and aspartic acid (p<0.05) linearly increased with the increasing dose rate of enzyme supplementation. In conclusion, supplementation of the diet with an enzyme complex containing amylase, protease and xylanase improved piglet performance. This is likely a result of improvement in nutrient digestibility, volatile fatty acid concentrations and bacteria ratio in the large intestine.

Effects of a specific blend of essential oils on apparent nutrient digestion, rumen fermentation and rumen microbial populations in sheep fed a 50:50 alfalfa hay:concentrate diet

  • Objective: An experiment was conducted to investigate the effects of a specific mixture of essential oils (MEO), containing thyme, clove and cinnamon EO, on rumen microbial fermentation, nutrient apparent digestibility and blood metabolites in fistulated sheep. Methods: Six sheep fitted with ruminal fistulas were used in a repeated measurement design with two 24-d periods to investigate the effect of adding MEO at 0 (control), 0.8, and 1.6 mL/d on apparent nutrient digestibility, rumen fermentation characteristics, rumen microbial population and blood chemical metabolites. Animals were fed with a 50:50 alfalfa hay:concentrate diet. Results: Ruminal pH, total volatile fatty acids (VFA) concentration, molar proportion of individual VFA, acetate: propionate ratio and methane production were not affected with MEO. Relative to the control, Small peptides plus amino acid nitrogen and large peptides nitrogen concentration in rumen fluid were not affected with MEO supplementation; while, rumen fluid ammonia nitrogen concentration at 0 and 6 h after morning feeding in sheep fed with 1.6 mL/d of MEO was lower (p<0.05) compared to the control and 0.8 mL/d of MEO. At 0 h after morning feeding, ammonia nitrogen concentration was higher (p<0.05) in sheep fed 0.8 mL/d of MEO relative to 1.6 mL/d and control diet. Ruminal protozoa and hyper ammonia producing (HAP) bacteria counts were not affected by addition of MEO in the diet. Relative to the control, no changes were observed in the red and white blood cells, hemoglobin, hematocrit, glucose, beta-hydroxybutyric acid, cholesterol, total protein, albumin, blood urea nitrogen and aspartate aminotransferase and alanine aminotransferase concentration. Apparent total tract digestibility of dry matter, crude proten, organic matter, and neutral detergent fiber were not influenced by MEO supplementation. Conclusion:The results of the present study suggested that supplementation of MEO may have limited effects on apparent nutrient digestibility, ruminal fermentation and protozoa and HAP bacteria count, blood cells and metabolites.

  • 백자실(柏子實)의 Amino 산(酸), 당(糖), 지방(脂肪), 무기물(無機物), vitamin을 분석(分析)하고 이 결과(結果)로 백서(白鼠)의 영양실험(榮養實驗)을 한 결과(結果), 단백질(蛋白質)의 소화율(消化率)은 87%, T.D.N.은 159.4%며 백자실중(柏子實中)에는 성장촉진(成長促進) 급(及) 사료효율(飼料效率)이 증대(增大)하는 U.G.F.의 존재(存在)를 확인(確認)하고 실험(實驗)한 백서(白鼠)를 해체(解體)한 결과(結果) 심장(心臟), 신장(腎臟), 비장(脾臟)이 약간(若干) 비대(肥大)하였음을 인정(認定)했다.

Higher Protein Digestibility of Chicken Thigh than Breast Muscle in an In Vitro Elderly Digestion Model

  • This study investigated the protein digestibility of chicken breast and thigh in an in vitro digestion model to determine the better protein sources for the elderly in terms of bioavailability. For this purpose, the biochemical traits of raw muscles and the structural properties of myofibrillar proteins were monitored. The thigh had higher pH, 10% trichloroacetic acid-soluble α-amino groups, and protein carbonyl content than the breast (p<0.05). In the proximate composition, the thigh had higher crude fat and lower crude protein content than the breast (p<0.05). Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of myofibrillar proteins showed noticeable differences in the band intensities of tropomyosin α-chain and myosin light chain-3 between the thigh and breast. The intrinsic tryptophan fluorescence intensity of myosin was lower in the thigh than in the breast (p<0.05). Moreover, circular dichroism spectroscopy of myosin revealed that the thigh had higher α-helical and lower β-sheet structures than the breast (p<0.05). The cooked muscles were then chopped and digested in the elderly digestion model. The thigh had more α-amino groups than the breast after both gastric and gastrointestinal digestion (p<0.05). SDS-PAGE analysis of the gastric digesta showed that more bands remained in the digesta of the breast than that of the thigh. The content of proteins less than 3 kDa in the gastrointestinal digesta was also higher in the thigh than in the breast (p<0.05). These results reveal that chicken thigh with higher in vitro protein digestibility is a more appropriate protein source for the elderly than chicken breast.

Phytase 첨가가 산란계의 생산성, 난질, 질소와 인의 배설량 및 회장 소화율에 미치는 영향 (Effects of Dietary Phytase on Performance, Egg Quality, Excretion of N and P, and Ileal Digestibility in Laying Hens)

  • 산란계의 생산성, 계란 품질, 질소와 인의 흡수량과 배설량 및 회장 소화율에 미치는 파이타제의 첨가 효과를 구명하고자 사료에 0, 300, 600 FTU/kg의 파이타제를 첨가하여 4주간 사양 시험 종료후 1주간 대사실험을 실시하였다. 공시동물은 산란율이 유사한 55주령 White Leghorn 산란계 162수$(1.6{\pm}0.3\;kg)$를 공시하였으며, 처리구는 첨가 수준에 따라 P0(대조구), P1(300 FTU/kg), P2(600 FTU/kg)의 3처리구로 나누고, 처리구당 6반복, 반복당 9수씩 완전임의 배치하였다. 산란율은 파이타제 첨가 수준에 따라 증가하였다. 계란 품질은 난중, 난각 두께, 하우유니트와 난황색 모두 처리구간에 차이를 보이지 않았으나(p>0.05), 난각 강도는 파이타제 처리구에서 4주째에 각각 3.86 $kg/cm^2$와 3.85 $kg/cm^2$로 나타나 파이타제 첨가에 따른 차이를 보였다(p<0.05). 질소 배설량과 흡수량은 처리구간에 차이가 없었으나(p>0.05), 인 배설량과 흡수량은 파이타제 첨가구에서 대조구에 비해 배설량이 크게 감소하고 흡수량은 증가하였다(p<0.05). 파이타제 첨가에 따른 영양소와 아미노산의 소화율은 조섬유, 조회분, 칼슘과 인, 필수 아미노산 중 lysine, methionine, phenylalanine, 비필수 아미노산 중 alanine, cystine, glutamic acid, glycine, tyrosine의 소화율을 유의적으로 증가시켰다(p<0.05). 이 실험 결과 산란계에 미생물유래 파이타제 급여는 산란율을 향상시키고, 인 배설량을 낮추며, 흡수량을 증가시키지만, 계란 품질에 미치는 영향이 없었고, 소화율에 대한 영향은 추후의 연구가 필요하다고 사료된다.

Replacement of Soyflakes with Cottonseed Meal in Diets of Angora Rabbits

  • This experiment was conducted with 30 adult wool producing Angora rabbits of either sex, to evaluate the effect of replacing soyflakes (SF), on equal protein basis, with low gossypol containing cottonseed meal (CSM) either alone or in combination with lysine and methionine; on the biological performance, total wool yield, wool yield per shearing, wool characteristics and mortality. Three experimental mash diets were prepared by incorporating SF (6%) as standard / control protein source $(T_1)$ and CSM (9%) as test protein source ($T_2$ and $T_3$). In $T_3$, amino acids-lysine and methionine (0.1 % each) were added. Animals were given the experimental diets about $150g{\cdot}day^{-1}{\cdot}head^{-1}$, for a period of 225 d or three shearing, and ad libitum Kudzu-vines. No significant effect of $T_2$ or $T_3$, on the body weight gain, total wool yield, wool yield per shearing and wool characteristics, was observed compared to $T_1$. However, the digestibility of dry matter, crude fibers, ether extract, acid detergent fibers, neutral detergent fibers, cellulose and hemicellulose was significantly (p<0.05) depressed in CSM based diets. Mortality of about 20% was recorded in $T_2$ and $T_3$, but not related to the addition of CSM or gossypol toxicity. More studies are needed to standardize the safe level of CSM, duration of safe feeding of CSM, and level of amino acids supplementation in CSM based diets.

PROTEIN SPARING EFFECT AND AMINO ACID UTILIZATION IN BROILERS FED TWO TYPES OF LYSINE

  • A growth experiment was conducted to evaluate the nutritivie values of supplemental lysine and methionine in broiler chicks. Two types of L-lysine, liquid and powder type, and DL-methionine were added to the diets at different levels of dietary protein with two growth phases, 0-3 weeks and 4-6 weeks named starter and grower, respectively. Six hundred seventy two chicks were allotted in 14 treatments; 3 controls by dietary CP level (starter-grower) with CP 23-21%, CP 21-19% and CP 20-18, 8 groups of liquid and powder lysine supplementation of 0.1, 0.2, 0.3 and 0.4%, and 3 groups of lysine and methionine supplementation. Body weight, feed intake, and excreta were measured and analyzed to determine growth performance, amino acid digestibilities, and the quantity of excreted nitrogen in feces. Chicks fed CP 23-20 with 3,200 ME kcal showed significantly better growth performance than those fed CP 21-18 for 6 weeks. The supplementation of 0.2% of either type of lysine to CP 21-19 diet improved weight gain and feed efficiecy to the extent that CP 23-21 diet was fed. Physical type of lysine did not affect chick's growth and amino acid digestibilities of the diets. The level of CP in the diet significantly affected nitrogen excretion in feces. Supplementation of lysine and methionine to CP 21-18 diet reduced fecal nitrogen by 10% compared to CP 23-21 diet. It was confirmed that 0.2% of supplemental lysine to the broiler diet spared the dietary protein by 3%, and also reduced nitrogen excretion in feces by 10%.

Amino acid, fatty acid, and carbohydrate metabolomic profiles with ginsenoside-induced insecticidal efficacy against Ostrinia furnacalis (Guenee)

  • Background: Previous studies have shown the insecticidal efficacy of ginsenosides. In the present study, we aimed to investigate the metabolic mechanism related to the inhibitory effect of panaxadiol saponins (PDSs) against the Asian corn borer Ostrinia furnacalis (Guenee). Methods: Third instar larvae of O. furnacalis were fed normal diets with different concentrations of PDSs for 4 days. The consumption index, relative growth rate, approximate digestibility, and conversion of ingested and digested food were recorded. A targeted gas chromatographye-mass spectrometry assay was performed to detect the profiles of amino acids, fatty acids, and carbohydrates in larvae of O. furnacalis. In addition, the activity of detoxification-related enzymes was determined. Results and Conclusions: PDSs decreased the consumption index, relative growth rate, approximate digestibility, and conversion of ingested and digested food in the 3rd instar larvae of O. furnacalis in a dose-dependent manner. PDSs decreased 15 free amino acids, 16 free fatty acids, and 5 carbohydrates and increased the levels of palmitoleic acid, palmitic acid, and 9-octadecenoic acid in the 3rd instar larvae. The activity of detoxification-related enzymes, such as acetylcholinesterase, glutathione S-transferase, cytochrome P450, carboxylesterase, trehalase, acid phosphatase, and alkaline phosphatase, was reduced in a dose-dependent manner in the 3rd instar larvae exposed to PDSs. These data confirmed the inhibitory effect of PDSs against growth, food utilization, and detoxification in the 3rd instar larvae of O. furnacalis and the potential for using PDSs as an efficient tool for insect pest management for O. furnacalis larvae.

붕어고음 잔사분말을 첨가한 Cookies의 품질특성 (Protein Qualities and textural Properties of Cookies Containing Crucian Carp Extraction Residue)

  • To find the possibility in utilizing the fish meat processing by-products, protein nutritional quality and textural properties of crucian carp extraction residue (CCER, feeze dired) incorporated into cookies were investigated. Moisture, ash and protein contents in cookies were increased with the higher residue treatments, but lipid contents were similar within all levels (3%, 9% and 15%). Major constitutional amino acids were revealed as glutamic acid, proline, leucine and arginine, and the sum of those amino acids was about 50% of total amino acid contents. Cookies with residue (CCER) had higher (80.74~84.50%) in vitro protein digestibility than standard cookies (83.32%), while slightly lower trypsin indigestible substrate (TIS) contents were showed in CCER containing cookies than control. CCER treatments resulted the decreased protein nutritional quality in C-PER (computed protein efficiency ratio) value from 2.41 (standard) to 1.15 (cinnamon flavored. 9% CCER), and those C-PER of all cookies were lower than ANRC casein (2.50). Lipophilic browning was developed steadily till 60 days storage and a significant (p<0.05) changes of browning ws not noteed between 60 days and 90 days storage. Color of cookies, expressed as L, a and b value, was significantly (p<0.05) lightened with the increased CCER. Similar trends by treatments were noted for hardness. Cookies containing 9% CCER were similar to control regarding textual and sensory properties.

The Effect of Feeding Processed Soy Protein on the Growth Performance and Apparent Ileal Digestibility in Weanling Pigs

  • For Exp. 1, one hundred twenty Duroc${\times}$ Yorkshire${\times}$Landrace pigs (6.34${\pm}$ 0.70 kg average initial BW) were used in a 35 d growth assay. There were six pigs per pen and four pens per treatment. Dietary treatments included 1) NC (negative control; basal diet), 2) PC (positive control; added 5% HP $300^{(R)}$, HAMLET PROTEIN A/S, as protein source), 3) PSP 2.5 (added 2.5% $Pepsoygen^{(R)}$, Genebiotech Co. Ltd., as protein source), 4) PSP 5.0 (added 5% $Pepsoygen^{(R)}$ as protein source), and 5) PSP 7.5 (added 7.5% $Pepsoygen^{(R)}$ as protein source). For d 21, ADFI was increased in pigs fed PSP diets compared with PC diet (p<0.05). ADG was tended to improve as concentration of PSP in the diets was increased. For d 21-35 and entire experimental period, pigs fed PSP diets had more feed intake than pigs fed NC or PC diets (p<0.05). For the whole period, ADG was improved in PSP treatments compared with NC treatment (Linear effect, p=0.05). On d 21 and d 35, digestibilities of DM and N were higher (p<0.05) for pigs fed PSP diets than pigs fed NC diet and PSP 2.5 treatment was the highest among the treatments. On d 21, digestibility of N was significantly improved in PSP treatments compared with PC treatment (p<0.05). For Exp. 2, five Duroc${\times}$Yorkshire${\times}$Landrace barrows (8.74${\pm}$0.22 kg average initial BW) were surgically fitted with a simple T-cannulas approximately 15 cm prior to the ileo-cecal junction. The experimental designs were 5${\times}$5 latin squares with pigs and periods as blocking criteria. Dietary treatments were same as Exp.1. Generally, apparent ileal digestibilities of DM and N were higher for pigs fed PSP diets than pigs fed NC or PC diets (p<0.05) and PSP 5.0 and 2.5 treatments were the highest among the treatments, respectively. Apparent ileal digestibilities of the whole amino acids were increased in PSP treatments compared with NC treatment (Quadratic effect, p${\leq}$0.05). Also, histidine, lysine and threonine digestibilities in essential amino acids and alanine, asparatic acid and glycine digestibilities in non essential amino acids were significantly higher for pigs fed PSP diets than pigs fed PC diets (p${\leq}$0.05). In conclusion, feeding processed soy protein to weanling pigs increased growth performance, nutrient digestibility and apparent ileal digestibilities of DM, N and most amino acids.
