Studies on the Food Components of Triploid Abalone, Haliotis discus hannai (3배체 참전복, Haliotis discus hannai의 식품 성분에 대한 연구)

  • This study analyzed the proximate compositions, total amino acids, and fatty acid compositions in muscle and viscera of 51-month-old triploid and diploid Pacific abalone, Haliotis discus hannai. Proximate composition analysis showed that the muscle of diploid abalones had a higher protein and ash ratio than that of triploid abalones, while there was no significant difference in the ratios of moisture and fat (P>0.05). The triploid abalones had a significantly higher ratio of carbohydrate in muscle than the diploid abalones (P<0.05), which suggests that the transference of carbohydrates from muscle to gonad is decreased in sterile triploid organisms. Amino-acid analysis indicated that the total content was 133.42 mg/g in diploid muscle and 151.46 mg/g in triploid muscle. Taurine, arginine, glutamine, glycine, glutamic acid, alanine, and leucine, which are the major amino acids in abalone, were measured as 84.64 mg/g in triploid muscle and as 73.92 mg/g in diploid muscle. The essential amino acid content of triploid muscle (38.83 mg/g) was significantly higher than that of diploid muscle (31.94 mg/g) (P<0.05). Saturated fatty acids and monoene fatty acids were abundant in triploid muscle and polyene fatty acids were abundant in diploid muscle. Therefore, triploid Pacific abalone appears to be a good nutritional food source.

The Effect of Dietary Ionized Water and Premixed Mineral on Fatty Acid and Amino Acid Composition in Finishing Pigs (이온수와 복합광물질 급여가 비육돈의 지방산 및 아미노산 조성에 미치는 영향)

  • This study was conducted to determine the effects of ion water and premixed mineral supplementation on the growth performance, carcass, and meat quality parameters in finishing pigs (LY$\times$D). Each 20 pigs were randomly allotted to three treatments; CON (basal diet), T1 (CON diet added active water), T3 (T1 diet added 1.0% premixed mineral). Used ion water and premixed mineral consisted mainly of Zn and Si, respectively. Cholesterol content (mg% meat) were not differ significantly in the samples, however, the ratios of cholesterol to fat in T1 and T2 was significantly lower than the control (p<0.05). The fatty acid qualities of palmitic acid, linoleic acid, arachidonic acid, SFA (saturated fatty acid), EFA (essential fatty acid), and EFA/UFA ratio were the highest significantly, however, those of the oleic acid, UFA (unsaturated fatty acid), and UFA/SFA ratio were the lowest significantly in T2 (p<0.05). The amino acid levels of glutamic acid, alanine, leucine, lysine, total amino acids, and EAA (essential amino acid) were the highest in T1 (p<0.05). Whereas, the levels of serine, valine, isoleucine were the highest in T2 (p<0.05). The results indicate that dietary ionized water and premixed mineral affects fatty acid composition and improves amino acid composition.

Seasonal variation in the body composition, amino acid, fatty acid and glycogen contents of triploid Pacific oyster, Crassostrea gigas in western coastal waters of Korea (서해안 3배체 굴 (Crassostrea gigas) 의 계절별 체성분, 아미노산, 지방산 및 글리코겐 함량 변화)

  • We examined the seasonal variation of the proximate, amino acid, fatty acid composition and glycogen contents of triploid Pacific oyster cultured in western coastal waters of Korea in 2015. The specimens were collected in February, May, August and November from off-bottom farming site. The lipid content was the highest in winter season (February, $2.10{\pm}0.06%$), the protein content was the highest in autumn season (November, $9.09{\pm}0.04%$) while the moisture and ash content was the highest in summer season (August, $86.00{\pm}0.12%$ and $2.67{\pm}0.03%$, respectively). The total amino acid contents was the highest in winter season (1,011.5 mg / 100 g), decreased gradually and was the lowest in summer season (486.7 mg / 100 g). The total amino acid composition was dominated by taurine, aspartic acid, glutamic acid, glycine and alanine. In the fatty acid composition, the two fatty acids, eicosapentaenoic acid and docosahexaenoic acid, which comprised that as much as 50% of the total fatty acid contents, were dominated during all season. The glycogen content was the highest in summer season ($1.26{\pm}0.03g$ / 100 g) although it was stable during the other season with the range of $0.69{\pm}0.04-0.86{\pm}0.01g$ / 100 g. Taken together, these results suggest that triploid pacific oysters have high commercial value not only in winter season but in summer season because they do not spawn and have high contents of glycogen comparing with those of diploid oyster in this period.

Bread Baking Characteristics of Korean Wheat Varieties seen from their Amino Acid Composition (아미노산 조성(組成)으로 본 국산소맥(國産小麥)의 제(製)빵특성(特性))

  • Protein content and amino acid composition of three prospective Korean wheat varieties (Youngkwang, Wonkwang and Chokwang) were studied from the viewpoint of baking performance. 1. Protein contents of Korean varieties were Less than 10% which were relatively lower than the U. S. baking standard. These levels were considered insuffcient for best baking performance. 2. Compared to U. S. baking standard, Korean wheat varieties showed slightly lower level of glutamic acid, which is considered to play an important role in baking performance of flour. Approximately 25% decrease in glutamic acid content was observed in Korean varieties when the absolute amount of glutamic acid per unit sample size was compared. 3. Korean wheat varieties showed similar levels of proline and cystine, slightly higher levels of lysine, ammonia and aspartic acid; and slightly lower level of essential amino acids compared to the U. S. baking standard. 4. Protein content and amino acid composition of Korean wheat varieties were considered to be intermediate of U. S. baking and cookie standards. From this viewpoint, Youngkwang showed the greatest potential for best baking performance among Korean varieties.

  • The amino acid analysis of ground rice and the crude protein isolated from rice were performed by the most recent method of Moore, Spackman and Stein, using Amberlite IR-120. The effect of carbohydrates on the hydrolysis of rice was found to be definite in losing several amino acids nevertheless Dustin's claim that large excess of hydrolyzing acid reduces the losses of amino acid. Consequantly the removal carbohydrates as much as possible is considered to facilitate the amino acid determination. The amino acid composition of the crude rotein of rice was very similar to that previously found by other investigators.

Chemical Composition of Glycoprotein from Urechis unicinctus (개불(Urechis unicinctus) 당단백질의 성분조성과 특성)

  • To confirm the food quality of Urechis unicinctus which have been favored as a special raw seafood in southern area in Korea, the chemical composition of Urechis unicinctus and those glycoprotein were studied. Freeze dried Urechis unicinctus was composed of more than 70% of crude protein and 15% of total carbohydrate. The amino acids such as glycine(18.69%), glutamic acid(12.50%) and aspartic acid(9.37%) were noted as major components of total amino acids. The predominant free amino acids were alanine(32.98%), glycine(27.50%), asparagine(19.65%) and taurine(8.29%), and the sum of them were more than 88% to total free amino acids, so they may cause unique taste of Urechis unicinctus with sweetness. As the basis of sugar composition analysis, 56.35% of glucose and 30.49% of N acetylglucosamine were contained respectively, and they might also play an important role in a sweet taste. The leading carbohydrate moiety of glycoprotein from Urechis unicinctus was identified as glucose and N acetylglucosamine similarly to raw muscle, and they occupied more than 50% of total sugar content. Fucose(18.32%) and D glucuronic acid(12.31%) also detected in higher levels com pared to raw muscle. The total amino acid profile of glycoprotein showed a similar trend to raw muscle protein but glycine was detected a significantly lower than that in raw muscle. The glycoprotein from Urechis unicinctus was composed of 4 kinds of subunits, i.e. 25kDa, 20kDa, 18kDa and 12.5kDa of molecular weights through the SDS polyacrylamide gel electrophoresis. On the basis of the IR spectrum of absorptions appeared in 1,240cm-1 and 850cm-1, the glycoprotein had sulfate groups.

  • Amino acid complex was made with the composition of amino acids quite similar to that of stratum corneum on the skin. In order to evaluate the efficacy of amino acid complex on the skin as an active cosmetic ingredient, we measured cytotoxicity. TIMP-1 expression, moisturizing effect and the amount of horny substance. In cytotoxicity assay, amino acid complex did not show any cytotoxicity. In moisturizing effect test using corneometer, it showed very good moisturizing effect. In TIMP-1 mRNA assay using RT-PCR, moo acid complex showed the increase of TIMP-1 expression, suggesting that amino acid complex have anti-wrinkle effect. Therefore, amino acid complex may be useful as an active ingredient for anti-aging.

Composition of Fatty Acid and Amino Acid in Water Extracted Material from Cockscomb Plant Root (맨드라미 뿌리의 물 추출물질의 지방산과 아미노산의 조성)

  • Composition of fatty acids and amino acids in water extracted material of cooks comb plant root was studied by HPLC and GC-MS. There are 14 kinds of fatty acids in water extracted material of cockscomb plant root, especially, hexadecadienoic acid$(C_{16:2})$ was contained. Tricoasnoic acid, lignoceric acid, cerotic acid and 1,1'-[3-(2-cuclopentylethyl:dene)-1,5-pentanediyl] bis-cyclopentane were identified by HPLC and GC-MS. There are 16 kinds of acids containing ${\alpha}-aminobutyric$ acid in water extracted material of cockscomb plant root.

  • In this study, three kinds of Korean snake wine (50 V% alcohol extracts) were determined by amino acid analyzer and were discussed as follows. 1. Salmosa Ju and Nungsa Ju were composed of 22 free amino acids: cysteic acid, aspartic acid, threonine, serine, glutamic acid, proline, glycine, alanine, cystine, valine, methionine, iso-leucine, allo-iso-leucine, leucine, nor-leucine, tyrosine, phenylalanine, ${\beta}-alanine$, lysine, arginine, histidine, trytophan. 2. Doksa Ju were composed of 21 free amino acids which were all same as above except missing histidine. 3. The free amino acid composition were almost identical in Doksa Ju, Salmosa Ju and Nungsa Ju quantitatively. 4. The contents of cysteic acid, glutamic acid, glycine, alanine, valine, leucine and lysine were relatively high, on the other hand, methionine, allo-iso-leucine, nor-leucine, and tryptophan were trace amount in every snake wine. 5. Eleven unknowns of ninhydrin positives were identified in the every snake wine. 6. The free amino acids in snake wines were various in kind as compared with in beer, Japanese sake and Korean Tack Ju. Especially cysteic acid, allo-iso-leucine, nor-leucine and ${\beta}-alanine$ in snake wines were missed in every cereal wine.

Protein Quality and Amino Acid Utilization in Chickens

  • It is well known that dietary protein affects the growth performance and carcass composition of poultry. Over the last several decades, numerous studies have been carried out to investigate to optimize the level of dietary protein since the protein is an important and expensive constituent in poultry feed. It is generally accepted that dietary protein should represent a balance of amino acids supporting the requirements for growth and maintenance of birds. A protein with balanced essential amino acids that matches a bird's requirement and sufficient non-essential amino acid nitrogen to enable the synthesis of all of the non-essential amino acids, is referred to as an 'ideal protein'. Feeding of excess protein or amino acids may result in an amount of nitrogen emission. Most common method to reduce nitrogen emission is using diet formulation which has lower dietary crude protein level and higher concentration of amino acid supplements. However, there are conflicting reports whether low protein diets supplemented with synthetic amino acids can obtain the growth performance equal to high protein diets. Excessive nitrogen excretion caused by amino acid imbalance also may influence the environment of poultry house due to ammonia production from uric acid. These environmental conditions may increase the incidence of skin problem or respiratory diseases of chickens. Various strategies based on comprehensive understanding should be tested to optimize nitrogen utilization and reduce nitrogen emission while maintaining the performance in poultry production.
