Objective: This study was conducted to evaluate the effects of inclusion level and amino acid (AA) supplementation on energy values of soybean oil (SO) as determined by difference method or regression method when fed to growing pigs. Methods: Thirty-six barrows (initial body weight: 28.0±1.3 kg) were randomly assigned to one of 6 dietary treatments, which included 2 control diets formulated using a basal diet with or without AA supplementation, and 4 experimental diets with 5% or 10% SO addition in the 2 control diets, respectively. All pigs were individually housed in metabolism crates for 19 d, and during the last 5 d, total urine and feces production were collected. The nutrient digestibility in diets and the digestible energy (DE) and metabolizable energy (ME) values of SO were determined using the difference method and the regression method, respectively. Results: Our results showed that there were no interaction effects (p>0.05) between AA supplementation and SO inclusion levels on energy values of SO and dietary nutrient digestibility. The DE and ME values of SO determined by the difference method were not affected (p>0.05) by AA supplementation, however, the ME value of SO increased (p<0.05) as the inclusion level of SO increased. Moreover, the energy values of SO determined using the regression method were close to those determined using difference method with 10% SO inclusion, but were greater than those obtained using difference method with 5% SO inclusion. Conclusion: We concluded that the DE and ME values of SO increased with the inclusion level but were not affected by AA supplementation in the range of 0% to 10%. The difference method can substitute for the regression method to determine the DE and ME values of SO when the inclusion level is 10%, but not at 5% inclusion level.
As a novel approach for disease control and prevention, nutritional modulation of the intestinal health has been proved. However, It is still unknown whether branched-chain amino acid (BCAA) is needed to maintain intestinal immune-related function. The objective of this study was to determine whether BCAA supplementation in protein restricted diet affects growth performance, intestinal barrier function and modulates post-weaning gut disorders. One hundred and eight weaned piglets ($7.96{\pm}0.26kg$) were randomly fed one of the three diets including a control diet (21% crude protein [CP], CON), a protein restricted diet (17% CP, PR) and a BCAA diet (BCAA supplementation in the PR diet) for 14 d. The growth performance, plasma amino acid concentrations, small intestinal morphology and intestinal immunoglobulins were tested. First, average daily gain (ADG) (p<0.05) and average daily feed intake (ADFI) (p<0.05) of weaned pigs in PR group were lower, while gain:feed ratio was lower than the CON group (p<0.05). Compared with PR group, BCAA group improved ADG (p<0.05), ADFI (p<0.05) and feed:gain ratio (p<0.05) of piglets. The growth performance data between CON and BCAA groups was not different (p>0.05). The PR and BCAA treatments had a higher (p<0.05) plasma concentration of methionine and threonine than the CON treatment. The level of some essential and functional amino acids (such as arginine, phenylalanine, histidine, glutamine etc.) in plasma of the PR group was lower (p<0.05) than that of the CON group. Compared with CON group, BCAA supplementation significantly increased BCAA concentrations (p<0.01) and decreased urea concentration (p<0.01) in pig plasma indicating that the efficiency of dietary nitrogen utilization was increased. Compared with CON group, the small intestine of piglets fed PR diet showed villous atrophy, increasing of intra-epithelial lymphocytes (IELs) number (p<0.05) and declining of the immunoglobulin concentration, including jejunal immunoglobulin A (IgA) (p = 0.04), secreted IgA (sIgA) (p = 0.03) and immunoglobulin M (p = 0.08), and ileal IgA (p = 0.01) and immunoglobulin G (p = 0.08). The BCAA supplementation increased villous height in the duodenum (p<0.01), reversed the trend of an increasing IELs number. Notably, BCAA supplementation increased levels of jejunal and ileal immunoglobulin mentioned above. In conclusion, BCAA supplementation to protein restricted diet improved intestinal immune defense function by protecting villous morphology and by increasing levels of intestinal immunoglobulins in weaned piglets. Our finding has the important implication that BCAA may be used to reduce the negative effects of a protein restricted diet on growth performance and intestinal immunity in weaned piglets.

Recent evidence has shown that methionine (Met) supplementation can improve milk protein synthesis under hyperthermia (which reduces milk production). To explore the mechanism by which milk protein synthesis is affected by Met supplementation under hyperthermia, mammary alveolar (MAC-T) cells were incubated at a hyperthermic temperature of 42℃ for 6 h in media with different concentrations of Met. While the control group (CON) contained a normal amino acid concentration profile (60 ㎍/mL of Met), the three treatment groups were supplemented with Met at concentrations of 10 ㎍/mL (MET70, 70 ㎍/mL of Met), 20 ㎍/mL (MET80, 80 ㎍/mL of Met), and 30 ㎍/mL (MET90,90 ㎍/mL of Met). Our results show that additional Met supplementation increases the mRNA and protein levels of BCL2 (B-cell lymphoma-2, an anti-apoptosis agent), and decreases the mRNA and protein levels of BAX (Bcl-2-associated X protein, a pro-apoptosis agent), especially at an additional supplementary concentration of 20 ㎍/mL (group Met80). Supplementation with higher concentrations of Met decreased the mRNA levels of Caspase-3 and Caspase-9, and increased protein levels of heat shock protein (HSP70). The total protein levels of the mechanistic target of rapamycin (mTOR) and the mTOR signalling pathway-related proteins, AKT, ribosomal protein S6 kinase B1 (RPS6KB1), and ribosomal protein S6 (RPS6), increased with increasing Met supplementation, and peaked at 80 ㎍/mL Met (group Met80). In addition, we also found that additional Met supplementation upregulated the gene expression of αS1-casein (CSN1S1), β-casein (CSN2), and the amino acid transporter genes SLC38A2, SLC38A3 which are known to be mTOR targets. Additional Met supplementation, however, had no effect on the gene expression of κ-casein (CSN3) and solute carrier family 34 member 2 (SLC34A2). Our results suggest that additional Met supplementation with 20 ㎍/mL may promote the synthesis of milk proteins in bovine mammary epithelial cells under hyperthermia by inhibiting apoptosis, activating the AKT-mTOR-RPS6KB1 signalling pathway, and regulating the entry of amino acids into these cells.
Objective: The objective of the present study was to investigate the effect of dietary betaine (BT) supplementation on the hepatic transcriptome profiles in broiler chickens raised under heat stress (HS) conditions. Methods: A total of 180 (21-d-old) Ross 308 male broiler chicks were allotted to 1 of 3 treatment groups with 6 replicated cages in a completely randomized design. One group was kept under thermoneutral conditions at all times and was fed a basal diet (PC). Other 2 groups were exposed to a cyclic heat stress condition. One of the 2 groups under heat stress conditions was fed the basal diet as a negative control (NC), whereas the other group was fed the basal diet supplemented with 0.2% BT. All chickens were provided with diets and water ad libitum for 21 d. Following the experiment, the liver samples were collected for RNA sequencing analysis. Results: Broiler chickens in NC and BT group had decreased (p<0.05) growth performance. In the transcriptome analysis, the number of differentially expressed genes were identified in the liver by HS conditions and dietary BT supplementation. In the comparison between NC and PC treatments, genes related to energy and nucleic acid metabolism, amino acid metabolism, and immune system were altered by HS, which support the reason why heat-stressed poultry had decreased growth performance. In the comparison between NC and BT treatments, genes related to lipid metabolism, carbohydrate metabolism, and immune system were differently expressed under HS conditions. Conclusion: HS negatively impacts various physiological processes, including DNA replication, metabolism of amino acids, lipids, and carbohydrates, and cell cycle progression in broiler chickens. Dietary BT supplementation, however, offers potential counteractive effects by modulating liver function, facilitating gluconeogenesis, and enhancing immune systems. These findings provide a basis for understanding molecular responses by HS and the possible benefits of dietary BT supplementation in broiler chickens exposed to HS.

An 8 week feeding trial was conducted to evaluate whether methionine and cysteine would effectively spare taurine supplementation on growth performance in juvenile rock bream Oplegnathus fasciatus. Triplicate groups of 25 fish averaging $2.74{\pm}0.04g$ ($mean{\pm}SD$) were fed one of the experimental diets. Five experimental diets including a fish meal based control diet were prepared by adding sulfur amino acid at fixed level of 1.0% and taurine at graded levels of 0%, 0.25%, 0.5% and 1.0% ($S+Tau_0$, $S+Tau_{0.25}$, $S+Tau_{0.5}$ and $S+Tau_{1.0}$, respectively). After the feeding trial, growth performance of fish fed the $S+Tau_{0.25}$, $S+Tau_{0.5}$ and $S+Tau_{1.0}$ diets were significantly higher (P < 0.05) than those of fish fed the Control diet. ANOVA test suggested that when sulfur amino acid were supplemented to the diets, the optimum taurine supplementation level could be 0.25% in the diet, and broken line analysis of weight gain indicated a level of 0.33%, for positive effects on growth and feed utilization. Fish whole-body protein content and taurine concentration steadily increased with the increase of dietary level in the presence of sulfur amino acid in the diets. On the other hand, whole-body lipid content significantly decreased with the incremental levels of dietary taurine. In conclusion, the results of the present study clearly indicated that dietary supplementation of methionine and cysteine at a level of 1% could spare 0.25 to 0.33% of taurine in juvenile O. fasciatus diets.
This study evaluated fermented soybean meal (FSM) as a fish meal (FM) replacement and determined the appropriate amount of FSM in juvenile olive flounder diet. Twenty-four aquaria with a flowing-water system were stocked with fish averaging 20.9 g at a density of 25 fish/tank. Five experimental diets were prepared replacing FM with 0, 10, 20, 30, or 40% FSM based on FM protein (designated $FSM_{0}$, $FSM_{10}$, $FSM_{20}$$FSM_{30}$, and $FSM_{40}$, respectively). Two additional diets were prepared that replaced 30 or 40% of the FM with FSM with added amino acids (methionine and lysine) (designated $FSM_{30+AA}$, and $FSM_{40+AA}$, respectively). Fish (triplicates) were fed one of the eight experimental diets (50% crude protein and 16.7 kJ available energy $g^{-1}$ diet) for 8 weeks. Survival did not differ among the treatments during the feeding experiment. There were no significant differences in weight gain (WG) or specific growth rate (SGR) among the fish fed diets with up to 30% of the FM replaced. However, fish fed $FSM_{40}$ or $FSM_{40+AA}$ had a reduced WG and SGR, as compared to $FSM_0$ (control) (P < 0.05). The feed efficiency and apparent digestibility showed a similar trend (P < 0.05). The proximate composition in the whole body of fish differed only between the control and $FSM_{40}$ for the crude protein level and between the control and $FSM_{30+AA}$ for the crude lipid level. The whole-body amino acid composition did not differ among treatments. No significant differences were found between the diet groups with and without amino acid supplementation, indicating that amino acid supplementation had no effect. The major finding of this study is that fermented soybean meal may replace up to 30% of fish meal without amino acid supplementation for normal growth of juvenile olive flounder.
The effects of arginine (Arg) and methionine (Met) supplementation on nutrient use in pigs were determined under hot season conditions. A total of five experimental diets including basal diet (CON) were supplemented with two types of amino acids (Arg and Met) and two different amounts of amino acids (0.2% and 0.4%). Under hot season condition, pigs fed with additional Arg were significantly higher in average daily gain (ADG) than the CON group and the ADG increased linearly (p < 0.05) with increasing Arg supplementation. But there was no significant difference with Met supplementation (p > 0.05). The apparent ileal digestibility (AID) of amino acids had no significant difference among treatments (p > 0.05), while d-reactive oxygen metabolites (d-ROMs) concentration in treatments with Arg supplementation, were significantly higher (p < 0.05) than other treatments. In conclusion, exposure of pigs to heat stress does not affect the AID of amino acid, whereas pig fed with additional Arg improved ADG and feed efficiency under heat stress condition.
This experiment was conducted to determine the effect of dietary supplementation with BCAA (branched-chain amino acids: leucine, isoleucine and valine) on improving the growth of rats in a malnutritional IUGR (Intrauterine Growth Retardation) model, which was established by feeding restriction. In the experimental treatment, rats were fed purified diets supplemented with BCAA (mixed) during the whole gestation period, while arginine and alanine supplementation were set as the positive and negative control group, respectively. The results showed that, compared to the effect of alanine, BCAA reversed IUGR by increasing the fetus weights by 18.4% and placental weights by 18.0% while fetal numbers were statistically increased. Analysis of gene and protein expression revealed that BCAA treatment increased embryonic liver IGF-I expression; the uterus expressed higher levels of estrogen receptor-$\alpha$ (ER-$\alpha$) and progesterone receptor (PR), and the placenta expressed higher levels of IGF-II. Amino acid analysis of dam plasma revealed that BCAA supplementation effectively enhanced the plasma BCAA levels caused by the feed restriction. BCAA also enhanced the embryonic liver gluconeogenesis by augmenting the expression of two key enzymes, namely fructose-1,6-biphosphatase (FBP) and phosphoenolpyruvate carboxykinase (PEPCK). In conclusion, supplementation of BCAA increased litter size, embryonic weight and litter embryonic weight by improving the dam uterus and placental functions as well as increasing gluconeogenesis in the embryonic liver, which further provided energy to enhance the embryonic growth.

Three hundred and fifty-two growing pigs were utilized in three growth trials to investigate the effects of crystalline amino acid supplementation on growing pigs' growth performance and feed consumption. In Exp. 1, diets were formulated on the basis of digestible amino acids. Utilization of rapeseed meal plus cottonseed meal (R + C) in diet supported similar average daily gain (ADG), feed/gain (F/G) and cost per gain (p > 0.05) to group of soybean meal (SBM). In Exp. 2, 14% CP diets containing equal amount of cottonseed meal and rapeseed meal were formulated with the addition of threonine and/or tryptophan. Supplementation with threonine improved ADG and F/G (p < 0.05). Supplementation with only tryptophan made no benefits for ADG of growing pigs. In Exp. 3, diets with different threonine level were formulated. Increasing dietary threonine to 0.54% improved (p < 0.05) ADG and F/G compared to 0.45% dietary threonine. Increase in dietary threonine did not improve ADG (p > 0.05) but decreased F/G (p < 0.05) because of a decrease in average daily feed intake. Pigs fed diet with dietary threonine/lysine ratio of 67.5% supported optimum growth performance.
