Nalidixic Acid Prodrugs: Amides from amino acid ester and nalidixic acid

  • Amides from amino acid ester and nalidixic acid were synthesized. The solubility characteristics and partition coefficient of the compounds were studied. The hydrolysis of the compounds was studied in the simulated gastric fluid and simulated intestinal fluid. Some compounds showed better antibacterial activity than nalidixic acid.

배건담치의 정미성분에 관한 연구 (THE TASTE COMPOUNDS OF BROILED DRIED SEA MUSSELS)

  • This study was attempted to evaluate the taste of broiled-dried sea mussel which is a traditional dried Sea food in Korea. The contents of such compounds as free amino acids, nucleotides and their related compounds, TMAO, betaine, and nonvolatile organic acids were analysed. From the results of analysis of nucleotides, the contents of AMP and ADP in broiled-dried Mytilus coruscus and Mytilus edulis appeared higher than other nucleotides and tended to increase slightly after cooking, drying, and storage which might be due to both their stability and the addition of degradation of nucleic acids during cooking. In the free amino acid composition of fresh samples abundant amino were taurine, glycine, serine, glutamic acid, alanine, arginine, threonine, tyrosine, lysine and valine in order. Such amino acids as histidine, leucine, methionine and isoleucine were poor and both proline and phenylalanine were merely trace. The free amino acid composition in the extract of Mytilus corucus and Mytilus edulis after broil-drying was not changed. These amino acids, taurine, glycine, feline, glutamic acid, alanine and arginine were abundant in dried samples as well as in the fresh. The total free amino acid was greatly reduced after cooking and drying ranging from $76.3\%$ to $79.7\%$ loss to that of tile fresh. The content of betaine shelved the same tendency as in total free amino acid while TMA slightly increased relating to TMAO decrease during broil-frying. Such nonvolatile organic acids as succinic, lactic, malic and fumaric acid were abundant in both fresh and broiled-dried samples whereas oxalic and pyruvic acid were poor. It is found that the taste compounds of broiled-dried Mytilus coruscus and Mytilus edulis were composed of amino acids as glycine, serine, alanine, glutamic acid, arginine, and betaine, TMAO, ADP, AMP, and organic acids such as succinic acid, lactic acid, malic acid and fumaric acid. No significant differences in the taste compounds between male and female as well as between Mytilus coruscus and Mytilus edulis was not observed.

바지락젓의 유리(遊離)아미노산(酸) (Changes in the Free Amino Acids During the Fermentation of Clam, Tapes japonica)

  • Clam, Tapes japonica, has been widely used and occupies an important position in the markets. More than thirty kinds of fermented fish and shellfish are presently available in the markets in korea. This study was attempted to establish the basic data for evaluating the taste compounds of the clam. The free amino acids as taste compounds during the fermentation of the clam, Tapes japonica, were analyzed by an amino acid autoanalyzer. Alanine (17.0%), glycine (15.5%), taurine (13.9%) were high in amount in the fresh clam and then glutamic acid, arginine, tyrosine, asparagine, lysine, threonine, serine and leucine were the next in order. Such amino acids as methionine, phenylalanine, histidine and cystine were low in amounts. The total free amino acid nitrogen in the fresh clam was 45.5% of its extract nitrogen. During the fermentation of the clam, the kinds of the free amino acids were the same with the fresh samples although the amounts were changed. It is believed that glutamic acid, aspartic acid, glycine, alanine, lysine, and leucine may play an important role as the taste compounds in the fermented clam.

아미노산 성분에서의 염소 소독부산물 생성 특성 (Characteristics of Chlorination Byproducts Formation of Amino Acid Compounds)

  • 20종의 아미노산 성분들에서의 염소 소독부산물 생성 특성을 조사한 결과, 단위 DOC당 THM 생성능은 Br 첨가 유무에 관계없이 tryptophan과 tyrosine에서 높게 나타났고 나머지 18종에서는 매우 낮은 생성능을 나타내었다. 단위 DOC당 HAA 생성능 조사결과, tryptophan, tyrosine, asparagine, aspartic acid 및 histidine에서 Br 첨가 유무에 관계없이 높은 생성능을 보였으며, asparagine, aspartic acid 및 histidine은 DCAA 생성능 조사결과, aspartic acid, histidine, asparagine, tyrosine 및 tryptophan에서 Br 첨가 유무에 관계없이 높은 생성능을 나타내었으며, aspartic acid는 660.2 ${\mu}$g/mg으로 나타나 다른 아미노산 성분들에 비해 월등히 높은 HAN 생성능을 나타내었으며, 반응성이 높은 6종의 아미노산 성분에서 생성되는 HAN 구성종의 대부분은 DCAN으로 나타났다. 단위 DOC당 chloral hydrate 와 chloropicrin의 생성능을 조사결과에서 chloral hydrate는 asparagine, aspartic acid, histidine, methionine, tryptophan 및 tyrosine에서 생성능이 높은 것으로 나타났고, chloropicrin의 경우는 20종의 아미노산에서 전체적으로 1${\mu}$g/mg 이하의 아주 낮은 생성능을 보였다. 아미노산의 작용기별 소독부산물 생성 특성은 THM의 경우 방향족(aromatic) 화합물인 tryptophan과 tyrosine에 반응성이 높았고, HAA는 aromatic, neutral, acidic, basic 순으로 반응성을 보였으며, HAN은 acidic, basic, neutral, aromatic 순으로 반응성을 나타내어 방향족 화합물보다 지방족 화합물이며 친수성인 물질에서 반응성이 더 높은 것으로 나타났다.

  • Changes of free amino acids as taste compounds during the fermentation of oyster were analyzed by amino acid autoanalyzer. In fresh oyster, taurine, glutamic acid and alanine were abundant amino acids and the amounts of taurine (731mg%, on moisture and salt free base), glutamic acid (365mg%) and alanine (345.4mg% ) were 63.8% of the total free amino acids. Cystine, isoleucine, phenylalanine, leucine and histidine were detected as less abundant free amino acids and the amount of those amino. acids ranged from 5. 5mg% (cystine) to 32.9mg% (histidine). The free amino acids analyzed in this experiment were not changed in composition hut changed in amounts during 124 days of fermentation. Aspartic acid and leucine were continually increased during 124 days of fermentation. Lysine, histidine, threonine, serine, glutamic acid, tyrosine and phenyalanine were increased unlit 68 days of fermentation and than decreased gradually. The increase of arginine, glycine, valine and isoleucine were fluctuated. Taurine were dramatically decreased during the 124 days of fermentation. It is believed that glutamic acid, alanine, lecuine, serine, Iysine and threonine play an important role as taste compounds in fermented oyster because those amino acids were most abundant in fermented oyster.

한국전통간장의 맛과 향에 관여하는 주요 향미인자의 분석(II) - 질소 화합물, 유리아미노산 및 핵산관련물질 분석 - (Analysis of Significant Factor in the Flavor of Traditional Korean Soy Sauce (II) - Analysis of Nitrogen Compounds, Free Amino Acids and Nucleotides and Their Related Compounds -)

  • This study was carried out in order to analyzed the changes in nitrogen containing taste compounds of three different types of traditional Korean soy sauce with varing ripening period, and to investigates correlation between sensory characteristics and taste compounds contents via contents assay and sensory evaluation of soy sauce samples. Total nitrogen, ammonia type nitrogen and amino type nitrogen contents showed the highest value in Kyupjang. 17 kinds of free amino acid was detected in Chungjangs and 16 kinds of free amino acid was detected in Kyupjang. Nucleotides and their related compounds detected were hypoxanthin, xanthin, IMP, AMP, Inosine, ADP. Free amino acid and nucleotides and their related compounds contents were highest in Kyupjang. Nitrogen related compounds content of high concentration soy sauce ripened over 150 days increased similarly with Kyupjang. In the sensory evaluation of soy sauce taste, Chungjang samples acquired the highest score in the offensive taste test while Kyupjang marked highest score for sweet taste, nutty taste, taste preference. High concentration soy sauce ripend over 150 days yielded the result similar to that of Kyupjang. Sweet taste showed positive correlation with nitrogen compounds. The materials that showed positive correlation with nutty and traditional soy sauce taste and taste preference were nitrogen compound, IMP, AMP. Except for tryptophan, all free amino acid showed positive correlation with nutty and traditional soy sauce taste. Particulary, taste preference correlated to lysine, aspartic acid and glutamic acid.

  • Six previously unreported N-acylated-DL-1-amino alkyl phosphonic acids were prepared; N-Acetyl-DL-1-amino-3-methyl butyl phosphonic acidN-Benzoyl-DL-1-amino-2-methyl propyl phosphonic acidN-Benzoyl-DL-1-amino-3-methyl butyl phosphonic acidN-Benzoyl-DL-1-amino-2-methyl butyl phosphonic acidN-Acetyl-DL-1-amino-2-methyl propyl phosphonic acidN-Acetyl-DL-1-amino-2-methyl butyl phosphonic acidThe first four compounds were characterized, and the last two compounds were obtained in the crude oil state. The above three DL-1-amino-alkyl phosphonic acid were synthesized from iso-valeric acid, iso-caproic acid and ${\beta}$-methyl valeric acid using Hell-Volhard-Zelinsky reaction, the condensation reaction with triethyl-phosphite and the modified Curtius Reaction. Iso-caproic acid and ${\beta$-methyl valeric acid were prepared by the conventional methods.

보리새우육 엑스분의 추출조건과 그 유리아미노산 조성 (Effect of Extraction Condition on Free Amino acid Composition of Naturally Grown and Cutured Prawn Meat Extracts)

  • As a series of investigation on the ex-N compounds, the amino acid composition of the naturally grown and cultured prawn meat extracts was analyzed according to the following extraction conditions: 70% ethyl alcohol extraction, alcohol treatment of the same concentration after hot water extraction for 30 min, and 5% TCA treatment. The Protein content and the level of pure protein was 22 .8% and 18.4% in naturally grown prawn muscle, and 21.8% and 15.3% in the cultured prawn muscle, respectively. None protein nitrogenous compounds of the extracts were 4.4% in the naturally grown prawn muscle and 6.5% in the cultured prawn muscle, respectively. According to the different extract condition, ex-N compounds and the amino acid composition and related compounds and the amino acid composition and related compounds showed high level from 5% trichloroacetic acid extraction. The content of ex-N compounds of the cultured prawn muscle was 1033.0mg%. whereas the content of amino acids and related compounds-N was 825.3mg%, nearly reached to 80% to the total extractive nitrogen. A common feature of the free amino acid composition in the extracts of the muscles of the naturally grown and cultured prawn was high contents of glycine, proline, taurine, alanine and hydroxyproline.

건조(乾燥)멍게의 정미성분(呈味成分) (The Taste Compounds of Sun Dried Ascidian, Cynthia roretzi)

  • Changes of free amino acids, nucleotides and their related compounds as taste compounds during sun drying of ascidian Cynthia roretzi, were analyzed by amino acid autoanalyzer and high speed liquid chromatography. In fresh ascidian, the results showed that 5'-UMP $(12.1\;{\mu}mole/g)$ was dominant and the content of cytosine, 2', 3'-CMP, 2', 3'-GMP, hypoxanhtine, 5'-AMP,5'-IMP were 5.8, 3.4, 3.1, 2,3, 1.7 and $1.3\;{\mu}mole/g$ ondry base respectively. 5'-IMP, 2', 3'-CMP and 2', 3'-GMP tended to degrade slowly and 5'-AMP, cytosine and 5'-UMP were decreased rapidly while hypoxanthine were increased remarkably during the sun drying. In dried ascidian, the content of hypoxanthine was the highest, 7.2 mole/g on dry base, whereas that of 5'-AMP $(0.5\;{\mu}mole/g)$) and 5'-IMP $(0.9\;{\mu}mole/g)$ were lower. Glutamic acid, alanine and serine were dominant amino acid in the fresh extracts, having 22.4% (611.3mg%, on dry qase), 19.8% (540.5mg%) and 14.8% (402.8mg%) of the total amino acid content respectively. The content of tyrosine, histidine, lysine, methionine, isoleucine and valine were low, and proline, phenylalanine were detected in trace amount. The free amino acid were not changed in composition but the increase of total free amino acid was approximately 116.8mg% during sun drying. In sun dried ascidian, glutamic acid (691.0mg, on dry base), alanine (641.3mg%), serine (469.5mg%), threonine (234.8mg%) and glycine (206.3mg%) were dominant amino acid. It is believed that glutamic acid, serine, alanine, threonine, glycine and hypoxanthine play an important role as taste compounds in sun dried ascidian.

  • Catabolism of amino acids, including sulfur-containing amino acids, can be responsible for the development of cheese flavor during ripening Since accelerating, intensifying, modulating cheese flavor development is of major economical interests, the identification of flavor compounds and enzymes contributing to cheese flavor development needs to be investigated. Generally, two different pathways, which are a transamination pathway catalyzed by aminotransferases and an elimination reaction catalyzed by lyases, potentially lead to conversion of amino acids into flavor compounds. In this review, enzymes and amino acid catabolic pathways responsible for cheese flavor formation will be discussed.
