Furfural from Pine Needle Extract Inhibits the Growth of a Plant Pathogenic Fungus, Alternaria mali

  • The antifungal effect of pine needle extract prepared by a distinguishable extraction method and the dry distillation method, was examined. The effect of this extract itself was insignificant. The chemical components of pine needle extract were then investigated by gas chromatographic analysis, and four, chemical components, acetol, furfural, 5-methyl furfural, and terpine4-ol, were identified. The antifungal effects of those four chemical components against Alternaria mali (A. mali), an agent of Alternaria blotch of apple, were then examined. It was observed that the minimum inhibitory concentrations (MICs) were 6.25, 0.78, 0.78, and 12.5 (mg/ml) of acetol, furfural, 5-methyl furfural, and terpine-4-ol, respectively. MICs of furfural and 5-methyl furfural had the same order of magnitude as that of an antifungal agrochemical, chlorothalonil. Although furfural itself can not be completely substituted for an antifungal agrochemical, a partial mixture of furfural and antifungal agrochemical may be used as a substitute. The use of agrochemicals for the prevention of plant disease caused by pathogenic fungus such as A. mali could be partially reduced by the application of this mixture.

Characterization and Pathogenicity of Alternaria burnsii from Seeds of Cucurbita maxima (Cucurbitaceae) in Bangladesh

  • In the course of survey of endophytic fungi from Bangladesh pumpkin seeds in 2011~2012, two strains (CNU111042 and CNU111043) with similar colony characteristics were isolated and characterized by their morphology and by molecular phylogenetic analysis of the internal transcribed spacer, glyceraldehydes-3-phosphate dehydrogenase (gpd), and Alternaria allergen a1 (Alt a1) sequences. Phylogenetic analysis of all three sequences and their combined dataset revealed that the fungus formed a subclade within the A. alternata clade, matching A. burnsii and showing differences with its other closely related Alternaria species, such as A. longipes, A. tomato, and A. tomaticola. Long ellipsoid, obclavate or ovoid beakless conidia, shorter and thinner conidial size ($16{\sim}60[90]{\times}6.5{\sim}14[{\sim}16]{\mu}m$) distinguish this fungus from other related species. These isolates showed more transverse septation (2~11) and less longitudinal septation (0~3) than did other related species. Moreover, the isolate did not produce any diffusible pigment on media. Therefore, our results reveal that the newly recorded fungus from a new host, Cucurbita maxima, is Alternaria burnsii Uppal, Patel & Kamat.

  • This study was conducted to find out environment-friendly disease control method to Alternaria blight caused by Alternaria panax of ginseng. For this study, 150 methanol extracts from medicinal plants were evaluated and the extract of Impatiens balsamina showed most strong antifungal activity against A. panax. The methanol extract of I. balsamina showed also stable antifungal activity against other ginseng pathogens such as Botrytis cinerea and Fusarium sp., when treated by heat or pH. In vivo, Alternaria blight incidence rate was low of 13% with the treatment of I. balsamina methanol extract compared to 35% of the non-treatment. The antifungal compound of I. balsamina was purified and identified by using a silica gel column chromatography, TLC and ESI-LC/MS/MS analysis. The compound which showed strong antifungal activity was identified as 2-methoxy-1,4-naphtoquinone ($C_{11}H_8O_3$).

  • 먹물버섯 Coprinellus congregatus의 버섯조직이 자가분해되는 시기에 발현되는 chitinase (Chi2)는 사람에게 질병을 일으키는 Cryptococcus neoformans의 세포벽 두께를 정상 세포벽의 32% 수준까지 감소시킬 뿐만 아니라, 세포 외 섬유상 물질도 제거하였다. Chi2 처리에 의하여 사람에게 알레르기를 유발하는 Alternaria alternata의 생장은 물론 생쥐 모델에서 이 균에 의한 피부알레르기 증상도 감소시켰다.

Alternaria yunnanensis sp. nov., a New Alternaria Species Causing Foliage Spot of Rubber Tree in China

  • A new species of Alternaria causing leaf spots on the rubber tree (Hevea brasiliensis) in Yunnan, China, was isolated, examined, and illustrated. Morphologically, it belongs to the section Porri of Alternaria, which produces relatively large conidia and a simple or branched, filamentous long beak. It is, however, characterized by conidiophores gradually enlarging near the apex into a clavate conidiogenous cell and long ellipsoid to obclavate, smoothwalled conidia with a long filamentous beak. Molecular phylogenetic analyses based on ITS rDNA, GAPDH, and TEF1-alpha sequences demonstrate that the phytopathogen falls in the clade of the section Porri, being most closely related to A. sidae, A. sennae, A. deseriticola, A. cyamopsidis, A. rostellata, A. nitrimali, A. crassa, and A. thunbergiae.

Occurrence of Alternaria cassiae in Seeds of Sickle Senna in Korea

  • Alternaria cassiae Jurair & Khan was recorded on seeds of sickle senna(Cassia tora L.). Seed infection ranged from $25{\sim}45%$ in two samples. Developing roots and shoots turn brown and died in case of severe infection. Detailed descriptions were given on the habit characters of the fungus and on the morphology of conidiophores and condia. This is the first report of A. cassiae on seeds of Cassia.

Interactions Between Leafminer Damage and Leaf Necrosis Caused by Alternaria alternata on Potato in the Sultanate of Oman

  • Four field experiments were carried out from 1998 to 1999 and from 1999 to 2000 growing seasons of potato to investigate the relationship between leafminers and Alternaria alternata. The experiments established differential susceptibility among potato varieties to alternaria leaf necrosis, relationship between leafminer infestation and the level of necrosis, use of insecticides to reduce leafminer and leaf necrosis damages, and use of polyester fleece to eliminate leafminer and eventually reduce leaf necrosis. Results of the study indicate that control of leafminer is of primary importance because this will automatically lead to control of the damage caused by A. alternata.

  • The alkaloid securinine was assessed against spore germination of some plant pathogenic and saprophytic fungi (Alternaria alternata, Alternaria brassicae, Alternaria brassicicola, Curvularia lunata, Curvularia maculans, Curvularia pallenscens, Colletotrichum musae, Colletotrichum sp., Erysiphe pisi, Helminthosporium echinoclova, Helminthosporium spiciferum, Heterosporium sp.). Spore germinations of all the tested fungi were inhibited. Alternaria brassicicola, C. lunata, C. pallenscens and H. spiciferum were highly sensitive as complete inhibition of spore germination was observed at very low concentrations (200 ppm).

A Novel Alternaria Species Isolated from Peucedanum japonicum in Korea

  • We isolated and examined a new Alternaria sp., which causes leaf spots on Peucedanum japonicum in Korea, by using molecular and morphological methods. Phylogenetic analysis based on a combined internal transcribed spacer region analysis and two protein-coding genes (gpd and Alt a1) demonstrated that the causal fungus was most closely related to A. cinerariae and A. sonchi, and relevant to A. brassicae. However, conidial morphology indicated that it is a novel species within the genus Alternaria, and therefore we have assigned the fungus a new name in this study.

Identification and Characterization of Alternaria iridiaustralis Causing Leaf Spot on Iris ensata in China

  • In 2016, a severe leaf spot disease was found on Iris ensata Thumb. in Nanjing, China. The symptom was elliptical, fusiform, or irregularly necrotic lesion surrounded by a yellow halo, from which a small-spored Alternaria species was isolated. The fungus was identified as Alternaria iridiaustralis based on morphological characteristics. The pathogenicity tests revealed that the fungus was the causal pathogen of the disease. Phylogenic analyses using sequences of ITS, gpd, endoPG, and RPB2 genes confirmed the morphological identification. This study is the first report of A. iridiaustralis causing leaf spots on I. ensata in China.
