Antioxidative Diarylheptanoids from the Fruits of Alpinia oxyphylla

  • The antioxidative activity of Alpinia oxyphylla was investigated through measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) and inhibitory activity for linoleic acid peroxidation. Two antioxidative diarylheptanoids, yakuchinone A (1) and oxyphyllacinol (2), were isolated from the fruits of A. oxyphylla using thin layer chromatography (TLC) autographic assays. The DPPH scavenging activities of the compounds ($IC_{50}=1$, $57{\pm}2.1\;{\mu}M$; 2, $89{\pm}3.1\;{\mu}M$) were lower than vitamin C ($IC_{50}=51{\pm}1.1\;{\mu}M$), but higher than butylated hydroxytoluene (BHT, $IC_{50}=99{\pm}2.2\;{\mu}M$). Also, inhibitory activities for linoleic acid peroxidation of the compounds ($IC_{50}=1$, $0.19{\pm}0.011\;mM$; 2, $0.31{\pm}0.009\;mM$) were higher than those of vitamin C ($IC_{50}=0.59{\pm}0.017\;mM$) and BHT ($IC_{50}=0.52{\pm}0.014\;mM$). In addition the $^{13}C-NMR$ data of oxyphyllacinol (2) have been first reported in this paper.

항톡소포자충 효과를 가진 천연물 유래 추출물에 관한 연구 (Anti-Toxoplasmosis effects of herbal extracts against Toxoplasma Gondii)

  • Toxoplasmosis is a parasitic disease caused by Toxoplasma gondii, with very few therapeutic treatment options. The choices for treatment are pyrimethamine and sulfadiazine, however their utility is limited because of drug toxicity and serious side effects. In this study, ethanol extracts of 13 traditional medicines used to treat Toxoplasma gondii were tested in vitro for their anti-Toxoplasma gondii cytotoxicity. The median effective concentration ($EC_{50}$) values for the herbal extracts ranged from 173 mg/mL to 1995.35 mg/mL. In HeLa cell, the selectivity of Alpinia oxyphylla (2.75), Mucunae Caulis (2.96), Dictamnus dasycarpus (7.52) which was higher than sulfadiazine (2.08). This indicates that Alpinia oxyphylla, Mucunae Caulis, Dictamnus dasycarpus extracts may be sources of new anti-Toxoplasma gondii compounds.

적백하오관중탕(赤白何烏寬中湯)의 기원(基源), 변천과정(變遷過程) 및 구성원리(構成原理) (The Origin, Changes and Compositive Principles of Jeokbaekhaogwanjung-tang)

  • 1. Objectives: This paper was written to understand the origin, changes and the constructive principles of Jeokbaekhaogwanjung-tang(Chibaihewu-tang; 赤白何烏寬中湯). 2. Methods: Jeokbaekhaogwanjung-tang and other related prescriptions were analyzed in terms of pathology, based on "Donguibogam(東醫寶鑑)", "Donguisusebowon Chobongwon(東醫壽世保元 草本卷)", "Donguisusebowon Gabobon(東醫壽世保元 甲午本)", "Donguisusebowon Sinchukbon(東醫壽世保元 辛丑本)" and "Donguisasangsinpyeon(東醫四象新編)" 3. Results: and Conclusions: 1) The origin of Jeokbaekhaogwanjung-tang, which inherited the spirit of Zhang, Zhongjing(張仲景)'s Sasim-tang(Xiexin-tang; 瀉心湯), is discovered in the prescriptions for Sun-qi(順氣), that is, Gwanjung-hwan(Kuanzhong-wan; 貫衆丸) and Mokhyangsungi-san(Muxiangshunqi-san; 木香順氣散). 2) The Jeokbaekhaogwanjung-tang was derived from Gangchulpajeok-tang(Jiangzhupoji-tang; 薑朮破積湯) of "Dongyisusebowon Gabobon", where the herbal medicines, Panax ginseng(人蔘) of Sasim-tang was replaced with Cynanchum wilfordii(白何首烏) and Allium sativum(獨頭蒜) was newly used too. Thereafter, Polygonum multiflorum(赤何首烏) and Alpinia oxyphylla(益智仁) were first added in Jeokbaekhaogwanjung-tang in "Sinchukbon". 3) The Jeokbaekhaogwanjung-tang, composed of 8 herbs except for Zizyphus jujuba(大棗), treats Taeumjeung(太陰證) of Soeumin(少陰人) through warming the Stomach(溫胃) of Cynanchum wilfordii, Polygonum multiflorum, Zingiber officinale(乾薑) and Alpinia officinarum(良薑) and downbearing the Yin(降陰) of Citrus reticulata(靑皮), Citrus unshiu(陳皮), Cyperus rotundus(香附子) and Alpinia oxyphylla.

두충을 포함하는 한방추출물(Mix)의 항노화, 항염, 미백 효능 활성에 관한 연구 (Studies on Antioxidant, Anti-inflammatory and Whitening Effects of Oriental Herbal Extracts (Mix) including Eucommiae cortex)

  • 최근 미세먼지, 내분비장애, 환경 호르몬등에 의해 피부 염증 질환 환자들이 증가함에 따라 화장품 소재 개발 연구는 스테로이드, 항히스타민제 등의 피부염 치료 합성화합물 보다 이를 대체할 수 있는 천연물 기반의 보다 안전한 소재 발굴에 관심이 더 증가되고 있다. 따라서 본 연구에서는 여러 가지 천연 약재 스크리닝을 거쳐 피부 효능 효과가 검증된 두충, 익지인, 백강잠의 한방추출물(Mix)을 선정하고, 이들의 새로운 피부효능을 규명하여 천연 화장품 소재를 개발하고자 하였다. 우선 한방추출물(Mix)의 항산화 효능을 검증하기 위해 DPPH assay를 수행한 결과 $400{\mu}g/mL$ 농도 조건에서 약 98.8% DPPH radical 소거 활성을 확인하였다. 한방추출물(Mix)의 미백효능을 확인하기 위해 B16F10 세포주를 활용하여 한방추출물에 의한 멜라닌 합성량을 측정하였다. $400{\mu}g/mL$ 농도 조건에서 ${\alpha}-MSH$ 처리군 대비 27.1% 수준까지 멜라닌 합성량이 감소하여 미백효능 활성에 대해 유의성 있는 결과를 확인하였다. 또한 한방추출물(Mix)의 항염증 효능을 확인하기 위해 LPS 자극으로 활성화된 RAW264.7 대식세포에서 NO(nitric oxide) 합성 및 iNOS, COX-2의 유전자 발현을 관찰한 결과 모두 유의적으로 감소되는 것을 확인하였다. 염증성 사이토카인으로 알려진 $IL-1{\alpha}$, $IL-1{\beta}$, IL-6, $TNF-{\alpha}$의 mRNA 발현을 분석한 결과에서도 한방추출물에 의해 효과적으로 억제되는 것을 확인하였다. 마지막으로 피부장벽 기능 강화를 확인하기 위해 인간 각질형성세포인 HaCaT을 이용해 tight junction 구성 단백질인 claudin 1 유전자의 발현 변화를 관찰한 결과 한방추출물에 의해 mRNA 발현이 농도 의존적으로 증가되는 것을 확인하였다. 따라서 본 연구를 통해 우리는 두충, 익지인, 백강잠을 함유한 한방추출물(Mix)이 항산화, 항염, 미백, 그리고 피부장벽 강화 등에 유효한 소재임을 확인하였고, 향후 피부 임상시험 등이 이뤄진다면 피부염 환자에 효과적인 천연 화장품 소재로 개발될 수 있을 것이다.

누에 복합 추출물의 면역 활성 증진 효과 (Evaluation of Immunopotentiation Activities of Combined Extract of Silkworm and Food material)

  • Objectives : Silkworm is known as immunomodulatory substances and contain various bioactive compounds such as serine, tyrosine and alanine. The aim of this study was to investigated the immunopotentiating activity of combine extract that silkworm and food materials (Eucommia ulmoides, Angelica gigas, Acanthopanax, Allium hookeri, Cinnamomum cassia, Liriope platyphylla, Curcuma longa, Achyranthes japonica, Alpinia oxyphylla, Adenophora triphylla). Methods : Among 10 kinds of food materials, to select food materials with the effect of enhancing the immune function mouse splenocyte proliferation ability was measured by 3-[4,5-dimethylthiazol-2-y]-2,5-diphenyl terazolium bromide (MTT) assay. Then, combine extract of silkworm and food materials were evaluated that mouse splenocyte proliferation ability by EZ-cytox cell viability assay. Morever, cytokines production such as IL-2, IL-4, IL10, IL12, $IFN-{\gamma}$ on mouse T lymphocyte stimulated with concanavalin A (ConA) was measured. Results : Eucommia ulmoides, Acanthopanax, Allium hookeri, Cinnamomum cassia, Liriope platyphylla has high proliferation ability of mouse splenocyte compared with Curcuma longa, Achyranthes japonica, Alpinia oxyphylla, Adenophora triphylla. The silkworm and food material combined extract has a relatively high proliferation ability of mouse splenocyte proliferation when the silkworm and food materials are used as a single material. In particularly, combined extract of silkworm and Cinnamomum cassia was stimulate cytokine production on T lymphocyte such as IL12, $IFN-{\gamma}$. Combined extract of silkworm and Liriope platyphylla was stimulate cytokine production on T lymphocyte such as IL2, IL4, IL10. Conclusion : In conclusion, the combined extract of the silkworm and Cinnamomum cassia or Liriope platyphylla may enhance immune function by regulating mouse splenocyte proliferation and stimulating cytokine production.

The Effect of the Water Extracts of Digestive Medicinal Plants on the Shelf-life of Pork Patties

  • This study was performed to examine the possibility of water extracts of several digestive medicinal plants (DMPEs), such as Amomum tasoko, Alpinia oxyphylla, Citrus unshiu, and Myristica fragrans, as a natural antioxidant. Total phenol contents of each extract were expressed as gallic acid equibalents (GAE) and those were significantly different among A. tasoko ($39.87{\pm}5.77$ mg GAE/g), A. oxyphylla ($30.28{\pm}3.36$ mg GAE/g), C. unshiu ($28.13{\pm}5.01$ mg GAE/g) and M. fragrans ($6.36{\pm}0.30$ mg GAE/g) (p<0.05), and extract of A. tasoko showed significantly higher antioxidative effect than butylated hydroxyanisole (BHA) on linoleic acid peroxidation at 72 h after incubation (p<0.05). Addition of extracts in pork patties did not affect the pH value and total microbes during cold storage. However, thiobarbituric acid reative substances (TBARS) of treated patties were lower in dose dependant manner than that of control as storage period increased (except patties treated with C. unshiu extract), and patties treated with 0.5% A. tasoko extract showed no significant difference with patties treated with 0.5% BHA at day 7.

Effect of Medicinal Plant Extract Incorporated Carrageenan Based Films on Shelf-Life of Chicken Breast Meat

  • This study was performed to examine the possibility of water extracts for several medicinal plants, such as Amomum tsao-ko, Alpinia oxyphylla, and Citrus unshiu, as an active packaging ingredient for prevention of lipid oxidation. Chicken breast meats were packed with medicinal plant extracts incorporated carrageenan based films and their physico-chemical and microbial properties during storage at $5^{\circ}C$ were investigated. In chicken meat samples packed with A. tsao-ko (TF) or A. oxyphylla (OF) extract incorporated carrageenan based films, pH value, thiobarbituric acid reactive substances (TBARS), and the population of total microbes were significantly lower than those of the negative control (film of no extract was incorporated, CF) after 5 d of storage (p<0.05). Especially, TBARS value of TF ($0.12{\pm}0.01$ mg malonaldehyde/kg meat) was significantly lower than chicken meat samples packed with positive control (ascorbic acid incorporated film, AF, $0.16{\pm}0.01$ mg malonaldehyde/kg meat) at 3 d of storage, and it means TF has enough antioxidative activity to prevent the lipid oxidation of chicken meat. However, there was no consistent effect on VBN values of chicken meats packed with medicinal plant extracts incorporated films during storage. Based on the obtained results, it is considered that A. tsao-ko extract has potential for being used as a natural antioxidant ingredient in active packaging areas.

Oriental medicines with anti-anaphylactic effect

  • The pharmacological mechanisms of most Oriental medicines have not been clearly defined in spite of their effective use in treating many diseases throughout the world. Many Oriental medicines have been used against various allergic diseases for generations, and still occupy an important place in traditional medicine in far eastern countries including Korea. It is also still unclear how Oriental drugs prevent allergic disease in vivo or in vitro models. Some Korean folk medicine inhibited the mast cell-mediated allergic reaction. This review summarizes the effective folk medicine in experimental effect on systemic or local anaphylaxis reaction. Potential anti-anaphylactic folk medicines include: Poncirus trifoliata; Siegesbeckia glabrescence; Solanum lyratum; Aquilaria agallocha; Ulmi radicis; Polygonum tinctorium; Hwanglyun-Haedok-Tang; Rehmannia glutinosa; Kum-Hwag-San; Syzygium aromaticm; Spirulina platensis; Sosiho-Tang; Sinomenium acutum; Schizonepta tenuifolia; Shini-San; Magnoliae flos; Sochungryong-Tang; Oryza sativa; Cryptotympana atrata; Salviae radix; Rosa davurica; Asiasari radix; Chung-Dae-San; Cichorium intybus; Perilla frutescens; Vitex rotundifolia; Terminalia chebula; Siberian Ginseng; Solanum melongena; Gahmi-Shini-San; Alpinia oxyphylla; Acanthopanax senticosus root; Prunella vulgaris; Allergina; Ixeris dentate; Acanthopanax senticosus stem; Tongkyutang; Salvia plebeia; Rubus coreanus; Sinpo- Tang; Dodutang; Forsythia fructus; Xanthii fructus; and Purple bamboo slat. Ensuring the effects and understanding the mechanisms of action for these Oriental medicines can permit drug development and laying of the ground-work for evaluating potential synergistic effects by addition and subtraction of prescriptions.

SUPPRESSION OF PHORBOL ESTER-INDUCED EXPRESSION OF CYCLLOOXYGENASE-2 AND INDUCIBLE NITRIC OXIDE SYNTHASE BY SELCTED CHEMOPREVENTIVE PHYTOCHEMICALS VIA DOWN-REGULATION OF NF-$\textsc{k}$B

  • A wide arry of naturally occurring substances particularly those present in dietary and medicinal plants, have been reported to possess substantial cancer chemopreventive properties. Certain phytochemicals retain strong antioxidative and anti-inflammatory properties which appear to contribute to their chemopreventive or chemoprotective activities. Inducible cyclooxygenase(COX-2) and nitric oxide synthase (iNOS) are important enzymes that mediate inflammatory processes. There is some evidence that expression of both COX-2 and iNOS is co-regulated by the eukaryotic transcription factor NF-$textsc{k}$B. Increased expression of COX-2 and/or iNOS has been associated with pathophysiology of certain types of human cancers as well as inflammatory diseases. Since inflammation is closely linked to tumor promotion, substances with potent anti-inflammatory activies are anticipated to exert chemopreventive effects on carcinogenesis, particularly in the promotion stage. An example is curcumin, a yellow pigment of turmeric (Curcuma longa L., Zingiberaceae), that strongly occurring diaryl heptanoids structurally related to curcumin have substantial anti-tumor promotional activities in two-stage mouse skin carcinogenesis. Thus, yakuchinone A [1-(4'-hydroxy-3'-methoxyphenyl)-7-phenyl-3heptanone] and yakuchinone B [1-(4'-hydroxy-3'methoxyphenyl)-7-phenylhept-1-en-3-one] present in Alpinia oxyphylla Miquel (Zingiberacease) attenuate phorbol ester-induced inflammation and papilloma formation in female ICR mice. These diarylheptanoids also suppressed phorbol ester-induced activation of epdermal ornithine decarboxylase and its mRNA expression when applied onto shaven backs of mice. Yakuchinone A and B as well as curcumin inhibited phorbol ester-induced expression of COX-2 and iNOS and their mRNA in mouse skin via inactivation of NF-$textsc{k}$B. Capsaicin, a major pungent ingredient of red pepper also attenuated phorbol ester-induced NF-$textsc{k}$B activation. Similar suppression of COX-2 and iNOS and down-regulation of NF-$textsc{k}$B activation for its DNA binding were observed with the ginsenosied Rg3 and the ethanol extract of Artemisia asiatica. We have also found that certain anti-inflammatory phytochemicals exert inhibitory effects on phorbol ester-induced COX-2 expression and NF-$textsc{k}$B activation in immortalized human breast epithelial (MCF-10A) cells in culture. One of the plausible mechanisms undelying inhibition by aforementioned phytochemicals of phorbol ester-induced NF-$textsc{k}$B activation involves interference with degragation of the inhibitory unit, I$textsc{k}$Ba, which blocks subsequent nuclear translocation of the functionally active p65 subunit of NF-$textsc{k}$B. the activation of epidermal NF-$textsc{k}$B by phorbol ester and subsequent induction of COX-2 hence appear to play an important role in intracellular signaling pathwasy leading to tumor promotion and targeted inhibition of NF-$textsc{k}$B may provide a new promising cancer chemopreventive strategy.
