Concentrations of Bioavailable Testosterone and Dihydrotestosterone Determined by Luminescence Immunoassay in Serum (혈청내 섬광면역측정법에 의한 활성적 Testosterone과 Dihydrotestosterone의 농도)

  • 혈액에서 생물학적 활성을 나타내는 (bioavailable) steroid hormone은 주로 비결합형(free form)과 알부민 결합형(albumin-bound form)으로 구성된다. 특히 Testosterone (T)과 5 alpha-Dihydrotestosterone (DHT)의 활성적 분획이 전체의 T, DHT 양에 비해 생리적 현상과 보다 잘 일치하는 것으로 알려지고 있다. 본 연구는 섬광면역측정법(Luminescence immunoassay, LIA)으로 혈청내 활성적 T 및 DHT의 농도의 측정에 이용하고져 하였다. 항체는 T- 또는 DHT-3-CMO-BSA를 항원으로 토끼에 면역주사하여 얻었다. 추적자는 T-3-CMO, DHT-3-CMO에 aminobutylethylisoluminol(ABEI)를 부착시켜 사용하였다. 항체중 IgG분획을 Protein-A-Sepharose CL-4B로 분리한 후 Immunobead(Bio-Rad)에 부착시켜 Solid-phase LIA를 실시하였다. 본 연구에서 LIA는 정확도(accuracy), 정밀도(precision), 감도(sensitivity), 교차반응도(specificity)등을 조사하고, 기존의 방사면역측정법(RIA)과 비교하여 만족할만한 결과를 얻었다. 혈청내 T및 DHT의 활성적 분획의 농도를 측정한 결과는 다음과 같았다. T의 경우는 남성에서 T의 전체량의 33% 이상으로 $7.1{\pm}1.5nmol/l$, 여성에서는 26% 이상으로 $0.28{\pm}0.05nmol/l$이었다. DHT의 활성적 분획은 남성의 경우 $601.7{\pm}85.8pmol/l$, 여성의경우 $52.4{\pm}19.9\;pmol/l$이었다. 이상의 결과를 보아 본 연구에서 이용된 LIA는 혈청내 활성적 농도를 측정하기에 충분하다고 사료된다. 또한 이 방법을 이용하여 여성의 Androgenicity 및 남성 정소기능등의 제어방법에 응용될 수 있을 것으로 판단된다.

Growth of α-Ga2O3 Epitaxial Films on Al2O3 by Halide Vapor Pressure Epitaxy

  • In this study, we investigated the growth of single-crystallinity α-Ga2O3 thin films on c-plane sapphire substrates using halide vapor pressure epitaxy. We also found the optimal growth conditions to suppress the phase transition of α-Ga2O3. Our results confirmed that the growth temperature and partial pressure of the reactive gas greatly influenced the crystallinity. The optimal growth temperature range was about 460~510℃, and the α-Ga2O3 thin films with the highest crystallinity were obtained at a III/VI ratio of 4. The thickness and surface morphology of the thin films was observed by scanning electron microscopy. The film thickness was 6.938 ㎛, and the full width at half maximum of the ω-2θ scan rocking curve was as small as 178 arcsec. The optical band gap energy obtained was 5.21 eV, and the films were almost completely transparent in the near-ultraviolet and visible regions. The etch pit density was found to be as low as about 6.0 × 104 cm-2.

Gas Sensing Characteristics of WO3:In2O3 Prepared by Ball-mill Time (볼밀시간에 의한 WO3:In2O3 가스센서의 감응특성)

  • [ $WO_3$ ]powders were ball-milled with an alumina ball for 0-72 hours. $In_2O_3$ doped $WO_3$ was prepared by soaking ball-milled $WO_3$ in an $InCl_3$ solution. The mixed powder was annealed at $700^{\circ}C$ for 30 min in an air atmosphere. A paste for screen-printing the thick film was prepared by mixing the $WO_3$:In2O3 powders with ${\alpha}$-terpinol and glycerol. $In_2O_3$ doped $WO_3$ thick films were fabricated into a gas sensor by a screen-printing method on alumina substrates. The structural properties of the $WO_3$:$InO_3$ thick films were a monoclinic phase with a (002) dominant orientation. The particle size of the $WO_3$:$InO_3$ decreased with the ball-milling time. The sensing characteristics of the $In_2O_3$ doped $WO_3$ were investigated by measuring the electrical resistance of each sensor in the test-box. The highest sensitivity to 5 ppm $CH_4$ gas and 5 ppm $CH_3CH_2CH_3$ gas was observed in the ball-milled $WO_3$:$InO_3$ gas sensors at 48 hours. The response time of $WO_3$:$In_2O_3$ gas sensors was 7 seconds and recovery time was 9 seconds for the methane gas.

The effect of Pd film evaporation condition on the kinetics of hydrogen absorption-desorption (Pd 박막 시료의 제작 조건이 수소 흡수-방출 동역학에 미치는 영향에 관한 연구)

  • This study was conducted to know to the effect of Pd film($180{\AA}$ thick) evaporation condition on the kinetics of hydrogen absorption-desorption. The activation energy of the forward reaction, the activation energy of the backward reaction, and the enthalpy were calculated by hydrogen absorption-desorption in ${\alpha}$-phase.($25{\sim}50^{\circ}C$ temperature) The activation energy of the forward reaction of Pd film, which is made at room temperature, is $6.4{\pm}0.4$ kcal/mol H and of the backward reaction $8.4{\pm}1.5$ kcal/mol H, which yields the reaction enthalpy -2kcal/mol H. The activation energy of forward reaction of Pd film, which is made at $300^{\circ}C$, is $-0.18{\pm}0.61$ kcal/mol H and of the backward reaction $-0.17{\pm}2.3$ kcal/mol H. The sample of $300^{\circ}C$ is more stable than the sample of room temperature in its struciural compactness and resistance value but standard error of result of $300^{\circ}C$ sample is higher than sample of room temperature do.

  • L-Thyroxine possessing a chiral center, the naturally occurring thyroid hormone has been used for the treatment of thyroid dysfunctions and marketed as levothyroxine (L-thyroxine) sodium salt. In this study, after extraction of levothyroxine tablet as a pre-treatment process, direct enantiomer separation of thyroxine on crown ether derived chiral columns for determination of optical purity was performed using reversed mobile phase with acid additive. The chromatographic method developed in this study was applied in the determination of optical purity of several current domestic and foreign commercialized levothyroxine tablets. Optical purity values of these commercialized L-thyroxine sodium tablets except one were higher than 99 percents.

Synthesis and Sintering Behavior of Zr2WP2O12 Ceramics (Zr2WP2O12 세라믹스의 합성과 소결거동 연구)

  • $Zr_2WP_2O_{12}$ powder, which has a negative thermal expansion coefficient, was synthesized by a solid-state reaction with $ZrO_2$, $WO_3$ and $NH_4H_2PO_4$ as the starting materials. The synthesis behavior was dependent on the solvent media used in the wet mixing process. The $Zr_2WP_2O_{12}$ powder prepared with a solvent consisting of D. I. water was fully crystallized at $1200^{\circ}C$, showing a sub-micron particle size. According to the results obtained from a thermal analysis, a $ZrP_2O_7$ was synthesized at a low temperature of $310^{\circ}C$, after which it was reacted with $WO_3$ at $1200^{\circ}C$. A new sintering additive, $Al(OH)_3$, was applied for the densification of the $Zr_2WP_2O_{12}$ powders. The cold isostatically pressed samples were densified with 1 wt% $Al(OH)_3$ additive or more at $1200^{\circ}C$ for 4 h. The main densification mechanism was liquid-phase sintering due to the liquid which resulted from the reaction with amorphous or unstable $Al_2O_3$ and $WO_3$. The densified $Zr_2WP_2O_{12}$ ceramics showed a relative density of 90% and a negative thermal expansion coefficient of $-3.4{\times}10^{-6}/^{\circ}C$. When using ${\alpha}-Al_2O_3$ as the sintering agent, densification was not observed at $1200^{\circ}C$.

  • The effect of $Al_2O_3+Y_2O_3$ additives on fracture toughness of ${\beta}-SiC-TiB_2$ composites by hot-pressed sintering was investigated. The ${\beta}-SiC-TiB_2$ ceramic composites were hot-press sintered and pressureless-annealed by adding 16, 20, 24 wt% ${\beta}-SiC-TiB_2$(6:4 wt%) powder as a liquid forming additives at low temperature(1800 $^{\circ}C$) for 4 h. Phase analysis of composites by XRD revealed mostly of ${\alpha}$-SiC(6H), $TiB_2$, and YAG($Al_5Y_3O_{12}$). The relative density was over 95-88 % of the theoretical density, and the porosity increased with increasing $Al_2O_3+Y_2O_3$ contents because of the increasing tendency of pore formation. The fracture toughness showed the highest value of 5.88 MPa${\cdot}m^{1/2}$ for composites added with 20 wt% $Al_2O_3+Y_2O_3$ additives at room temperature. The electrical resistivity showed the lowest value of $5.22{\times}10^{-4}\;{\Omega}\;{\cdot}\;cm$ for composite added with 20 wt% $Al_2O_3+Y_2O_3$ additives at room temperature, and was all positive temperature coefficeint resistance(PTCR) against temperature up to 900 $^{\circ}C$.

A Study on Plasma Etching of Tungsten Thin Films using $SF_6$ and $SF_6-N_2$ gases ($SF_6$$SF_6-N_2$ 가스를 이용한 텅스텐 박막의 플라즈마 식각에 관한 연구)

  • The plasma etching of tungsten thin films has been studied with $SF_6$ gas in RIE system. The etch rate of ${\alpha}$-phase W film with $SF_6$ gas plasma has been showed to depend strongly on process parameters ($SF_6$, $SF_6-N_2$ gas). Effect of $N_2$ addition and etching selectivity between W film and photoresist have also been studied in detail. Etching profiles between W film and photoresist were investigated by SEM. The compounds on W surface after $SF_6-N_2$ gas plasma treatment were examined by XPS and the concentration of F ions was detected by OES during plasma on.

Bioconversion of Piceid to Piceid Glucoside Using Amylosucrase from Alteromonas macleodii Deep Ecotype

  • Resveratrol, or its glycoside form piceid, is a dietary antioxidant polyphenolic compound, found in grapes and red wine that has been shown to have protective effects against cardiovascular disease. However, very low water solubility of the compound may limit its application in the food and pharmaceutical industries. The amylosucrase (AMAS) of Alteromonas macleodii Deep ecotype was expressed in Escherichia coli and showed high glycosyltransferase activity to produce the glucosyl piceid when piceid was used as an acceptor. The conversion yield of piceid glucoside was 35.2%. Biotransformation using culture of the E. coli harboring the amas gene increased the yield up to 70.8%. The transfer product was purified by reverse phase chromatography and recycling preparative HPLC, and the molecular structure of the piceid glucoside was determined using NMR spectroscopy. The piceid glucoside was identified as glucosyl-${\alpha}$-($1{\rightarrow}4$)-piceid. The solubility of glucosyl piceid was 5.26 and 1.14 times higher than those of resveratrol and piceid, respectively. It is anticipated that dietary intake of this compound is more effective by enhancing the bioavailability of resveratrol in the human body because of its hydrophilic properties in the intestinal fluid.

Effect of vitamin C on azoxymethane (AOM)/dextran sulfate sodium (DSS)-induced colitis-associated early colon cancer in mice

  • BACKGROUD/OBJECTIVES: The objective of this study was to investigate the effects of vitamin C on inflammation, tumor development, and dysbiosis of intestinal microbiota in an azoxymethane (AOM)/dextran sulfate sodium (DSS)-induced inflammation-associated early colon cancer mouse model. MATERIALS/METHODS: Male BALB/c mice were injected intraperitoneally with AOM [10 mg/kg body weight (b.w)] and given two 7-d cycles of 2% DSS drinking water with a 14 d inter-cycle interval. Vitamin C (60 mg/kg b.w. and 120 mg/kg b.w.) was supplemented by gavage for 5 weeks starting 2 d after the AOM injection. RESULTS: The vitamin C treatment suppressed inflammatory morbidity, as reflected by disease activity index (DAI) in recovery phase and inhibited shortening of the colon, and reduced histological damage. In addition, vitamin C supplementation suppressed mRNA levels of pro-inflammatory mediators and cytokines, including cyclooxygenase-2, microsomal prostaglandin E synthase-2, tumor necrosis $factor-{\alpha}$, Interleukin $(IL)-1{\beta}$, and IL-6, and reduced expression of the proliferation marker, proliferating cell nuclear antigen, compared to observations of AOM/DSS animals. Although the microbial composition did not differ significantly between the groups, administration of vitamin C improved the level of inflammation-related Lactococcus and JQ084893 to control levels. CONCLUSION: Vitamin C treatment provided moderate suppression of inflammation, proliferation, and certain inflammation-related dysbiosis in a murine model of colitis associated-early colon cancer. These findings support that vitamin C supplementation can benefit colonic health. Long-term clinical studies with various doses of vitamin C are warranted.
