Theoretical Calculation on Alpha Track Density by Using an Electrostatic Ion Spectrometer (정전기분광분석방법에 의한 알파입자비적밀도의 이론적 계산연구)

  • To develop a technique of theoretical alpha track density calculation for comparison with measured track density, an electrostatic ion spectrometer was specially designed and fabricated. The mobility spectrum of first radon daughter(Po-218) in the range of $0.07{\sim}5.0cm^2/V\;s$ from the radon chamber was measured using-the electrostatic ion spectrometer. Measurement was taken in a radon chamber operated using dry particle free air passed through silica gel, activated charcoal and molecular sieve filters. The mobility of a new-born Po-218 ion measured by the electrostatic ion spectrometer was determined to be $1.92cm^2/V\;s$. A comparison of the theoretical and measured alpha track densities was completed and uncertainties concerning the shape of the spectrum were analyzed. It was found that the discrepancies in track densities are primarily Que to the neglect of wall loss of ions in the theoretical track density calculation.

Sll0396 regulates transcription of the phycocyanin genes in Synechocystis sp. PCC 6803

  • An olive-green mutant was generated in Synechocystis sp. strain PCC 6803 by inactivation of the sll0396 gene. Whole-cell absorption spectra of the mutant revealed the missing of phycocyanin peak. An investigation of the low-temperature fluorescence emission spectra revealed that the $sll0396{\Omega}$ mutant has a reduced amount of phycocyanin. Western blot analysis showed that the mutant contained less phycocyanin ${\beta}$- and ${\alpha}$-subunits and lacked the 30- and 32-kDa linker polypeptides, and northern blot analysis revealed that the transcription of the 1.4-kb cpcBA gene encoding the phycocyanin ${\beta}$- and ${\alpha}$-subunits was lower in the mutant. The Sll0396 protein has a DNA-binding motif and shares homology with known response regulators. Our results indicate that Sll0396 plays a regulatory role in the transcription of the phycocyanin genes during phycobilisome synthesis.

Quality Control of Radiation Counting Systems and Measurement of Minimum Delectable Activity (방사선 계측기의 품질관리 및 최소검출방사능 측정)

  • Various radiation counters have been using to determine radioactivity of radwastes for disposal. A radiation counting system was set up using a radiation detector chosen in this study and its stability was investigated through the periodic determination of background and counting efficiencies in accordance with a quality control program to increase the confidence level. The average background level for the $\gamma$-spectrometer was 1.59 cps and the average counting level for the standard sample was 45248 Ops within $2{\sigma}$ confidence levels. The average alpha background level for the low background ${\alpha}{\beta}$ counting system was 0.31 cpm and the efficiency for alpha counting was 34.38%. The average beta background level for the ${\alpha}{\beta}$ counting system was 1,30 cpm and the efficiency for beta counting was 46.5%, The background level in the region of 3H and 14C for the liquid scintillation counting system was 2.52 and 3.31 cpm and the efficiency for alpha counting was 58.5 and 95.6%, respectively. The minimum detectable activity for the$\gamma$-spectrometer was found to be 3.2 Bq/$m\ell$ and 3.8 Bq/$m\ell$ for the liquid scintillation counter, and 20.5 and 23.0 Bq/$m\ell$, respectively for the $\alpha$ and $\beta$ counting system.

Analysis of Binding Trimethylamine with Rice-washed Solution using Electronic Nose Based on Mass Spectrometer (Mass spectrometer 기반의 전자코를 이용한 트리메틸아민과 쌀뜨물간의 결합 분석)

  • The effectiveness of a technique for binding rice-washed solution with trimethylamine (TMA) was investigated in this study. The mixtures of TMA and rice-washed solution were quantified using an electronic nose based on the mass spectrometer. After 7 min of reaction in a model system, the binding of TMA to the rice-washed solution was detected. As the concentration of rice-washed solution increased, the levels of TMA detected in the headspace decreased, thereby indicating an increase in the binding of TMA to the rice-washed solution. The binding effect of the rice-washed solution was comparable to those of starch, ${\alpha}$-cyclodextrin, or amylose, and superior to that of flour. The results of this study demonstrate the potential for reduction of off-flavors using the rice-washed solution.

  • Using N-type HPGe gamma spectrometer, uranium analysis technique of soil sample is developed where the chemical preprocessing is not a necessity. The results of uranium activities using the method presented in this paper were compared with those results with conventional alpha spectrometer and two results were similar from within uncertainty range. Therefore, this new method will be applied in uranium activity analysis of soil sample.

  • Antioxidant, tentatively named PA-29B substance was isolated from the fermentation broth of rare Actinomycetes. It was isolated by means of silica gel column chromatography and obtained as colorless plates, mp 155-157$^{\circ}C$. The structure of PA-29B substance was assigned to be $\alpha$-phenyl acetamide by $^1$HNMR spectrometer and mass spectrometer.

  • We studied on the ultra thin L- $\alpha$ -DLPC by LB method. The $\pi$ -A isotherm of the L- e -DLPC was measured at the air-water interface varying with the compressing speed and amounts of solutions for spreading. The molecular arrangement of deposited films were evaluated by measuring the absorption with the UV spectrometer. And we made structures of metal(Au)/L- $\alpha$ -DLPC/Metal(Au) and examined electron through L- $\alpha$ -DLPC LB films by means of current-voltage(I- V) measurement

Synthesis of Transglucosylated Xylitol Using Cyclodextrin Glucanotransferase and Its Stimulating Effect on the Growth of Bifidobacterium. (Cyclodextrin Glucanotransferase를 이용한 당전이 Xylitol의 합성과 비피더스균 생육증식 효과)

  • Several transglucosylated xylitols were synthesized using intermolecular transglucosylation reaction of cyclodextrin glucanotransferase (CGTase) and their bifidogenic effects were investigated. The CGTase from Thermoanaerobacter sp. showed the highest transglycosylation activity on xylitol compared to those obtained from other strains. Extruded starch was identified to be the most suitable glucosyl donor for transglucosylation reaction on xylitol molecule by CGTase. The optimum reaction conditions for transglucosylation were also studied using extruded starch as a glucosyl donor. The transglucosylated xylitols were purified by activated carbon column chromatography with ethanol gradient elution from 0 to 18%, and their chemical structures were analyzed by fast atom bombardment mass spectrometer, $\^$13/C-nuclear magnetic resonance spectrometer, and enzyme digestion method. Two transglucosylated xylitol, F-I and F-II, which had one or two glucose molecules attached to maternal xylitol by ${\alpha}$-1,4-linkage, were mainly obtained. F-II showed increased stimulation effect on the growth of Bifidobacterium breve compared to xylitol, indicating the possibility utilized as a new functional alternative sweetners having bifidogenic effects.
