Evaluation of Testosterone Metabolites/Dehydroepiandrosterone As the Indicators of Testosterone Administration in Horse Doping (경주마 약물검사에서 testosterone 투여 여부표지자로서의 testosterone 대사체들에 대한 dehydroepiandrosterone의 비율 평가)

  • The metabolism of testosterone ($17{\beta}$-hydroxy-androst-4-en-3-one) was confirmed in horse after a single intramuscular administration of testosterone cypionate (750 mg). Solvent extracts of urine obtained with enzymatic hydrolysis and methanolysis were analyzed by GC/MS after oxime t-butyldimethylsilyl (oxime-TBDMS) derivatization. The structures of four urinary metabolite after testosterone administration in horse were determined based on EI mass spectra and $5{\alpha}$-androstane-$3{\beta}$, $17{\alpha}$-diol and $5{\alpha}$-androstane-$3{\beta}$-ol-17-one as major was confirmed with authentic standard. Also the concentrations of $5{\alpha}$-androstane-$3{\beta}$, $17{\alpha}$-diol, $5{\alpha}$-androstane-$3{\beta}$, $17{\beta}$-diol, dehydroepiandrosterone (DHEA), $5{\alpha}$-androstane-$3{\beta}$-ol-17-one and testosterone were determined in the urine of normal subjects and the urine after administration. The recovery and detection limit in the most drugs were 86.3~94.7% and 1~3 ppb, respectively. Correlation coefficients for calibration were in the range of 0.984~0.999. Excretion profile of testosterone presents the rapid and large increasement up to maximum values at days 5 after administration and the slow regression. The relative ratios of testosterone, its metabolites over DHEA were determined for indication of testosterone administration in horse doping.

A Study on Correlation Between Subjective Assessment and EEG for Essential Odors (주관적 향의 선호도와 뇌파 반응과의 상관관계)

  • In the present study, the correlation between subjective assessment and EEG for essential odors were investigated quantitatively. EEG signals were measured from 19 electrodes according to the International 10-20 system (Fp1, Fp2, F3/4, F7/8, Fz, C3/4, Cz, P3/4, Pz, T3/4, T5/6, O1/2) from 8 healthy males subjects for four odor(Rose oil bulgarian, Lemon oil misitano, jasmin abs, Laverder oil france (KIMEX co. Ltd) conditions. The result of the subjective assessment shows the most pleasant odor for each subject. and the power spectrum of ${\alpha}/({\alpha}+{\beta})$ of EEG signals from the most pleasant odor was compared with those from the control condition, which has no odor at all. Power spectrum of ${\alpha}/({\alpha}+{\beta})$ of EEG from the most pleasant odor was increased significantly at T4, T6 compared to the control condition. This result implies that the parameter, power spectrum of ${\alpha}/({\alpha}+{\beta})$, could be an important index for signifying the levels of pleasantness for odors.

Compounds from Non-polar Fraction of the Feces of Trogopterus xanthipes (오령지 저극성 분획으로부터의 화합물 분리)

  • Thirteen compounds were isolated from n-hexane layer of the extracts of feces of Trogopterus xanthipes. Their chemical structures were elucidated as lupeol (1), lupenone (2), simiarenol (3), epitaraxerol (4), taraxerone (5), fatty acid esters of 11-oxo-${\beta}$-amyrin (6), 12-oleane-3,11-dione (7), $5{\beta}$-stigmastan-$3{\alpha}$-ol (8), $5{\beta}$-stigmastan-$3{\beta}$-ol (9), $5{\alpha}$-stigmastan-3-one (10), $5{\beta}$-stigmastan-3-one (11), $5{\beta}$-cholestan-$3{\alpha}$-ol (12), and 2-methoxyphenanthrene (13) on the basis of spectroscopic data. Even though all the isolated compounds are known, to the best of our knowledge, all the compounds (1-13) are reported from this species for the first time.

  • Five steroids and two norsesquiterpene glycosides were isolated from the methanol extract of H. japonicus. Their structures were determined by means of physio-chemical and spectral data to be friedelin (1), stigmast-5-en-3-${\beta}$-ol (${\beta}$-sitosterol) (2), 7-keto-${\beta}$-sitosterol (3), 6${\beta}$-hydroxy-4-stigmasten-3-one (4), 7${\alpha}$-hydroxy-${\beta}$-sitosterol (5), 3-hydroxy-4,4-dimethyl-4-butyrolactone (6), daucosterol (7), (6S, 9S)-roseoside (8), and (9S)-drummondol-9-O-${\beta}$-D-glucopyranoside (spinoside B) (9). The compounds 1, 3, 4, and 6 - 9 were first isolated from this plant source.

Quercetin Inhibits ${\alpha}3{\beta}4$ Nicotinic Acetylcholine Receptor-Mediated Ion Currents Expressed in Xenopus Oocytes

  • Quercetin mainly exists in the skin of colored fruits and vegetables as one of flavonoids. Recent studies show that quercetin, like other flavonoids, has diverse pharmacological actions. However, relatively little is known about quercetin effects in the regulations of ligand-gated ion channels. In the previous reports, we have shown that quercetin regulates subsets of homomeric ligand-gated ion channels such as glycine, 5-$HT_{3A}$ and ${\alpha}7$ nicotinic acetylcholine receptors. In the present study, we examined quercetin effects on heteromeric neuronal ${\alpha}3{\beta}4$ nicotinic acetylcholine receptor channel activity expressed in Xenopus oocytes after injection of cRNA encoding bovine neuronal ${\alpha}3$ and ${\beta}4$ subunits. Treatment with acetylcholine elicited an inward peak current ($I_{ACh}$) in oocytes expressing ${\alpha}3{\beta}4$ nicotinic acetylcholine receptor. Co-treatment with quercetin and acetylcholine inhibited $I_{ACh}$ in oocytes expressing ${\alpha}3{\beta}4$ nicotinic acetylcholine receptors. The inhibition of $I_{ACh}$ by quercetin was reversible and concentration-dependent. The half-inhibitory concentration ($IC_{50}$) of quercetin was $14.9{\pm}0.8\;{\mu}M$ in oocytes expressing ${\alpha}3{\beta}4$ nicotinic acetylcholine receptor. The inhibition of $I_{ACh}$ by quercetin was voltage-independent and non-competitive. These results indicate that quercetin might regulate ${\alpha}3{\beta}4$ nicotinic acetylcholine receptor and this regulation might be one of the pharmacological actions of quercetin in nervous systems.

Evaluation of the Estrogenic Activity by Yeast Two-hybrid Assay and Enzyme-linked Immunosorbent Assay in Sewage Treatment Plant (하수처리장의 내분비계장애물질에 대한 Yeast Two-hybrid Assay와 Enzyme-linked Immunosorbent Assay에 의한 에스트로겐활성도 평가)

  • Several endocrine disrupting chemicals(EDCs) were monitored to evaluate the estrogenic activities and the concentrations by yeast two-hybrid assay and enzyme-linked immunosorbent assay(ELISA) in sewage treatment plant(STP) which consist of industrial and domestic line. In the influent of domestic line, estrone, 17$\beta$-estradiol, 17$\alpha$-ethinylestradiol and alkylphenolethoxylate(APE) were detected up to 167.1, 39.7, 7.3 and 145.4 ng/L, respectively. The average removal efficiency of 17$\beta$-estradiol after the activated sludge process was 77.5% and further removed to 80.8% after the sand filtration-ozonation step. These results suggests that the activated sludge process has limited potential to remove the estrogenic activity effectively. The contributions of the estrogenic chemicals to the estrogenic activities were 70.7, 23.3, 3.7 and 2.3% for estrone, 17$\beta$-estradiol 17$\alpha$-ethinylestradiol and APE, respectively, in the domestic line effluents. Therefore, 17$\beta$-estradiol and estrone contributed most of the estrogenic activity in the domestic line effluents.

Identification and Growth Activity to Bifidobacterium spp. of Locust Bean Gum Hydrolysates by Trichoderma harzianum ${\beta}$-mannanase (Trichoderma harzianum 유래 ${\beta}$-mannanase에 의한 Locust Bean Gum 가수분해 올리고당의 동정 및 Bifidobacterium spp.에 대한 생육활성)

  • This study was performed to elucidate substrate specificity to the locust bean gum galactomannan by Trichoderma harzianum ${\beta}-mannanase$. The medium composition for enzyme production were determined 3% cellulose, 3% corn steep liquor, 1% $KH_2PO_4$, 0.2% $(NH_4){_2}SO_4$, and incubated for 115 hr at $28^{\circ}C$. The ${\beta}-mannanase$ exhibited maximum activity at pH 4.5 and $60^{\circ}C$. Locust bean gum galactomannan was hydrolyzed by the ${\beta}-mannanase$, and then hydrolysates separated by activated carbon column chromatography. The main hydrolysates were composed of D.P 4 and 7 galactosyl mannooligosaccharides by TLC. For the elucidate the structure of D.P 4 and 7 oligosaccharides, methylation analysis was performed. D.P 4 and 7 were identified as M-M-M-M and M-M-M-M-M (G- and M-represent ${\alpha-1,6-D-galactosidic\;and\;{\beta}-1,4-mannosidic$ linkages, respectively). //G-G To investigate the effects of locust bean gum galactosyl mannooligosaccharides on the in vitro growth of B. longum, B. bifidum, B. infantis, and B. breve, Bifidobacterium spp. were cultivated individually on the modified-MRS medium containing carbon source such as D.P 4 and 7 galactosyl mannooligosaccharides, respectively. B. longum grew up 3.4-fold and 4.3-fold more effectively by the replacement of D.P 4 and 7 galactosyl mannooligosaccharides as the carbon source in a comparasion of standard MRS.

The Effect of Gyejibokryunghwan on Fracture Union in Tibia-fractured Rats (계지복령환(桂枝茯苓丸)이 흰쥐의 골절유합에 미치는 영향)

  • Objectives The object of this study was to assess the effect of Gyejibokryunghwan (GBH) on anti-oxidant and anti-inflammatory activities in RAW 264.7 cells and on factors associated with fracture union in tibia-fractured rats. Methods The 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity was measured to assess anti-oxidant activity. The production of nitric oxide (NO), interleukin-6 (IL-6), interleukin-$1{\beta}$ ($IL-1{\beta}$) and tumor necrosis factor-${\alpha}$ ($TNF-{\alpha}$) in the RAW 264.7 cells were measured to assess anti-inflammatory activity. The production of osteocalcin, calcitonin, carboxy-terminal telepeptides of type II collagen (CTXII), transforming growth factor-${\beta}$ ($TGF-{\beta}$), bone morphogenetic protein-2 (BMP-2) in serum of tibia-fractured rats were measured to assess the effects of fracture union. X-rays were taken every two weeks from 0 to 4th week to assess fracture union effect. Results DPPH radical scavenging activity of GBH was increased according to concentration of GBH in RAW 264.7 cell. NO, prostaglandin $E_2$ ($PGE_2$), IL-6, $IL-1{\beta}$ and $TNF-{\alpha}$ were significantly decreased, indicating anti-inflammatory effect. Osteocalcin, calcitonin, $TGF-{\beta}$ were significantly increased in the experimental groups. CTXII was significantly decreased in the experimental groups. BMP-2 was not significantly changed in the experimental groups. The X-ray showed that the experimental group has better healing effects on tibia-fractured rats than control group. Conclusions From above result, GBH has an effect on anti-oxidant, anti-inflammatory activities in RAW 264.7 cells. GBH showed significant results in factors related with fracture union and radiologic examination. In conclusion, GBH can help fracture union and it well be expected to be used actively in clinics.

  • The ${\alpha}-and\;{\beta}$-isomer of 4-nitroazoxybenzenes have been separated by liquid chromatography and their U.V. spectra were examined. The n${\to}{\pi}^{\ast}$ transition band of the compounds did not appear, likewise the cases of other compounds of the series. Transition band of the new isomer were as usual as those of other azoxy-compounds, whereas the ${\pi}{\to}{\pi}^{\ast}$ transition band of the other isomer which is reported in the literature shown peculier hypochromic shift and hypochromic effect. From the spectroscopic point of view it is very likely that the new isomer (m.p. $184-5^{\circ}C$) is ${\alpha}$-isomer and the other one (m.p. $152^{circ}C$) is ${\beta}$-isomer contrary to the literature.

  • Five biflavones and sevenflavonolglycosideswereisolatedfromtheleaves of Ginkgo biloba. They were sciadopitysin(1), ginkgetin(2), isoginkgetin(3), bilobetin(4), amentoflavone(5), kaempferol 3-O-[$6'-O-{\rho}-coumaroyl-{\beta}-_D-glucopyranosyl(1{\rightarrow}2)-{\alpha}-_Lrhamnopyranoside$](6), quercetin 3-O-[$6'-O-{\rho}-coumaroyl-{\beta}-_D-glucopyranosyl(1{\rightarrow}2)-{\alpha}-_Lrhamnopyranoside$](8), rutinosides of kaempferol(7), isorhamnetin(9), quercetin(10), laricitrin(11), and kaempferol 3-O-($2',6'-{\alpha}-_L-dirhamnopyranosyl-{\beta}-_{D}-glucopyranoside$)(12). The structures were established by spectroscopic and chemical methods.
