Growth Period Effects on the Protective Properties of Aloe vera Against t-BHP-Induced Oxidative Stress in Chang Cells

  • Aloe vera has been used in traditional medicine for the therapy of a variety of disorders, such as wounds and burns. However, few studies have examined the antioxidant capacities of A. vera plants during different growth periods. In order to investigate the effects of growth on antioxidant activity, A. vera was prepared from 2-, 4-, 6-, 8-, and 12-month-old aloe. The extracts from 6-month-old A. vera showed the highest contents of flavonoids (9.750 mg catechin equivalent/g extract) and polyphenols (23.375 mg gallic acid equivalent/g extract) and the highest ferric reducing antioxidant power (0.047 mM ferrous sulfate equivalent/mg extract). The extract from 6-month-old A. vera exhibited the highest free radical scavenging potential, and the lowest IC50 values were found for 2,2-diphenyl-1-picrylhydrazyl (0.26 mg/ml) and alkyl radicals (0.50 mg/ml). In addition, the extract from 6-month-old A. vera showed the greatest effects on cell viability in normal liver cells. Based on these findings, the extract from 6-month-old A. vera was examined further in order to determine its protective potential against tert-butyl hydroperoxide (t-BHP)-induced oxidative stress. The extract from 6-month-old A. vera at a concentration of 0.25 mg/ml showed the highest protective activity against t-BHP-induced reactive oxygen species production. These findings suggested that harvesting regimens were critical in the regulation of effects of the bioactive potential of A. vera on antioxidant activity.

In vivo Angiogenic Activity of Dichloromethane Extracts of Aloe vera Gel

  • The angiogenic activity of Aloe vera (Aloe baradensis), known as a good healing plant, was investigated. We have extracted and fractionated dichloromethane extract (G1M1D1) and methanol soluble fraction of dichloromethane extract (G1M1D1M1) which contain low-molecular weight substances of Aloe vera gel. G1M1D1 and G1D1M1 fractions induced a radially arranged, spoke-sheel-like vasculature in chick embryo chorioallantoic membrane (CAM) assay. The angiogenic activity was dose-dependent and the angiogenic pattern in the CAM assay. The angiogenic activity was dose-dependent and the angiogenic pattern in the CAM assay was very similar to that of phorbol 12-myristate-13-acetate (PMA) used as a positive control. The modified CAM assay, e simple and accurate quantitating method, was used to quntitate the angiogenic activity of G1D1M1 fraction. Application of G1M1D1M1 fraction ($100\mug/egg$) resulted in much more intense angiogenesis than in control while slightly less intense angiogenesis than in PMA (100 ng/egg).

Screening for Chemosensitizers from Natural Plant Extracts through the Inhibition Mechanism of P-glycoprotein

  • P-gp plays a critical role in drug disposition and represents a mechanism for the development of multidrug resistance. Flavonoids, a major class of natural compounds widely present in foods and herbal products, have been shown to inhibit P-gp. Therefore, the aim of this study was to identify new candidate chemosensitizers by screening various plant extracts. The ability of natural plant extracts to inhibit P-gp activity was assessed by measuring cellular accumulation of calcein AM, daunorubicin and vincristine in P-gp overexpressing MDCKII-MDR1 cells. Among more than 800 plant extracts, eight were found to inhibit P-gp activity. Curcuma aromatica extract produced greatest inhibition, followed by Curcuma longa and Dalbergia odorifera extracts. Extracts of Aloe ferox, Curcuma zedoariae rhizome, Zanthoxylum planispinum, and Ageratum conyzoides showed moderate inhibitory effects. Curcumin and quercetin exhibited similar inhibition of P-gpmediated efflux of daunorubicin and vincristine, and flavones had a lesser effect. When chemosensitizing effect was evaluated by measuring daunorubicin sensitivity to MDCKII-MDR1 cells in the presence of natural plant extracts, Curcuma aromatica showed the most potent chemosensitizing effect based on daunorubicin cytotoxicity. In conclusion, natural plant extracts such as Curcuma aromatica can potently inhibit P-gp activity and may have potential as a novel chemosensitizers.

Aloe vera Callus 현탁배양 생성물의 항염증 활성 (Anti-inflammation Activities of Cultured Products from Suspension Culture of Aloe vera Callus)

  • 알로에의 유효성분인 다당의 효율적 생산연구일환으로 수행되었던 aloe callus의 현탁배양과 관련하여, 본 연구에서는 이의 배양생성물(세포외 다당)과 callus의 용매추출물에 대한 항염증 활성을 조사하였다. 세포외 다당(ACP)과 callus (AC)의 물 및 에탄올 추출물의 대식세포(Raw 264.7) 세포생존율을 MTT assay로 조사한 결과, AC-DMSO의 $400{\mu}g/mL$ 첨가구에서 97%로 미미한 증식저해를 보였으나 나머지 시료처리구는 모두 증식 촉진효과를 나타내었다. 또, LPS에 의해 활성화된 Raw 264.7 세포 배양액의 알로에 추출물 처리에 따른 NO 생성 저해효과를 조사한 결과, AC-DW와 AC-DMSO는 오히려 양성대 조구인 LPS 단독보다 더 많은 NO를 생성하였다. 특히, 세포외 다당 분획인 ACP-DW는 NO 생성을 강력히 억제하여 $80-100{\mu}g/mL$의 비교적 낮은 농도의 첨가로 대조구 수준까지 억제하였다. 또, Raw 264.7 세포에 LPS로 자극을 주고 알로에 현탁 배양 생성물의 처리에 따른 COX-2단백질의 발현정도를 조사한 결과, ACP-DW 처리구는 단독으로 COX-2 단백질을 발현시키지 않았으며, LPS 자극에 따른 COX-2의 발현감소는 미미하였다. 아울러, 활성화된 대식세포로부터 생성된 염증 사이토카인인 IL-$1{\beta}$와 TNF-${\alpha}$의 발현량을 RT-PCR로 조사한 결과, ACPDW 처리구에서 TNF-${\alpha}$의 발현이 강력하게 억제되었다. 이상의 결과들은 알로에 현탁배양 유래의 생성물들이 비교적 우수한 항염증활성을 나타냄을 보여주는 결과로, 그동안 삽목번식의 재배방법에 의존하였던 알로에 유효성분의 생산을 대체할 수 있는 효율적이고, 새로운 방법으로서의 알로에 callus의 현탁배양 가능성을 보여주었다.

알로에 배지에서 재배한 표고버섯 추출물의 종양괴사인자로 유도된 유방암세포 MDA-MB231에 대한 전이 저해 활성 (Inhibitory Effect of Shiitake Mushroom Extracts Cultured in Aloe-Supplement on Invasion/Migration of TNF-α-Induced MDA-MB-231 Breast Cancer Cells)

  • 알로에를 첨가한 표고버섯 배양 추출물(ESA)의 항암효과를 검정하기 위하여 인간 유방암 세포주인 MDA-MB-231에 ESA를 처리하여 생육저해 활성과 세포 이동 억제 효과를 확인하였다. MDA-MB-231 세포에 ESA를 처리하여 72시간 후 관찰한 결과 약간의 생육저해 활성을 나타내었으며, 또한 transwell에서 TNF-${\alpha}$가 처리된 세포에 ESA를 처리해 본 결과 migration/invasion 정도를 상당히 저해함을 알 수 있었다. 이러한 세포 이동능의 저해 기작은 웨스턴 블럿 분석을 통해 확인해 본 결과 p-ERK 신호를 통해 일어나며, ICAM-1 단백질의 발현을 억제함으로써 나타나는 것을 알 수 있었다. 따라서 본 실험에서는 알로에 첨가 담자균 배양추출물(ESA)은 인간 유방암세포에 대해 잠재적인 항암효과가 있음을 확인하였다. 이후 표고버섯 외 꽃송이버섯과 상황버섯에서의 알로에 배양액에 대한 항암효과에 대한 연구를 수행하고 있어 그 결과를 보고하고자 한다.

약용식물 추출물의 아토피성 피부염에 대한 항염증 및 항알레르기 효과 (제 2 보) (Anti-inflammatory and Anti-allergic Effects of Herbal Extracts on Atopic Dermatitis ( Part II ))

  • 아토피성 피부염은 만성 재발성 염증성 피부질환으로 피부장벽기능의 이상과 환경유발인자에 대한 피부과민성과 연관되어 있다. 이전의 연구에서는 아토피성피부염에 효과적인 약용식물 추출물을 발굴하기 위하여 노회, 자회지정, 석류, 석곡 추출물들의 세포독성, 항산화, 항염, 항알레르기 효과를 검토하였다. 본 연구에서는 지질다당류로 활성화시킨 대식세포 RAW26.7 에 대한 약용식물 추출물들의 항염작용을 보다 상세하게 검토하여 항염작용의 근본적인 분자기전을 확인하고자 하였다. 역전사중합효소연쇄반응분석(reverse transcription polymerase chain reaction analysis) 결과, 석류, 석곡, 노회는 염증성 사이토카인인 IL-6 와 IL-$1{\beta}$ 유전자발현을 현저하게 억제시켰으며 자화지정은 영향이 없었다. 형질주입과 발광효소분석(transfection and luciferase analysis) 결과, 약용식물 모두가 전사 핵인자 카파비(NF-${\kappa}B$)의 활성화를 억제시켰다. 웨스턴 블럿 분석(western blot analysis) 결과, 노회는 JNK MAP 인산화효소의 활성화를 차단하였지만 p38 MAP 인산화효소의 활성화는 차단하지 못하였다. 반면에 자화지정, 석류, 석곡은 JNK MAP 인산화효소뿐만 아니라 p38 MAP 인산화효소의 활성화도 차단하였다. 이들 실험결과들은 노회, 자화지정, 석류, 석곡은 항염효능을 가지고 있으며 따라서 아토피성 피부염의 증상을 경감 또는 완화시키는 잠재력이 있음을 보여 준다.

붉나무 캘러스 유도 및 그 추출물로부터 피부미백 효과에 관한 연구 (Induction and Proliferation of Callus in Rhus chinensis Mill. and Its Effect on Skin Whitening)

  • The objective of this study was to optimize the condition for induction and proliferation of callus from Rhus chinensis Mill. and investigate the skin-brightening effect of Rhus chinensis callus (RCC). It was confirmed that the most proper plant growth regulator (PGR) for callus induction is 1.0 mg/L of 2,4-dichlorophenoxyacetic acid (2,4-D). The most optimal condition of PGR, medium and additives for callus proliferation were 2,4-D (1.0 mg/L), MS medium and citric acid, respectively. Inhibitory activities of tyrosinase were higher at 50 and 100 ㎍/mL of RCC extracts (41.86 and 75.56%, respectively) than arbutin (27.32%). As the results of measuring melanin inhibition in B16F1 melanocyte and B16F10 melanoma cell, RCC extracts increased its inhibitory activities concentration-dependently, and were found to have higher whitening effect than arbutin at a concentration of 100 ㎍/mL. Therefore, it is suggested that RCC can be used as an effective material for skin-brightening cosmetics.

시중에서 유통되는 건강식품의 종류에 관한 연구 -매스미디어와 인터넷 미디어를 중심으로- (A Study on the Classification of Health Food Circulated in the Market -Surveyed on Mass Media and Internet-)

  • This study was performed to investigate the kinds of health food advertised and sold through media like printed matter, TV broadcasting or internet. It was found that fifty three percent of the health food was circulated through internet, 27% through printed matter, 9% through TV Broadcasting. When the health foods was classified, the proportion of special purpose nutritious foods was 36.1%, health aid foods 33.9%, uncooked powdered foods 18.5%, functional foods for health 4.9%, Ginseng products 3.9%, and tea 0.9%. The special purpose nutritious foods was composed of 59% of nutrition supplementary foods, 24% of dietary fiber foods, 9% of weaning foods, 7% of milk formula and 2% of HCA. Vegetable extracts occupied 21.1% of health aid foods, yeast 7.5%, mushroom extracts 7.0%, chitosan 6.2%, aloe 5.3%. Dried powdered type health food occupied the highest proportion. The other type were capsule(18.8%), tablet(18.1%), and liquid type(16.4%). When the health food was classified with health claim, the proportion of "nutrition supplementation" was the highest (23.9%), "diet" 14.9%, "ergogenic"(18.8%), "promoting bowel movement" 7.8%, "growth" 5.7%, "regulation of blood sugar" 4.5%, "improving of immunity"(2.4%) and anti-aging effect(2.4%).

Research Trends in the Development of Cosmetic Ingredients for Skin Barrier Improvement

  • In 2022, the domestic production performance of functional cosmetics in South Korea reached 4.6 trillion won, accounting for 33.85% of the total cosmetics production. The number of functional cosmetics reviewed increased by about 7.5% from the previous year, totaling 974 items. Especially with the increasing importance of the skin barrier function due to skin sensitivity caused by various environmental pollutants, domestic cosmetic companies are showing interest in the development of new ingredients and products related to this area. This study aims to analyze academic research trends related to in vitro experiments for the development of cosmetics improving the skin barrier, to provide practical information for the cosmetic industry. The findings are as follows: Academic research mainly focused on the efficacy of natural ingredients in improving the skin barrier, but there is a significant lack of quantitative accumulation of research. For the development of skin barrier-improving cosmetic ingredients, efficacy evaluation indicators were set, including hyaluronic acid production, expression of filaggrin gene, loricrin, formation of cornified envelope (CE), and expression of ceramide synthesis enzyme genes. Moreover, effective cosmetic ingredients for improving the skin barrier included lemongrass and perilla leaf extracts, flavonoids, Lactococcus lactis subsp. lactis, Exosomelike Nanovesicles derived from apple callus, Eleutherococcus sessiliflorus, Acanthopanax sessiliflorus, Eleutherococcus gracilistylus, Acer okamotoanum extracts, Aloe vera adventitious root extract, ethanol extract of Aruncus dioicus, and organic solvent fraction of Dracocephalum argunense.

Nematocidal Screening of Essential Oils and Herbal Extracts against Bursaphelenchus xylophilus

  • Five essential oils and 15 herbal extracts were evaluated to control Bursaphelenchus xylophillus in laboratory. The essential oils from clove plant (Syzygium aromaticum), mustard (Brassica integrefolia), thyme (Thymus vulgaris), and Pelargonium inquinans were found to be highly promising and gave excellent control of the nematodes at all the time of exposure. Among them, the least one gave 91.3% mean mortality rate at 24 hours of exposure time, which is highly significant from the control. While in the second study, most of the methanol (Desmodium caudatum, Paulownia coreana, Auckulandia lappa, Sophota flavescens, Aloe sp., Rheum palmatum, Zingiber officinale, Magnolia officinalis, and Eugenia caryophyllata), hexane (Torreya nucifera, Pharbitis nil, Prunus mume, Melia azedarach, and Xanthium strumarium), and hot water (Cinnamomum cassia) herbal extracts killed the nematodes, but in varying degrees compared to the control. Only one extract was found to be promising viz Magnolia officinalis which found to be statistically different from the control and gave mean mortality of 72, 82.3, and 85.3 % for 24, 48, and 72 hours exposure, respectively. Further screening was conducted for M. officinalis with concentrations of 1,000, 100, and 10 ppm against the same species of nematode with the same time of exposure. However, it gave an excellent result for 1,000 ppm for all time of exposure, whereas for the 100 and 10 ppm it gave mean mortality of 39.5 and 25.8% for the time 72hrs, respectively that were statistically different from the control.
