Effect of Allium cepa (red) and Angelica gigas Nakai on Hair-growth Promotion in C57BL/6 Mice (C57BL/6 마우스에서 모발성장 촉진에 대한 Allium cepa (red)와 Angelica gigas Nakai의 효과)

  • This study was conducted to evaluate the hair-growth promotion effect in C57BL/6 mice of a new substance mixed with Allium cepa (red)-bioconversion extract and Angelica gigas Nakai. The ethanol extract of Allium cepa (red) was bioconverted through the use of the Bacillus subtilis KJ-3 (BS3) strain, which was named Red-BCQ. The quercetin content of Red-BCQ increased by about 7.4-fold after bioconversion. Angelica gigas Nakai extract (Agnex) contains a large amount of coumarins such as decursin (D) and decursinol angelate (DA). A 1 mg portion of Agnex contained 0.4146 mg of D and 0.3659 mg of DA. Minoxidil has been known to promote hair growth. In this study, the hair-growth promotion effects of Red-BCQ, Agnex, and a mixture of both Red-BCQ and Agnex were compared with 5% minoxidil. Twenty-five mice were divided into five experimental groups including saline (CON), 5% minoxidil (PCON), Red-BCQ (RA), Agnex (AG), and a Red-BCQ-Agnex mixture (RAG)-treated group. Samples were administered orally once a day at a fixed time for 4 weeks. Hair growth was monitored by photograph at 7, 14, 21, and 28 days. We also observed 5α-reductase, alkaline phosphatase (ALP), γ-glutamyl transpeptidase (γ-GT), insulin-like growth factor (IGF)-1, transforming growth factor (TGF)-β1, antioxidant enzyme, and the hair follicles of the skin tissue. In all the results, the RAG-administered group showed greater antioxidative and hair-growth promotion effects than the other groups. These data suggest that RAG has potent stimulating activity on hair growth in C57BL/6 mice.

Marker-assisted Genotype Analysis of Bulb Colors in Segregating Populations of Onions (Allium cepa)

  • Bulb color in onions (Allium cepa) is an important trait whose complex inheritance mechanism involves epistatic interactions among major color-related loci. Recent studies revealed that inactivation of dihydroflavonol 4-reductase (DFR) in the anthocyanin synthesis pathway was responsible for the color differences between yellow and red onions, and two recessive alleles of the anthocyanidin synthase (ANS) gene were responsible for a pink bulb color. Based on mutations in the recessive alleles of these two genes, PCR-based markers for allelic selection were developed. In this study, genotype analysis of onions from segregating populations was carried out using these PCR-based markers. Segregating populations were derived from the cross between yellow and red onions. Five yellow and thirteen pink bulbs from one segregating breeding line were genotyped for the two genes. Four pink bulbs were heterozygous for the DFR gene, which explains the continuous segregation of yellow and pink colors in this line. Most pink onions were homozygous recessive for the ANS gene, except for two heterozygotes. This finding indicated that the homozygous recessive ANS gene was primarily responsible for the pink color in this line. The two pink onions, heterozygous for the ANS gene, were also heterozygous for the DFR gene, which indicated that the pink color was produced by incomplete dominance of a red color gene over that of yellow. One pink line and six other segregating breeding lines were also analyzed. The genotyping results matched perfectly with phenotypic color segregation.

Comparison of the antioxidant properties and flavonols in various parts of Korean red onions by multivariate data analysis

  • To compare the antioxidant properties and flavonols in various parts; dry skin (DS) and edible portion (EP), of 8 red onions (Allium cepa L, ROs), total content of phenolics (TPC), flavonoids (TFC), and anthocyanins (TAC) and DPPH radical scavenging properties were estimated and the content of six flavonols were quantified by HPLC-PDA analysis. The major component of DS and EP of RO was quercetin and quercetin-4'-glucoside, respectively. Score plots of the PCA and PLS-DA were segregated by flavonols content and antioxidant properties according to the EP and DS of ROs. Loading plot of the PCA showed that the quercetin and sum of flavonol content were highly correlated with antioxidant activity of ROs. Therefore, flavonol content and antioxidant activity can be used as markers for distinct parts of ROs.

Bulb Storability of Red and Yellow Onion (Allium cepa L.) Cultivars Grown in Korea

  • The bulb storability of eight red and ten yellow onion (Allium cepa L.) cultivars grown primarily in Korea was evaluated. During storage, sprouting occurred earlier in red cultivars than in yellow cultivars. In addition, the ratio of sprouted to unsprouted bulbs increased more rapidly in red cultivars than in yellow cultivars. However, not all yellow cultivars had strong storabilities. Bulb storability of three yellow cultivars was as poor as that of red cultivars, suggesting only a slight possibility of a pleiotropic effect of color and bulb storability. Meanwhile, $F_3$ lines of a red cultivar selected based on stronger storabilities showed intermediate storabilities between those of the red and yellow cultivars, implying that strong storability could be obtained by successive selection. In contrast, $F_1$ hybrids from crosses between yellow and red breeding lines showed poor storability compared with the yellow cultivars, indicating that poor storability may be dominant over strong storability. The relationships between color and other traits that potentially affect storability, such as fresh and dry weight, water content, and firmness of bulbs, were evaluated. No significant differences in these traits were observed between the two colors, although the water content of yellow cultivars and the dry weight of red cultivars were slightly higher than their counterparts. Correlation analyses between bulb storability and other traits, including weight, water content, and firmness, also showed no significant correlations. In this study, no correlations were identified between bulb color, storability, and other traits. However, based on these results, red cultivars with stronger storabilities could be developed through successive selection of bulbs with stronger storabilities.

Physiological Activities of Bioconversion Products Using Bacillus Subtillis KJ-3 and Their Mixtures (Bacillus Subtilis KJ-3를 이용한 생물전환물 및 그 혼합물의 생리활성)

  • This research was performed to develop a new material consisting of a mixture of Red Allium cepa (RA) Cucurbita moschata duch (CM), and Angelica gigas Nakai (AG). RA and CM have low storage stability because of their high moisture content. Therefore, their major components were extracted and used for the research after a content analysis. In order to overcome these limitations, the quercetin from RA, ${\beta}-carotene$ from CM, and decursin/decursinol angelate (D/DA) from AG were separately extracted, and the biochemical activity of each extract and mixture was compared. RA was bioconverted by the Bacillus subtillis KJ-3 (BS3) after ethanol extraction. After bioconversion, the quercetin content of RA was increased by 128.9%. ${\beta}-carotene$ was detected in the CM ethanol extract and its content was very low concentrations at 0.2 mg/g. The AG ethanol extract (1 mg) contained 0.4146 mg and 0.3659 mg of D/DA, respectively. The purity of the D/DA was found to be about 78%. The flavonoid and polyphenol content of each extract and their mixtures (mixture 1 (RA:CM:AG = 5:2:3), mixture 2 (RA:CM: AG = 3:5:2), and mixture 3 (RA:CM:AG = 3:2:5)) were measured. In addition, the cell survival rate, anti-inflammatory activity, and antioxidant ability were also evaluated. In all the results, the antioxidant activity of mixture 3 was most effective. Therefore, these findings provide basic data for future food development using a 3:2:5 mixture of RA, CM, and AG.

Development of Functional Markers for Detection of Inactive DFR-A Alleles Responsible for Failure of Anthocyanin Production in Onions (Allium cepa L.)

  • Inactivation of the gene coding for dihydroflavonol 4-reductase (DFR) is responsible for the color difference between red and yellow onions (Allium cepa L.). Two inactive DFR-A alleles, DFR-$A^{PS}$ and DFR-$A^{DEL}$, were identified in our previous study. A functional marker was developed on the basis of the premature stop codon that inactivated the DFR-$A^{PS}$ allele. A derived cleaved amplified polymorphic sequences (dCAPS) primer was designed to detect the single nucleotide polymorphism, an A/T transition, which produced the premature stop codon. Digested PCR products clearly distinguished the homozygous and heterozygous red $F_2$ individuals. Meanwhile, to develop a molecular marker for detection of the DFR-$A^{DEL}$ allele in which entire DFR-A gene was deleted, genome walking was performed and approximately 3 kb 5' and 3' flanking sequences of the DFR-$A^R$ coding region were obtained. PCR amplification using multiple primers binding to the extended flanking regions showed that more of the extended region of the DFR-A gene was deleted in the DFR-$A^{DEL}$ allele. A dominant simple PCR marker was developed to identify the DFR-$A^{DEL}$ allele using the dissimilar 3' flanking sequences of the DFR-A gene and homologous DFR-B pseudogene. Distribution of the DFR-$A^{PS}$ and DFR-$A^{DEL}$ alleles in yellow onion cultivars bred in Korea and Japan was surveyed using molecular makers developed in this study. Results showed predominant existence of the DFR-$A^{PS}$ allele in yellow onion cultivars.

Major constituents and antioxidant activities of domestic onion (Allium cepa L.) cultivars (국내 육종 양파 품종별 주요성분의 함량 및 항산화활성 비교)

  • The content of major constituents and antioxidant activities of nine onion cultivars, namely yellow domestic cultivars Meapsihwang, Sinsunhwang, Yeonsinhwang, Yeongbohwang, and Yeongpoonghwang, red domestic cultivar Eumjinara, green domestic cultivar Sweetgreen, yellow-introduced cultivar Sunpower, and red-introduced cultivar Cheonjujeok, were compared. Yeongbohwang and Cheonjujeok had the highest total phenolic and total flavonoid contents among the six yellow and two red cultivars, respectively. The red cultivars displayed the highest DPPH and ABTS+ radical scavenging activities (DRSAs and ARSAs, respectively) and reducing power (RP). Among all cultivars, Cheonjujeok and Eumjinara had the highest DRSA, while both red cultivars had similar ARSAs. Among the yellow cultivars, Sinsunhwang and Yeongbohwang showed excellent DRSAs. The RP of the domestic cultivars was slightly higher than that of the introduced cultivars. Thus, the domestic cultivar Yeongbohwang exhibited excellent characteristics among the yellow cultivars, while the red cultivars Eumjinara and Cheonjujeok were similar.

Mild taste and early maturing red onion cultivar 'Eumjinara' (매운맛이 적고 숙기가 빠른 자색양파 '엄지나라')

  • Red onion cultivar 'Eumjinara' with mild taste and early maturation was developed in 2013 by the Allium vegetable crop research team, National Institute Crop Science, Rural development Administration (RDA). 'Eumjinara' was originally selected from red onion breeding line, 'IS1115', It was preliminarily selected and designated 'Mon21-31-3' and advanced yield trial was performed from 2008 to 2009. Regional yield trial test was conducted in six regions from 2010 to 2012. Foliage morphology and leaf color were similar to those of a check cultivar, 'Cheonjujeok', but its shape was comparatively broad ovate. Foliage of 'Eumjinara' lodged seven days earlier than check cultivar on May 31 and the average bulb weight ($234{\pm}49.1g/bulb$) was also higher than that of check cultivar. The new cultivar was relatively resistant to clod hardness and downy mildew infection in the field condition. Average yield of 'Eumjinara' in three-year the regional yield trials was 6,999 kg/10a. The total soluble solid contents and quercetin concentrations were $41.43{\mu}g/g$ FW and $556.11{\mu}g/g$ FW, respectively.

Effects of Presowing Seed Treatments on Improvement of Seed Germination and Seedling Emergence of Onion (양파의 발아 및 입묘 향상을 위한 종자처리의 효과)

  • Transplantings of onion (Allium cepa L.) seedlings are required for higher germinability and uniformity. This study was done to determine effects of various seed treatments (seed cleaning, priming, GA$_3$, prechilling, drying, and light quality during seed drying) on germination and seedling emergence. Nongwoodaego and Changnyungdaego were used as cultivars for checking seed germination, and two cultivars and Cheonjuguhyeonghwang were investigated with seedling production rate. Seed cleaning using water prior to the other treatments greatly increased germination rate. Priming with 200mM Ca(NO$_3$)$_2$ solution somewhat alleviated the germination rate while GA$_3$ did not. Prechilling had the highest rate among the treatments. Seed drying after prechilling enhanced the rate compared to non drying, and during drying the prechilled seeds red light illumination showed the greatest rate in comparison with dark, blue, and far-red ones. Seedling production rate was enhanced in sequential treatments of seed cleaning, prechilling for 3 to 5 days, and red light treatment during 6 hour seed drying.

Effects of Onion Vinegar on the Cerebral Blood Flow and the Safety Examination (양파식초가 뇌혈류 및 안전성에 미치는 영향)

  • The aim of this study is to investigate the effects of onion vinegar on the cerebral blood flow by measuring the changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) and by observing the recovery of focal ischemic brain injury in rats. Rats are divided into thee groups depending on the medication; control group (no medication), 8.8-OV group (vinegar using 8.8 brix onion medication), 14.6-OV group (vinegar using 14.6 brix onion medication). The medication of onion vinegar significantly increased rCBF but decreased MABP. This result suggests that onion vinegar significantly increased rCBF by dilating arterial diameter. In addition, focal ischemic brain injury is induced in rats by middle cerebral arterial occlusion. The recovery from focal ischemic brain injury is more significantly improved in the groups using onion vinegar compared to the control group. The amount of recovery is measured by the GAP-43 and the medication of onion vinegar significantly increased GAP-43. This result suggests that onion vinegar is effective on the nerve regeneration. After the medication, the change of body weight, outcomes of renal and liver function test, and outcomes of CBC are analysed for safety examination. There are no statistical differences among control group and all experimental groups in the body weight, renal and liver function test, and CBC. In conclusion, these results suggest that onion vinegar can increase rCBF in normal state, and improve the stability of rCBF in ischemic state.
