Antianaphylactic Effect of Sinpo-tang by Anal Therapy (肛腸療法에 의한 辛蒲湯의 抗아나필락시 效果)

  • Objective : Anal Therapy is another way of taking medicine and applies to each field of clinical treatment extensively. Sinpo-tang(SPT) has been used for the treatment of a allergic rhinitis. In this study, the auther investigated the anti anaphylactic action of Sinpo-tang by anal therapy was investigated on cutaneous allergic reaction models. Methods : Results : 1. Sinpo-tang (0,001-0.1 g/kg) dose-dependently inhibited the compound 48/80-induced ear swelling response in mice. Inhibitory effect of Sinpo-tang was significant (P < 0.05) at the doses of 0.01, 0.1 g/kg. 2. Sinpo-tang (0.001-0.1 g/kg) inhibited the cutaneous allergic reaction activated by anti-dinitrophenyl IgE in rats. Of special note, Sinpo-tang (0.1 g/kg) inhibited the cutaneous allergic reaction by $68\%$. 3. This inhibitory effect of Sinpo-tang was confirmed by observation of alcian blue/nuclear fast red stained-mast cells in the cutaneous tissue. 4. Sinpo-tang (0.01-1 g/L) dose-dependently inhibited the compound 48/80-induced histamine release from the peritoneal mast cells. Conclusions :These results indicate that anal therapy of Sinpo-tang may be beneficial in the treatment of mast cell-mediated anaphylaxis by inhibition of histamine release from mast cells in vivo and in vitro.

Effect of Juglandis Semen BL13·BL17 Aqua-acupuncture and Acupuncture on the Allergic Response (폐유(肺兪)·격유(膈兪)의 호도약침(胡桃藥鍼) 자극이 알레르기 반응에 미치는 영향)

  • Objective & Methods : Experimental studies were done to research the clinical effects of Juglandis Semen aqua-acupuncture and acupuncture($BL_{13}{\cdot}BL_{17}$) on the anti-allergic response. anaphylaxis provoked by the compound48/80, delayed type hypersensitivity response to picryl chloride and SRBC and inflammation response to egg albumin. Results : The following results have been obtained; 1. Juglandis Semen aqua-acupuncture and acupuncture($BL_{13}{\cdot}BL_{17}$) group were increased the survival rate in compound 48/80 induced systemic anaphylactic reaction. 2. Picryl chloride induced contact dermatitis and delayed type hypersensitivity in SRBC challenged mouse were significantly decreased in Juglandis Semen aqua-acupuncture and acupuncture($BL_{13}{\cdot}BL_{17}$) group. 3. Inflammation response-WBC, CRP and Nitric Oxide in egg albumin induced allergic rat were significantly decreased in Juglandis Semen aqua-acupuncture and acupuncture($BL_{13}{\cdot}BL_{17}$) group. Conclusions : According to the above results, Semen aqua-acupuncture and acupuncture($BL_{13}{\cdot}BL_{17}$) both depress the allergy reaction.

Inhibitory Effects of Acute and Chronic Allergic Reaction by Anal Therapy of Gamiseunggal-tang (加味升葛湯의 肛腸療法에 의한 急慢性 알레르기 反應 抑制效果)

  • Gamiseunggal-tang(G-tang) has long been used for the treatment of acute and chronic allergic diseases(especially, urticaria) in oriental medicine. But, It's mechanism of action is not well investigated. Anal therapy is another way of taking medicine, which is not used in common situation but available for the treatment of colon and anal diseases. It is also used in GI tract diseases, Gynecological diseases, and pediatric diseases and so on. It is especially benefitable for patients who are in such situations as coma, severe vomiting, and swallowing difficulty. In this study, the author investigated the effects of G-tang by anal therapy on acute and chronic allergic reactions in murine model. The results obtained are as follows: 1. G-tang(0.01, 0.1 g/kg) inhibited the compound 48/80-induced ear swelling response in mice. Inhibitory effects of G-tang was significant (P<0.05) at the dose of 0.01 g/kg. 2. G-tang(0.001-0.1 g/kg) inhibited the cutaneous allergic reaction activated by anti-dinitrophenyl(DNP) IgE in rats. Especially, G-tang 0.01, 0.1 g/kg inhibited the cutaneous allergic reaction significantly. 3. G-tang(0.01-1 g/L) dose-dependently inhibited the compound 48/80-induced histamine release from the peritoneal mast cells. 4. G-tang(0.001-0.1 g/L) had a inhibitory effect on anti-DNP IgE-induced tumor necrosis factor-${\alpha}$ production. Above results indicate that anal therapy of G-tang may be beneficial in the treatment of acute and chronic allergic diseases.

Anti-allergic effects of Perilla frutescens var. acuta Kudo 30% ethanol extract powder

  • Perilla frutescens var. acuta Kudo (PF) is a traditional Korean medicinal herb for allergic reaction regulation. In the present study, we investigated the effect of 30% ethanol extract powder of PF (EPPF) and rosmarinic acid (RA), the active compound of EPPF on various allergic reactions using in vivo and in vitro models. EPPF and RA significantly inhibited compound 48/48-induced systemic anaphylactic reaction and histamine release (P < 0.05). In addition, EPPF and RA significantly inhibited passive cutaneous anaphylaxis (PCA) in a dose-dependent manner (P < 0.05). These effects were stronger than those of disodium cromoglycate, the reference drug tested. EPPF and RA also significantly inhibited production of inflammatory cytokines, tumor necrosis factor-a interleukin (IL)-6, and vascular endothelial growth factor on the PCA reaction and phorbol 12-myristate 13-acetate and calcium ionophore A23187-stimulated human mast cell line, HMC-1 cells (P < 0.05). Moreover, EPPF and RA showed an inhibitory effect on lipopolysaccharide (LPS)-induced IL-4 production from whole spleen cells. Finally, EPPF and RA significantly decreased IL-4-dependent IgE production by LPS-stimulated whole spleen cells (P < 0.05). In conclusion, these results indicate that EPPF has potent anti-allergic activities.

A Comparison study of the Multiple Allergen Simultaneous Test Results by Sasang Constitution (다중 알레르기 항원 동시 검사 결과의 사상체질 간 비교 연구)

  • Objectives The purpose of this study was to explore the differences in allergic reactions by type of Sasang constitution using multiple allergen simultaneous test(MAST). Methods This study was conducted using the big data of Korean medical health checkups. Subjects with a history of allergic disease, Sasang constitution type information, and MAST results were included. Subjects who had taken drugs before MAST that affected allergic reactions were excluded. We compared total immunoglobulin E(IgE) concentrations, and the number of positive antigens by Sasang constitution using analysis of variance. The presence or absence of a positive reaction and specific IgE concentration classes of 92 antigens in MAST were analyzed by Fisher's exact test or Chi-squared test. Results The distribution of Sasang constitutional types of Tae-Yang, Tae-Eum, So-Yang, and So-Eum among 147 subjects was 2.7%, 42.9%, 38.1%, and 16.3%, respectively. There was no statistically significant difference in the total IgE concentration, the number of positive antigens, and specific IgE concentration classes for each antigen. There was a statistically significant difference in MAST-positive reaction for alder and pine antigens by type of Sasang constitution, and Tae-Eum had the most. Conclusions Tae-Eum showed a relatively strong positive reaction to a large number of antigens compared to the other types, but no statistically significant difference was observed. It is necessary to continuously investigate allergic reactions to provide evidence for the treatment of allergic diseases by Sasang constitution.

Experimental Studies on the Inhibitory Effect of Immediate-Type Allergic Reaction of Tongku-tang (통규탕의 즉각형 알레르기 반응 억제 효과에 관한 실험적 연구)

  • This report describes an inhibitory effect of Tongku-tang(TKT) on mast cell-mediated immediate-type allergic reactions. TKT is an Oriental herbal prescription, which has been successfully applied for the treatment of allergic disorders, mainly skin anaphylactic diseases in eastern medicine. TKT has concentration-dependently inhibited the ear swelling response induced by intradermal injection of non-specific mast cell degranulator compound 48/80 in mice. TKT also inhibited mast cell-dependent passive cutaneous anaphylaxis activated by dinitrophenyl (DNP)-IgE antibody in rats. I studied the effect of TKT on the histamine and β-hexosaminase release from the rat peritoneal mast cells by compound 48/80. TKT did not inhibit significantly the histamine and β-hexosaminase release from the rat peritoneal mast cells by compound 48/80. However, TKT inhibited both TNF-α and IL-1β secretion induced by phorbol 12-myristate 13-acetate and A23187 respectively. These results provide evidence that TKT may be beneficial in the treatment of immediate-type allergic reaction.

Effects of Gunjung-tang on Allergic Reaction (알레르기 반응에 미치는 건중탕의 효과)

  • Gunjung-tang(GJT) has been reported at Shanghanlun(傷寒論), which has been used for the treatment of general weakness, digestive organ disease and the treatment of allergic disorders in clinical medicine. However, its effect in experimental models remains unknown. In a rat and mouse model, the role of GJT was examined in mast cell-dependent allergic reactions and secretion of inflammatory cytokines. GJT inhibited anti-dinitrophenyl IgE antibody-induced passive cutaneous anaphylaxis reaction by Intraperitoneal pretreatment. GJT inhibited both IL-1β and TNF-α secretion on egg albumin induced allergic tissues and compound 48/80 induced thymus. Furthermore, GJT enhanced Band T lymphocytes proliferation. Our findings provide evidence that GJT inhibits the IgE-dependent allergic reactions and inflammatory cytokines secretion, and enhanced immune response.

Inhibitory Effect of Immediate-Type Allergic Reaction by Magnoliac flos (辛夷花에 의한 卽時型 알레르기의 反應 抑制 效果)

  • The mast cell plays a pivotal role in initiating allergic response by secreting intracytoplasmic granular mediators such as histamine. Magnoliae flos has been used for treatment of allergic disease in Korea. However, its effect in experimental models remains unknown. The present report describes an inhibitory effect of Magnoliae flos on mast cell-mediated immediate-type allergic reactions. Topical application of compound 48/80 can induce an ear swelling response in normal($WBB6F_1-+/+$) mice but not in the congenic mast cell-deficient $WBB6F_1-W/W^v$ mice. Magnoliae flos inhibited concentration-dependently mast cell-dependent ear swelling response induced by compound 48/80 by topical application. Magnoliae flos inhibited concentration-dependently passive cutaneous anaphylaxis induced by anti-dinitrophenyl (DNP) IgE in rats by topical application. Magnoliae flos also inhibited concentration-dependently the histamine release from the rat peritoneal mast cells by compound 48/80 or anti-DNP IgE. Moreover, Magnoliae flos had a significant inhibitory effect on compound 48/80-induced systemic anaphylactic reaction. These results indicate that Magnoliae flos inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.

Ginsenoside Rg3 suppresses mast cell-mediated allergic inflammation via mitogen-activated protein kinase signaling pathway

  • Background: Ginsenoside Rg3 (G-Rg3) is the major bioactive ingredient of Panax ginseng and has many pharmacological effects, including antiadipogenic, antiviral, and anticancer effects. However, the effect of G-Rg3 on mast cell-mediated allergic inflammation has not been investigated. Method: The antiallergic effects of G-Rg3 on allergic inflammation were evaluated using the human and rat mast cell lines HMC-1 and RBL-2H3. Antiallergic effects of G-Rg3 were detected by measuring cyclic adenosine monophosphate (cAMP), detecting calcium influx, and using real-time reverse transcription polymerase chain reaction, enzyme-linked immunosorbent assay, Western blotting, and in vivo experiments. Results: G-Rg3 decreased histamine release from activated mast cells by enhancing cAMP levels and calcium influx. Proinflammatory cytokine production was suppressed by G-Rg3 treatment via regulation of the mitogen-activated protein kinases/nuclear factor-kappa B and receptor-interacting protein kinase 2 (RIP2)/caspase-1 signaling pathway in mast cells. Moreover, G-Rg3 protected mice against the IgE-mediated passive cutaneous anaphylaxis reaction and compound 48/80-induced anaphylactic shock. Conclusion: G-Rg3 may serve as an alternative therapeutic agent for improving allergic inflammatory disorders.

The Effects of Tongkwansan on the Changes of Granulocytes and Nasal Tissue in Allergic Rhinitis Mouse Model (통관산(通關散)이 알레르기성 비염 동물 모델의 과립구 및 조직학적 변화에 미치는 영향)

  • Background & Objectives : Rhinitis is an inflammation of nasal mucosa and the symtoms are watery rhinorrhea, sneezing, itchy nose, and nasal obstruction. Rhinitis is classified into allergic rhinitis and nonallergic. Allergic rhinitis is an immune reaction by allergen, and vasomotor rhinitis which is nonallergic noninfectious hypersensitive reaction. The incidence of allergic rhinitis has increased and the rate of vasomotor rhinitis is high. However there have been no studies about vasomotor rhinitis compared with allergic rhinitis. And there have been no studies so far performed on the effect of Tongkwansan. Therefore this study is aimed to find out the effects of Tongkwansan on allergic rhinitis and vasomotor rhinitis. Materials and Methods : Fifteen BALC/c mouses divided into three groups : normal group, control group and sample group. To induce the allergic rhinitis in control group and sample group, mouses were sensitized intrapertioneally 0.1 % ovalvumin solution three times at intervals of 1 week. Then intranasal sensitization was performed by diffusing 0.1 % ovalbumin solution 3 times at intervals of 2 days. After that time, mouses in the sample group were oral and administration treated by Tongkwansan for 28 days. We observed changes in nasal mucosa and submucosa; also changes in the segment of leucocyte, erythrocyte, neutrophil, lymphocyte, monocyte, eosinophil, IL-4 and $IFN-{\gamma}$ in blood. We used the statistical methods of ANOVA test(p<0.05). Results : There were no significant changes statistically in leucocyte, erythrocyte, neutrophil, lymphocytem, monocyte, eosinophil, IL-4 and $IFN-{\gamma}$ in blood(p<0.05). Hypertrophy of epithelium in nasal mucosa and expansion of glandular cells in nasal submucosa were decreased in treated group when compared with control group. Conclusion : According to above results, it is supposed that Tongkwansan has significant effects on vasomotor rhinitis which is nonallergic and noninfectious.
