This study was designed to investigate clinical features, allergy-inducing substances as well as foods, and nutritional behavior of allergic patients. Sixty-eight adult male and 78 adult female patients were surveyed by using the self-developed questionnairs. The results were as follows: 1. The common target organs of allergic symptoms included bronchus, skin and nose. 2. 37.7% of the subjects did not recognize their allergy inducing substances. Among the known substances, house-dust mite, food, temperature and weather change, pollens, smoke, air pollution materials, drugs and even stress provoked symptoms. 3. As for the food allergy, a number of foods were found to be implicated in allergic reactions. These foods included pork, chicken, seafoods such as mackerel, crab & shrimp, milk, egg, peanut, apple and pupa. Symptoms and signs of food reactions included urticaria, asthma, and rhinitis. 4. The nutrition adequacy ratio (NAR) of energy was below 0.75 for male patients and about 0.80 for female patients. And 24.0% of the subjects were regarded as excellent, 31.5% as fair, and 44.5% as poor in food related behavior evaluation.
Antigen delivery systems play critical roles in determining the quality and quantity of Ab responses in vivo. Induction of protective antibodies by B cells is essential in the development of vaccines against infectious pathogens, whereas production of IgE antibodies is prerequisite for investigation of allergic responses, or type 1 hypersensitivity reactions. Virus-like particles (VLPs) are efficient platforms for expression of proteins of interest in highly repetitive manners, which grants strong Ab responses to target antigens. Here, we report that delivery of hen egg lysozyme (HEL), a model allergen, through VLP could provoke strong HEL specific IgE Ab responses in mice. Moreover, acute allergic responses were robustly induced in the mice sensitized with VLPs that express HEL, when challenged with recombinant HEL protein. Our data show that antigen delivery in the context of VLPs could function as a platform for sensitization of mice and for subsequent examination of allergic reactions to molecules of interest.
Objective : To investigate the effect between Fructus Immaturus Ponciri (FIP, the immature fruit of Poncirus trifoliata) and Fructus Ponciri (FP, the ripe fruit of Poncirus trifoliata) on mast cell-mediated immediate-type allergic reactions. Methods : We performed anaphylactic reaction, histamine release, cAMP, $TNF-{\alpha}$, IgE. Results : The aqueous extract of FIP dose-dependently inhibited systemic and local allergic reaction was induced by compound 48/80 or anti-dinitrophenyl (DNP) IgE in a murine model. FIP also significantly inhibited mast cell-dependent ear swelling response induced by topical application of compound 48/80. When mice were orally pretreated with FIP, the plasma histamine levels were reduced in a dose-dependent manner. FIP dose-dependently inhibited histamine release from the rat peritoneal mast cells (RPMCs) was activated by compound 48/80 or anti-DNP IgE. The level of cAMP in RPMCs, when FIP was added, increased compared with that of a normal or control. In addition, FIP had a significant inhibitory effect on anti-DNP IgE-induced tumor necrosis factor-a ($TNF-{\alpha}$) production from the RPMCs and IgE produced by lipopolysaccharide-stimulated murine whole spleen cells or U266B1 as human IgE-bearing B cells. However, FP showed the lower inhibition rate than those of FIP in above all allergic reactions. Conclusion : These data have important implications for our understanding of the clinical effects of FIP and FP on allergic diseases, and FIP is more effective than FP on the allergic reaction.
Perilla frutescens var. acuta Kudo (PF) is a traditional Korean medicinal herb for allergic reaction regulation. In the present study, we investigated the effect of 30% ethanol extract powder of PF (EPPF) and rosmarinic acid (RA), the active compound of EPPF on various allergic reactions using in vivo and in vitro models. EPPF and RA significantly inhibited compound 48/48-induced systemic anaphylactic reaction and histamine release (P < 0.05). In addition, EPPF and RA significantly inhibited passive cutaneous anaphylaxis (PCA) in a dose-dependent manner (P < 0.05). These effects were stronger than those of disodium cromoglycate, the reference drug tested. EPPF and RA also significantly inhibited production of inflammatory cytokines, tumor necrosis factor-a interleukin (IL)-6, and vascular endothelial growth factor on the PCA reaction and phorbol 12-myristate 13-acetate and calcium ionophore A23187-stimulated human mast cell line, HMC-1 cells (P < 0.05). Moreover, EPPF and RA showed an inhibitory effect on lipopolysaccharide (LPS)-induced IL-4 production from whole spleen cells. Finally, EPPF and RA significantly decreased IL-4-dependent IgE production by LPS-stimulated whole spleen cells (P < 0.05). In conclusion, these results indicate that EPPF has potent anti-allergic activities.

Since the prevalence of allergies is increasing, food allergy is a major concern for consumers, as well as for the food industry. The foods that account for over 90% of all moderate to severe allergic reactions to food are milk, eggs, peanuts, soybeans, fish, shellfish, wheat, and tree nuts. Of these food allergens, milk is one of the major animal food allergens in infants and young children. Milk is the first food that an infant is exposed to; therefore, the sensitization rate of milk in sensitive individuals is understandably higher. The mechanisms involved in allergic reactions caused by this hypersensitivity are similar to those of other immune-mediated allergic reactions. The reactions occur in the gastrointestinal tract, skin, and respiratory tract, with headaches and psychological disorders occurring in some instances. The major allergenic proteins in milk are casein, ${\beta}$-lactoglobulin, and ${\alpha}$-lactalbumin, while some of the minor allergenic proteins are lactoferrin, bovine serum albumin, and immunoglobulin. Reliable allergen detection and quantification are essential for compliance with food allergen-labeling regulations, which protect the consumer and facilitate international trade.
The occupational skin diseases in an industry of silk reeling located in Daegu city of Korea were analyzed by inspection through a mass survey for improving the workers' health, November, 1975. The subjects, 177 persons were all females, and were distributed at 18-38 years old. About two thirds were under 25 years old. The meanage was 23.6 years. The incidence of skin diseases was about 53%. It was generally divided into three groups;- the epidermic changes of those were most frequent, 47.5%, the allergic reactions were 14.7%, and athlete's foot was only four cases, 2.3%. Most frequent signs of epidermic changes was the worn-out nail, 32.3%; and the keratolysis, 28.8%; and the callus, 15.3% were followed in order. Theses were significant in five per cent level between the sections of the silk reeling and the manufacturing;-Allergic reactions and worn-out nail were more frequent in the manufacturing group than the silk reeling, and keratolysis and callus were inverted proportionally. Allergic reactions were occurred within a few months from first entrance time, and it was more frequent in the group exposed to same allergen in the past than the continuing group from first. Most of the appearing sites of theses were hands, and infrequently, it was occurred in the face, neck, trunk, knee anfeet. These epidermic changes except allergic phenomena were appeared in the fingers, palmal and dorsal surface of hands frequently contacted. But these changes and allergic reactions were generally combined with two or more kinds.
Gagam-danguiemja (GDGJ), a traditional Korean prescription, has been used as therapeutics for atopic allergic diseases such as atopic dermatitis. To evaluate the atopic allergic effect of modified GDGJ, we investigated a possible effect of GDGJ on mast cell-mediated allergic reaction, cytokinases secretion and mRNA expression in vivo and in vitro. Mast cells are a potent source of mediators that regulate the inflammatory response in allergic reaction. In mice orally administered by GDGJ (0.01, 0.1 and 1.0 g/kg) for 1 h, compound 48/80-induced ear oedema was significantly reduced. TNF-${\alpha}$, IL-8, and IL-6 secretion were inhibited by GDGJ in the human mast cell line (HNC-1). But TNF-${\alpha}$, IL-8, and IL-6 mRNA expression were not inhibited by GDGJ at the dose of 0.01 mg/ml. These findings may help in understanding the mechanism of action of this herbal medication, leading to the control of mast cells in atopic allergic reaction like AD.
Mast cell-mediated allergic disease is involved in many diseases such as anaphylaxis, asthma and atopic dermatitis. The discovery of drugs for the treatment of allergic disease is an important subject in human health. In the present study, the effect of water extract of Gleditsiae Spina (WGS) (Leguminosae), on compound 48/80-induced systemic allergic reaction, anti-DNP IgE antibody-induced local allergic reaction, and histamine release from human mast cell line (HMC-1) cells were studied. In addition, the effect of WGS on phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187 (A23187)-induced gene expression and secretion of pro-inflammatory cytokines were investigated using HMC-1 cells. WGS was anally administered to mice for high and fast absorption. WGS inhibited compound 48/80-induced systemic allergic reaction. WGS dose-dependently decreased the IgE-mediated passive cutaneous anaphylaxis. WGS reduced histamine release from HMC-1 cells. In addition, WGS decreased the gene expression and secretion of pro-inflammatory cytokines in PMA plus A23187-stimulated HMC-1 cells. These findings provide evidence that WGS could be a candidate as an antiallergic agent.
Mast cells play an important role in early and late phase allergic reactions through allergen and IgE-dependent release of histamine, proteases, prostaglandins, and several multifunctional cytokines. In this study, we investigated whether Cudrania tricuspidata extract (CTE) suppresses IgE-mediated allergic responses in mast cells, an allergic animal model, and its mechanism of action in mast cells. We found that CTE inhibited IgE-mediated degranulation and cytokine production in rat basophilic leukemia (RBL)-2H3 mast cells and bone marrow-derived mast cells (BMMC), as well as passive cutaneous anaphylaxis (PCA) in mice. With regard to its mechanism of action, CTE suppressed the activating phosphorylation of spleen tyrosine kinase (Syk), a key enzyme in mast cell signaling processes and that of LAT, a downstream adaptor molecule of Syk in $Fc{\varepsilon}RI$-mediated signal pathways. CTE also suppressed the activating phosphorylation of mitogen-activated protein (MAP) kinases and Akt. The present results strongly suggest that the anti-allergic activity of CTE is mediated through inhibiting degranulation and allergic cytokine secretion by inhibition of Syk kinase in mast cells. Therefore, CTE may be useful for the treatment of allergic diseases.

The effect of aqueous extract of Stachys riederi var. japonica Miq. (Labiatae) (SRAE) on the immediate-type allergic reactions was investigated. SRAE was found to exhibit a inhibitory activity on the compound 48/80-induced systemic anaphylaxis in mice. SRAE inhibited the plasma histamine release induced by compound 48/80 in mice. In addition, SRAE also inhibited the passive cutaneous anaphylaxis reaction induced by IgE/anti-IgE in mice. The effect of SRAE on the histamine release from rat peritoneal mast cells (RPMC) was studied. SRAE inhibited the histamine release induced by compound 48/80 in RPMC. To clarify the mechanism of these inhibiting reactions, we investigated the effects of SRAE on cyclic AMP (cAMP). The level of cAMP in human leukemia cell line, HMC-1, when SRAE (1 mg/ml) was added, significantly increased compared with that of basal cells. These results indicate that SRAE may be beneficial in the treatment of immediate-type allergic reaction.
