Catalytic performance of Al-MCM-48 molecular sieves in the isopropylation of phenol with isopropyl acetate (isopropyl acetate을 이용한 페놀의 isopropylation 반응의 Al-MCM-48 분자체 촉매반응 특성)

  • Al-MCM-48 molecular sieves (Si/Al = 25, 50, 75 and 100) were synthesized hydrothermally using cetyltrimethyl ammonium bromide as the structure directing template. The orderly arrangement of mesopores was evident from the low angle X-ray diffraction patterns and TEM images. The catalytic performance was evaluated in the vapour phase isopropylation of phenol with isopropyl acetate. Phenol conversion decreased with increase in the Si/Al ratio of the catalysts. The major reaction product was 4-isopropyl phenol (selectivity: 78%). Delocalization of phenolic oxygen electron pair over the aromatic ring promoted para-selective alkylation. Such delocalization could be aided by the hydrophilic surface of the molecular sieves. Though ester was used as the alkylating agent, phenyl isopropyl ether was not formed

Thermally Stable and Processible Norbornene Copolymers

  • Processible norbornene copolymers were realized by judiciously designing norbomene comonomers, which were themselves prepared by the Diels-Alder reaction of cyclopentadiene and benzoquinone followed by the isomerization and alkylation of alcohols. The norbornene copolymers containing these derivatized comonomers, prepared by [Pd($x_{2}CH-{3} $)$_{4}$][$SbF_{6}$]$_{2}$ catalyst, exhibited excellent solubility in many organic solvents as well as good thermal stability, as evidenced by their high glass transition ($T_{g}$) and decomposition ($\∼$350$^{circ}C$) temperatures. In addition, fairly strong adhesion to substrates such as glasses and silicon wafers was also achieved with these copolymers to overcome the limitations experienced by polynorbornene homopolymers and to make them attractive for many important industrial applications.

  • For the development of new synthetic method for unnatural amino acid esters, N-aryl phenylglycine Ο-alkyl esters 4a∼i were synthesized through base-catalyzed hydrolysis of 1,5-diphenylhydantoins 1a∼b and Ο-alkylation in 16∼87% yield. An efficient and practical route for final 4a∼i was that the starting materials 1a∼b were heated in dil-methanol for 30 minute using sodium hydroxide or potassium hydroxide and evaporated. In addition, reaction mixture were refluxed for 1 h in DMF. All synthetic process from hydantoin to N-aryl phenylglycine Ο-alkyl esters 4a∼i could be carried out in one-pot without isolation of intermediates.

A Versatile Synthesis of α-Keto (cyanomethylene)triphenylphosphorane Ylides from Alkyl Halides Utilizing a Noble Phenylsulfonyl Reagent

  • A noble phenylsulfonyl reagent 8 having ${\alpha}$-oxo (cyanomethylene)triphenylphosphorane ylide subunit readily condensed with various alkyl halides under basic conditions to afford ${\beta}$-alkyl-${\alpha}$-oxo-${\beta}$-phenylsulfonyl (cyanomethylene)triphenylphosphorane ylides 9 in excellent yields. These sulfonyl ylides 9 were then reductively desulfonylated with $Na(Hg)/Na_2HPO_4$ in the presence of methanol to provide ${\alpha}$-keto (cyanomethylene)-triphenylphosphorane ylides 2' in good to excellent yields. Our new synthetic approach offers an expeditious access to various ${\alpha}$-keto (cyanomethylene)triphenylphosphorane ylides from alkyl halides utilizing a new phenylsulfonyl reagent as the key reagent under mild reaction conditions in good overall yields.

  • Starting with readily available p-tert-butylcalix[4]arene 3 tert-butyl groups are removed by $AlCl_3$-catalyzed de-alkylation reaction, and the calix[4]arene 4 formed is converted into the tetraacyl esters. These compounds undergo Fries rearrangement to yield p-acylcalix[4]arenes. p-Acetyl, p-propionyl, p-butyryl, and p-benzoylcalix[4]arene 10, 11, 12 and 14 are synthesized in 70-80% yields by treatment of the corresponding esters 5, 6, 7 and 9 with $AlCl_3$ in nitrobenzene. When the tetraisobutyryl ester 8 was treated with the same condition, only two isobutyryl groups were rearranged to the para-positions of calix[4]arene, and remaining two groups were simply cleaved.

  • A stereocontrolled synthesis of (${\pm}$)-isocomene (1) via selective monoketalization of tricyclo[6.3.0.$0^{1.5}$]undeca-4,7-dione(13) was reported. Grignard reaction of bicyclic enone 10, which was prepared from 2-methyl-1,3-cyclopentadione, gave the 1,4-addition product 11. The subsequent aldol condensation product 12 was converted to mesyl derivative 13. Transformation from 13 to the desired product 19 was achieved by a series of reactions, i.e., the selective monoketalization at C-4 carbonyl group, the elimination of a mesyl group, Birch alkylation, methylation at C-6, the reduction of carbonyl group, the dehydration of alcohol 18, and hydrolysis of the ketal group.

  • Quarternized poly(4-vinyl pyridine)s have been prepared by the reaction of poly (4-vinyl pyridine)s (Mw=50 kg/mol and 200 kg/mol) and alkylating agents varying the carbon numbers of the alkyl groups (m):dimethyl sulfate (m=1) as well as bromoalkane (m= 5, 8, 12, 16, and 22) was used as an alkylating agent. The degree of alkylation was determined by using an elemental analysis and NMR spectroscopy. As a result, polyelectrolytes were obtained by the almost full alkylation of poly (4-vinyl pyridine)s. The critical aggregation concentration (CAC) was determined by measuring the change of turbidity occurred by addition of sodium dodecyl sulfate (SDS) into aqueous solution of quarternized poly-(4-vinyl pyridine)s, and the dependence of molecular weight of polymer, the length of N-alkyl group and concentration of NaCl upon CAC was investigated. As a result, as the molecular weight or the length of alkyl group was increased, less amount of SDS Gould induce the aggregation.

  • Six, heretofore undescribed, $5^I-Methyl-5^I-(5-Substituted uracil-1-ylmethyl)-2^I-oxo-3^I-methylenetetrahydrofurans(F, Cl, Br, l, CH_3, H)(6a-f)$were synthesized and evaluated against three cell lines (FM-3A, P-388 and U-937). For the preparation of .alpha.-methylene-.gamma.-butyrolactone bearing 5-substituted uracils (6a-f), the effcient Reformatsky type reaction was employed which involves the treatment of ethyl .alpha.(bromomethyl) acrylate and zinc with the respective 5-substituted uracil-1-ylacetones (5a-f). The acetone derivatives (5a-f) were directly obtained by the respective alkylation reaction of 5-substituted uracils with chloroacetone in the presence of $K_{2}$$CO_{3}$(or NaH). These lactone compounds 6a-f exhibited moderate to significant activity in all of the three cell lines, and 6b, 6c and 6e showed significant antitumor activities (inhibitory concentrations ($IC_{50}$) ranged from 1.3-3.8 .mu.g/ml.

Design, Synthesis and Preliminary Biological Evaluation of a Biotin-S-S-Phosphine Reagent

  • Biotin-S-S-Phosphine was designed and synthesized as a potential tool for a proteomic study of O-GlcNAcmodified proteins. This reagent features a disulfide linker between a triarylphosphine moiety, which allows selective conjugation to azide-containing proteins, and a biotin moiety that can allow easy isolation through its strong affinity toward avidin-coated solid beads. The disulfide linkage within this reagent can allow the easy release of the bound molecules of interest, which is difficult to achieve when a biotin:avidin pair is used alone, by reducing the disulfide bond of the reagent with DTT. Preliminary in vitro biological assays with azidelabeled and unlabeled cell lysates and a pure protein Nup62 showed that the Biotin-S-S-Phosphine reagent is highly reactive toward the free thiol groups of proteins. When a molecular tool with a disulfide linker is applied to the enrichment of the molecules of interest from other species, it is important to block the free-thiols of the sample using exhaustive alkylation prior to the Staudinger ligation reactions to restore the bioorthogonal nature of this reaction.

Synthesis and Cation Binding Properties of Triester Calix[4]arenes and Calix[4]quinones

  • The complexes M(CO)4-1,2-(PPh2)2-1,2-C2B10H10 (M=Cr 2a, Mo 2b, W 2c) have been prepared in good yields from readily available bis-diphenylphosphino-o-carboranyl ligand, closo-1,2-(PPh2)2-1,2-C2B10H10 (1), by direct reaction with Group Ⅵ metal carbonyls. The infrared spectra of the complexes indicate that there is an octahedral disposition of chelate bis-diphenylphosphino-o-carboranyl ligand around the metal atom. The crystal structure of 2a was determined by X-ray diffraction. Complex 2a crystallizes in the monoclinic space group P21/n with cell parameters a = 12.2360(7), b = 17.156(1), c = 16.2040(6) Å, V = 3354.1(3) Å3, and Z =4. Of the reflections measured a total of 2514 unique reflections with F2 > 3σ(F2) was used during subsequent structure refinement. Refinement converged to R1 = 0.066 and R2 = 0.071. Structural studies showed that the chromium atom had a slightly distorted pseudo-octahedral configuration about the metal center with two phosphine groups of o-carborane occupying the equatorial plane cis-orientation to each other. These metal carbonyl complexes are rapidly converted to the corresponding metal carbene complexes, [(CO)3M=C(OCH3)(CH3)]-1,2-(PPh2)2-1,2-C2B10H10 (M= Cr 3a, Mo 3b, W 3c), via alkylation with methyllithium followed by O-methylation with CF3SO3CH3.
