Grape is one of the most popular and widely cultivated fruits in the world. Although grape skin and seeds are waste product of the winery and grape juice industry, these wastes contain large amounts of phytochemicals such as flavonoids, phenolic acids, and anthocyanidins, which play an important role as chemopreventive and anticancer agents. We evaluated efficacies of grape skin and seeds on hepatic injury induced by dimethylnitrosamine (DMN) in rats. Treatment with DMN significantly increased levels of serum alanine transaminase, aspartate transaminase, alkaline phosphatase, and bilirubin. Diet supplementation with grape skin or seeds (10% daily for 4 weeks) prevented these elevations. The grape skin and seeds also restored serum albumin and total protein levels, and reduced the hepatic level of hydroxyproline and malondialdehyde. Furthermore, grape skin and seeds reduced DMN-induced collagen accumulation, as estimated by histological analysis of liver tissue stained with Sirius red. Grape skin and seeds also reduced hepatic stellate cell activation, as assessed by ${\alpha}$-smooth muscle actin staining. In conclusion, grape skin and seeds exhibited in vivo hepatoprotective and antifibrogenic effects against DMN-induced liver injury, suggesting that grape skin and seeds may be useful in preventing the development of hepatic fibrosis.
Tunicamycin, an endoplasmic reticulum (ER) stress inducer, specifically inhibits N-glycosylation. The cyclic AMP (cAMP) response element-binding protein H (CREBH) was previously shown to be regulated by UPR-dependent proteolytic cleavage in the liver. On the other hand, the role of CREBH in other tissues is unknown. In the present study, tunicamycin increased the level of CREBH activation (cleavage) as well as mRNA expression in osteoblast cells. Adenoviral (Ad) overexpression of CREBH suppressed BMP2-induced expression of alkaline phosphatase (ALP) and osteocalcin (OC). Interestingly, the BMP2-induced OASIS (structurally similar to CREBH, a positive regulator of osteoblast differentiation) expression was also inhibited by CREBH overexpression. In addition, inhibition of CREBH expression using siRNA reversed the tunicamycin-suppressed ALP and OC expression. These results suggest that CREBH inhibited osteoblast differentiation via suppressing BMP2-induced ALP, OC and OASIS expression in mouse calvarial derived osteoblasts.
On the inter-order protoplast fusants of Lentinus edodes and Ganoderma lucidum was the antitumor activity test performed and the fusant P22 was selected. The hot water extract of the cultured mycelia of P22 were purified by DEAE-cellulose chromatographya and the resulting purified fraction was designated as P22A. It was found to be a proteoglycan whose molecular weight was 47 kDa. When examined for immunopotentiation activity, P22A increased the number of colony forming unit in the bone marrow stem cells to 3-folds. It also potentiated the secretion of nitric oxide in activated macrophages to 2-folds. In humoral immune response, it increased the activities of the alkaline phosphatase in differentiated B cells to 1.6-folds and the number of plaque forming cells to 1.8-folds. In cellular immune response, it restored the depressed response of delayed type hypersensitivity in tumor bearing mice to normal level. These results suggest that P22A have potential to restore the decreased immune activity of the tumor bearing mice to normal level.
This paper aimed to investigate the effect of MWCNTs on properties of slag Geopolymeric mortar. Geopolymeric matrices containing different MWCNTs concentrations (0.0, 0.1, 0.2, 0.3 and 0.4 % by weight of the used binder) were synthesized. The Geopolymer mortar composed of aluminosilicate slag to sand (1:2), while the alumino silicate source binder composed of 50% air cooled slag and 50%water cooled slag both passing a sieve of $90{\mu}m$, while the sand passing a sieve of 1 ml. The materials prepared at water/binder ratios in a range of 0.34-0.39% depending on the added MWCNT, whereas the Gelenium Ace-30 superplasticizer used in the ratio of 1.4-2.2% from the total dry weight for better dispersion of MWCNT under sonication for 15 min. Alkaline activation of the Geopolymer mortar was carried by using of 6% NaOH. Curing was performed under temperature of $40^{\circ}C$ and 100% R.H. Results showed that the addition of MWCNTs enhanced the resulting amorphous geopolymer structure with marked decrease in the drying shrinkage as well as water absorption specially when using 0.1% MWCNT, while further increase in MWCNTs results in agglomeration in MWCNT within the matrix and so hinder the propagation of Geopolymerization reaction and negatively affect the formed geopolymer structure.
We isolated two soluble forms of glutamate dehydrogenase isoproteins, GDH I and GDH II, from bovine brain. The regulation of GDH I and GDH II by phosphorylation and dephosphorylation has been examined in various conditions. There were dose- and time- dependent activation of the GDH isoproteins when phosphorylated by cAMP-dependent protein kinase. The phosphorylated GDH had 1.1 mol of covalently bound phosphate/mol of subunit and a 2-fold increased specific activity. The phosphorylated amino acid was identified as serine. When treated with alkaline phosphatase, the activities of the phosphorylated GDH isoproteins were reduced in dose and time dependent manner and returned to those of unphosphorylated enzymes. There were no significant differences between GDH I and GDH II in their sensitivities to the action of phosphorylation and dephosphorylation demonstrating that the microenvironmental structures of the phosphorylation site in GDH isoproteins are similar to each other, These results results suggest that the inter-conversion between less active form of brain GDH isoproteins and more active form is regulated by phosphorylation through cAMP-dependent protein kineses.
From $70\%$ ethanol extract of the roots of Smilax bockii warb., seven flavonoids, kaempferol (1), $kaempferol-7-O-\beta-D-glucopyranoside$ (2), quercetin (3), isorhamnetin (4), (+)-dihydrokaempferol (5), engeletin (6), isoengeletin (7), and $n-butyl-\beta-D-fructopyranoside$ (8), caffeic acid n-butyl ester (9) were isolated and identified by means of chemical and spectroscopic. Compounds 2, 4, and 6-9 were isolated for the first time from the roots of S. bockii and compounds 2, 8, and 9 were firstly isolated from the genus Smilax. In addition, using the SEAP (Secreted alkaline phosphatase) assay system, we investigated the in vitro anti-inflammatory activity of the $70\%$ ethanol extract of the roots of S. bockii, which showed moderate activity in inhibiting $TNF-\alpha-induced NF-{\kappa}B$ activation with an $IC_{50}$ value of $166.6 {\mu}g/mL$.

Bone health is maintained by balance between bone resorption and bone formation, and bone homeostasis requires balanced interactions between osteoblasts and osteoclasts. Most of drugs and functional foods for bone health have been developed as bone resorption inhibitors, which maintain bone mass by inhibiting the function of osteoclasts. The recent studies have shown beneficial effects of marine natural products on bone health. Therefore, this review is aimed to study effects of marine-derived natural substances on osteoarthritis inhibition via attenuation of MMPs and osteoblastic differentiation via activation of alkaline phosphatase (ALP), osteoclacin (OC), bone morphogenic protein-2 (BMP-2) as an important factor for bone formation, and mineralization. The present review can provide new insights in the osteoblastic differentiation of marine natural products and possibility for their application in bone health supplement.
The Laminaria japonica Aresch (Sea tangle) belongs to the brown algae and has a long history as a food material in Asia, including Korea. Recent studies have found that the fermented Sea tangle extract (FST) inhibited the differentiation of osteoclasts and protected osteoblasts from oxidative damage. This study aims to explore the possibility that FST can induce the differentiation of osteoblasts and identify the responsible mechanism. According to our results, FST induced differentiation into osteogenic cells in the presence of osteoblastic MC3T3-E1 cells under non-toxic conditions.. This finding was confirmed by phalloidin staining, increased alkaline phosphatase activity, and calcium deposition. Additionally, it was found that this process was achieved by increasing the expression of key factors involved in osteoblast differentiation, such as runt-related transcription factor-2, osterix, β-catenin, and bone morphogenetic protein-2. Moreover, FST increased autophagy, which may contribute to the maintenance of the bone formation homeostasis, and is associated with the activation of the phosphatidylinositol 3-kinase/Akt and mitogen-activated protein kinase signaling pathways. Although further research about the bioactive substances contained in FST and the tests of their efficacy are required, the results of this study indicate that FST has incredible applicability as a functional material for maintaining the bone homeostasis.
Disposal of waste glass derived from bottle or packaging glass, flat glass, domestic glass is one of the major environmental defies. Moreover, the remnants of bricks resulting from the remnants of buildings are also considered an important factor in polluting the environment due to the difficulty of filling or getting rid it. The aim of this study is to valorize these wastes through chemical activation to be an environmentally friendly material. The Microstructure, compressive strength, setting time, drying shrinkage, water absorption of different pastes produced by clear glass (CG), green glass (GG) and brick waste (BP) activated were tested and recorded after curing for 3, 7, 28 and 365 days. Five samples of pastes were mixed in proportions represented by: 100% GP (GP), 100% GGP (GGP), 100% BP (BP), 90% GP + 10% BP (GPB) and 90% GGP + 10% BP (GGPB). Various parameters considered in this study include sodium hydroxide concentrations (10 mol/l); 0.4 as alkaline liquid to binder ratio; 2.5 as sodium silicate to sodium hydroxide ratio and cured at 60℃ for 24 hours. Experimental results revealed that the addition of 10% of BP resulted in an increased strength performance of geopolymer paste especially with GGPB compared to GGP in 365 days. In addition, the 10% amount of BP increases the absorption and shrinkage rate of geopolymer pastes (GPB and GGPB) by reducing the setting time. SEM results revealed that the addition of BP and GP resulted in a dense structure.

Bone homeostasis is regulated by constant remodeling through osteogenesis by osteoblasts and osteolysis by osteoclasts and osteoporosis can be provoked when this balance is broken. Present pharmaceutical treatments for osteoporosis have harmful side effects and thus, our goal was to develop therapeutics from intrisincally safe natural products. Fucoidan is a polysaccharide extracted from many species of brown seaweed, with valuable pharmaceutical activities. To intensify the effect of fucoidan on bone homeostasis, we hydrolyzed fucoidan using AMG, Pectinex and Viscozyme. Of these, fucoidan biotransformed by Pectinex (Fu/Pec) powerfully inhibited the induction of tartrate-resistant acid phosphatase (TRAP) activity in osteoclasts differentiated from bone marrow macrophages (BMMs) by the receptor for activation of nuclear factor-κB ligand (RANKL). To investigate potential of lower molecular weight fucoidan it was separated into >300 kDa, 50-300 kDa, and <50 kDa Fu/Pec fractions by ultrafiltration system. The effects of these fractions on TRAP and alkaline phosphatase (ALP) activities were then examined in differentiated osteoclasts and MC3T3-E1 osteoblasts, respectively. Interestingly, 50-300 kDa Fu/Pec suppressed RANKL-induced osteoclasts differentiation from BMMs but did not synergistically enhance osteoblasts differentiation induced by osteogenic agents. In addition, this fraction inhibited the expressions of NFATc1, TRAP, OSCAR, and RANK, which are all key transcriptional factors involved in osteoclast differentiation, and those of Src, c-Fos and Mitf, as determined by RT-PCR. In conclusion, enzymatically low-molecularized 50-300 kDa Fu/Pec suppressed TRAP by downregulating RANKL-related signaling, contributing to the inhibition of osteoclasts differentiation, and represented a potential means of inducing bone remodeling in the background of osteoporosis.
