고등어 surimi를 효율적으로 생산하기 위한 연구의 일환으로 감압수세장치를 설계 제작하고 이 장치를 이용하여 각각 상압과, 560 및 660 mmHg의 감압 하에서 각각 1, 3, 5 및 7회 알칼리 수세하고 압력 및 수세횟수에 따른 고등어 surimi의 품질 특성을 검토하였다. 설계 제작한 감압수세장치는 연속수세가 가능하였으며 양질의 surimi를 효율적으로 제조할 수 있었다. 전체의 수세조건을 통하여 surimi의 수분함량은 $72.0{\sim}72.9%$, 조지방 $4.8{\sim}5.7%$ (수세하지 않은 것, 7.0%), pH $6.9{\sim}7.0$ (수세하지 않은 것, 6.0), VBN $6.9{\sim}7.0\;mg/100\;g$ 및 가압드립 $6.7{\sim}8.3%$이었으며, 단백질 추출성은 560 mmHg, 5회 수세 시 염용성, 수용성 및 기질 단백질추출성이 각각 3,694, 6,036 및 1,424 mg/100 g으로써 각각 가장 높았다. $Mg^{2+}-$ 및 $Ca^{2+}-ATPase$ 활성은 560 mmHg, 5회 수세 시 각각 0.25 및 $0.17\;{\mu}mol\;Pi/min/mg$ actomyosin으로써 가장 높았다. Setting gel의 TGase 활성은 560 mmHg, 5회 수세 시 3.932 nmol/mg이었으며 gel 강도는 setting gel 및 cooked gel에서 각각 420 g cm (상압, 320 g cm) 및 485 g cm (상압, 412 g cm)로써 각각 가장 높았다. 전반적으로 보아 고등어 surimi 제조를 위한 가장 적합한 수세조건은 760 mmHg, 660 mmHg 및 560 mmHg 압력 중 560 mmHg의 감압 하에서 5회 반복 수세하는 것이었다.
수도수(TW)와 0.5% sodium metabisulfite(0.5% SMS)를 대조구로 하고 강산성 전기분해수(SAEW: strong acidic electrolyzed water, pH 2.58, ORP 1,128 mV, HClO 105.0 ppm)와 약알칼리 전기분해수(LAEW: law alkaline electrolyzed water, pH 8.56, ORP 660 mV, HClO 73.8 mV)를 연근의 박피 후 침지액으로 하여 랩포장과 진공포장하여 $5^{\circ}C$ 저장 중 품질 변화를 비교.검토하였다. 0.5% SMS와 전기분해수 침지로 TW 침지 박피 연근에 비해 1 log cyle의 초기 균수 감소 효과와 저장중 상대적으로 낮은 균수를 나타내었다. 침지 처리에 의한 초기 및 저장중 경도의 차이를 나타내었으며 뚜렷한 PPO 활성을 저해하는 효과는 확인할 수 없었다. 전기분해수 침지로 표면색도와 색차는 0.5% SMS 침지와 유사한 억제 효과를 나타내었고 관능적 특성의 변화에서도 TW 침지 박피 연근에 비해 0.5% SMS 침지 박피 연근과 전기분해수 박피 연근이 다소 높게 평가되었다. 포장 방식에 있어서는 랩포장에 비해 진공포장이 품질 유지에보다 효과적이었다.
건강웰빙식품의 하나로서 수요가 증가하고 있는 새싹채소는 미생물학적 관점에서 안전성 확보방안이 요구되고 있다. 따라서 새싹채소의 병원성 미생물을 제거하기 위해 적무종자의 발아 전 침지동안 종자 소독처리에 따른 미생물 제어 효과와 발아율을 평가하였다. L. monocytogenes ATCC 19111가 3~4 log CFU/g의 농도로 접종된 적무 종자는 20,000 ppm calcium hypochlorite, 50 ppm과 100 ppm 염소수, 오존수, 전해산화수 및 전해환원수로 처리하였으며, 증류수에 침지시킨 종자를 대조군과 비교하였다. 발아율은 각 처리군에 대해 48시간 동안 조사하였다. 일반세균과 L. monocytogenes 모두 20,000 ppm calcium hypochlorite에 침지 후 3 logs 이상의 가장 큰 감소효과를 보였고, 염소수, 전해산화수와 전해환원수 침지 후 일반세균과 L. monocytogenes 수는 각각 3 logs와 2 logs 감소되었으며, 염소수와 오존수를 제외하고는 온도변화에 따른 유의적인 차이는 없었다. 종자 발아 후 일반세균 수는 모든 처리군에서, L. monocytogenes 수는 염소수 50 ppm과 오존수를 제외한 모든 처리군에서 대조군보다 유의하게 낮았으며, 염소수 100 ppm을 제외하고는 온도변화에 따른 유의적인 차이는 없었다. 발아율은 48시간 후 calcium hypochlorite(82.3~84.8%)를 제외하고 다른 모든 소독제 처리 시 93.5~97.7%로 나타났으며 대조군과 유의한 차이가 없었다. 이 연구에서 사용된 소독제 처리가 적무 종자에 접종된 L. monocytogenes를 완전히 사멸시키지는 못했지만, 발아 동안 재배수의 소독처리가 추가 병행된다면 생존한 균의 급속한 성장을 지연시킬 수 있을 것으로 사료된다.
This experiment was focused to determine the effect of preharvest treatment of alkaine-reduced water (ARW) and $CaCl_2$ on the storage of oyster mushroom. ARW was prepared through electrolysis of water with $CaCl_2$ (0.1%) as salts and $CaCl_2$ (0.1%) was dissolved in deionized water. Mushrooms were harvested after two sprays of each solutions and quality factors including firmness and browning was determined during storage. The severity of browning was more greater when stored 20 days. Unlike $CaCl_2$, ARW containing 139 ug/L calcium was positive on maintaining freshness in terms of delay of firmness loss and respiration drop, keeping soluble carbohydrate content such as trehalose, and inhibiting the occurrence of browning. Browning, however, seemed to be more influenced by the developmental stage of mushroom at harvest. Even no significant difference was found, the level of ${\beta}$-glucan was low in ARW treatment compared to that of control. Further research including application and preparation methods of ARW is required to clarify the potential of ARW on keeping freshness of oyster mushroom through inhibiting tissue browning.
The optimum dosage of quicklime in producing organic fertilizer using livestock wastes vith a greater than 80% water content was analysed. After one day had elapsed to allow for the organic fentilizer to dry, the quicklime dosage and the composition of the organic fertilizer were analysed. Any from done to the organic fertilizer was also assessed. The amount of the quicklime required to stabilize livestock wastes was determined by water content of livestock wastes. For J farm(slurry style) of which livestock wastes have 94.6% of water concentration, less than 3% of total amount of livestock wastes, for H farm (scraper style) of which livestock wastes have 85% of water concentration, less then 4% of total livestock wastes and Y farm(traditional style) of which livestock wastes have 80% of water concentration, less then 5% of total livestock wastes. Generally, in order to pack the organic fertilizer, water containing quicklime0stabilized livestock wastes should be less than 35%. It takes 9 days to keep this water content for the wastes from H and Y farms(less than 85% in water content), and 12 days for the wastes from J (94.6% in water content). According to the classification standard for compost constitution by Higgins, the crude fertilizers from all 3 farms had high grade $K_2O$ and CaO, the middle grade T-N and middle or low grade $P_2O_5$. Stabilization by quicklime is known to inhibit bacterial decomposition of organic matter and the activity of pathogenic organisms. In this study, more then 99.99% of coliform group, fecal group and viable cell count were reduced. Our results indicate that livestock wastes of greater 80% water content could be used to produce organic fertilizer without the addition of a material for moisture control.
Sodium hypochlorite alkaline was tested against Pseudomonas tolaasii causing bacterial blotch on cultivated oyster mushroom (Pleurotus ostreatus). The minimum inhibitory concentration of sodium hypochlorite against P. tolaasii contained active chlorine (AC) at 1.4 mg/l on plate assay. The highest cultivation yield was obtained from the treatment of AC 5.7 mg/l. Treatment of sodium hypochlorite at the rate of higher than AC 11.4 mg/l resulted in reduced yields at the harvest. However, the population of total bacteria on the bed surface treated with AC 5.7 mg/l of sodium hypochlorite was maintained to some extent. Inhibitory concentration against total bacteria on the bed surface was over AC 22.8 mg/l. Mushroom mycelium was damaged and its growth strongly inhibited at the concentration of AC 200 mg/l. Mushroom caps showed yellowish symptom by chemical injury by treatments of AC 74.1 mg/l or higher. Sporocarps infected by P. tolaasii were irrevocable at any concentration of sodium hypochlorite. Routine watering with AC 5.7 mg/l from mushroom initiation to the end of picking resulted in reduced bacterial blotch incidence of 40% and 86% at two mushroom farms. The treatment resulted in higher quality mushroom production compared to that conventionally watered with tap water alone.
The removal of protozoa in the coagulation process was evaluated under the different pH and turbidity using the jar test after the addition of polyaluminium chloride (PAC) as a coagulant. Two well-known protozoa of Cryptosporidium parvum and Giardia lamblia were tested at the same time with turbidity, the critical water quality parameter of the water treatment process. Both protozoa were removed about 1log (and up to 2log) at the optimum injection of PAC. The source water turbidity and pH affected the removal of protozoa and turbidity. At neutral and alkaline pH, 1.3-1.7log removal of protozoa for low turbid water with 5NTU, and 1.6-2.3log removal for high turbid water with 30NTU were achieved. However, at acidic pH, maximum 0.8-1.0log and 1.1-1.2log were removed for low and high turbid water, respectively, at the optimum PAC injection of 15mg/L. The relation of protozoa and turbidity removals were expressed as the 1st order equation (significantly positive relation) in the most of the tested conditions. In addition, the relation of protozoan removals with residual turbidity were also expressed the 1st order equation (significantly negative relation), although the significance of the equations were reduced at acidic pH. Therefore, residual turbidity could be a good index of efficient protozoan removal in the coagulation process, probably except at the low pH condition.
Wet Scrubber reacts the incoming pollutant gas with cleaning water (water + absorbent) to absorb pollutants and release the clean air to the atmosphere. Wet scrubbers and packed tower scrubbers using this principle are widely used in businesses that emit acid gases. In particular, in the etching process using hydrochloric acid (HCl), alkaline washing water (NaOH) having a pH of about 8 to 11 is used to absorb a large amount of acid gas. However, These salts are attached to the injection nozzle (nozzle), filling material (packing), and the demister (Demister), causing air pollution, human damage, and inoperability due to clogging and acid gas discharge. Therefore, In this study, an improvement plan was proposed to manage the washing water with pH 3~4 acidic washing water. The test method takes samples from the Wet Scrubber flue measurement laboratory twice a month for 1 year. Hydrogen chloride (HCl) concentration (ppm) was measured, and nozzle clogging and scale conditions were measured, compared, and analyzed through a differential pressure gauge and a pressure gauge. As a result of the check, it was visually confirmed that the scale was reduced to 50% or less in the spray nozzle, filler, and demister. In addition, the emission limit of hydrogen chloride in accordance with the Enforcement Regulation of the Air Quality Conservation Act [Annex 8] met 3 ppm or less.
