  • The current definition of Arismatis Rhizoma is the tuberous root of Alisma orientale Juzepzuk in the Korean Pharmacopoeia, but there is still no marker compound. So it has difficulties in quality control. Therefore, in this study, we have established a method to analyze alisol B and alisol B acetate using HPLC as a marker compounds of Arismatis Rhizoma. As the result of the analysis, alisol B content was ranged from 0.02% to 0.50% and alisol B acetate content was ranged from 0.12% to 0.25% in 40 samples. The stability of alisol B and alisol B acetate was investigated during 24 months. As a result, alisol B acetate was significantly decreased. The marker compound of Arismatis Rhizoma is alisol B acetate and the content of 0.05% or more is suitable by applying the stability results.

  • Near infrared spectroscopy (NIRS) combined with multivariate analysis was used to discriminate the geographical origin of Alisma orientale from Korea (n=94) and China (n=72). Two-thirds of samples were selected randomly for the training set, and one-third of samples for the test set. Second derivative was used for the pretreatment of NIR spectra. Partial least square discriminant analysis (PLS-DA) models correctly discriminated 100% of the Korean and Chinese A. orientale samples. These results demonstrate the potential use of NIR spectroscopy combined with multivariate analysis as a rapid and accurate method to discriminate A. orientale according to their geographical origin.

  • Alismatis Rhizoma is an oriental medicine originated from the rhizome of Alisma orientale or Alisma plangtago-aquatica var. orientale (Alismataceae). As an standard compound of this plant, alisol B 23-acetate was isolated from the dichloromethane fraction of Alisma orientale and identified by the spectroscopic evidences. A Quantitative analysis of alisol B 23-acetate using HPLC method showed that the average content was 0.47$\pm$0.11% in 33 samples throughout the various regions of Korea.

  • Objectives: The aim of the study was to evaluate the anti-asthmatic effect of alismatis rhizoma and alisol acetate B combination therapy in a murine asthma model. Methods: C57BL/6 mice were sensitized to and challenged with a mixture of ragweed, dust mite, and aspergillus to induce an asthma animal model. Alismatis rhizoma extract and alisol acetate B combination therapy was co-administered only in the experimental group. To evaluate the anti-asthmatic effect of the combination therapy, inflammatory cell counts in bronchoalveolar lavage (BAL) fluid were determined, and tissue was examined histologically with hematoxylin and eosin (H & E) and periodic acid-Schiff (PAS) stains, by enzyme-linked immunosorbent assay (ELISA) of IgE, IL-4, and IL-5, and with reverse transcription polymerase chain reaction (RT-PCR) of IL-5, IL-33, MUC5AC. Results: Alismatis rhizoma and alisol acetate B combination therapy reduced the number of inflammatory cells, alleviated histologic features, and down-regulated all the investigated asthma mediators, IgE, IL-4, IL-5, IL-33, and MUC5AC. Conclusions: According to the above results, alismatis rhizoma and alisol acetate B combination therapy may have therapeutic potential for asthma.

  • Alismatis Rhizoma is originated from the rhizome of Alisma plantago-aquatica L. var. orientale Samuelson or A. canaliculatum A. Br. et Bouche (Alismataceae). Prostane-type triterpenes, guaiane-type sesquiterpenes, and kaurane-type diterpenes have been reported as the main canstituents from these plants. Four prostane-type triterpenes. alisol B 23-acetate (1). alisol C 23-acetate (2), alisol B (3). and alisol A 24-actate (4). were isolated from the EtOAc-soluble fraction of this dried rhizome. • Lectin was purified by using $(NH_4)_2SO_4$, DEAE-cellulose ion-exchange chromatography and Sephadex G-150 column chromatography from Alismatis Rhizoma(AR). The specific activity of AR lectin was 50, 441units/mg, and purification folds were 114. The AR lectin agglutinated human erythrocytes of all types(A, B, O, AB). The molecular weight of AR lectin was estimated about 90, 500 daltons by gel filtration and each subunits were 42,000, 27,000 and 22,500 daltons on SDS-PAGE respectively. The hemagglutinating activity of the lectin was inhibited by sialic acid, glucose, ribose, galactose, sucrose, and lactose. It was also inhibited by cations such as $Hg^{++},\;Fe^{++},\;Cu^{++}\;and\;Pb^{++}$.

  • This experiment was conducted for the production of alismatis rhizoma (Alisma plantago-aquatica var. orientale Samuelsson) with high root tuber yield and alisol-B monoacetate content by treatment of different fertilizers levels. The results obtained were as follows: Shoot growth tended to increase as the fertilizers levels increased. Root tuber yield increased by 25% and 20% with 20-10-20 and 30-15-30 treatment, respectively. Alisol-B monoacetate contents increased as fertilizers levels increased, being the highest in 30-15-30 treatment.

  • Alismatis rhizoma (AR), the dried rhizome of Alisma orientale (Sam.) Juzep, is a well-known, traditional medicine that is used for the various biological activities including as a diuretic, to lower cholesterol and as an anti-inflammatory agent. The present study was carried out to investigate the potential toxicity of the Alismatis rhizoma aqueous extract (ARAE) following 90-day repeated oral administration to Sprague-Dawley rats. ARAE was administered orally to male and female rats for 90 days at 0 (control), 500, 1,000 and 2,000 mg/kg/day (n = 10 for male and female rats for each dose). Additional recovery groups from the control group and high dose group were observed for a 28-day recovery period. Chromatograms of ARAE detected main compounds with four peaks. Treatment-related effects including an increase in the red blood cells, hemoglobin, hematocrit, albumin, total protein, and urine volume were observed in males of the 2,000 mg/kg/day group (p < 0.05). However, the diuretic effect of ARAE was considered, a major cause of hematological and serum biochemical changes. The oral no-observed-adverse-effect level (NOAEL) of the ARAE was > 2,000 mg/kg/day in both genders, and no target organs were identified.

  • Traditional Korean medicines may be managed more scientifically, through the development of logical criterion to verify their cultivation region. It contributes to advance the industry of traditional herbal medicines. Volatile compounds were obtained from 14 samples of domestic Taeksa and 30 samples of Chinese Taeksa by steam distillation. The metabolites were identified by NIST mass spectral library in the obtained gas chromatography/mass spectrometer (GC/MS) data of 35 training samples. The multivariate statistical analysis, such as Principal Component Analysis (PCA), Partial Least Squares Discriminant Analysis (PLS-DA), and Orthogonal Partial Least Squares Discriminant Analysis (OPLS-DA), were performed based on the qualitative and quantitative data. Finally trans-(2,3-diphenylcyclopropyl)methyl phenyl sulfoxide (47.265 min), 1,2,3,4-tetrahydro-1-phenyl-naphthalene (47.781 min), spiro[4-oxatricyclo[5.3.0.0.(2,6)]decan-3-one-5,2'-cyclohexane] (54.62 min), 6-[7-nitrobenzofurazan-4-yl]amino-morphinan-4,5-epoxy (54.86 min), p-hydroxynorephedrine (55.14 min) were determined as marker metabolites to verify candidates for the origin of Taeksa. The statistical model was well established to determine the origin of Taeksa. The cultivation areas of test samples, each 3 domestic and 6 Chinese Taeksa were predicted by the established OPLS-DA model and it was confirmed that all 9 samples were precisely classified.
