Nutrient losses, especially nitrogen and phosphorus, in agricultural runoff can contaminate surface and ground water, leading to eutrophication. Thus, erosion control is crucial to minimizing nutrient losses from agricultural land. Assessments of various erosion control practices were carried out under various cropping system, soil management practices, and slope conditions by means of a lysimeter study and under artificial rainfall. Soil and nutrient losses were monitored in a small agricultural field to evaluate the soil conservation practices. Nutrient losses occur in runoff and leachate (dissolved nutrient) and in sediments (particulate nutrient). Dissolved nitrates accounted for the majority (about 90%) of nitrate transport within the soil. Particulate phosphate in sediments represented the majority (60% to 67%) of phosphate transport. Recently, engineering and agronomic erosion-control practices haver been used to reduce erosion problems in fields on slopes. These practices reduced soil loss, runoff, and nutrient loss to 1/6, 1/2,and 1/3 their original levels, respectively. Bioavailable particulate phosphate in sediments represents a variable but longterm source of phosphate for algae. Dissolved nitrate and phosphate are immediately available for algal uptake, so reducing fluxes of these nutrients should also reduce the risk of eutrophication.
The automatic water quality monitoring system (AWQMS) have been installed to immediately response to any pollution incident. It also make it possible to conduct the task efficiently regarding water quality control. The purpose of this study is to enhance reliability by securing accuracy of automatic water quality data through quality assessment (QA) for temperature, pH, dissolved oxygen (DO), electric conductivity (EC), total organic carbon (TOC). The result of comparison between manual and automatic data, relative accuracy of general items (temperature, pH, EC, DO) and TOC were mostly satisfied with guideline (i.e. less than 20%). On the other hand, relative accuracy of DO between sampling site and housing site was somewhat against the guideline. The contamination by attaching algae and microorganism in the pipeline is considered as main cause. After backwashing the pipeline, DO concentration was increased up to 53%. Therefore, pipeline management is recognizable as important thing to secure reliability of water quality data.
To optimize the natural chemical agents against nuisance phytoplankton, we examined algal removal activity (ABA) of Plant-Mineral Composite (PMC), which already developed by our teams (Kim et al., 2010), on various conditions. The PMC are consisted of extracted-mixtures with indigenous plants (Camellia sinensis, Quercusacutissima and Castanea crenata) and minerals (Loess, Quartz porphyry, and natural zeolite), and characterized by coagulation and floating of low-density suspended solids. A simple extraction process was adopted, such as drying and grinding of raw material, water-extraction by high temperature-sonication and filtering. All tests were performed in 3 L plastic chambers varying conditions; six different concentrations ($0{\sim}1.0\;mL\;L^{-1}$), six light intensities ($8{\sim}1,400\;{\mu}mol\;m^{-2}s^{-1}$), three temperatures ($10{\sim}30^{\circ}C$), four pHs (7~10), five water depths (10~50 cm), and three different waters dominated by cyanobacteria, diatom, and green algae, respectively. Results indicate that the highest ABA of PMC was seen at $0.05\;mL\;L^{-1}$ in treatment concentrations, where showed a reduction of more than 80% of control phytoplankton biomass, while $1,400\;{\mu}mol\;m^{-2}s^{-1}$ in light intensity (>90%), $20{\sim}30^{\circ}C$ temperature (>60%), 7~9 in pH (>90%), below 50 cm in water depth (>90%), and cyanobacterial dominating waters (>80%), respectively. Over the test, ABA of PMC were more obvious on the algal biomass (chlorophyll-${\alpha}$) than suspended solids, suggesting a selectivity of PMC to particle size or natures. These results suggest that PMC agents can play an important role as natural agents to remove the nuisant algal aggregates or seston of eutrophic lake, where occur cyanobacterial bloom in a shallow shore of lake during warm season.
A family of calcium-dependent protein kinases (CDPKs) is a unique enzyme which plays crucial roles in intracellular calcium signaling in plants, algae, and protozoa. CDPKs of malaria parasites are known to be key regulators for stage-specific cellular responses to calcium, a widespread secondary messenger that controls the progression of the parasite. In our study, we identified a gene encoding Plasmodium vivax CDPK4 (PvCDPK4) and characterized its molecular property and cellular localization. PvCDPK4 was a typical CDPK which had well-conserved N-terminal kinase domain and C-terminal calmodulin-like structure with 4-EF hand motifs for calcium-binding. The recombinant protein of EF hand domain of PvCDPK4 was expressed in Echerichia coli and a 34 kDa product was obtained. Immunofluorescence assay by confocal laser microscopy revealed that the protein was expressed at the mature schizont of P. vivax. The expression of PvCDPK4-EF in schizont suggests that it may participate in the proliferation or egress process in the life cycle of this parasite.
In this study, we investigate the antimicrobial and antioxidant activities of extracts of Enteromorpha intestinalis. To evaluate the antimicrobial and antioxidant activities, three solvents (hexane, chloroform and methanol) were applied to obtain extracts. The extraction yields are hexane (A) 1.11%, chloroform (B) 0.94%, and methanol (C) 8.2%. Also, the contents of total phenolic compounds of extract A, B, C are 4.03%, 8.15%, and 2.33%, respectively. In the results of antimicrobial activity, extract A and B have higher activity than that of extract C. Especially, Vibrio vulificus, Pseudomonas aeruginosa, Bacillus subtilis, Escherichia coli are more sensitive than others. In antioxidant activity, extract A and B present the higher DPPH activity than that of control (BHA and ascorbic acid). The DPPH radical scavenging activity and SOD-like activity are ordered as B>C>A. However, extract C show high value in ferric reducing antioxidant power assay. In nitrite scavenging activity, extract A is relatively higher than others. However, the antioxidant activities are lower than that of controls (ascorbic acid and BHA). The antioxidant activities are presented the increasing pattern of increasing by the increasing of extract concentration.

Recently, algae has been considered as a potential anti-inflammatory source due to its distinctive habitat environment exposing to light and high oxygen concentration. In present study, anti-inflammatory effect of brown alga, Sargassum fullvellum ethanol extract (SFEE), was examined. SFEE inhibited not only the production of nitric oxide and pro-inflammatory cytokines (IL-6, IL-$1{\beta}$, TNF-${\alpha}$) but also the expression of inducible nitric oxide synthase and cyclooxygenase 2 in LPS-induced RAW 264.7 cells without affecting cell viability. SFEE also suppressed the expression of nuclear factor kappa B (NF-${\kappa}B$), suggesting that SFEE could affect the expression of inflammation related cytokines and proteins through the regulation of NF-${\kappa}B$. Furthermore, formation of edema of the ear was 40% lower in mice treated with the highest dose (250 mg/kg) of SFEE than in the control mice. Thus, our study showed that SFEE may be a potential therapeutic anti-inflammatory drug.
Control of Sediment is very important in prawn farm due to the eruption of toxic material such as W1ionized H2S, NH3 and NO2-. In this study, column test study, column with filter media such as activated carbon, zeolite, oyster shell and iron chloride to evaluate the reduction of toxicity from sediment ammonia-N(NH3) was effectively removed by Zeolite and oyster shell. It was indicated that ammonium ion(NH4+) was removed by ion exchange of zeolite. And the ammonia in the column of oyster shell was existed as the form of NH4+, which is not toxic for prawn because oyster shell was stably kept around pH 8. Therefore, some of ammonia(NH3) was reduced by oyster shell. Hydrogen sulfide and COD were effectively removed by adsorption of activated carbon and a partial removal of hydrogen sulfide was accomplished by Oyster shell. Phosphorous was removed by activated carbon, oyster shell and iron chloride. In prawn farm, the concentration of ammonia was increased with increase of pH by algae photosynthesis in the column of activated carbon, zeolite and iron chloride, but it was revealed that pH was stably kept in the column of oyster shell.
In 2015, white cottony tufts were observed on gametophytes of the kelp Saccharina japonica. Wet mount and histopathology examination revealed numerous fungal hyphae and mycelium around the gametophytes. The gametophytes surrounded by fungal hyphae were generally round and empty. A specific 610-bp fragment of the internal transcribed spacer (ITS)-5.8S rDNA-ITS gene of fungi was amplified by polymerase chain reaction and the nucleotide sequence showed 100% identity with those of Acremonium sclerotigenum, Acremonium sp. and Ascomycota sp. When fungus-infected gametophytes were mixed with healthy gametophytes, a high transmission rate (100%) resulted. This is the first report of mycosis of gametophytes in Korea.
Seaweeds produce antioxidants to counteract environmental stresses, and these antioxidant genes are regarded as important defense strategies for marine algae. In this study, the expression of Pyropia yezoensis (Bangiales, Rhodophyta) ascorbate peroxidase (PyAPX) and manganese-superoxide dismutase (PyMnSOD) was examined by qRT-PCR in P. yezoensis blades under abiotic stress conditions. Furthermore, the functional relevance of these genes was explored by overexpressing them in Chlamydomonas. A comparison of the different expression levels of PyAPX and PyMnSOD after exposure to each stress revealed that both genes were induced by high salt and UVB exposure, being increased approximately 3-fold after 12 h. The expression of the PyAPX and PyMnSOD genes also increased following exposure to $H_2O_2$. When these two genes were overexpressed in Chlamydomonas, the cells had a higher growth rate than control cells under conditions of hydrogen peroxide-induced oxidative stress, increased salinity, and UV exposure. These data suggest that Chlamydomonas is a suitable model for studying the function of stress genes, and that PyAPX and PyMnSOD genes are involved in the adaptation and defense against stresses that alter metabolism.
Scenedesmus spp. were cultured for 51 days in newly developed medium, KEP I (Kim and Ecopeace: initials of corresponding author and environmental company) made with Bacterio-Mineral-Water (3%, v/v) that had been bio-reacted with swine urine medium to 10% (v/v) Bold's Basal medium, and investigated for cancer chemopreventive potential by measuring the induction of quinone reductase (QR), glutathione S-transferase (GST), and reduced glutathione (GSH), and inhibition of cytochrome P450 (CYP) 1A1 activity. The activitives of QR and GST of Scenedesmus spp. cultured in KEP I medium were increased by 3.0-fold and 1.5-fold, respectively. However, Scenedesmus spp. cultured in control medium (CT) increased the activitives of QR and GST by 1.8-fold and 1.3-fold, respectively. Scenedesmus spp. in KEP I medium strongly inhibited CYP 1Al activity. These results show that Scenedesmus spp. in KEP I medium has cancer chemopreventive potential and may be a candidate for further development as a chemopreventive agent.
