Since toxic Alexandrium catenella and non-toxic A. fraterculus are morphologically similar, they are difficult to discriminate under the light microscope. However, a novel technology, such as fluorescein isothiocyanate (FITC)-conjugated lectin probes enables easy and rapid differentiation. Toxic A. catenella bound seven different lectins, whereas the non-toxic A. fratercuzus did not bind Arachis hypogaea (PNA) lectin. In addition, Pseudo-nitrschia species in this study were also difficult to identify to species level with light microscope techniques, but it was possible to classify them using fluorescent lectins. Pseudo-nitzschia multistriata, P. subfraudulenta and P. pungens bound Canavalia ensiformis (ConA), whereas P. subpaclfica did not, and P. pungens also bound Ricinus communis (RCA). These results imply that lectin could be used as a critical tool in the differentiation of P. multistriata, P. subfraudulenta and P. pungens. However, P. subpacifica was not differentiated by the lectins tested. Therefore, it isconcluded that lectin probes are useful for discriminating toxic A. catenella from non-toxic A. fraterculus, and for the identification of some Pseudo-nitzschia species. In addition, this method has a great potential to speed and detection between non-toxic and toxic harmful algal blooms (HABs) in Korean biotoxin monitoring systems.
본 연구에서는 와편모조류 Alexandrium affine(LIMS-PS-2345)의 생장에 미치는 용존태 무기 및 유기 영양염의 영향을 조사하였다. 영양염 흡수 동력학 실험에서 A. affine의 최대흡수속도(ρmax)와 반포화상수(Ks)는 질산염에서 77.0 pmol/cell/hr과 17.6 μM, 인산염에서 15.5 pmol/cell/hr과 3.88 μM로 산출되어, 무기영양염에 대하여 높은 요구량 및 낮은 친화성을 가지고 있는 것으로 나타났다. 유기 영양염에 따른 A. affine의 생장속도를 확인한 결과, 유기 질소 urea, glycine와 유기 인 adenosine triphosphate(ATP), glycerol phosphate(Glycerol-P) 첨가구에서 무기 영양염 첨가구의 70 % 이상 생장속도를 보였다. 따라서 낮은 무기 영양염 환경에서 A. affine의 우점화와 종간경쟁에서 우위를 위해서는 용존태 유기 영양염의 이용이 필요할 것으로 생각된다.
1993년 진해만 일원의 마비성패독 발생의 원인규명을 위한 모니터링의 일환으로 원인생물의 독생산과 성분조성을 조사하기 위하여 양식장 인근해역의 저서 휴면포자를 발아시켜 분리한 무균주의 독성분을 분석하였다. 분리된 전체주중에서 수정리산 (St. 1) 5주, 욱곡리산 (St. 2) 3주 및 대곡리산 (St. 4) 11주의 독조성 및 독함량을 비교하였을 때, 각 지점별 평균 독량은 약 54-70 fmol/cell 높은 량을 나타내었고, 동일 휴면포자의 clone 분리주 뿐만 아니라 전체 분리주에서 개체별 독함량의 차이가 크게 나타났다. 독조성은 C1/C2 (epiGTX8/GTX8), GTX1/GTX4 및 neoSTX가 주요 구성성분을 이루었고, GTX2/GTX3, GTX5, C4, dcSTX 및 STX성분은 미량 또는 산발적으로 출현하였다. 주요 성분중에서 neoSTX는 $5-54mol\%$로 변동이 컸으나, 전체 분리주의 절반은 출현을 보이지 않아 이 지역에서 조성이 다른 개체군의 출현이 시사되었다 한편, 상대적으로 독성이 강한 GTX1-4 및 neoSTX와 같은 Carbamate군의 성분이 세 정점에서 각각 $57\%,\;54\%$ 및 $67\%$의 높은 평균치를 나타내어 이 지역에서의 높은 독화율과 독화 가능성의 잠재력이 큰 것으로 시사되었다.
한국 남해연안해역에서 분리한 유독와편모조류 $Alexandrium$$tamarense$와 $Alexandrium$$catenella$의 성장에 영향을 미치는 수온과 염분의 특성을 실내 배양실험을 통해 살펴보았다. 수온은 10, 15, 20, 25, $30^{\circ}C$의 5단계, 염분은 10, 15, 20, 25, 30, 35 psu의 6단계를 조합한 총 30단계의 조건에서 성장속도 변화를 관찰하였다. 최대성장속도를 보이는 수온과 염분은 $A.$$tamarense$에서 $15^{\circ}C$와 30 psu, $A.$$catenella$에서 $25^{\circ}C$와 30 psu로 나타났다. 또한 최적성장조건은 $A.$$tamarense$에서 $10-20^{\circ}C$, 25-35 psu이고, $A.$$catenella$에서 $20-30^{\circ}C$, 25-35 psu였다. 얻어진 수온과 염분의 조건을 이용하여 중회귀분석에 의해 예측 모델식을 계산한 결과, $A.$$tamarense$는 ${\mu}=0.04+0.0193T-0.0339S-0.0005T^2+0.0021S^2+0.00073TS-0.000022T^3-0.000038S^3+0.00000086TS^2-0.0000255T^2S$이며, $A.$$catenella$는 ${\mu}=1.01-0.1288T-0.0778S+0.0067T^2+0.0038S^2+0.00204TS-0.0001T^3-0.000059S^3-0.0000131TS^2-0.0000392T^2S$로 나타났으며, 예측값과 실험값 사이에는 높은 상관관계를 보였다. 본 연구결과는 수온과 염분의 현장관측 값을 이용하여 성장속도를 추정할 수 있기 때문에, $A.$$tamarense$와 $A.$$catenella$의 개체군 확대를 이해하는데 도움이 될것으로 기대된다.
와편모조류 Alexandrium affine의 생장에 대한 수온, 염분 및 광 조건 실내실험을 통해서 살펴보았다. A. affine의 최대성장속도($0.69day^{-1}$)는 수온 $25^{\circ}C$, 염분 25 psu로 나타났으며, 최적생장(최대성장속도의 80 %)은 수온 $20{\sim}25^{\circ}C$의 염분은 20 ~ 35 psu로 나타났다. 따라서 A. affine는 비교적 협온성이며, 광염성의 생리학적 특성을 가지고 있는 것으로 생각된다. 광량에 따른 생장 실험결과에서 유도된 생장식은 ${\mu}=0.75(I-4.25)/(I+65.47)$로, 보상광량($I_c$)은 $4.25{\mu}mol\;m^{-2}s^{-1}$, 반포화광량($K_I$)은 $57.0{\mu}mol\;m^{-2}s^{-1}$로 나타났다. 결론적으로 A. affine는 하계에 낮은 광, 염분의 변동 폭이 큰 연안해역에서도 우점화 할 수 있는 유리한 생리적 특성을 보이는 것으로 판단된다.
The marine dinoflagellate genus Alexandrium, which produces PSP toxins, has a global distribution. As human-assisted dispersal of the species has been suggested, it is important to develop molecular tools to trace the dispersal pathway. To screen population-specific DNA sequences that differentiate Korean and Japanese A. catenella, we targeted the area downstream of the chloroplast psbA gene using PCR with population-specific DNA primers followed by RFLP (restriction fragment length polymorphism) analysis and sequencing. The RFLP patterns of the PCR products divided Korean and Japanese A. catenella regional isolates into three types: Korean, Japanese, and type CMC3, isolated from Korea. We sequenced the PCR products, but found no similar gene in a homology search. The molecular phylogeny inferred from the sequences separated the Korean and Japanese A. catenella strains, as did the RFLP patterns. However, the Japanese isolates included two slightly different sequences (types J and K), while the Korean sequence was the same as the Japanese K type. In addition, a unique sequence was found in the Korean strains CMC2 and CMC3. Population-specific PCR amplification with Japanese A. catenella type-specific PCR primers designed from the type J sequence yielded PCR products for Japanese strains only, showing that the unknown gene can be used for a population analysis of Korean and Japanese A. catenella.

Monospecific red tide by a toxic dinoflagellate belonging to the genus Alexandrium occurred at Chindong Bay in the southern coast of Korea and continued from April 6th to 15th in 1997. The ratio of its cell number to total phytoplankton cell number was much higher than $95\%$. This organism was identified as Alexandrium tamarense, although slight morphological differences were found comparing to the original and successive descriptions of the species. We found neither anterior nor posterior attachment pores in these cells of the bloom population. The occurrence of red tide caused by A. tamarense was first reported in Korea. Its plate formula is Po, Pc, 4', 6"c, 8s, 5"' and 2"". Thecal plates are thin with pore-like ornamentation. In those plates, the anterior part of the first apical plate (1') is narrower and its posterior end has sometimes a block-like accessory, but this variation was considered within the range of the morphological variability of this taxon. The cell density during the red tide exhibited a wide range of variation by the depth of water column, ranging from $2\times10^6$ cells$l^{-1}$ to $5\times10^6$ cells·$l^{-1}$. Water temperature varied from 11.8 to $12.3^{\circ}C$. Toxicity of A. tamarense during red tide was measured as $8.8\times10^5$. $MU\;\cdot\;cell^{-1}$ by mouse bioassay.
국내 남해안의 적조발생지역에서 채집한 Prorocentrum minimum, P. micans, P. triestinum, P. balticum, Gymnodinium sanguineum, Alexandrium catenella, Scrippsiella trochoidea, Heterosigma akashiwo를 순수배양하고 이로부터 PCR을 이용해 약 700 bp에 해당되는 24S rRNA 유전자의 D1과 D2 변이부위를 증폭하고 클로닝한 후 염기서열을 분석하였다. 분석된 염기서열들을 이미 밝혀진 적조원인종들의 것과 ClustalW 프로그램을 사용하여 배열하고 계통수를 구성한 결과 Prorocentrum과 Alexandrium 속의 것들은 형태적인 분류결과와 종수준까지 거의 일치함을 보여주었다. 이러한 연구결과는 그 24S rRNA 유전자의 염기서열분석이 국내에서 발생하는 적조원인종들에 대한 종 수준에서의 신속한 확인에 이용될 수 있음을 제시한다.
휴안포자를 형성하는 과편모조류중 우리나라 남해동부와 동해남부의 연안수역 특히 진해만에서 1982년 이래 3월부터 9월에 걸쳐 적조를 일으킨 종은 8종이었으며 이들은 6종의 과편모조류와 2종의 녹색편모조류로 구성되었다. 계절별로는 이른 봄철에 Scrippsiella trochoidea와 Heterocapsa triquetra종이, 늦봄에 Heterosigma akashiwo가 그리고 여름철에는 Gyrodinium instriatum과 Pheopolykrikos hartmannii가 적조를 일으켰다. 한편 Cochlodinium sp.와 Alexandrium affine는 늦여름부터 가을철에 적조를 일으켰다. 이중 Heterosigma akashiwo는 매년 같은 곳에서 같은 시기에 발생하는 경향을 뚜렷히 나타냈으며 Cochlodinium sp.에서도 이와 같은 특성을 발견할 수 있었다. 한편 적조발생시의 밀도는 $10^3\~10^5\;cells/ml$ 수준이었으며, 밀도변화는 환경특성 보다는 오히려 세포의 크기에 따라 변화하고 있었다.
알렉산드륨 적조생물의 리보소옴 알엔에이 유전자의 ITS1, 2 및 5.8S부위를 대상으로 종간 혹은 종내의 유전적 다양도를 조사하기 위하여 지리적으로 격리된 33 스트레인 유전자의 염기서열를 비교했다. 진해만에서 분리된 AT-2, AT-6, AT-10, AT-A, AT-B 5클론은 일본종 OFX151-A과 동일한 유전자임을 발견했다. ITS부위에서 가장 짧은 종은 A. margalefi로 481 bp이며 가장 긴 종은 A. affine으로 528 bp로 나타났다. ITS1과 ITS2 염기서열에 대한 상호관계는 역으로 나타낸 반면에, G+C 함량에 대한 상호관계는 플러스로 나타났다. 유전적 변이율은 0.3% (1 bp)에서 53% (305 bp)였다. A. tamarense과 가장 적게 유전적 변이율을 보인 종은 A. fundyense(1.2-2.3% = 6-12 bp)인 반면에, A. catenella와는 큰 변이율 (19.8% = 102 bp)을 보였고, A. catenella와 A. fundyense은 19.7% 상이하였다. 알렉산드륨 적조생물의 bootstrap은 약하게 지지되는 데도 불구하고, A. catenella 분리종은 독립적인 그룹으로 형성하여 상호간에는 강력한 bootstrap 값은 PAUP과 NJ 분석에서 보였다. A. cohorticula와 A. frateculus 적조생물은 항상 sub-group 내에서 높은 bootstrap을 가졌다. 결론적으로 ITS부위의 염기서열 분석은 알렉산드륨 적조생물의 집단내 혹은 집단간의 계통분류을 밝히는데 유용한 것으로 보였다.
