패독발생의 원인규명을 위한 모니터링의 일환으로 원인생물의 출현과 휴면포자의 발아에 의한 현장 개체군의 유입가능성을 조사하기 위하여 1993년 9월부터 진해만을 중심으로 Alexandrium속의 발생현황, 원인생물의 잠재력을 제공하는 저질중의 휴면포자의 분포 및 실내 발아율을 조사하였다. 만내의 조사된 모든 정점에서 1월부터 Alexandrium속이 출현하여 대곡리 정점(칠천도)에서 3월에 533 개체/l의 최고치를 보였으며, 다른 지점에서는 100개체 이하를 나타냈으나 수온상승기인 4월에는 출현량이 감소하였다. 저질중의 Alexandrium속 휴면포자의 분포밀도는 채집한 정점간의 차이가 많아 100-800 개체/$cm^3$의 밀도를 보였으나, 채집지역의 저질상부층$(0\~4cm)$에서 $700\~800$ 개체/$cm^3$의 분포 지점이 많았다 휴면포자의 발아율은 수정리 정점에서 1월$(72.5\%)$, 4월$(68.0\%)$, 7월$(44\%)$ 및 10월$(9.0\%)$ 순으로 채집 계절에 따른 차이를 나타냈으며, 1월 채집한 만내의 다른 2 정점의 휴면포자도 $71\~97|%$의 높은 발아율을 보였다. 한편, 수심이 얕은 다대포 지점의 휴면포자는 6월$(86.5\%)$과 10월$(70.0\%)$에도 높은 발아율을 보여 진해만의 각 지점과는 대조적인 결과를 보였다. 이상의 결과로부터 Alexandrium속 휴면포자의 발아는 지역환경에 따라 차이를 보일 수 있으며 비교적 수심이 깊은 지역에서는 온도 등의 환경요인의 변동 외에 내재적 발아리듬의 영향이 클 것으로 추정되었다. 따라서 진해만에서는 저수온기일지라도 휴면포자의 발아에 의한 Alexandrium속의 출현이 놀게 나타나며, 이 지역의 패독발생의 원인생물로 기여할 가능성이 높은 것으로 시사되었다.
적조는 연안 해역에서 양식어류를 폐사 시키는 등 많은 경제적 피해를 입혀왔다. 이러한 적조의 광역 분포를 효과적으로 파악하기 위하여 원격탐사가 활용되어 왔는데, 적조 해수가 가지는 흡광 특성은 원격탐사에 중요한 요소인 해수반사도를 이해함에 있어서 핵심적인 역할을 한다. 본 연구에서는 2017년 8월 통영 인근 해역에서 출현한 Alexandrium affine 종이 우점한 해수에 대한 흡광 특성을 분석하였다. 선박 관측을 통하여 총 20개 정점에서 획득한 시료에서 색소 흡광, 부유물 흡광, 용존유기물 흡광을 측정하였고, 이를 수질 변수인 엽록소 농도, 부유물 농도와 관련하여 분석하였다. 분석 결과, 규조류 우점 해수와는 다르게 Alexandrium 우점 시료는 400 nm 이하에서 강한 흡광을 나타내었고, 또한 400 nm - 500 nm 에서 굴곡이 크게 나타남을 확인하였다. 색소 흡광의 집적 효과 또한 규조류와 다르게 나타남을 확인하였고, 부유물 및 용존유기물 흡광 모델 (adg(λ)=adg(λ0)e-s(λ-λ0))에서의 지수부 계수(s) 역시 기존 규조류에서 일반적으로 사용하는 0.015와는 달리 0.01이 더 적합함을 보였다.
New PCR primers (N=18) were designed for the isolation of complete SSU to LSU rDNA sequences from the dinoflagellate Alexandrium tamarense. Standard PCR, employing each primer set selected for amplifications of less than 1.5 kb, successfully amplified the expected rDNA regions of A. tamarense (Korean isolate, HY970328M). Complete SSU, LSU rDNAs and ITS sequences, including 5.8S rDNA, were recorded at 1,800 bp, 520 bp and 3,393 bp, respectively. The LSU rDNA sequence was the first report in Alexandrium genus. No intron was found in the LSU rRNA coding region. Twelve D-domains within the LSU rDNA were put together into 1,879 bp (44.4% G+C), and cores into 1514 bp (42.8% G+C). The core sequence was significantly different (0.0867 of genetic distance, 91% sequence similarity) in comparison with Prorocentrum micans (GenBank access. no. X16108). The D2 region was the longest in length (300 bp) and highly variable among the 12 D-domains. In a phylogenetic analysis using complete LSU rDNA sequences of a variety of phytoplankton, A. tamarense was clearly separated with high resolution against other species. The result suggests that the sequence may resolve the taxonomic ambiguities of Alexandrium genus, particularly of the tamarensis complex.
진해만에서 분리한 무독성 Alexandrium tamarense와 이미 보고된 독성을 가진 A. tamarense종 간의 접속공, 제1상판, 후속공의 형태를 상호 비교하였다. 형태적으로 독성종과 거의 일치되지만, HPLC나 생체실험에서 진해만에서 새롭게 분리한 종은 독성이 전혀 나타나지 않았다. lectin반응결과 무독종 A. tamarense은 PNA와 강한 반응을 보여, 세포표면에 lactose나 galactose와 같은 물질이 많이 분포하고 있음을 보였다. 또한, 단백질 전기영동시 무독종은 유독종과 달리 21 kDa 정도 부위에서 종 특이적 밴드를 보였다. 따라서 PNA lectin이나 면역학적 방법을 이용하면 무독종 A. famarense을 신속하게 동정하는데 이용될 수 있다.

The amount of CO2 absorbed by the oceans continues to rise, resulting in further acidification, altering some functional traits of phytoplankton. To understand the effect of elevated partial pressures of CO2 (pCO2) on functional traits of dinoflagellates Alexandrium affine and A. pacificum, the cardinal temperatures and chain formation extent were examined under two pCO2 (400 and 1,000 µatm) over the range of temperature expected to be associated with growth. The growth rate and chain formation extent of A. affine increased with higher pCO2, showing significant changes in cardinal temperatures and a substantial increase in middle chain-length (4-8 cells) fractionation under elevated pCO2 condition. By contrast, there were no significant differences in specific growth rate and any chain-length fractionation of A. pacificum between ambient and elevated pCO2 conditions. The observed interspecies variation in the functional traits may reflect differences in ability of species to respond to environmental change with plasticity. Moreover, it allows us to understand the shifting biogeography of marine phytoplankton and predict their phenology in the Korea Strait.

We analyzed the paralytic shellfish poisoning (PSP) toxins of the toxic marine dinoflagellate Alexandrium tamarense collected from Dadaepo and Gaduck-do in Busan and from Sujeong-ri in Jinhae Bay, Korea, in April 2003. We also analyzed the PSP toxin of mussels (Mytilus galloprovincialis) and oysters (Crassostrea gigas) collected around Busan and Jinhae Bay. PSP toxin analyses were conducted by high performance liquid chromatography (HPLC). Fifteen cultured A. tamarense isolates contained 2.78 to 57.47 fmol/cell, with nearly identical toxin profiles: major components C2, GTX4; minor components C1, GTX1, NEO; and trace components GTX2, GTX3, STX. PSP toxin contents were 0 to $492\;\mu{g}$ STXeq/100 g in mussels and 0 to $48\;\mu{g}$ STXeq/100 g in oysters. Mussels at Gijang and Sujeong-ri contained the most PSP toxin contents ($492\;\mu{g}\;STXeq/100\;g\;and\;252\;\mu{g}\;STXeq/100\;g,\;respectively$), exceeding the quarantine level ($80\;\mu{g}$ STXeq/100 g). Their dominant toxin components were C2, C1, GTX2, and GTX3; the minor components GTX1, GTX4, GTX5, and NEO were sporadically detected. Phytoplankton contained 0.774 fmol/L seawater and 1.228 fmol/L seawater at Gijang and Sujeong-ri in April. At that time, Alexandrium cells were present in the water column at Gijang at 2,577 cells/mL and at Sujeong-ri at 6,750 cells/mL. Overall, we found the high and similar PSP toxin contents in AZexandrium isolates and mussels, and a correlation between occurrence of toxic Alexandrium cells in the water column and mussel intoxification. High densities of toxic Alexandrium cells in the water column immediately preceded shellfish intoxification at Gijang and Sujeong-ri in April.
독성이 있는 특이한 편모충인 Alexandrium tamarense의 구별하기 위한 종특이적인 형광 DNA탐침 AT1이 서로 다른 3가지의 고정액에서, 그리고 배양기간의 차이와 whole cell hybridization을 사용하는 광학 현미경 또는 형광 현미경에 의한 세포밀도 측정에 있어서 hiding activity를 비교하는 방법으로 여러 다른 종의 실험에 사용되어졌고, 그의 결과를 보고하는 바이다. 형광 DNA탐침 AT1은 특이적으로, A. tamarense에 결합했지만, 형태학적으로 유사한 다른 편모충에는 결합하지 않았다. 특히 형태적으로 A. tamarense에 유사한 A. catenella는 형광 DNN탐침 AT1에 결합하지 않았다. A. tamarense은 다른 세가지 고정액을 처리하였을 때, 고정액과 상관없이 강한 형광신호를 발산하였다. 추가해서 말하면, 배양기간에 관계없이 형광 DNA탐침 AT1의 biding activity는 강했다. 광학 현미경에 의해서 측정 되어지기 보다는 형광현미경에 의해서 세포밀도가 측정되었다. 형광 DNA탐침 AT1을 이용한 A. tamarense의 동정과 계산은 이 분야에서 새로운 생물독성 감시와 예측 시스템에 기여 할 것이다.
본 연구는 마비성 독소 표준물질의 안정적인 공급을 위해 친환경적이고 경제성이 우수한 발광다이오드 광원의 파장을 이용하여, PST의 원인종 Alexandrium catenella와 A. pacificum의 독 함량을 증대시킬 수 있는 가능성을 살펴보았다. 파장별 광량과 생장속도의 관계를 보면, 반포화상수(Ks)는 두 종 모두 적색파장에서 가장 높게 나타났고, 청색파장에서 가장 낮았다. 빛 파장에 따른 독함량의 경우, 적색 파장이 A. catenalla은 최고 약 8배 그리고 A. pacificum은 약 3.4배 다른 파장보다 높았다. 그리고 광량에 대한 친화성을 나타내는 Ks와 독농도와 밀접한 관련이 있는 것으로 나타났다. 따라서 세포당 독농도 증가를 위한 배양법은 초기에 세포수량의 확보 및 빠른 생장속도를 유도하기 위해서 청색파장을, 대수생장기 후기에는 적색파장을 주사하여, 세포내 독함유량을 증가시킬 수 있는 2단계 배양법을 제시할 수 있을 것으로 생각된다.

Severe damages will result in human society, when several different critical natural phenomena coincide. One example relates to the resting cysts of Alexandrium species (dinoflagellates that cause paralytic shellfish poisoning), which are preserved in surface sediments throughout Osaka Bay, Japan. These cysts have been found to accumulate particularly densely in shallow areas in the inner parts of Osaka Bay, where a tsunami caused by an earthquake could occur any time. Damage by a tsunami could cause a change of the coastal ecosystems at Osaka Bay including the resuspension of surface sediments containing resting Alexandrium tamarense cysts and the subsequent redistribution of the cysts in newly deposited sediment. Under certain environmental conditions, these cysts could germinate and form dense blooms, leading to paralytic shellfish poisoning. Such a scenario could also affect other coastal areas, including the southern coast of the Korean Peninsula.

The marine dinoflagellate genus Alexandrium has been recognized as the most representative toxic phytoplankton on account of production of paralytic shellfish poisoning (PSP) throughout the world. PSP producers, generally A, tamarense and A. catenella, within the genus Alexandrium have caused high level intoxicauon of fisheries products and even death of human. In addition, more recent increasing of geographical range of this deleterious species has given rise to alarming tension. The study presented here aimed construction of the molecular phylogenetic relationships through sequences-determination from 16 morphotypic species (containing newly sequenced 3 morphotypic species, A. tamiyavainchii, A. fraterculus and A. pseudogonyaulax) in LSU rDNA D1-D2 and 12 morphotypic species (containing newly sequenced 6 - morphotypic species, A. catenella, A. tamiyavanichii, A. fraterculus, A. affine, A. insuetum and A. pseudogonyaulax) in SSU rDNA region, and the sequences were subjected to comparative-analysis in respect to regional population using functionally expressed rDNA genus and pseudogenes. And we discussed on genetic differentiation between A. tamarense and A. catenella together with putative PSP divegence of the genus Alexandrium. The results of phylogenetic analysis showed the robust monophyletic 14 distinct classes of A. tamarense, A. excavatum, A. catenella, Tasmanian A. tamarense, A. affine (and/or A. concavum), Thai A. tamarense, A. tamiyavanichii, A. fraterculus, A. margalefii, A. andersonii, A. ostenfeldii, A. minutum (and/or A. lusitanicum), A. insuetum, and A, pseudogonyaulx clade. A. fraterculus and A. tamiyavanichii were sister relationship and they were positioned independently between A, affine cluster and those of A. margalefi, A. andersonii, A. ostenfeldii, A. minutum and A. insuetum. A. pseudogonyaulax appeared to be an ancestral taxon among Alexandrium.
