Isolation and Identification of Aldehyde Producing Methanol Utilizing Yeast (메탄올 자화성 효모의 분리, 동정 및 Aldehyde 생산)

  • Hansenula nonfermentans KYP-l was selected and identified from 19 methanol utilizing yeasts isolated from soil samples by the enrichment culture technique. This strain showed a high cell concentration and a high aldehyde production. Aldehyde production was carried out in a resting cell system using methanol utilizing yeast as a biocatalyst. The molar yield of acetaldehyde was the highest among the aldehyde investigated, and the maximum amount of aldehyde was produced by cells obtained from a 40 hours' culture.

Seasonal Analysis of Odorous Compounds Emitted From the Chemical Plant (계절별 악취물질의 배출량 분석 및 평가에 관한 연구)

  • In this study, the concentrations of offensive odorous compounds seasonally emitted from the chemical plant at Chongju industrial complex in Korea were determined by the analytical methods of gas chromatography, high performance liquid chromatography and uv/vis spectroscopy. The kinds of offensive odorous compounds examined are formaldehyde, acetaldehyde, butyl aldehyde, propion aldehyde, n-valeric aldehyde, iso-valeric aldehyde, hydrogen sulfide, methyl mercaptan, dimethyl sulfide, dimethyl disulfide, trimethyl amine and ammonia. The seasonally emission levels of all odorous compounds except dimethyl sulfide at 13 sampling points of plant were lower than those of the regulation standard levels of the industrial complex in Korea. The levels were the highest in June, and lowest in December. The propion aldehyde and iso-valeric aldehyde in June and December, butyl aldehyde in December, and n-valeric aldehyde were not detected in all the three seasons at any sampling points of the plant examined. But in June, dimethyl sulfide was emitted up to 16 times than that of the regulation level.

  • The concentrations of offensive odorous compounds emitted from the two chemical plants in Chongju and Yeosu industrial complex in Korea were determined by uv/vis spectroscopy, gas chromatography, and high performance liquid chromatography. The odorous compounds examined in this study are ammonia, trimethyl amine, formaldehyde, acetaldehyde, propion aldehyde, butyl aldehyde, n-valeric aldehyde, iso-valeric aldehyde, hydrogen sulfide, methyl mercaptan, dimethyl sulfide and dimethyl disulfide. The concentrations of those were determined from the 10 sampling points of the two plants, respectively. The emission concentrations of all odorous com-pounds examined in the two plants were lower than those of the regulation standard levels of industrial complex in Korea, respectively. The propion aldehyde, n-valeric aldehyde, methyl mercaptan and dimethyl disulfide in Chongju and Yeosu plants, and butyl aldehyde and iso-valeric aldehyde in Yeosu plant were not detected at any sampling points examined.

  • Aliphatic aldehyde polyoxyethylene glycol acetals were synthesised through the reaction of aliphatic aldehydes such as caproic aldehyde, capryl aldehyde, capric aldehyde and lauric aldehyde with excess diethylene glycol, triethylene glycol and tetraethylene glycol, respectively. The acetal formation, in which water was azeotropically distilled by adding benzene to the reaction system, was gained a good yield of acetal type compounds. This reaction is found pseudo first order reaction at various temperatures such 70, 80, 90 and $97^{\circ}C$. Also these activation energies of reaction of acetal type products such as caproic aldehyde diethylene glycol acetal, capryl aldehyde diethylene glycol acetal, capric aldehyde diethylene glycol acetal, lauric aldehyde diethylene glycol acetal, caproic aldehyde triethylene glycol acetal and caproic aldehyde tetraethylene acetal were 17.3, 19.6, 21.2, 21.6, 15.5 and 14.7 Kcal/mole.

  • The present study was undertaken to elucidate the effect of Serotonin and Norepinephrine on Aldehyde Oxidase activity in rabbit liver, in vitro. The results were as follows; 1. Aldehyde Oxidase was measured optimum substrate concentration at $5{\times}10^{-4}M$ and incubation time for 10 minutes. 2. Aldehyde Oxidase were inhibited by Serotonin and Norepinephrine. 3. It was observed that relationship between biogenic amines and substrate were competitive inhibition on Aldehyde Oxidase.

  • The effects of metallothionein and cadmium ion on the hepatic aldehyde oxidase activity and brain lipid peroxidation were tested in vitro. The content of brain lipid peroxide at the condition of normal or $300{\mu}M$ Fe(II)-induced was remarkably reduced by the addition of metallothionein in the incubation mixture. The induced content of lipid peroxide by cadmium $(30{\mu}g/ml)$ ion was reduced by metallothionein $(100{\mu}g/ml)$. The activity of aldehyde oxidase was not affected by metallothionein, but cadmium ion $(8.38{\mu}g/ml)$ increased the activity of aldehyde oxidase about 80% compared to the control. The cadmium-induced activity of aldehyde oxidase was restored to the control level by metallothionein or penicillamine.

  • 1. 전복, 소라, 북방조개, 가리비의 중요한 인지질은 choline phosphatides, ethanolamine phlosphatides, serine phosphatides, cardiolipin, sphingomyelin 및 lyso choline phosphatides 였다. 2. 이들 plasmalogen은 거의 대부분이 ethanolamine phosphatides에 존재하고, choline phosphatides에는 소량존재하였으며, serine phosphatide에는 전연발견되지 않았다. 3. 전복의 ethanolamine plasmalogen 함량은 1월에 채취한 것에는 $3.5\%$, 7월에 채취한 것에는 $17.2\%$였다. 또, 소라, 북방조개에는 각각 그 함량이 $20.7\%,\;15.1\%$였다. 가리비에는 ethanolamine phosphatides에 plasmalogen이 $18.8\%$ 존재하였다. 4. 전복 및 소라의 중요한 plasmalogen의 aldehyde 조성은 stearyl aldehyde $(C_{18:D})$, palmityl aldehyde $(C_{16:0})$, Palmitoleyl aldehyde $(C_{16:1})$ 및 oleyl aldehyde $(C_{18:1})$ 인데 대하여, 2매패인 북방조개, 가리비에는 eicosamonoenal $(C_{20:1})$$14-22\%$ 정도함유 되어 있어 권패와 대조적이였다.

  • 알데히드는 여러 가지 악취물질중의 하나로서 공기 중에 미량으로 존재하면서 냄새를 일으키며, 건강 위해성도 큰 물질이다. 본 연구에서는 휘발성이 강하면서 건강에 유해하여 문제가 되고 있는 이러한 aldehyde 화합물을 간편하게 분석할 수 있는 방법을 개발하고저 하였다. 기존의 aldehyde 분석에서는 알데히드를 DNPH와 같은 유도체로서 변환시킨 뒤에 주로 HPLC로 분석하는 방법을 사용하였다. 이 방법은 보편적으로 많이 알려져 있으나 HPLC가 갖고 있는 단점(크로마토그파피의 낮은 분해능, 긴 분석시간, 다량의 고가 HPLC용매 소모)을 안고 사용해야 하는 불편한 점도 있다.   • The roots of Cynanhum caudatum (Asclepiadaceae) have been used in folk medicine in Japan and China for the prevention and treatment of various geriatric diseases and also as a cardiotonic agent. Constituents of this plant have mainly been examined for glycosides: besides two steroidal alkaloids, gagaminine and gagamine which was firstly isolated by us, more than 35 polyoxypregnane glycosides and aglycones have also been identified. Gagaminine inhibits potently the hepatic aldehyde oxidase activity and lipid peroxidation in vitro. The present work deals with the comparison of antioxidative activities of gagamine, a new pregnane alkaloid, three isolated polyoxypregnanes containing a keto group at C-20 with those of gagaminine, a potent antioxidant, in order to explain the structure-activity relationships. The results of this study further prove that the cinnamoyl group of gagaminine is very important for the inhibition on the aldehyde oxidase activity while the nicotinoyl group is necessary for anti-lipid peroxidation. Besides that, the keto compounds having no ester group at C-12 were found to be more active than the others except gagaminine.
