Allyl Alcohol Found in Heated Garlic is a Potent Selective Inhibitor of Yeasts

  • Allyl alcohol (2-propen-l-ol), found in considerable amounts in heated garlic, was able to discriminate yeasts from bacteria and was approximately three orders of magnitude more inhibitory towards yeasts than bacteria. The average minimum inhibitory concentration (MIC) of allyl alcohol for bacteria and yeasts was 5.0% and 0.0056%, respectively. The unsaturated primary alcohols, including allyl alcohol and 2-buten-l-ol, seemed to work differently from all the other saturated alcohols and unsaturated secondary alcohols in inhibiting various yeasts. An alcohol dehydrogenase-negative (ADH$^-$) strain of Saccharomyces cerevisiae was as resistant to allyl alcohol as various bacteria, exhibiting an MIC of 5.0%. The unsaturated primary alcohols were apparently oxidized into the corresponding unsaturated aldehydes before they inhibited the yeasts.

효모와 초기 당도를 달리하여 개발한 수박 양조주의 기호도에 관한 연구 (A Study on Preferences of Watermelon-Wine Manufactured with Different Kinds of Yeasts and Different Starting Sugar Contents)

  • The principal objective of this study was to prepare watermelon wine with watermelon juice. For the manufacture of watermelon wine, we determined the contents of starting sugar content ($20^{\circ}Brix$, $25^{\circ}Brix$, $30^{\circ}Brix$, $35^{\circ}Brix$) and three different kinds of yeasts (EC-1118, K1-V1116, D-47). The main additives of the watermelon wine included $K_2S_2O_5$, $NH_4H_2PO_4$, bentonite, yeasts, sugar, mixed acid, and $C_6H_8O_2$. The watermelon wine was fermented at $15^{\circ}C$ until the remaining alcohol content was 5%. We assessed customers' preference regarding the manufactured watermelon wine via sensory evaluation. Three yeasts were shown to result in significantly different alcohol fermentation, among which K2-V1116 was selected for the preparation of the watermelon wine. In our sensory evaluation, the preference of the starting sugar contents was highest in the watermelon wine with $25^{\circ}Brix$ of sugar content. The preference of alcohol content was found to be higher in the watermelon wines prepared with sugar contents of 5% and 7%. The preference test between watermelon wines and purchased white wines demonstrated that the watermelon wine with an alcohol content of 5% was the most preferred wine, and the Chilean Late Harvest wine was the second most preferred.

Screening Wild Yeast Strains for Alcohol Fermentation from Various Fruits

  • Wild yeasts on the surface of various fruits including grapes were surveyed to obtain yeast strains suitable for fermenting a novel wine with higher alcohol content and supplemented with rice starch. We considered selected characteristics, such as tolerance to alcohol and osmotic pressure, capability of utilizing maltose, and starch hydrolysis. Among 637 putative yeast isolates, 115 strains exhibiting better growth in yeast-peptone-dextrose broth containing 30% dextrose, 7% alcohol, or 2% maltose were selected, as well as five ${\alpha}$-amylase producers. Nucleotide sequence analysis of the 26S rDNA gene classified the strains into 13 species belonging to five genera; Pichia anomala was the most prevalent (41.7%), followed by Wickerhamomyces anomalus (19.2%), P. guilliermondii (15%), Candida spp. (5.8%), Kodamaea ohmeri (2.5%), and Metschnikowia spp. (2.5%). All of the ${\alpha}$-amylase producers were Aureobasidium pullulans. Only one isolate (NK28) was identified as Saccharomyces cerevisiae. NK28 had all of the desired properties for the purpose of this study, except ${\alpha}$-amylase production, and fermented alcohol better than commercial wine yeasts.

인삼성분이 효모의 생육 및 대사에 미치는 영향 (The Effects of Korean Ginseng (Panax ginseng C.A. Meyer) Extracts and Their Fractions on the Growth and Metabolism of Saccharomyces cerevisiae and Saccharomyces uvarum)

  • This study was conducted to investigate the effects of Korean ginseng extracts and their fractions on the growth of Saccharomyces cerevsiae and Saccharomyces uvamm, their glucose consumption and alcohol production. The growth of both yeasts were stimulated by ginseng extracts and their water soluble fractions, but were supressed by ether extracts and an n-butanol extracts. Their growth were enhanced considerably by low molecular weight fractions (< 1,000) in water solubles. Similar results were also obtained with glucose consumption by yeasts. Substances increasing the growth and glucose consumption by yeasts proved to be a low molecular weight fractions (<1,000) in water solubles not saponins. The production of n-propyl alcohol by yeast was enhanced by adding ginseng extracts into the media, but that of ism-butyl alcohol was suppressed at same condition.

효모의 종류에 따른 탁주의 품질특성 변화 (Quality Characteristics of Takju by Yeast Strain Type)

  • Investigation of the quality characteristics of Takju brewing mashes prepared with several different yeasts was carried out during the 42-day brewing process, including examination of titratable acidity, amino acid content, sugar contents, alcohol contents, pH, reducing sugar, yeast population, lactic acid bacteria, and sensory evaluation. Fermivin yielded the highest titratable acidity, while la parisienne had the lowest. Fermivin also had the highest value of amino acid content (14.33%), while other yeasts shows period tended to increase over exclude Instaferm and Song chun. The amount of sugars in instaferm ($11.33^{\circ}Bx$) and instaferm red ($10.53^{\circ}Bx$) were higher than those in La parisienne red and Song chun ($9.57^{\circ}Bx$). The alcohol contents of instaferm was the highest value (15.20%) while the alchol content of the other yeasts tended to increase the fementation period. Fermivin had the highest pH at $12^{th}$ day among the yeasts(4.02), Instaferm red had the lowest pH among the yeasts (3.87) while safbrew wb-06 had the highest reducing sugar content(117.60 mg/mL). Instaferm obtained the highest preference in the sensory evaluation for all items.

고추장의 발효미생물에 관한 연구 2 (Microbiological Studles of Red pepper paste Fermentation (II))

  • Among 50 yeasts of red pepper paste isolated and identified in the previous report, 12 strains of yeast which possess osmophilic and non-film characteristics were examined their growth rates and alcohol fermentation ability in red pepper medium. 5 strains of yeast possessing powerful growth rates and excellent alcohol fermentation ability were selected as excellent yeasts in this experiment. And their cultural conditions, salt resistance and red pepper concentration upon the growth were examined in this report. The results obtained were as follows ; 1) Among 12 strains of yeast, Saccharomyces cerevisiae group II(strain $D_1$), Saccharomyces oviformis($D_2$), Saccharomyces steineri($M_3$), Saccharomyces rouxii($D_9$) and Saccharomyces mellis($G_8$) were selected as excellent yeasts. 2) Among 5 strains of yeast selected, the strain $D_1$ and $D_2$showed the best alcohol fermentation ability and the best fermentative flavour. 3) The optimum temperature for growth of the strain $D_1$ and $D_2$ was $25^{\circ}C$$M_3$, $D_9$ and $G_8$ $30^{\circ}C$$D_1$ and $D_2$ was $62^{\circ}C$$M_3$,$D_9$ and $G_8$ ws 60.deg.C with treatment for 10 minutes. 4) The optimum pH for growth of all strains was around 5.0. 5) The strain $D_1$ and $M_3$were grown exceedingly well on the emdia containing 3 percent of NaCl and $D_2$,$D_9$ and $G_8$ on the media containing 6 percent of NaCl. 6) In the case of 5 percent of red pepper liquor added 5 percent of glucose was more nutritional than in the case of 5 percent glucose only. However, the growth rates of yeasts were restrained by the increase of red pepper concentration.

재래누룩에서 분리한 N4와 N9 효모의 증류식 소주 양조특성 (Soju brewing characteristics of yeast strains N4 and N9 isolated from Korean traditional Nuruk)

  • 재래누룩에서 분리한 효모인 N4와 N9에 대해 쌀 증류식 소주제조 특성을 살펴보았다. 원료 쌀(한아름 품종)의 일반성분은 수분 14.7, 조단백질 6.8, 조지방 0.9, 조회분 0.4, 탄수화물 76.5 g/100 g이었다. 제조된 입국의 총산 함량은 2.92%(citric acid, dry base)였으며 효소활성에 있어 당화력은 926.72, ${\alpha}$-amylase 52.40, gluco-amylase 887.71, ${\alpha}$-glucosidase 0.14, acidic carboxypeptidase 17,335.73, ${\beta}$-glucosidase 174.46 U/g dry base로 나타났다. 술덧의 알코올 함량은 재래누룩 분리 효모가 15.37-16.58%로 상업용 효모 12.33-13.19%보다 16.7-36.0% 높은 것으로 나타났다. 반면, 환원당 함량은 각각 2.04-3.92, 7.92-8.78 g/100 mL로 상업용 효모의 당 이용률이 누룩분리 효모보다 낮았다. 이러한 결과로 증류 후 원주의 획득량이 누룩분리 효모에서 25.2-52.7% 높았다. 쌀 증류식 소주(알코올 25%)에서 41개의 휘발성 성분이 검출되었으며, 주성분 분석 결과 누룩분리 효모와 상업용 효모는 i-BuOH, isobutanal diethyl acetal, ethyl caprate, tetradecanoic acid 성분의 함량 차이가 있는 것으로 나타났다.

  • 사과주내의 과도(過度)한 산(酸)을 malo-alcohol 발효(醱酵)에 의해 감소(減少)시키기 위하여 사과산 분해능(分解能)이 강(强)한 효모(酵母) Schizosaccharomyces japonicus var. japonicus St-3의 이용성(利用性)을 양조학적(釀造學的) 측면(側面)에서 Schizosaccharomyces pombe 0-77과 비교(比較) 검토(檢討)한 결과(結果)는 다음과 같다. 사과산 분해효모(分解酵母) Schiz. pombe와 Schiz. japonicus var. japonicus 를 첨가(添加)한 구(區)는 발효(醱酵) 13일(日)동안에 약 80%의 사과산을 분해(分解)하는데 반하여 무첨가구(區)는 발효(醱酵) 40일동안에 32%정도의 사과산을 분해(分解)하므로 malo-alcohol 발효효모(醱酵酵母)의 첨가(添加)가 사과산분해(分解)에 효과적이었다. 또한 천연배지(天然培地)에서는 Schiz. pombe 가 우수하였고 합성배지(合成培地)에서는 Schiz. japonicus var. japonicus가 양호(良好)하였다. 사과산 분해효모(分解酵母)에 의(依)한 malo-alcohol 발효(醱酵)에 가장 알맞는 온도(溫度)는 $20{\sim}30^{\circ}C$였으며 $SO_2$는 200 ppm까지 발효(醱酵)에 아무런 영향을 주지 않았다. $Mg^{2+}$ 또는 특정(特定) 아미노산, 비타민, 유기산 등(等)의 첨가(添加)는 사과산의 분해(分解)를 촉진(促進)시켰다.

충남 서천군 갈대밭 주변 토양에서 야생효모의 분리 및 미기록종 특성 (Isolation of Wild Yeasts from Soils of Reed Fields in Seocheon-gun County, Chungcheongnam-do, South Korea, and Characterization of Unrecorded Yeasts)

  • 충남 서천군 한산면 신성리 금강하구 갈대밭 토양의 야생효모 분포특성을 알아보기 위하여 갈대밭 토양 20점을 채취하여 야생효모들을 분리하여 총 11종 20균주의 야생효모를 동정하였다. Candida속의 효모가 10균주로 가장 많이 분리되었고, 그 중 Candida subhashii 균이 6균주로 가장 많았다. 분리된 균주 중에서 Candida tropicalis (Y5-1,Y5-2, Y6-1), Candida subhashii (J7-1, J7-3, J8-1, Y9-1, Y9-2, Y9-3), Lachancea thermotolerans (MJS 007, MJS 019) 균주 등 12균주들이 알코올 발효능을 보였다. Bullera japonica YJ10-1, C. subhashii J7-1, Kluyveromyces yarrowii YJ11-1, Ustilago shanxiensis Y10-1등 4균주들이 국내 미기록 효모 균주들로 최종 선별되었다. 이들 국내 미기록 균주들은 구형으로 Candida subhashii J7-1만이 자낭포자와 의균사를 형성하였다. Kluyveromyces yarrowii YJ11-1은 비타민을 함유하지 않은 배지에서도 생육하였고 Candida subhashii J7-1는 YM과 YPD 배지 및 5% NaCl을 함유한 YPD 배지에서 생장하지 않았다.

Inhibition of Yeast Film Formation in Fermented Vegetables by Materials Derived from Garlic Using Cucumber Pickle Fermentation as a Model System

  • Film-forming yeasts generate an undesirable yeasty flavor in fermented vegetables such as kimchi in the presence of oxygen. Antimicrobial materials including garlic oil (GO), heated garlic (HG), and allyl alcohol (AA) were investigated for use as alternative natural food preservatives to inhibit the growth of film-forming yeasts in fermented vegetables. Using the fermentation of cucumber pickles as a model system, GO, HG, and AA were effective in preventing film formation at concentrations of 0.006, 3.0, and 0.02%, respectively. The effectiveness of HG in preventing the growth of a film yeast, Hansenula anomala, was not influenced by pH, while that of potassium sorbate, a typical anti-yeast food preservative, was highly dependent on pH. All tested materials were effective when added at the beginning of fermentation due to their negligible inhibitory activity toward lactic acid bacteria.
