Bactericidal Effect of Waterless Alcohol Gel Hand Washing Agent (물 없이 사용하는 알코올 젤의 손소독 효과에 관한 연구)

  • The purpose of this study was to compare the hand disinfection effect of waterless alcohol gel hand washing agent with that of soap and water, 4% chlorhexidine gluconate, and 10% povidone-iodine. Hands of fourty subjects were artificially contaminated with Acinetobacter baumannii $5m{\ell}$ and randomly distributed to each hand washing methods. Samples were collected from gloved hand by glove juice sampling procedure. Mean log reduction after hand washing were compared with baseline values. Number of microorganisms were converted to log and tested by ANOVA in SPSSWIN 10.0. Mean log reduction of soap and water, alcohol gel, 4% chlorhexidine gluconate, 10% povidone-iodine were $2.76{\pm}0.62$, $2.97{\pm}0.56$, $4.66{\pm}1.70$, $4.60{\pm}0.91$, respectively. The bactericidal effect of alcohol gel was similar to that of soap and water, but the effect was much less than chlorhexidine gluconate and povidone-iodine(p<0.001). In terms of microorganism reduction, the efficacy of waterless alcohol gel was almost the same as soap and water hand washing. Further evaluation of the bactericidal effect of waterless alcohol gel is needed because waterless alcohol gel is simple, convenient, and non-irritating hand washing agent and also very effective in busy hospital environment.

Analyzing Image Quality and Disinfection Effectiveness of Alcohol Gel Application in Ultrasound Scans (초음파 스캔에서 알코올 겔 적용 시 화질과 소독효과 분석)

  • To analyze the image quality and disinfection effectiveness of alcohol gel application in ultrasound scans, we varied the application of alcohol gels with different alcohol content from ultrasound gels. The image quality analysis compared the Dead zone, axial and lateral resolution, sensitivity, and gray scale changes using the ATS-539 phantom, while the disinfection effectiveness analysis was performed by swabbing the three cover faces of convex probe used in clinical practice, collecting specimens and enrichment culture them for 24 hours before identification with Gram stain. image quality analysis showed that all items, regardless of the alcohol content, produced images that were not different from the ultrasonic gel application images, and pathogens such as staphylococcus aureus, streptococcus, and enterococcus were detected on all probes, and all were extinction by the application of alcohol gel. The application of alcohol gel during ultrasound examination was meaningful in terms of image quality maintenance and disinfection effect, and if the results of this study are further studied and commercialized, the application of alcohol gel as an alternative to ultrasound gel will be very useful in clinical examinations because it can consider not only image quality but also infection prevention.

  • A genomic library of Zymomonas mobilis DNA was constructed in Escherichia coli using plasmid pUC9 Allyl alcohol was used to screen a genomic clone expressing alcohol dehydrogenase. The plasmids isolated from two clones, which were sensitive to allyl alcohol, were found to be related and to share a common 2.6 kb fragment encoding alcohol dehydrogenase II identified as one of two isozymes in Z. mobilis by staining for alcohol dehydrogenase activity on polyacrylamide gel and spectrophotometric analysis of several substrate oxidations.

Comparison of Antimicrobial Effect of Alcohol Gel according to the Amount and Drying Time in Health Personnel Hand Hygiene (의료종사자 손 소독용 알코올 젤의 사용량과 건조시간에 따른 항균효과의 비교)

  • Purpose: The purpose of this study was to compare the effect of alcohol gel according to the amount and drying time in health personnel hand hygiene and to promote in their practice adequate and effective hand hygiene. Methods: The crossover experimental study was performed with 14 volunteers. Hands were artificially contaminated with 5 mL of $10^8$ CFU/mL of Serratia marcescens (ATCC 14756) and four different alcohol gel hand hygiene methods varying by the amount of alcohol gel (2 mL vs. 1 mL) and drying time (complete vs. incomplete) were compared. Samples were collected by glove juice sampling procedures. Results: Mean log reduction values of the four different hand hygiene methods were $2.22{\pm}0.36$, $1.26{\pm}0.53$, $1.49{\pm}0.60$, $0.89{\pm}0.47$ respectively for the 4 groups: adequate amount (2mL) and complete dry (30 seconds rubbing followed by 2 min air-dry), inadequate amount (1 mL) and complete dry, adequate amount and incomplete dry (15 seconds rubbing and no air-dry), and inadequate amount and incomplete dry. The difference was statistically significant in the adequate amount and complete dry group compared to other three groups (p<.001). Conclusion: Only alcohol gel hand hygiene with adequate amount and complete drying was satisfactory by U.S. FDA-TFM efficacy requirements for antiseptic hand hygiene products.

  • Cetostearyl alcohol has been incorporated into cosmetic or pharmaceutial emulsions to give them an appropriate consistency and a long term stability. It is well known that cetostearyl alcohol forms a gel phase with a nonionic surfactant in an aqueous system, and the properties of the gel phase depend on several factors such as the ratio of fatty alcohols. The aim of the present Paper is to investigate the effect of the structural difference of the lipophilic part of surfactants on the stability of the gel phase in the nonionic surfactant / cetostearyl alcohol / water ternary systems using differential scanning calorimeter. It is concluded that the gel phase formed by the surfactant having the bended alkyl chain is more unstable than that formed by the case of the straight alkyl chain, and we discussed the reason why the former is more unstable after long term storage by means of the measurements of the ${\Delta}$H.

Proton Conducting Behavior of a Novel Composite Based on Phosphosilicate/Poly(Vinyl Alcohol)

  • A series of proton conductive composite membranes based on poly(vinyl alcohol) and phosphosilicate gels powders were successfully prepared. The proton conductivity of these composite was attributed to the phosphosilicate gel, which derived from tetraethoxysilane and phosphoric acid by sol-gel process at a molar ratio of P/Si = 1.5. The proton conductivity increased with increasing both the content of phosphosilicate gel and relative humidity. Temperature dependence of conductivity showed a Vogel-Tamman-Fulcher type behavior, indicating that proton was transferred through a liquidlike phase formed in micropores of phosphosilicate gel. The high conductivity of 0.065 S/cm with a membrane containing 60 wt$\%$ of the gel was obtained at $60^{\circ}C$ at $90\%$ relative humidity.

  • Poly(vinyl alcohol) (PVA) and gelatin (GEL) blend membranes were prepared by solution casting method under a high electric field. SEM observation of the membrane showed that gelatin rich domains were elongated and oriented to the direction of the applied electric field in PVA matrix. This can be attributed to the electrostatic emulsifying effects due to a reduction in interfacial tension. In addition, it was observed through WAXD and swelling measurements that the degree of crystallinity of membranes increased with applied electric field strength. This may be interpreted to be caused by the orientation effect of GEL domains in the blend membrane, and the self-annealing effect due to some heat generated from high electric field during casting.

  • Polyvinyl alcohol (PVA) hydrogels were prepared by the irradiation and heating. Irradiation and heating processes were carried out to improve the heat resistance of PVA hydrogels at high temperature. The physical properties such as gel content, degree of swelling and gel strength for the synthesized hydrogels were examined. The structure variations were investigated using the following techniques: differential scanning calorimetry (DSC) and X-ray diffraction (XRD). Gel content and gel strength of the hydrogels were higher when the two steps of irradiation followed by heat treatment were used rather than only with the irradiation. The hydrogels prepared by the irradiation and the two steps had good heat resistance at high temperature.

  • The alcohol dehydrogenase (ADH) isozyme of the four strains of Drosophila melanogaster in Korea was studied by the starch gel electrophoresis and the results are given below: 1. The numbers of bands, staining intensity and mobility are considerably variable in the three strains, Chunchun, Jinju and Sinchon-Seoul, but relatively uniform in the Taijun strain. 2. It is suggested that the Taijun strain may be homogeneous and the other three strains, on the contrary, heterogeneous for the ADH constitution. 3. The electrophoretic patterns are observed to be different between males and females in all strains used which is hard to intereret. 4. The starch gel electrophoresis is to have a resolving power superior to cellulose acetate electrophoresis.

  • The Polyvinyl alcohol (PVA) oxidase is a key enzyme involved in degradation of PVA with PVA hydrolase. The PVA oxidase has been purified to homogeneity from the culture broth of PVA grown Pseudomonas cepacia G5Y strain by ammonium sulfate precipitation, DEAE-cellulose column chromatography, and Sephadex G-150 gel filtration. The molecular weight of the purified enzyme was estimated about 60, 000 daltons by SDS-polyacrylamid gel electrophoresis. The enzyme is most active at 45$\circ$C and at pH 8.5, and is stable below 55$\circ$C and between pH 6 and 9. The enzyme activity was strongly inhibited by Ag$^{2+}$ and Hg$^{2+}$.
