Schwanniomyces castellii에 의한 전분의 직접 알콜발효 (Direct Alcohol Fermentation of Starch by Schwanniomyces castellii)

  • Schwanniomyces castellii NRRL Y-2477에 의한 전분의 직접 알콜발효에 대한 연구를 수행하였다. 이 균주의 150g/l glucose에 대한 알콜발효능력은 사용기질의 94%를 이용하여 ethanol를 63.9g/l를 생성하였으며, 이때의 ethanol yeild를 0.45g-ethanol/g-glucose로서 일반적인 알콜발효 효모인 Sacch. cerevisiae보다는 약간 낮았다. 전분이 상온에서 최대로 용해될 수 있는 20g/l전분의 직접알콜 발효에서는 통기인 경우 ethanol의 생성없이 세포성장만이 이루어졌으며, 통기를 하지 않은 경우는 9.1g/l의 ethanol를 생성하여 ethanol yeild는 이론값의 88%를 나타내었다. 반면 100g/l의 dextrin을 이용한 알콜발효에서는 통기에 의하여 ethanol의 생성이 1.8배 증가하였으며, 또한 $CaCO_3$첨가에 의한 pH를 조절한 경우 조절하지 않은 경우보다 ethanol생성이 2.1배 증가한 34.5g/l의 농도로서 이때의 기질에 대한 발효능력은 82%이고 ethanol yeild는 이론값의 약 84%였다. 또한 150g/l dextrin의 알콜발효에서는 약 79%의 기질을 소모하면서 52.1g/l의 ethanol을 생성하였다. 따라서 Schwanniomyces castellii NRRL Y-2477 에 의한 전분의 직접 알콜발효에서의 율속단계는 전분의 가수분해 과정임을 확인하였다.

대규모의 주정발효 과정에서 생성된 불순물과 그 효율적 분리 (Impurities formed from ethanol fermentation process among different materials and it′s effective separation in large scale)

  • 주정발효공정에서 생성되는 불순물의 분리를 위한 공정개선으로 주정원료별 불순물의 양을 알아보고 이를 super-allospas식 연속증류기에서 집적하여 효율적인 분리를 하였다. 주정발효공정중에 생성되는 불순물 중 methanol은 절간고구마가 가장 많이 생성되었고 그 다음이 타피오카였다. fusel oil중 n-propanol은 쌀, 절간고구마, 옥수수, 쌀보리 및 타피오카의 순서이고 iso-butanol은 타피오카, 옥수수, 쌀, 절간고구마 및 쌀보리의 순서였다. 그리고 iso-amyl alcohol은 타피오카, 쌀. 절간고구마 옥수수, 쌀보리의 순이었다. 한편 supper-allospas식 연속증류기에 의한 fusel oil의 분리시 alcohol함량에 따른 집적율을 보면 n-propanol은 alcohol이 78.25%(v/v)일 때 37.9%. iso-butanol은 alcohol이 68.54% (v/v)일 때 28.6%, n-butanol은 alcohol 50% (v/v)일 때 37.4% iso-amyl alcohol은 alcohol이 50% (v/v)일 때 56.1%의 가장 높은 집적율을 나타내었다. 이와 같이 집적된 불순물은 side cut 하여 후류탑으로 유입시킨 후 재차 농축하여 탑저부의 붕(plate)에서 fusel oil을 side cut하여 일부는 직접분리하고 나머지는 다시 fusel oil분리기에 넣어 alcohol이 20%(v/v) 되도록 가수하여 분리하였다.

Characterization of Alcohol Fermentation and Segregation of Protoplast Fusant of Saccharomyces cerevisiae and Pichia stipitis

  • A study was conducted to investigate the characteristics of segregation and alcohol fermentation of intergeneric fusants. The protoplast fusion of both Pichia stipitis CBS 5776 and Saccharomycess cerevisiae STV 89 was carried out. The fusion frequency was $5\times10^{-8}$ and among fusants selected, a fusant F5 showed the best results in ethanol production by sucrose and xylose fermentations. The performance of xylose fermentation by this fusant was better than that of P. stipitis CBS 5776 and fusant F5 exhibited sucrose fermentation patterns intermediate to the two parent strains. The fusant F5 was segregated into a pair of parental strains during the several culture passages. In the average, 91$%$ of colonies had a similar characteristics of P. stipitis while 7$%$ of colonies resembled S. cerevisiae. Only 2$%$ of colonies had the characteristics of the original fusants. At the sixth passage, all segregants resembled P. stipitis. From these results it is suggested that intergeneric protoplast fusion led to an integration of S. cerevisiae genes, rather than whole chromosomes, within the entire genome of P. stipitis.

으뜸도라지 식초의 저장기간별 품질 특성 및 항산화 활성 (Quality Characteristic and Antioxidant Activities of Vinegar Added with Etteum Bell Flower Root)

  • The aim of this study was to investigate the characteristics and antioxidant activities of vinegar made with Etteum bell flower root (0.5%, 1%, 1.5% and 2%) during two-step fermentation. Acetic acid was fermented at $30^{\circ}C$ for 16 days and samples of vinegar were extracted at 2, 4, 6, 8, 10, 12 and 16 days. The pH of Etteum bell flower root vinegar did not significantly differ among the samples, but the acidity increased during fermentation. Alcohol content decreased at 16 days of fermentation and less than 1% alcohol was shown in all samples after fermentation. The pure acetic acid yield was 88.85~98.97%, whereby the total phenolic compound content and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities increased as the ratio of the Etteum bell flower root increased. The sensory scores of vinegar fermented with 1.5% Etteum bell flower root are greater than those of vinegar prepared by other treatments. Therefore, vinegar with 1.5% Etteum bell flower root added is considered to be the most suitable for manufacturing.

  • Experiments of alcohol fermentation of the yeast,K. fragi1is CBS 1555 were performed to obtain the following results. In these experiments, the initial concentrations of sugar which was composed of inulin and fructose as weight ratio of one to one were 30, 50, 75, 100 and 150g/l and the initial densities of the microorganism were less than 0.5g/l, 10g/1 to 15g/1, and 50g/l. The functional relationship among specific growthrates, sugar concentrations, and alcohol concentrations could be expressed by Aiba-shoda equation and the specific growth rate represented the trend that decreased with increase in the initial concentration of the microorganism. Also, $\mu$max and Ks of Monod's equation could be expressed as the function of initial cell concentration like the following equations. $\mu$max=0.8-0.008X Ks=0.54X+8 In the region that sugar, alcohol and cell concentrations were 10g/1 to 120g/l, 0g/l to 60g/l and 0.5g/l to 50g/l respectively, the differences between the experimental values and the calculated ones for specific growth rate approached to 40% with respect to experimental values at the worst cases, but in most cases, those were distributed in the range of less than 20%.

물의 경도가 알코올발효에 미치는 영향 연구 -오존의 영향을 중심으로- (Study of Hardness Effects of Water in Alcohol Fermentation to Focus Effect of Ozone)

  • 알코올 발효에서 물의 경도가 중요한데 오존을 사용하게 되면 경도를 줄이면서 알코올의 생성량을 증가시키는데 중요한 역할을 한다. 지하수에 오존을 처리하는 경우, 오존의 산화반응으로 알코올의 품질에 긍정적인 효과를 보이는 것으로 나타났으며 담금 공정에서의 처리효과를 분석한 결과는 다음과 같다. 1) 오존의 주입시간에 따라 에탄을 발생량이 다르게 나타났으며 오존주입시간을 4시간 처리한 경우가 미 오존 처리한 경우와 비교하여 수중의 경도는 85% 이상 감소되며 전기전도도는 50% 이상 증가하였지만 수중의 산소농도는 1.2% 미만으로 큰 차이가 나타나지 않았다. 오존주입시간과 발효시간 경과에 따라 에탄을 발생량은 초기와 비교하여 최대 300 % 이상 차이가 나는 것으로 나타났다. 2) 오존 처리시 수중의 오존의 농도는 1 ppm 이하의 규정을 유지한 상태에서 오존주입을 증가시키게 되면 오존 증가와 함께 에탄올의 생성량은 1차 담금 4일째부터 20% 이상 증가하는 것으로 나타났다.

Fermentation properties of beer produced from Korean two-row barley or malt (Gwangmaek) supplemented with Korean red ginseng extracts and Bokbunja (Rubus coreanus Miquel) juice

  • This study involved the production of specialty lager beers supplemented with Korean red ginseng extracts or Bokbunja (Korean black raspberry, Rubus coreanus Miquel) juice. The effects of the Korean red ginseng extracts or Bokbunja juice on the specific gravity, pH, yeast viability, free amino nitrogen content, reducing sugar content, color, alcohol content, turbidity, and sensory evaluation were evaluated. The alcohol content of the beers containing the extracts or juice were within the standard alcohol amounts (3.63-4.0%, v/v). The pH values of the three samples containing Bokbunja juice were lower than that of the control values. The sensory evaluation showed that the addition of Bokbunja juice was superior to the ginseng extracts, and the optimal addition time was before or after the secondary fermentation. These data indicate that the flavor and odor of the Bokbunja juice are more persistent than that of the ginseng extracts.

고정화된 Zygosaccharomyces rouxii BH-90과 Candida versatilis BH-90를 이용한 Column 형 reactor 에서 간장의 연속적 속성발효 (Continuous Rapid Fermentation of Soy Sauce by Immobilized Zygosaccharomyces rouxii BH-90 and Candida versatilis BH-91 Using Column Type Reactor)

  • The aim of this study was to find out the rapid fermentation of soy sauce from koji hydrolyzate using column type reactor packed with immobilized yeast cells. Each immobilized cell of Zygosaccharomyces rouxii BH-90 and Candida versatilis BH-91 in the packed column type reactor produced 2.8% ethyl alcohol and 18mg/l 4-ethylguaiacol over 96 hours under the optimal condition. Continuous fermentation was performed by immobilized Z. rouxii BH-90 packed in column type reactor. Immobilized Z. rouxii BH-90 produced 2.30~2.85% ethyl alcohol during 30 days, and decreased gradually from 40 days to 80 days. Also C. versatilis BH-91 produced 4-ethylguaiacol at the constant rate of 16~18mg/l and decreased gradually after 40 days. Final product of soy sauce contained 2.8% ethyl alcohol and 18mg/l 4-ethylguaiacol. However, amino acid compositions of final products were consisted of predominantly glutamic acid, leucine, arginine, aspartic acid, lysine and valine, which were more than 50% of total amino acid.

반 연속식 배양에 의한 효모 K. fragilis의 알콜발효 특성에 관한 연구 (Investigation of the Ethanol Fermentation Characteristics of K. fragilis by Semicontinuous Culture)

  • K. fragilis를 이용한 돼지감자의 반연속알콜발효에서 유효희석속도 및 주입기질 당농도의 변화가 정상상태에서의 알콜농도 및 알콜생산성에 미치는 영향을 시험을 통해 규명하였으며 그 결과를 요약하면 다음과 같다. 주입기질의 치환시간 간격을 1시간을 했을 경우 유효희석속도는 비증식속도와 일치하여 연속발효실험의 희속속도와 같음을 알 수 있다. 유효의석속도가 0.425$^hr-1$에서도 wash out이 발생되지 않았으며 최대 알콜 생산성은 5.5g/l.hr이였고 이 경유 유효희석속도는 0.25$hr^hr-1$, 주입기질의 당농도는 85g/lsowl 135g/l사이에 분포되어 있다.

  • Antioxidant activity of dropwort fermented extract (DFE) was measured according to fermentation period, and liver protective effects were examined using Sprague-Dawley rats. Total polyphenol and flavonoid contents as well as DPPH and ABTS radical scavenging activities increased up to 60~80 days and then decreased slightly. Proper fermentation time for DFE was more than 60 days and less than 80 days. Administration of alcohol to rats for 10 days at 10 mL/kg/day raised serum AST, ALT, total cholesterol, and triglyceride (TG) levels, which were then lowered by DFE and sugar liquid with the same soluble solids. While sugar liquid increased the blood lipid profile, especially TG levels, DFE had no effect due to its antioxidant activity. When TBARS content of the DFE group in liver tissue significantly decreased in a concentration-dependent manner compared to that of the ALH group (p<0.05). Liver damage was recovered by DFE treatment and was confirmed by hamatoxylin-eosin staining. These results suggest that DFE has a protective effect against alcohol-induced hepatotoxicity in SD rats.
