The Extract of Limonium tetragonum Protected Liver against Acute Alcohol Toxicity by Enhancing Ethanol Metabolism and Antioxidant Enzyme Activities

  • The protective effect of EtOAc fraction of Limonium tetragonum extract (EALT) against alcohol-induced hepatotoxicity was assessed following acute ethanol intoxication in Spraque-Dawley rats. EALT (200 mg/kg p.o.) was administrated once before alcohol intake (8 g/kg, p.o.). Blood ethanol concentration, and the activities of alcohol metabolic enzymes, alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) in the liver were measured. Also, the formation of malondialdehyde (MDA) and the activities of antioxidant enzymes, superoxide dismutase (SOD), glutathione peroxidase (GSH-px), catalase were determined after acute alcohol exposure. Pretreatment of rats received ethanol with EALT significantly decreased blood ethanol concentration and elevated the activities of ADH and ALDH in liver. The increased MDA level was decreased, and the reduced activities of SOD, GSH-px and catalase were markedly preserved by the treatment with EALT. This study suggests that EALT prevent hepatic injury induced by acute alcohol which is likely related to its modulation on the alcohol metabolism and antioxidant enzymes activities.

Analysis of Alcohol Drinking Cessation Programs on Worksites (사업장 절주 프로그램 분석 - 사업장 건강증진운동 우수사례집을 중심으로 -)

  • Purpose: The purpose of this study was to analyze alcohol drinking cessation programs to promote health status for workers on worksites. Method: Data were collected from the excellent 10 cases which were selected from "competition of health promotion programs on worksites" from 1999 to 2007 held by Korean Occupational Safety and Health Agency. Result: There were three main alcohol drinking cessation programs on worksites: health education, individual health services to change life style, and formation of supportive environments. Health education and individual health services were intervened in nine worksites. Building supportive environments for alcohol drinking cessation were implemented in 10 worksites. The most popular indicators for program evaluation were health diagnosis and percentage of alcohol drinkers. Problems to implement alcohol drinking cessation programs were low voluntary participation of labors, difficulty of applying programs to labors that had a shift duty, and complexity to evaluate the effectiveness of alcohol cessation programs due to deficits of standard instruments. Conclusion: These findings suggest that many worksites did not have alcohol drinking cessation programs and also these programs were not implemented effectively. Thus, employers, professionals and policy makers of occupational health should develop and support effective alcohol drinking cessation programs for l workers on worksites.

  • The regularities of immobilization of natural medicinal substances - alchidine on gels of poly(vinyl alcohol) - are investigated. It is shown that the polymer components of alchidine interact with nonionic polymer with formation of interpolymer complexes. The influence of the different factors on the properties of complexes is considered and the constants of alchidine distribution in the system gel - water are calculated.

  • For the urethane formation reaction of isocyanate with alcohol, orientation of approach of alcohol toward isocyanate was investigated theoretically by means of MO method. As a result of EHT and CNDO/2 calculations, it was found that the reaction proceeding via 4-centered mechanism, in which ROH is coplanar with isocyanate plane was energetically favored. It was also shown that the oxygen of ROH attacks the isocyanate carbon prior to the formation of 4-center complex. The reaction was found to be a charge controlled one.

Effect of Alcohol Addition on Back-Extraction of BSA and Cytochrome c Using AOT Reverse Micellar System

  • The protein back-extraction processes were discussed from the viewpoint of the micelle-micelle interaction. Bovine serum albumin (BSA) suppressing the cluster formation of reverse micelle (positive value of ${\beta}pr)$ has the high back-extra cted fraction (Eb), but cytochrome c enhancing the formation of reverse micelle (negative value of ${\beta}pr)$ has the low back-extracted fraction, relatively. We have also examined quantitatively the effects of alcohol addition and protein solubilization on the percolation process of reverse micelle. The alcohols suppressing the formation of micellar cluster (high values of ${\beta}t)$, remarkably improved the back-extraction rates of BSA and cytochrome c. The values of ${\beta}t$, defined by the variation of percolation process, and the back-extraction behavior of proteins have a good linear correlation. These results indicate that the micelle-micelle interaction or micellar clustering plays an important role in the back-extraction process of proteins.

  • Counterion-specific helix formation and ion-selective transport of alkali metal chlorides (LiCl, NaCl, KCl, CsCl) were investigated for a poly(L-glutamic acid)(PLGA)/poly (vinyl alcohol)(PVA) blend membrane immersed in aqueous ethanol. The counterion specificity for helix formation of PLG alkali metal salts in the membrane was Li>Na>K>Cs. This specificity is ascribed to a contact ion-pair formation between the PLG carboxyl anion and the bound counterion, which depends on the energy balance between the electrostatic interaction and the desolvation. In aqueous ethanol, an appreciable ion-selectivity was observed for the permeability coefficient, i.e. Li$^{+}{\cdot}$Cl$^{-}$) formation between counterion and coion, and the latter to a specific interaction of diffusing counterions with polymer charges.

Effects of Chungganhaeju-tang(Qingganjiejiu-tang) on Alcohol Metabolism and Alcoholic Liver Damages (청간해주탕이 알코올대사 및 손상간에 미치는 영향)

  • Objectives: This experiment was conducted to verify the effects of Chungganhaeju-tang(Qingganjiejiu-tang) on the alcohol metabolism and liver functions, by measuring the activity levels of ADH and ALDH, as well as glucose, triglyceride, and BUN. Damage of the liver cells caused by alcohol was determined through the examination of serum levels of AST, ALT, ALP, LDH, and uric acid. Methods: Sprague-Dawley rats were used in this experiment and the rats were divided into control and experiment groups. Chungganhaeju-tang(Qingganjiejiu-tang) extract was orally administered in the experiment group for three weeks. Each group was further classified into two sub-groups, and control group's blood was taken without oral ingestion of alcohol, while the experiment group' s blood was withdrawn after ingestion of alcohol. Evaluation of damage level was done considering the presence of extract and alcohol. Results: In this experiment, Chungganhaeju-tang(Qingganjiejiu-tang) significantly suppressed the activity of ADH which is a precursor enzyme of acetaldehyde, but didn't cause significant changes in the activity of ALDH which is a catabolic enzyme. Decreased glucose level due to alcohol consumption was recovered back to the normal level and increased levels of triglyceride, BUN, AST, ALT, ALP, LDH, and uric acid were significantly reduced. Conclusions: These experiment results suggest that Chungganhaeju-tang(Qingganjiejiu-tang) inhibits the formation of acetaldehyde in the metabolism of alcohol, and affects the recovery of weakened liver functions due to alcohol.

  • Low-salted and fermented squid product, squid jeotkal was prepared with the addition of 10% salt and fermented for 50 day at 1$0^{\circ}C$. During fementation of squid, sensory evaluation and changes of volatile components were examined. Volatile flavor components in raw squid and low-salted squid jeotkal were extracted using a rotary evaporating system. The volatile concentrates were identified by GC and GC-MS. Major volatile components of raw squid were methional and 2-methyl-2-propanol. However, alcohols such as propanol, isoamyl alcohol, methionol and phenylethyl alcohol increased during the period of fermentation. The model reaction using microorganism was carried out, in order to confirm formation mechanism ofvolatile flavor compounds of the squid during fermentation. The main volatile components of Pseudomonas sp. D2 model system were isoamyl alcohol and acetoin. Those of Staphylococcus xylosus model system were isoamyl alcohol and phenylacetaldehyde.

Transdermal Delivery of Diclofenac Using Microemulsions

  • A transdermal preparation containing diclofenac diethylammonium (DDA) was developed using an O/W microemulsion system. Of the oils tested, lauryl alcohol was chosen as the oil phase of the microemulsion, as it showed a good solubilizing capacity and excellent skin permeation rate of the drug. Pseudoternary phase diagrams were constructed to obtain the concentration range of oil, surfactant and cosurfactant for microemulsion formation, and the effect of these additives on skin permeation of DDA was evaluated with excised rat skins. The optimum formulation of the microemulsion consisted of 1.16% of DDA, 5% of lauryl alcohol, 60% of water in combination with the 34.54% of Labrasol (surfactant)/ethanol (cosurfactant) (1:2). The efficiency of formulation in the percutaneous absorption of DDA was dependent upon the contents of water and lauryl alcohol as well as Labrasol: ethanol mixing ratio. It was concluded that the percutaneous absorption of DDA from microemulsions was enhanced with increasing the lauryl alcohol and water contents, and with decreasing the Labrasol:ethanol mixing ratio in the formulation.

  • Silver nano-particles have been synthesized by liquid phase reduction method and alcohol reduction process. Silver nano-particles of the size 30 ~ 40 nm were formed successfully by alcohol reduction process. The formation, structure, morphology and size of silver nano-particles have been studied using FE-SEM, TEM, XRD, UV-visible spectroscopy. In particular high dispersion stability of the synthesized silver nano-particles could be obtained by PVP binding. Antibacterial activity of Ag/PET master batch sample made from its nano-silver particles showed excellent antibacterial activity against S. aureus and E. coli.
