BACKGROUND/OBJECTIVE: Oxidative stress plays a key role in neuronal cell damage, which is associated with neurodegenerative disease. The aim of present study was to investigate the neuroprotective effects of perilla oil (PO) and its active component, alpha-linolenic acid (ALA), against hydrogen peroxide $(H_2O_2)$-induced oxidative stress in SH-SY5Y neuronal cells. MATERIALS/METHODS: The SH-SY5Y human neuroblastoma cells exposed to $250{\mu}M$$H_2O_2$ for 24 h were treated with different concentrations of PO (25, 125, 250 and $500{\mu}g/mL$) and its major fatty acid, ALA (1, 2.5, 5 and $25{\mu}g/mL$). We examined the effects of PO and ALA on $H_2O_2$-induced cell viability, lactate dehydrogenase (LDH) release, and nuclear condensation. Moreover, we determined whether PO and ALA regulated the apoptosis-related protein expressions, such as cleaved-poly ADP ribose polymerase (PARP), cleaved caspase-9 and -3, BCL-2 and BAX. RESULTS: Treatment of $H_2O_2$ resulted in decreased cell viability, increased LDH release, and increase in the nuclei condensation as indicated by Hoechst 33342 staining. However, PO and ALA treatment significantly attenuated the neuronal cell death, indicating that PO and ALA potently blocked the $H_2O_2$-induced neuronal apoptosis. Furthermore, cleaved-PARP, cleaved caspase-9 and -3 activations were significantly decreased in the presence of PO and ALA, and the $H_2O_2$-induced up-regulated BAX/BCL-2 ratio was blocked after treatment with PO and ALA. CONCLUSIONS: PO and its main fatty acid, ALA, exerted the protective activity from neuronal oxidative stress induced by $H_2O_2$. They regulated apoptotic pathway in neuronal cell death by alleviation of BAX/BCL-2 ratio, and down-regulation of cleaved-PARP and cleaved caspase-9 and -3. Although further studies are required to verify the protective mechanisms of PO and ALA from neuronal damage, PO and ALA are the promising agent against oxidative stress-induced apoptotic neuronal cell death.

The occlusal relationship of the maxillary first molar to the mandibular first molar of the subjects were classified as follows; Group I : Group of neutrocclusion relationship. Group II : Group of distocclusion relationship. Group III : Group of mesiocclusion relationship. The obtained results were as follows; 1) The positional relationship of maxillary cuspid to ala-lacrimal duct line were as follows; (Male) Type A: $68.5%{\pm}1.58m%$, Type B: $22.5%{\pm}2.30m%$, Type C: $9.1%{\pm}\1.58m%$ (Female) Type A : $64.5%{\pm}2.94m%$, Type B: $26.5%{\pm}2.71m%$, TypeC: $8.9%{\pm}1.73m%$ 2) Ala-lacrimal duct line located more distally in group II and group III than group I at the point of the positional relationship of upper and lower first molar. 3) Average distance and standard deviation in type A were as follows; (Male) Right: $2.17mm.{\pm}1.01mm.$ Left: $2.27mm.{\pm}1.02mm.$ (Female) Right: $2.06mm.{\pm}0.70mm.$ Left: $2.24mm.{\pm}0.80mm.$ 4) Average distance and standard deviation in type C were as follows; (Male), Right: $1.47mm.{\pm}0.85mm.$ Left: $1.52mm.{\pm}0.97mm.$ (Female) Right: $1.04mm.{\pm}0.44mm.$ Left: $1.26mm.{\pm}0.89mm.$ The results of 3) and 4) had no significant difference between the male and female.
This study was conducted to evaluate effect of ${\alpha}$-linolenic acid (ALA) and bovine serum albumin (BSA) on viability, acrosome reaction and mitochondrial intact in frozen-thawed boar sperm. The boar semen was collected by gloved-hand method and cryopreserved using freezing extender containing 3 ng/mL ALA and/or $20\;{\mu}g/mL$ BSA. Cryo-preserved boar sperms were thawed in $37^{\circ}C$ water-bath for 45 sec to analysis. Viability, acrosome reaction, and mitochondrial intact were analyzed using flow cytometry. In results, viability of frozen-thawed boar sperm was significantly higher in only ALA+BSA supplement group than control group (p<0.05), whereas there was no difference either in ALA or BSA supplement. However, acrosome reacted sperm in both of live and all sperm population were significantly decreased in all treatment groups than control (p<0.05). Interestingly, mitochondrial intact of boar sperm was enhanced in ALA and ALA+BSA groups compared with control (p<0.05). In this study, we showed that supplementation of ALA and BSA in freezing extender enhanced the sperm viability, mitochondrial intact and decrease acrosomal membrane damage. In conclusion, our findings suggest that quality of frozen-thawed sperm in mammalians could improve by using of ALA and BSA.

Alpha-lipoic acid (ALA) is a naturally occurring antioxidant and has been previously used to treat diabetes and cardiovascular disease. However, the autophagy effects of ALA against oxidative stress-induced dopaminergic neuronal cell injury remain unclear. The aim of this study was to investigate the role of ALA in autophagy and apoptosis against oxidative stress in the SH-SY5Y human dopaminergic neuronal cell line. We examined SH-SY5Y phenotypes using the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay (cell viability/proliferation), 4′,6-diamidino-2-phenylindole dihydrochloride nuclear staining, Live/Dead cell assay, cellular reactive oxygen species (ROS) assay, immunoblotting, and immunocytochemistry. Our data showed ALA attenuated hydrogen peroxide (H2O2)-induced ROS generation and cell death. ALA effectively suppressed Bax up-regulation and Bcl-2 and Bcl-xL down-regulation. Furthermore, ALA increased the expression of the antioxidant enzyme, heme oxygenase-1. Moreover, the expression of Beclin-1 and LC-3 autophagy biomarkers was decreased by ALA in our cell model. Combined, these data suggest ALA protects human dopaminergic neuronal cells against H2O2-induced cell injury by inhibiting autophagy and apoptosis.
ALA is an essential intermediate in the tetrapyrrole biosynthesis pathway and has applications as a biodegradable herbicide and insecticide as well as medical applications including photodynamic therapy of cancers. For the development of mass production process of ALA it is necessary to on-line monitor some metabolites such as glycine, succinate, LA and ALA. In this work, medium compositions, pH conditions, induction and addition of LA were investigated to increase the production of ALA by recombinant E. coli. A 2-dimensional fluorescence sensor was used to monitor the processe and the fluorescence spectra were correlated with on- and off-line data like ALAS, ALAD, cell mass etc.

A simple and general method that selectively introduces metal binding sites into a protein can greatly increase the ability to design and biosynthesize artificial metalloproteins. Here, we report the incorporation of a phenanthroline-containing amino acid (Phen-Ala) into proteins in Escherichia coli by using the $tRNA{^{Tyr}}_{CUA}$ and tyrosyl aminoacyl-tRNA synthetase pair (BpyRS) from Methanococcus jannaschii, which was originally developed for a bipyridine-containing amino acid (Bpy-Ala). The incorporation efficiency of BpyRS for Phen-Ala was comparable to that for Bpy-Ala. Because of its high metal-binding ability and characteristic spectral properties, Phen-Ala can be a useful alternative to the existing metal-chelating amino acids for the design and synthesis of artificial metalloproteins.
5-Aminolevulinate (ALA) is well-known as an essential biosynthetic precursor of all tetrapyrrole compounds, which has been suggested to improve plant salt tolerance by exogenous application. In this work, the gene encoding aminolevulinate synthase (ALA-S) in yeast (Saccharomyces cerevisiae Hem1) was introduced into the genome of Arabidopsis controlled by the Arabidopsis thaliana HemA1 gene promoter. All transgenic lines were able to transcribe the YHem1 gene, especially under light condition. The chimeric protein (YHem1-EGFP) was found co-localizing with the mitochondria in onion epidermal cells. The transgenic Arabidopsis plants could synthesize more endogenous ALA with higher levels of metabolites including chlorophyll and heme. When the $T_2$ homozygous seeds were cultured under NaCl stress, their germination and seedling growth were much better than the wild type. Therefore, introduction of ALA-S gene led to higher level of ALA metabolism with more salt tolerance in higher plants.
