Assessment of plant genetic resources is applicable when breeding improved plant caltivars. In Korea, early maturing wheat germplasm is anticipated for wheat-rice double-cropping. In this study, we assessed five agronomical traits including days of heading after sowing(HD), days of maturity after sowing(MD), culm length(CL), ear length(EL), and thousand grain weight(TW) of 500 wheat germplasms collected form 10 different countries (Mexico, the United States, Afghanistan, Ethiopia, India, Japan, China, Turkey, Pakistan and Portugal), and grown in Korea. Besides, the correlation between the agronomical traits was analyzed using XLSTAT softerware version 2019 (Addinsoft, NY, USA). The result showed wide-ranging maturity period. Among the entire population, 2 accessions (K256306/JPN/breeding line and K256328/JPN/breeding line) that matured early were identified. Furthermore, HD showed strong correlation with MD (r=0.684) and CL (r=0.610), and weak correlation with the rest two agronomical traits (EL and TW). Overall, the results of our study provides wide spectrum of prospects, and could be applicable to breed new wheat varieties with early maturity.
This study investigated the diversity of 323 mung bean accessions from Korea, China, and India, along with six cultivars, using 22 agronomical traits. The standardized Shannon-Weaver index (H') for the qualitative traits ranged from 0.11 (terminal leaflet shape) to 0.98 (pubescence density of pod). Likewise, the coefficient of variation for the quantitative traits ranged from 8.76% (days to maturity (DM)) to 79.91% (lodging rate (LR)), indicating a wide genetic variance. Hypocotyl color, pod color, seed shape, and seed coat surface lust showed different distributions among Korean, Indian, and Chinese accessions. Chinese accessions had the highest average germination rate, DM, days from flowering to maturity, and one-hundred seeds weight, followed by Korean and Indian accessions, while the number of seeds per pod (SPP) displayed the opposite trend, with all except SPP showing significant variation (p < 0.05). Similarly, plant height, days to flowering, and number of pods per plant increased in the order of India > Korea > China, with LR showing the opposite trend (p < 0.05). The mung bean accessions were grouped into four major clusters using hierarchical cluster analysis supported by principal component analyses, and all of the quantitative traits showed significant variations between the clusters (p < 0.05). Generally, the mung bean accessions investigated in this study exhibited wide phenotypic trait variations, which could be beneficial for future genomics studies. Moreover, this study identified 77 accessions that outperformed the controls. Consequently, these superior accessions could provide a wide spectrum of options during the development of improved mung bean varieties.

Mungbean(Vigna radiata (L.) R. Wilczek) is one of legume cultivated in south and east Asia. It well cultivated in high temperature and drought condition. In order to establish pure line for genomic studies, the agronomical characteristics of Korean mungbean landraces were evaluated. A total of 324 acessions derived from National Agrobiodiversity Center were selfed two times in 2021 nersery. Tree times of selfed from 324 accessions were characterized. The color of seed coat was divided into three groups: green, yellow and brown. Among them green color was the most common with 91.7%. Flower colors were divided into four groups: yellow, purplish yellow, greenish yellow and purplish green. Among them purplish yellow color had the highest with 77.8%. Out of 324 resources, only 4 showed compound leaves, and all resources had a heart-shaped leaf. The days to first flowering was 31 to 78 days. About 63% of the accessions bloomed 35 to 50 days after sowing. 324 Korean landraces are expected to be used as a population of the Korean mungbean core collection and serve as a basis for genomic breeding materials for mungbean.

Mung bean(Vigna radiata (L.) R. Wilczek var. radiata) is a legume that originated in India. It is the third most cultivated legume in Korea after soybean and adzuki bean. Recently, the use of mung bean seeds and sprouts in trendy foods such as rice noodles and Chinese-style stir-fry is expanding thereby increasing its demand. Subsequently, improvement of mung bean varieties is also being actively conducted. In this study, the important agricultural characteristics of 324 mung bean germplasm were recorded and statistically investigated. Seeds of the mung bean germplasm were cultivated at an experimental field located in the National Agrobiodiversity Center (Jeonju, Korea) and 10 quantitative agricultural traits were investigated. Basic statistics, correlation analysis, and principal component analysis were then performed. The results showed significant variations of the quantitative traits among the germplasms (p < 0.05). The days to flowering, maturity, and growth were in the ranges of 31~80, 22~72, and 57~110 days with means of 45, 47, and 92 days, respectively. The highest frequency (f = 192) was for lodging score with 11~50%, while simultaneous maturity (f = 182) was below 50%. Other quantitative traits related to yield including the number of seeds per pod (CV = 10.9%), number of pods per plant (CV = 41.2%), and one-hundred seeds weight (CV = 36.6%) also showed significant variations. Correlation analysis showed positive correlations between the days to maturity and one-hundred seeds weight (r = 0.41) and the days to growth and simultaneous maturity (r = 0.39). In contrast, one-hundred seeds weight was negatively correlated to the number of pods per plant (r = -0.41) and the days to flowering (r = -0.29). Similarly, the days to growth and the number of pods per plant had a negative association with each other (r = -0.29). The principal component analysis revealed the number of days to maturity as the most influential variable along the first principal component (23.7%). In general, this study revealed wide variations in quantitative traits among the studied mung bean germplasm, which could provide several options for cultivar development.
Soybean [Glycine max (L.) Merr.] seed is an important dietary source of protein, oil, carbohydrate, isoflavone and other various nutrients for humans and animals. However, there are anti-nutritional factors in the raw mature soybeans. Kunitz trypsin inhibitor (KTI) protein and stachyose are the main anti-nutritional factors in soybean seed. The ${\alpha}^{\prime}$-subunit of ${\beta}$-conglycinin protein exhibit poor nutritional and food processing properties. The genetic removal of the KTI and ${\alpha}^{\prime}$-subunit proteins will improve the nutritional value of the soybean seed. The objective of this research was to develop a new soybean strain with KTI and ${\alpha}^{\prime}$-subunit protein free ($titicgy_1cgy_1$ genotype) and proper agronomic traits. A breeding population was developed from the cross of the Bl-1 and 15G1 parents. A total of 168 $F_2$ seeds from the cross of the BL-1 and 15G1 parents were obtained. The segregation ratios of 9: 3: 3: 1 ($104Ti\_Cgy_{1\_}:\;30Ti\_cgy_1cgy_1:\;21cgy_1cgy_1Ti\_:\;13titicgy_1cgy_1$) between the Ti and $Cgy_1$ genes in the $F_2$ seeds were observed (${\chi}^2=5.12$, P=0.5-0.10). Two $F_4$ plant strains with proper agronomical traits and $titicgy_1cgy_1$ genotype (free of both KTI and ${\alpha}^{\prime}$-subunit protein) were selected and harvested. 2 strains (S1 and S2) had yellow seed coats and hilum. The plant height of the S1 strain was 65 centimeters. The 100-seed weight was 29.2 g. The plant height of the S2 strain was 66 centimeters and 100-seed weight was 26.2 g. The two strains selected in this research will be used to improve the new cultivar that will be free of the KTI and ${\alpha}^{\prime}$-subunit proteins.

Soybean [Glycine max (L.) Merr.] is one of the major food sources of protein, oil, carbohydrates, isoflavones, and other nutrients for both humans and animals. However, soybean seeds contain antinutritional factors, such as lectin protein, Kunitz Trypsin Inhibitor (KTI) protein, and stachyose. The objective of this research was to stack recessive alleles for development a triple recessive genotype, titilelers2rs2, with low KTI protein, lectin protein, and stachyose contents. Three parents (Gaechuck#2, PI200508, and 14G20) were used to develop the breeding population. The presence or absence of the lectin and KTI proteins was detected by western blotting. The stachyose content in mature seeds was determined by HPLC. Agronomic traits, such as plant type, plant height, maturity date, lodging, seed quality, and 100-seed weight, were evaluated for the four $F_3$ plant strains. One $F_4$ plant strain with the desired agronomical traits was selected. One new strain with the triple recessive titilelers2rs2 genotype was developed. The plant height of the new strain was 51 cm and the 100-seed weight was 31.0 g. The new strain had a yellow seed coat and yellow hilum. The stachyose content of the new strain was 3.8 g/kg. One strain developed in this research will be used to produce improved yellow soybean cultivars that are free of lectin and KTI proteins and low in stachyose content.

Mutation breeding is the useful tool to improve agronomic traits in various crop species. Soybean is most important crop and is rich in protein and oil contents. Despite of the importance as economic value and various genetic resource of soybean, there have been limited studies of genetic relationship among mutant resources through radiation breeding. In this study, the agronomical phenotype for selecting various genetic resources was evaluated in 528 soybean mutant lines. As a result, 210 soybean mutants with their original cultivars were selected with various traits. We named 210 selected lines as Mutant Diversity Pool (MDP). The genetic diversity and the relationship of the MDP were investigated using TRAP and TE-TRAP markers. In TRAP analysis, sixteen primer combination (PC)s were used and a total of 551 fragments were amplified. The highest (84.00%) and the lowest (32.35%) polymorphism levels were showed in PC MIR157B+Ga5 and B14G14B+Ga3, respectively. The mean of PIC values was 0.15 ranging from 0.07 in B14G14B+Sa12 to 0.23 in MIR157B+Sa4. Phylogenetic and population structure analysis indicated that the 210 MDP lines dispersed to four groups among the wild types and their mutants. The highest genetic diversity among populations was observed between lines Paldal and 523-7 (Fst=0.409), whereas the lowest genetic diversity was between population KAS360-22 and 94seori (Fst=0.065). AMOVA showed 11.583 (21.0%) and 43.532 (79.0%) variations in inter and intra mutant population, respectively. Overall, the genetic similarity of each intra mutant populations was closer than that of inter mutant population. A total of 408 fragments were amplified in the 210 MDP using twelve PCs of TE-TRAP markers that were obtained from a combination of three TIR sequence of transposable elements (MITE-stowaway; M-s, MITE-tourist; M-t, PONG). The highest (77.42%) and the lowest (56.00%) polymorphism levels were showed in PONG+Sa4 and PONG+Sa12, respectively. The mean of PIC values was 0.15 ranging from 0.09 in M-s+Sa4 and M-s+Ga5 to 0.21 in M-t+Ga5. AMOVA of M-s showed 2.209 (20%) and 8.957 (80%) variations in inter and intra mutant population, respectively. AMOVA of M-t showed 2.766 (18%) and 12.385 (82%) variations in inter and intra mutant population, respectively. AMOVA of PONG showed 3.151 (29%) and 7.646 (71%) variations in inter and intra mutant population, respectively. According to our study, the PONG had higher inter mutant population and lower intra mutant population. This mean was that for aspect of radiation sensitivity, M-s and M-t showed higher mobility than that of PONG. Our results suggest that the TRAP and the TE-TRAP markers may be useful for assessing the genetic diversity and relationship among soybean MDP and help to improve our knowledge of soybean mutation/radiation breeding.

Soybean [Glycine max (L.) Merr.] seeds contain an average of 40% protein on a dry weight basis, but they also contain antinutritional elements such as lectin, Kunitz trypsin inhibitor (KTI), and 7S α'- subunit protein. The objective of this research was to develop a new soybean genotype with triple recessive alleles for these elements. Three parents (Gaechuck#2, PI506876, and Le-16) were used to develop the genetic population, and the presence of lectin and KTI protein was detected using Western blot while 7S α' subunit protein was detected using SDS-PAGE. One F3 plant strain with proper agronomical traits such as type, height, seed quality, and 100-seed weight was selected. The genotype of the developed strain is titilelecgy1cgy1, that is KTI, lectin, and 7S α' subunit protein free. The new strain has a purple flower, determinate growth habit, and light yellow pods at maturity. The seed has a buffer hilum and is yellow in color. The new strain's height was 58 cm compared to the Daewonkong cultivar at 46 cm, and its 100-seed weight was 27.1 g, smaller than the Daewonkong at 29.0 g. This is the first new soybean strain with the titilelecgy1cgy1 genotype, and it can be used to improve yellow soybean cultivars of high quality and function.

Seed coat color and seed weight are among the key agronomical traits that determine the nutritional quality of soybean seeds. This study aimed to evaluate the contents of total protein, total oil and five prominent fatty acids in seeds of 49 soybean varieties recently cultivated in Korea, and assess the influences of seed coat color and seed weight on each. Total protein and total oil contents were in the ranges of 36.28-44.19% and 13.45-19.20%, respectively. Likewise, individual fatty acid contents were in the ranges of 9.90-12.55, 2.45-4.00, 14.97-38.74, 43.22-60.26, and 5.37-12.33% for palmitic, stearic, oleic, linoleic, and linolenic acids, respectively. Our results found significant variations of protein, oil and fatty acid contents between the soybean varieties. Moreover, both seed coat color and seed weight significantly affected total oil and fatty acid contents. Total protein content, however, was not significantly affected by any factor. Among colored soybeans, pale-yellow soybeans were characterized by a high level of oleic acid (30.70%) and low levels of stearic (2.72%), linoleic (49.30%) and linolenic (6.44%) acids, each being significantly different from the rest of colored soybeans (p < 0.05). On the other hand, small soybeans were characterized by high levels of all individual fatty acids except oleic acid. The level of oleic acid was significantly high in large seeds. Cluster analysis grouped the soybeans into two classes with notable content differences. Principal component analysis also revealed fatty acids as the prime factors for the variability observed among the soybean varieties. As expected, total oil and total protein contents showed a negative association with each other (r = -0.714, p < 0.0001). Besides, oleic acid and linoleic acid showed a tradeoff relationship (r = -0.936, p < 0.0001) which was reflected with respect to both seed coat color and seed weight. In general, the results of this study shade light on the significance of seed coat color and seed weight to distinguish soybeans in terms of protein, oil and fatty acid contents. Moreover, the soybean varieties with distinct characteristics and nutritional contents identified in this study could be important genetic resources for consumption and cultivar development.
