Specific bovine antibody response against a new recombinant Cryptosporidium parvum antigen containing 4 zinc-finger motifs

  • A Cryptosporidium parvum sporozoite and oocyst λgt11 cDNA library was screened with a hyperimmune rabbit serum that was developed against insoluble fragments of ultrasonicated oocysts. A clone named Cp22.4.1 encoding a protein of 231 amino acids with 4 zinc-finger domains characterized by a Cys-X2-Cys-X4-His-X4-Cys motif was isolated and characterized. There was a complete match between the sequencing data of the coding region of Cp22.4.1 and the corresponding gene at chromosomal level. Cloning in a pBAD-TOPO-TA expression vector permitted to evaluate the antigenicty of the recombinant His-tagged antigen. This antigen was recognized by 2 out of 5 sera from Cruptosporidium immune calves and not by sera from parasite naive animals.

  • An expert system for cucumber disease and nutritional disorders(ESCD) was developed using HCLIPS in which several interface programs were added to CLIPS developed in NASA. The knowledges for constructing knowledge base was obtained from interviews with farmers, agrochemical vendors and employees of agrochemical company. The ESCD is able to diagnose eleven diseases and twelve nutritional disorders which farmers, agrochemical vendors and employees of agrochemical company. The ESCD is able to diagnose eleven diseases and twelve nutritional disorders which could frequently found in fields and greenhouses. Validation of ESCD by twenty -four experts proved a possibility of its practical use for diagnosing cucumber diseases and nutritional disorders in the future.

Effect of Prochloraz on Electrolytic Leakage and Spore Germination of Puccinia recondita Causing Wheat Leaf Rust

  • The effects of prochloraz on membrane permeability and germination of uredospores of Puccinia recondita were investigated to determine its potential mode of action on wheat leaf rust control activity. Disease control activity of ergosterol biosynthesis inhibitors (EBIs) and their activities on uredospore membrane permeability and germination were examined with wheat leaf rust pathogen, both in vitro and in vivo. While wheat leaf rust was not controlled by prochloraz, electrolytic leakage and spore germination of P. recondita uredospore was the highest with the use of prochloraz among the eight fungicides tested. Prochloraz stimulated uredospore of P. recondita to germinate at a higher ratio. Although certain EBIs, such as hexaconazole, showed excellent control activity, their effects on uredospore membrane permeability and germination was much inferior to prochloraz. Therefore, results of this study suggest that effects of EBIs on membrane permeability and germination of uredospore are not always correlated with their disease control activity.

  • The present study relates to a matrix-typed, sustained-releasing agent comprising agrochemical effective ingredients being capable of recognizing a content of moisture, and a preparation method thereof. The curdlan solution added to acid was formed matrix which has unique network structure. The matrix treated by heat lost water solubility. The network structure of matrix was opened in the aquatic condition but closed again in dry condition. Therefore, in the sustained-releasing formulation system, an agrochemical effective ingredient was released from the formulation only in the aquatic condition. Use of the composition according to the product can control a manifesting time of effects of agrochemicals and can provide agrochemicals with reduced harmful damages.
