Analysis of Systemic Pesticide Imidacloprid and Its Metabolites in Pepper using QuEChERS and LC-MS/MS (QuEChERS 전처리와 LC-MS/MS를 이용한 고추 중 침투성농약 Imidacloprid 및 대사물질 동시분석법)

  • Imidacloprid is a systemic insecticide which act as an insect neurotoxin. It used for control of pest such as aphids and other sucking insects in fruits and vegetables. Systemic pesticides move inside a crop following absorption by the plant, and these were converted into a variety of metabolites. Sometimes these metabolites make a problem about safety of agricultural products. So a simultaneous determination method of pesticide and its metabolites is needed, to monitor their presence in agricultural product and study on the fate of pesticide in a plant. This study's aim is to investigate simultaneous analysis method of imidacloprid and its metabolites, imidacloprid guanidine, imidacloprid olefin, imidacloprid urea, and 6-chloronicotinic acid in red pepper using QuEChERS method and LC-MS/MS systems. QuEChERS method was modifed beacuase $MgSO_4$ salts decreased the recoveries of 6-chloronicotinic acid in extraction procedure. Imidacloprid and its metabolites were extracted by acetonitrile with 1% glacial acetic acid and the extracts were purified through QuEChERS with primary secondary amine (PSA) and $C_{18}$ and analyzed with LC-MS/MS in ESI positive mode. Standard calibration curves were made by matrix matched standards and their correlation coefficients were higher than 0.999. Recovery studies were carried out on spiked pepper blank sample at four concentration levels (0.01, 0.04 and 0.1, 0.4 mg/kg). The average recoveries of imidacloprid and its metabolites were in the range of 70~120% with < 20% RSD. This result indicated that the method using QuEChERS and LC-MS/MS was suitable for the simultaneous determination of imidacloprid and its metabolites in red pepper.

Medium optimization for keratinase production by a local Streptomyces sp. NRC 13S under solid state fermentation

  • Thirteen different Streptomyces isolates were evaluated for their ability to produce keratinase using chicken feather as a sole carbon and nitrogen sources under solid state fermentation (SSF). Streptomyces sp. NRC 13S produced the highest keratinase activity [1,792 U/g fermented substrate (fs)]. The phenotypic characterization and analysis of 16S rDNA sequencing of the isolate were studied. Optimization of SSF medium for keratinase production by the local isolate, Streptomyces sp. NRC13S, was carried out using the one-variable-at-a-time and the statistical approaches. In the first optimization step, the effect of incubation period, initial moisture content, initial pH value of the fermentation medium, and supplementation of some agro-industrial by-products on keratinase production were evaluated. The strain produced about 2,310 U/gfs when it grew on chicken feather with moisture content of 75% (w/w), feather: fodder yeast ratio of 70:30 (w/w), and initial pH 7 using phosphate buffer after 8 days. Based on these results, the Box-Behnken design and response surface methodology were applied to find out the optimal conditions for the enzyme production. The corresponding maximal production of keratinase was about 2,569.38 U/gfs.

  • Pesticides are regarded as one of indispensable materials used in agriculture to protect pests and to keep the agriculture productions steadily. However, food contamination arisen from their application has brought about public concerns for safety suspicion to human health. A brief presentation is given in this paper on what has been conducted against pesticide residues to evaluate and ensure the safety of agro-products. The commonest countermeasures to keep safety would be the establishment of MRLs and safe-use guidelines of pesticides according to good agricultural practices. Korean MRLs have bun established on 38 chemicals with 56 commodities and in the case of safe-use guidelines 344 formulation items have been done by the government. On the other hand, no satisfactory information is still available to evalute the actual residue levels in domestic agro-products, but a cautious proposal could be suggested through the fragmentary monitoring data of pesticide residues, so that the current residue levels would be far from potent hazard enough to cause chronic impact on human. The most important thing is to have the farmers keep safe use guidelines at pesticide application for the production of safe agriculture commodities.

Chemical Composition of Cactus Pear Seed Oil: phenolics identification and antioxidant activity

  • Objectives: The chemical composition of cactus pear seed oil (Opuntia ficus-indica [L.] Mill.) was analyzed in terms of its fatty acid composition, tocopherol content, phenolic identification, and the oil's phenolic-rich fraction antioxidant power was determined. Methods: Fatty acid profiling was performed by gas chromatography coupled to an FI detector. Tocopherols and phenolic compounds were analyzed by LC-FLD/UV, and the oil's phenolic-rich fraction antioxidant power was determined by phosphomolybdenum, DPPH assay and β-carotene bleaching test. Results: Fatty acid composition was marked by a high unsaturation level (83.22 ± 0.34%). The predominant fatty acid was linoleic acid (66.79 ± 0.78%), followed by oleic acid (15.16 ± 0.42%) and palmitic acid (12.70 ± 0.03%). The main tocopherol was γ-tocopherol (172.59 ± 7.59 mg/kg. In addition, Tyrosol, vanillic acid, vanillin, ferulic acid, pinoresinol, and cinnamic acid were identified as phenolic compounds in the analyzed seed oil. Moreover, the oil's phenolics-rich fraction showed a significant total antioxidant activity, scavenged DPPH up to 97.85%, and effectively protected β-carotene against bleaching (97.56%). Conclusion: The results support the potential use of cactus pear seed oil as a functional food.

Effect of Thermophilic Bacteria on Degradation of Food Wastes (음식물 쓰레기 분해에 대한 고온성 미생물의 영향)

  • Food wastes were decomposed into the Mugri (Isung Engineering, Korea), a food waste reduction machine, with adding sawdust of cryptomeria. Degradation effects were better when the machine worked at over 45$^{\circ}C$ than those at the lower temperature. Thermophilic bacteria were isolated from cryptomeria sawdust and the food waste products degraded by the machine. The isolates from cryptomeria sawdust were classified into 3 genera (Acinetobacter baumannii, Enterobacter sp. and Erwinia cypripedii) and almost all the isolates from the degraded products were partially identified as Bacillus sp. by 16S rDNA sequence analysis. The isolated thermophilic bacteria showed degradative enzyme activities. In the case of addition of the 30 thermophilic bacteria into the machine, degradation rate of food wastes was almost twice as high with increasing process temperature up to 6$^{\circ}C$.

An Analysis of Factors Affecting Intention of Farmers to Participate in Integrated Export Organizations

  • Purpose: The integrated export organization for agricultural products is an export marketing organization established by joint participation of famers, farmer organizations, and exporters. The purpose of this study is to analyze the factors affecting the intention of farmers to participate in integrated export organizations. Research design, data and methodology: The multiple regression analysis was used to identify factors affecting intention of pear and mushroom farms to participate in the integrated export organizations. Results: It is shown that the lower farm age, the higher the need for consolidation of export companies, and the lower the government support for the integrated export organizations, the higher the willingness to participate in integrated export organizations. However, export experience, export scale, awareness level of integrated export organizations, and check price level did not significantly affect the intention to participate in integrated export organizations. Conclusions: It is desirable to promote export organizations of agricultural products among relatively young farmers and to reform their consciousness through emphasizing education for them. In addition, integrated export organizations should avoid disorderly competition by consolidating export companies. Lastly, governmental policies should be pursued in a direction that enables farmers and export companies to strengthen their competitiveness on their own.

  • Currently, the overall tendency for green and sustainable economic development is creating a circular economy. In actuality, agricultural output is currently benefiting greatly from the growth of the circular economy. The creation of a circular economy helps address resource scarcity, save the environment, combat climate change, and increase economic efficiency. Vietnam's economy can grow quickly and sustainably by shifting to a circular economy production model. Comparing prior growth techniques to the digital age and implementing circular economic development connected with high technology will be a fantastic opportunity to boost growth efficiency. In actuality, Vietnam currently has a large number of agricultural circular economy models. These are models: Creating and using gas from waste and wastewater in livestock and farming; model combining cultivation, livestock, and aquaculture; agro-forestry model; garden-forest model; Circular model using agricultural by-products as a catalyst or creating other valuable products; model of moderation, linked to reducing the use of growth hormones, veterinary medications, pesticides, and artificial fertilizers in agriculture and animal husbandry. Unfortunately, there have been few studies and applications of the aforementioned models, which has made it difficult to build the agricultural sector sustainably. In this paper, we outline the current situation and propose solutions to develop a circular economy model in agriculture in Vietnam for sustainable development.

  • Termites are destructive to agriculture, forestry, and buildings, but they can also promote agro-ecosystem balance through the degradation of lignocellulose. Termite-triggered cellulose digestion may be clarified through microbial metabolism of cellulose products. In the present study, we characterized the activities of cellulase and its three components synthesized by the cellulase-producing fungal strain HDZK-BYTF620 isolated from the gut of Odontotermes formosanus. The protein components of cellulases were synthesized by strain HDZK-BYTF620, which were isolated and characterized using polyacrylamide gel electrophoresis, and the expression of the cellulases was studied at the proteome level.

  • In Brazil canned 'Biuti' peach is a very popular form of this sub-tropical fruit. This production represents an important economic agro-activity in Minas Gerais, Brazil during the summer period, in preparation for the Christmas celebrations. The aim of this work was to characterize the 'Biuti' peach polyphenoloxidase (PPO), since peach products show enzymatic oxidation of the polyphenols by oxidative enzymes, which affects the products during their shelf life. Two different hypothesis for the browning problem in processed peaches were studied: the inadequacy of the blanching treatment and the presence of a latent phenolase in the peaches. The PPO was characterized: pH optimum (5.5) and stability (5.5-6.5); optimum temperature at $20^{\circ}C$ and 80% of the activity retained after 30 min at $15-40^{\circ}C$. The test for the presence of latent PPO in the processed and canned peaches was negative. Ascorbic acid, ${\beta}$-mercaptoethanol, sodium metabisulfite, and cysteine were efficient in inhibiting the PPO.

The Current Status and Prospect of Sericultural Byproduct Industry in China

  • Sericulture is a traditional agro-industry, which involves mulberry cultivation and silkworm rearing, has made great contributes to the human civilization. With the development of national economy and modem technology, mulberry and silkworm are being used to develop products with functionality besides the traditional cocoon production in China. In this paper, we brief the current developing situation of sericultural byproducts with functionality in the following aspects. (1) Functional products from silkworm larvae: silkworm powder, white muscardine silkworm, isolation and purification of anti-bacterial proteins from the larvae and production of medically valuable substances by Bombyx mori nuclear polyhedrosis virus (BmNPV) vector. (2) Utilization of silkworm feces: for pillow and for isolation of chlorophyll etc. (3) Production of valuable Chinese traditional medicine like Cordyceps sinensis with pupae, functional utilization of pupa protein and chitin. (4) Silk as additives to cosmetics, silk food and medical materials. (5) Functional utilization of mulberry: cultivation of edible fungus on mulberry shoots as medium, mulberry fruit drinks, mulberry tea, etc. The prospect of sericultural byproduct industry in China is also discussed.
