Effectiveness of Rhizobacteria Containing ACC Deaminase for Growth Promotion of Peas (Pisum sativum) Under Drought Conditions

  • A series of experiments were conducted to assess the effectiveness of rhizobacteria containing 1-aminocyclopropane-1-carboxylate (ACC) deaminase for growth promotion of peas under drought conditions. Ten rhizobacteria isolated from the rhizosphere of different crops (peas, wheat, and maize) were screened for their growth promoting ability in peas under axenic condition. Three rhizobacterial isolates, Pseudomonas fluorescens biotype G (ACC-5), P. fluorescens (ACC-14), and P. putida biotype A (Q-7), were selected for pot trial on the basis of their source, ACC deaminase activity, root colonization, and growth promoting activity under axenic conditions. Inoculated and uninoculated (control) seeds of pea cultivar 2000 were sown in pots (4 seeds/pot) at different soil moisture levels (25, 50, 75, and 100% of field capacity). Results revealed that decreasing the soil moisture levels from 100 to 25% of field capacity significantly decreased the growth of peas. However, inoculation of peas with rhizobacteria containing ACC deaminase significantly decreased the "drought stress imposed effects" on growth of peas, although with variable efficacy at different moisture levels. At the lowest soil moisture level (25% field capacity), rhizobacterial isolate Pseudomonas fluorescens biotype G (ACC-5) was found to be more promising compared with the other isolates, as it caused maximum increases in fresh weight, dry weight, root length, shoot length, number of leaves per plant, and water use efficiency on fresh and dry weight basis (45, 150, 92, 45, 140, 46, and 147%, respectively) compared with respective uninoculated controls. It is highly likely that rhizobacteria containing ACC deaminase might have decreased the drought-stress induced ethylene in inoculated plants, which resulted in better growth of plants even at low moisture levels. Therefore, inoculation with rhizobacteria containing ACC deaminase could be helpful in eliminating the inhibitory effects of drought stress on the growth of peas.

Development of CFD model for Predicting Ventilation Rate based on Age of Air Theory using Thermal Distribution Data in Pig House (돈사 내부 열환경 분포의 공기연령 이론법 적용을 통한 전산유체역학 환기 예측 모델 개발)

  • The tracer gas method has an advantage that can estimate total and local ventilation rate by tracing air flow. However, the field measurement using tracer gas has disadvantages such as danger, inefficiency, and high cost. Therefore, the aim of this study was to evaluate ventilation rate in pig house by using the thermal distribution data rather than tracer gas. Especially, LMA (Local Mean Age), which is an index based on the age of air theory, was used to evaluate the ventilation rate in pig house. Firstly, the field experiment was conducted to measure micro-climate inside pig house, such as the air temperature, $CO_2$ concentration and wind velocity. And then, LMA was calculated based on the decay of $CO_2$ concentration and air temperature, respectively. This study compared between LMA determined by $CO_2$ concentration and air temperature; the average error and root mean square error were 3.76 s and 5.34 s. From these results, it was determined that thermal distribution data could be used for estimation of LMA. Finally, CFD (Computational fluid dynamic) model was validated using LMA and wind velocity. The mesh size was designed to be 0.1 m based on the grid independence test, and the Standard $k-{\omega}$ model was eventually chosen as the proper turbulence model. The developed CFD model was highly appropriate for evaluating the ventilation rate in pig house.

Penicillifer diparietisporus: a New Record from Field Soil in Korea

  • A fungus was isolated from field soil collected from Daegu, Korea. The colony of the isolated fungus showed short, branched, and light to dark yellow pigments with hyaline, yellowish red to orange brown aerial mycelia. In addition, the fungus produced solitary to aggregated perithecia, ovoid to pyriform, short neck, and asci as well as biseriately arranged ascospores. Phylogenetic analysis using the internal transcribed spacer region and translation elongation factor $1-{\alpha}$ sequences and morphological characteristics identified the isolated fungus as Penicillifer diparietisporus, which belongs to the family Nectriaceae. To our knowledge, this is the first report of Penicillifer diparietisporus in Korea.

First Report of Anthracnose Disease Caused by Colletotrichum gloeosporioides in Stonecrop

  • In year of 2013, anthracnose disease symptoms were observed on stem of stonecrop in a plant resource field, Southern Forest Resource Research Center, Jinju, South Korea. The disease symptoms were appeared 2-5 cm smudged irregular black spot color on the plant stems. Suspected pathogen was isolated from the lesion with surface sterilization method and identified it as Colletotrichum gloeosporioides by morphological characteristics and phylogenetic analyses. Artificially inoculated the isolated pathogen led to the similar disease symptom as originally observed in the field. The fungal pathogen was re-isolated from the inoculated lesion and it exhibited the same characteristics compared to the original isolate. This is the first report of anthracnose of stonecrop grown in open fields, caused by Colletotrichum gloeosporioides in Korea.

Effect of Ridging System and Mulch Types on Growth, Yield, and Profitability of Potato (Solanum tuberosum L.) in Spring Cropping (감자(Solanum tuberosum L.) 봄재배 시 작휴와 멀칭이 생육, 수량 및 경제성에 미치는 영향)

  • BACKGROUND: This study was conducted to determine effects of ridging systems and plastic film mulch types on growth, yield, and profitability of potato (Solanum tuberosum L.) in spring cropping using paddy field and to suggest profitable ridging system and plastic mulch type.METHODS AND RESULTS: Two potato cultivars ('Goun', and 'Atlantic') were grown at paddy field located in Gangneung in spring. For treatments, ridging systems were one-row ridge (OR) and two-row ridge (TR). Mulch types were no-mulch (NM), black plastic film mulch (BPM), and transparent plastic film mulch (TPM). Emergence of sprout was affected by the mulch types and the fastest level was shown at TPM treatment. Accumulative soil temperature during sprouting was 16.2℃ higher at TR than at OR and also higher at TPM than at BPM or NM. Stem length was not affected by ridging systems or mulch types. Leaf area index (LAI) was influenced by mulch types, while not by ridging systems. The highest LAI was shown at TR with TPM and OR of BPM. There was no significant difference in specific gravity or dry matter rate by ridging systems and mulch types. Tuber yield was significantly influenced by ridging systems and mulch types. Mean tuber weight was heavier at OR treatment than at TR and also the heaviest at TPM among mulch types. The highest marketable yield was found at OR with BPM. Based on marketable tuber yield and market price, the highest income ratio in two cultivars was found at OR with BPM and it was 20~82% higher than the ratio at TR with TPM.CONCLUSION: In spring potato cropping using paddy field, OR with BPM is better for high yield and is more profitable for farmer's income than the conventional cultivation method, TR with TPM.

Quality and Yield Characteristics of Potato (Solanum tuberosum L.) Grown at Paddy Field in Spring Season

  • This study was conducted to determine the characteristics of quality and yield in potatoes grown at paddy field before rice transplantation during the spring season. Three potato cultivars ('Jowon', 'Haryeong', and 'Goun') were grown in Gangneung (asl 5 m) and Seocheon (asl 20 m). In both locations, weather condition belonged to the fourth zone (spring cropping) in potato production location's distribution of Korea. Daily mean soil temperature in both the locations was $0.2-0.6^{\circ}C$ lower than air temperature, while soil moisture was adequate level to potato growth in spite of spring drought. TR ratio was not affected by location, but by cultivar. Specific gravity, starch content, dry matter rate, and yield were significantly influenced by location and by cultivar. There was no difference in total tuber number by location, however there was a large gap in marketable tuber yield according to locations and cultivars. There were high negative relationships between yield and main qualities such as dry matter rate and starch content, while high positive correlation was observed between main qualities. It was possible to produce potato before rice transplanting at drained paddy fields located in representative two locations of potato spring cropping and their characteristics in growth and quality were similar to those generally well known in upland cultivation. Paddy field was thought to be more favorable than upland in terms of available soil moisture supply against spring drought. Further research, however, was needed to increase soil temperature and also preliminary review on proper cultivar according to location seemed to be needed for high yield.

Effects of Electric Current Stimuli and High-Voltage Electric Field Treatments on Brown Rice Germination (전류자극 및 전기장 처리가 현미 발아에 미치는 영향)

  • This study was conducted to investigate the effects of electric current stimuli and high-voltage electric field treatments on brown rice germination. The brown rice stimulated by electrical current stimuli, functional electrical stimuli of a pulse type, and high-voltage electric field treatments were observed (Type I, II and III). Treatment Type I was a method of semi-soaking brown rice with electric current stimuli of 0.13 V/cm, 0.19 V/cm, and 0.25 V/cm into Petri-dishes for 72 hours. Type II was a method of semi-soaking brown rice with functional electrical stimuli of a pulse type(DC 1 V, 1 Hz, 5%, and duty cycles of 5%, 20%, and 35%) into Petri-dishes for 72 hours. Type III was a method of water-soaking with high-voltage electric field treatments for 60 hours. High-voltage electric field treatments at 15 kV/cm were also conducted for 2.5 min, 7.5 min, and 10 min, respectively. The germination rate and the sprout growth of brown rice germinated by electric current stimuli with 0.13 V/cm, 0.19 V/cm, and 0.25 V/cm were increased by about 10-15% compared with those of the control group. The germination rate and the sprout growth of brown rice germinated by functional electrical stimuli of pulse type(DC 1 V, 1 Hz, 5% duty cycle) were increased by about 10∼15% compared to those of the control group. Also, the best effective treatment among high-voltage electric field treatments was the 10 min group at 15 kV/cm. The germination rate and the sprout growth of brown rice germinated by this treatment of 10 min at 15 kV/cm were increased by about 10∼20% compared to those of the control group. The treatments of electric current stimuli and high-voltage electric field accelerated the germination rate and sprout growth of brown rice by about 10∼15% compared to those of the control group.

Effect of Seed Potato Size on Plug Seedling Growth and Field Performance

  • Potato (Solanum tuberosum L.) tubers of 10, 20, 30, 40, and 50 g were planted in plug trays with vermiculite-based root medium on 10 August 2000 and grown for 15 days in a glasshouse to determine the effects of mini-tuber size on plug seedling growth and field performance of plug seedlings. For a control, common potato tubers weighing 50 g were also planted. As size of seed tubers planted increased from 10 to 50 g, seedling height decreased from 24.6 to 20.0cm while shoot number per seedling increased from 2.0 to 3.5, main stem diameter from 4.3 to 6.1mm, and fresh weight of root + top from 9.3 to 19.4 g/seedling. At 90 days after transplanting plug seedlings, the total number of tubers per plant increased from 3.62 to 4.72, average tuber weight from 62.9 to 72.8g, and total tuber yield 20.5 to 23.6 t/ha with increase in seed tuber size. Plug seedlings raised from 50g tubers produced 22% more tubers per plant and had 21 % higher>80g tuber yield than the directly planted potatoes.

  • The environment of agriculture in Korea is changed very rapidly. Since the labours in fruit cultivation field are getting older, the resources and the aid materials for farming need to be multi functional and easy to use. Therefore, They have to use various kind of aid materials in field of fruit cultivation like as fruiting bag. The development of practical utilization way of embossing process for manufacturing of fruiting bag and wrapping paper of agricultural products was main purpose of this study. Embossing roll was designed in Ginyong Embo(Co.) for the embossing process of fruiting bag. The embossing treated fruiting bag was manufactured in Agro(Co.) at mill scale. The mechanical properties of embossing paper was investigated and operation efficiency of bagging was tested at field. The properties of embossed paper was satisfied for fruiting bags for cultivation of apple and pear.

Research Trends in Organic Farming Technology by Journal Article Analysis (논문 분석을 통해 본 유기농업 기술 연구 동향)

  • To investigate the technology trends in organic farming using journal article analysis, total 2,378 published articles in organic farming between 1974 and 2014 were analyzed. The number of the articles published in organic farming has been steadily increased since 2006. As for category, articles in the field of producing the safe organic products and management (A) were published relatively dominantly, especially division of nutrient and soil management (AA). In the foreign countries, lots of articles in environmental assessment and safety field (B) were published than Korea. the main institutes published the articles in organic farming were Swedish University, Sweden and US department of Agriculture, USA. In case of Korea, the articles in the field of environmental assessment and safety (B) were relatively merely published, especially division of environment in organic farming (BA). With this study, more research based on the environment and safety fields in organic farming should be focused on.
