The strategy and current status of Brassica rapa genome project (배추 유전체 염기서열 해독 전략과 현황)

  • Brassica rapa is considered an ideal candidate to act as a reference species for Brassica genomic studies. Among the three basic Brassica species, B. rapa (AA genome) has the smallest genome (529 Mbp), compared to B. nigra (BB genome, 632 Mbp) and B. oleracea (CC genome, 696 Mbp). There is also a large collection of available cultivars of B. rapa, as well as a broad array of B. rapa genomic resources available. Under international consensus, various genomic studies on B. rapa have been conducted, including the construction of a physical map based on 22.5X genome coverage, end sequencing of 146,000 BACs, sequencing of >150,000 expressed sequence tags, and successful phase 2 shotgun sequencing of 589 euchromatic region-tiling BACs based on comparative positioning with the Arabidopsis genome. These sequenced BACs mapped onto the B. rapa genome provide beginning points for genome sequencing of each chromosome. Applying this strategy, all of the 10 chromosomes of B. rapa have been assigned to the sequencing centers in seven countries, Korea, UK, China, India, Canada, Australia, and Japan. The two longest chromosomes, A3 and A9, have been sequenced except for several gaps, by NAAS in Korea. Meanwhile a China group, including IVF and BGI, performed whole genome sequencing with Illumina system. These Sanger and NGS sequence data will be integrated to assemble a draft sequence of B. rapa. The imminent B. rapa genome sequence offers novel insights into the organization and evolution of the Brassica genome. In parallel, the transfer of knowledge from B. rapa to other Brassica crops would be expected.

  • This research estimates the necessity of a better governance plan on the purpose of fulfillment energy recovery by building resource recycling system for biomass resources and waste resources that derive from agricultural and mountain village areas. The utilization of new renewable energy technology which uses waste and biomass sources diverse as variety of resources, collecting method, operator etc. and is structurally complicated the formation of policy is also very difficult. There is failure because of the problems which occurs from the policy led by government. Biomass Town Development Project should be made through the central government and the local government integrated support system and should be formed a consultative group in order to process the project mutually with these two department including the experts from the related areas. This consultative group, while government organizations carry out the hub function of strategic knowledge management, should carry out the control tower function to be able to be net working transfer the information with the cooperation of private and government so vitalize the communication area among the related actors. And to be able to increase the participation rate of the local people the consistent and various educations should be given so a smooth business promotion progress will be desired through the change of perception and coactive participation of people.

The assessment of conservation value for agricultural pond wetland using the weighted function of modified RAM (modified RAM의 기능별 가중치 부여를 통한 농촌지역 연못형습지의 보전가치 평가)

  • The pond wetlands in agricultural landscape are important natural resources that carry out the function of bio-diversity conservation. However, recently, those have been gradually embedded as their utility value were disappeared. And, the assessment methods used for pond wetlands are insufficient. Therefore, this study was conducted to examine the conservation value of pond wetlands by using the modified RAM, and present the improvement of assesment methods. The study sites, a total of 32 pond of 4 types by land use, were selected on the basis of Ramsar Convention. Through the analysis of precedent studies, the weighted 8 functions were adjusted. According to the assessment results, pond wetlands made the largest contribution to Fishery and Herpetile Habitat function. In addition, it also made large contribution to Floral Diversity, Wildlife Habitat, and Water Quality Protection function. On the other hand, it made a small contribution to Aesthetics and Recreation, Runoff Attenuation, Shoreline /Stream Bank Protection, and Flood/Storm Water Storage function due to the characteristics of small-scale pond wetlands. In the assessment of 8 functions, house type showed the worst assessment result, and mountain type showed the best assessment result. It is thought that those are due to land use type in terms of vicinity. 10 items among 52 of the modified RAM showed the same assessment results in all land use types. Accordingly, it is required to be deleted and modified the assessment method. On the other hand, it is required to add age, interference, and water use to the assessment method. It is thought that these results can be utilized for the development and modification of assessment methods focused on pond wetlands in rural area.

Interspecies Transfer and Regulation of Pseudomonas stutzeri A1501 Nitrogen Fixation Island in Escherichia coli

  • Until now, considerable effort has been made to engineer novel nitrogen-fixing organisms through the transfer of nif genes from various diazotrophs to non-nitrogen fixers; however, regulatory coupling of the heterologous nif genes with the regulatory system of the new host is still not well understood. In this work, a 49 kb nitrogen fixation island from P. stutzeri A1501 was transferred into E. coli using a novel and efficient transformation strategy, and a series of recombinant nitrogen-fixing E. coli strains were obtained. We found that the nitrogenase activity of the recombinant E. coli strain EN-01, similar to the parent strain P. stutzeri A1501, was dependent on external ammonia concentration, oxygen tension, and temperature. We further found that there existed a regulatory coupling between the E. coli general nitrogen regulatory system and the heterologous P. stutzeri nif island in the recombinant E. coli strain. We also provided evidence that the E. coli general nitrogen regulator GlnG protein was involved in the activation of the nif-specific regulator NifA via a direct interaction with the NifA promoter. To the best of our knowledge, this work plays a groundbreaking role in increasing understanding of the regulatory coupling of the heterologous nitrogen fixation system with the regulatory system of the recipient host. Furthermore, it will shed light on the structure and functional integrity of the nif island and will be useful for the construction of novel and more robust nitrogen-fixing organisms through biosynthetic engineering.

The Effect of Field-Based Participation Design on Agricultural Direct Sales Facilities: Process Change of Awareness on Participant (농특산물 직거래판매시설에 관한 현장중심의 참여디자인 효과: 참여자의 의식변화를 중심으로)

  • A demand for user's participation in the designing process, which is called the participation design, is increasing with social change. Practically, however, there is lack of researches on the improvement of the problem in actual design development and participation design effect. In order to understand the field-based participation design effect in view of user's change of awareness on participation design, this study carried out a survey on the change of awareness through a design workshop in which all stakeholders have participated on the subject of agricultural direct sales facilities as a part of participation design programs. Many references were reviewed first to understand the participation design principles and their current research trend. Next, the questionnaire was developed using an effectiveness verification model for an effective analysis. A design workshop as one of the participation design programs was organized and applied for the field response. In order to analyze the effect, the survey on participants' change of awareness was carried out before and after the program. The survey questionnaires were about motivation, knowledge, recognition, function, and participation and asked the design study graduates. The participation design effect was supplemented by additional open questions. As a consequence, the participants in the participation design program showed the noticeable effect in knowledge and function about the agricultural direct sales facilities. It was found that they were also highly satisfied with the participation design result and they recognized an importance of collaboration. This study also showed that a sufficient time schedule considering agricultural village life should be given for a smooth operation of the program and also a working environment is necessary for designing work.

  • The purpose of this study was to analysis the education effects of web-based on-line cyber program mesaured by Kirkpatrick’s evaluation process. The average score on satisfaction of the program was 4.28(.59), which was designed to evaluate the level 1, reaction. To test level 2, learning, the average score that students achieved was calculated and it was 86.87(std.=7.05) in the term examinations. The level 3, job months. It was reported that most employees who took the course are utilizing the knowledge that they acquired from the course(mean=3.80, std.=.77). To identify the level 4, business results, the mean score of the number of accidents and near misses that happened in their factories for 3 months before and after the course were compared. There was statistically significant difference between the number of accidents that happened 3 months before the course and 3 months after the course, at the significance level of .01, which was tested by Paired t-test.

Factors Affecting the Extent of Economic Empowerment of Women in Farm Households: Experiences from Rural Bangladesh

  • The study identifies gender stereotypes, examines the level of women's economic empowerment at the household level and explores the influence of factors on it. Data were collected from 159 randomly selected farm women using both qualitative and quantitative survey methods between January and March 2007 from three villages of the Mymensingh District of Bangladesh. Four key informants (2 local leaders and 2 development personnel) were questioned to elicit views in the light of boosting women's empowerment. Five constructs of empowerment covering 30 indicators were aggregated together to develop a cumulative economic empowerment index (CEEI) to obtain multidimensional views of women's empowerment. The findings show that there were some prejudices against women in allocating divisions of labour and access to education, food, property, decision-making and institutions. The distribution of the CEEI demonstrates that the majority of the respondents (86%) had a low to moderate level of empowerment. A multiple regression analysis showed positive significant effects of education, training, media contact and freedom of mobility on women's CEEI, while domestic abuse restrained it. It is concluded that interventions by development agencies in co-ordination with the local community was necessary to attain women's self-reliance in the study area. Development actors can undertake some core strategies to enhance women's level of awarencess, knowledge, skills, and productive resources through providing training, loans, and information. To change traditional beliefs, it is important to create awareness of various gender issues amongst rural people through different methods and media.

A study to improve the national technical qualification practical evaluation method of National Competency Standards in the field of organic agriculture

  • The purpose of this study is to suggest an improvement plan for the National Technical Qualification Practical Evaluation focusing on National Competency Standards. First, in order to strengthen practical techniques such as field abilities and expertise, practical evaluation methods were applied to the National Competency Standards, referring to a learning module focused on field performance. Second, an expert advisory committee in the field of organic agriculture was utilized to compare, analyze, and match the competency unit and competency unit elements of the National Competency Standards with the national technical qualification examination standards in field of organic agriculture. In addition, in order to identify work in progress in the industrial field, share professional practice skills, and analyze facilities and equipment, we visited an organic agriculture industry site. Through this, a practical evaluation exam was developed. Subsequently, in order to confirm the adequacy of the developed exam, the pilot test assessment was conducted for those majoring in organic agriculture. Finally, as a feasibility study, a survey was conducted. Based on the results, greater technical job competency in the field of organic agriculture will contribute to strengthening knowledge in this field by suggesting an improvement plan for the national technical qualifications practical evaluation method in the field of organic agriculture.

A study of the improvement of the national technical qualification practical evaluation method for National Competency Standards in the seed and horticulture industry

  • The purpose of this study was to suggest an improvement plan ultimately to develop practical evaluation methods for national technical qualifications in the field of seeds and horticulture based on the National Competency Standards. Through this, national technical qualifications can be strengthened in terms of professionalism in the seeds and horticulture field. In order to conduct this study, the national technical qualification currently in use was matched to certain competency units and competency unit elements of National Competency Standards. We then visited an industrial site to understand the practical factors related to the technical, facility and equipment aspects of the seeds and horticulture industrial field, after which a practical evaluation exam was developed. Also, for a feasibility study, a pilot test assessment was conducted for students majoring in seeds and horticulture, with a survey also conducted with field experts and an advisory committee consisting of experts in seeds and horticulture. Based on the results, by suggesting an improvement plan for a practical evaluation method leading to better national technical qualifications in the field of seeds and horticulture, it was shown to be possible to improve knowledge, technical, and literacy aspects. This can lead to the training of outstanding technical professionals. Ultimately, the results here are expected to contribute to the improvement of the seeds and horticulture industry.

  • Capacity building for bioinformatics could be achieved with the systematic training of research staff and higher degree students in the current best practices for analysis of data from 'omic-type experiments. It is anticipated that the KOICA-University of the Philippines Los Baños - International Rice Research Insitute Agricultural Genomics Research Center activities will focus on the use of next generation sequencing technology for genome sequencing and annotation, genome variant discovery for use in GWAS and QTL mapping, and transcriptome analysis of organisms important to agriculture and food security. Such activities require that researchers have high levels of knowledge and skills in bioinformatics in order to gain insights from the results of the experiments performed. In this talk the bioinformatic tools/solutions and online training materials already available will be presented, as well the upcoming resources under development in support of the project.
