Analysis on Growth and Yield of Cherry Tomato Grown in a Two-Story Bed System Adapted to Strawberry Cultivation as Affected by the Planting Time during the Uncultivated Period (딸기 재배용 2단 베드 시스템에서 휴작기 이용 방울토마토 재배 시 정식 시기에 따른 생육과 생산성 분석)

  • This study was conducted to determine the yield of cherry tomato (Solanum lycopersicum Mill.) grown at three planting times during the uncultivated period of strawberry. Cherry tomato was planted under condition filled with strawberry dedicated culture medium on a two-story bed with April 20, April 30, and May 10 at 2015. Fruit harvest was completed on July 31. The supply concentration of nutrient solution at the time of transplanting was started as EC $1.2dS{\cdot}m^{-1}$ and it was gradually increased to EC $2.5dS{\cdot}m^{-1}$ after blooming of the first flower cluster. Netherlands PBG solution was supplied for one minute six times per day. The heights of cherry tomato plants planted at earlier were significantly greater than those of tomato plants planted later. The heights of cherry tomato plants grown at the bottom of the bed were greater than those grown in the upper bed. The yield of cherry tomatoes planted on April 20 at the bottom of the bed was greatest with an average of 2,954 g of tomatoes per plant. There were no significant differences in the average weight and sugar content of fruit according to planting times and bed position. The yield of cherry tomato plants planted on April 20 was 18% and 34% higher than that of plants planted on April 30 and May 10. We confirmed to increase the yield of the cherry tomato when early plants planted on two-story bed. These results indicate that farmers can choose the best period of producing cherry tomato during the un-cultivated period of strawberry under two-story bed conditions.

Rapid Analysis of Nitrate Concentration in Different Growth Stages and Plant Parts of Paprika Leaf Using On-site Rapid Detection Kit (신속분석기기를 이용한 파프리카 생육단계 및 부위별 엽내 질산태질소 농도 신속분석)

  • This research was aimed to establish rapid analysis technique for the determination of nitrate ($NO_3{^-}$) concentration in the leaves of paprika, which has key role for the stable vegetative and reproductive growth. Leaf petiole and blade sap of two paprika cultivars ('Raon red' and 'Raon yellow') were used for the determination of $NO_3{^-}$ concentration, separately using rapid detection kit (RQ-flex) and spectroscopy quantification methods. In addition, two paprika cultivars namely, 'Nicole' and 'TP2001' were used to determine the status of $NO_3{^-}$ concentration in leaf of each fruiting group. $NO_3{^-}$ concentration in leaf blade sap and the content in leaf showed significant correlation ($R^2=0.8628$), analysed by RQ-flex and spectroscopy methods, respectively. Furthermore, leaf petiole sap and the content in leaf also showed significant correlation ($R^2=0.6734$) but the relationship was poor compared to leaf blade sap and the leaf content. $NO_3{^-}$ concentration in petiole sap decreased in all the cultivars from early to late fruiting group. The higher concentration in the lower leaves and the continuous decrease towards the upper leaves in the both years were found through the analysis of $NO_3{^-}$ concentration in different leaf position. In addition, daily short-term fluctuation of $NO_3{^-}$ in petiole sap could be rapidly monitored. These results showed that long-term or short-term monitoring by test strip-based rapid analysis technique might be useful tool for the diagnosis of nutritional status for the stable of nutritional management in paprika.

Effects of Selenium Supplying Methods on the Growth and Se Uptake of Hydroponically Grown Tomato Plants (Selenium공급방법이 수경재배 토마토의 생장과 Se 흡수에 미치는 영향)

  • This study was conducted to clarify the effects of supplying methods of selenium on the growth and Se uptake of hydroponically grown tomato plants. Tomato seeds (Lycopersicum esculentum Mill. cv. Momotaro T-93, Daki Seed Co.) were sown in plug tray with fifty holes, and raised for sixty days. Tomato seedlings transplanted to coco fiber slabs were supplied with the nutrient solutions adjusted to EC $2.3dS{\cdot}m^{-1}$ and pH $5.8\~6.2$ recommended by the Japanese Horticultural Experiment Station. Selenium forms used were inorganic $SeO_2$ (here in after referred to Se) and organic selenium chlenium with sugar fatty acid ester (here in after referred to chelated-Se). 10 ppm selenium solutions were treated to tomato plants with foliar applications, drenching, and foliar application plus drenching. Growth characteristics in terms of plant height, number of leaves, leaf area and chlorophyll content were significantly increased in the plot of foliar application ot Se, and in the plot of foliar application plus drenching of chelated-Se than other plots, respectively. Transported contents of selenium into the tomato fruits were highest as 0.302 ppm in the plot of foliar application plus drenching of chelated-Se. Also, it had tended to be higher in the plot of foliar application plus drenching than in the plots of foliar application or drenching in both of Se and chelated-Se. Foliar application and drenching of organic chelated-Se were effective to produce the functional tomato fruits.

Changes in the Physicochemical Characteristics of Green Wheat during Maturation (성숙과정 중 녹색밀의 이화학적 특성변화)

  • In order to evaluate the utilization of immature green wheat kernels as food processing material, these experiments were carried out to analyze general composition, free sugars, free amino acids, mineral content and color of 3 wheat varieties according to their heading dates. Contents such as moisture, crude protein, crude lipid and ash were gradually decreased according to the days after heading of 3 wheat varieties, while crude fiber and starch were gradually increased. Maltose and maltotriose contents in immature green wheat kernels were high but its rapidity decreased by closing to ripeness. Contents of free amino acids showed a tendency to decrease, among which alanine content was the highest around 25 days after heading, and then glutamic acid, GABA, glycine in order. Contents of total free amino acid were decreased by closing to ripeness. Among 3 wheat varieties, its contents of Guru wheat was the highest. As wheat matured, ${\beta}-glucan$ contents were decreased from 1.5% to 0.28% on 43 days after heading. Mineral contents of 3 wheat varieties were generally increased by closing to ripeness after coming into ears, and K, Mg were especially prominent. As wheat matured, lightness was decreased, while redness and yellowness were increased. Considering chemical composition and color, the immature green wheat were produced through blanching the spikes harvested before the yellow ripe stage. The harvested green wheat cereals is able to eat raw wheat or cook it as food processing material.

Identification and Characterization of Three Isolates of Cucumber mosaic virus Isolated from Weed Hosts (잡초에서 분리한 3종 Cucumber mosaic virus의 동정과 특성)

  • Three isolates of Cucumber mosaic virus (CMV) were isolated from weed hosts showing typical mosaic symptoms, and some properties of the viruses were investigated. CMV isolates, designated as Is-CMV, Jd-CMV and Pla-CMV from Isodon inflexus, Jeffersonia dubia and Phryma leptostachya var. asiatica, respectively, were identified and characterized by biological reaction in several host plants, serological property, dsRNA analysis, reverse transcription-polymerase chain reaction (RT-PCR), restriction fragment-length polymorphism (RFLP). All isolates systemically infected in Nicotiana benthamiana, Cucurbita pepo cv. Black beauty and Cucumis sativus, and did not reveal any differences in these host plants between the isolates. However, remarkable difference in the symptoms was found between the CMVs in Capsicum annuum. Is-CMV induced an asymptomatic symptoms, while Jd-CMV and Pla-CMV produced severe mosaic symptoms in C. annuum plants. In dsRNA analysis, all isolates revealed four major bands with estimated molecular size of 3.4, 3.2, 2.1 and 1.0 kbp. The cDNAs of coat protein gene of the isolates were amplified by RT-PCR using a genus-specific single pair primers that designed to amplify a DNA fragment of approximately ranging from 938 to 966 bp. By restriction mapping analysis using RFLP of the RT-PCR products as well as by serological properties of gel diffusion test, the CMV isolates belong to a typical members of CMV subgroup IA. This is the first report on the occurrence of CMV in the three weed hosts.

Effect of Red LED, Blue LED and UVa Light Sources on Coloration of Paprika Fruits (적색 및 청색 LED광과 UVa광 처리가 착색단고추 과실의 착색에 미치는 영향)

  • This study was conducted to compare the effect of various LED and UVa light to enhanced coloration of paprika fruits cultivars; 'Special', 'Helsinki'. Immatured paprika fruits were irradiated with red LED, blue LED and UVa lights which were high intensity (red: $50{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, blue: $70{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, UVa: $3{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and low intensity (red: $20{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, blue: $40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, UVa: $0.25{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$). The coloration of 'Special' cultivar was increased the fastest in high intensity UVa light for 2 days after treatment. There were no significantly difference among treatments except the low intensity UVa light. The coloration of 'Helsinki' was increased the fastest in blue LED light, and followed by red LED light and UVa light for 2 days after treatment. The $a^*/b^*$ value of 'Special' and $b^*$ value of 'Helsinki' that indicated the paprika fruit coloration were generally higher high intensity LED and UVa lights than low intensity LED and UVa lights. We couldn't find out any significantly different coloration effect among all treatments in final days of coloration. and rather than UVa treatment fruit shrunk the surface of paprika fruits.

Reaction of Near-Isogenic Lines with Resistance to Bacterial Blight to Korean and Japanese isolates of Xanthomonas oryzae pv. oryzae (한국 및 일본 균주에 대한 벼흰잎마름병 저항성 근동질 유전자 계통의 반응)

  • To develop durable and stable breeding strategies far the control of rice bacterial blight (BB) disease, the near-isogenic lines (NILs) with diverse resistance genes to BB isolates were evaluated in plant at three different growth stages using four Korean ($K_1,\;K_2,\;K_3,\;K_{3a}$) and three Japanese isolates (I, II, III). The resistance of the NILs to seven BB isolates tended to increase with plant aging. At the seedling stage, the NILs with single resistance genes were mostly resistant to $K_1$ race but they showed partial or no resistance to other isolates. A NILs (IRBB5) possessing xa5 had full resistance to the four Korean isolates, illustrating that it is a useful source to give enhancement to Korean breeding program. At the maximum tillering stage, resistance of NILs to $K_2,\;K_3$, I and II isolates considerably increased while resistance to $K_1,\;K_{3a}$ and III were similar to those of seedling stage. NILs with resistance gene of Xa4, xa5 and Xa7 proved to be the most stable to BB isolates at the maximum tillering stage. At the heading stage, most resistance genes of NILs were effective against BB isolates, and xa5 showed the consistent resistance to all the BB isolates including $K_{3a}$ and III isolates, demonstrating that resistance genes of Xa4, xa5 and Xa7 can be used either alone or combined to enhance resistance to BB disease in Korea.

Effect of LED as Light Quality on the Germination, Growth and Physiological Activities of Broccoli Sprouts (LED 광질이 브로콜리 새싹의 발아, 생장 및 생리활성에 미치는 영향)

  • This study was carried out to investigate into the effect of light-emitting diode (LED) for the light quality as a light source on the broccoli seed germination and the physiological activity of vegetable sprouts. We have also germinated seeds of the broccoli and applied LED as a light quality such as blue, green, red, white, yellow and red + blue color lights to their sprouts for 14 hours and kept dark for 10 hours at the temperature of $25^{\circ}C$ (day)/$18^{\circ}C$ (night). Broccoli sprouts were extracted by methanol and their physiological activities were examined. All broccoli seeds were germinated at 3 days after seeding regardless of the light color. Total sprout fresh weight were mostly became highest by 0.389g (10 plants) at 8 days after seeding when their sprouts were grown under blue color light. Total phenol compound contents in broccoli sprouts were extremely increased by $83.0\;mg{\cdot}L^{-1}$ under the white light, and total flavonoid contents were most much more by $72.6\;mg{\cdot}L^{-1}$ under the blue light. DPPH radical scavenging activity at $2,000\;mg{\cdot}L^{-1}$ were most highest by 93.5% in broccoli sprouts grown under the white light. Nitrite radical scavenging activity at the concentration of $500\;mg{\cdot}L^{-1}$ in sprout extracts were the most increased by 66.9% under the yellow light, and tyrosinase inhibition activity at $2,000\;mg{\cdot}L^{-1}$ in sprout extracts were by 14.5% under red light.

Changes in Major Constituents by Extracting of Acanthopanax koreanum Root with Water and Ethanol Solution (탐라오가피 뿌리의 에탄올 추출 중에 유용성분의 변화)

  • In older to prepare functional food materials from Acanthopanax koreanum, changes of major constituents by extracting with water and ethanol were investigated Extracting 300 g of below 0.5 cm size dried sample in 7.5 L of water or $30{\sim}95%$ ethanol for 9 hr at $100^{\circ}C$ were carried out pH during extraction was between 4.0 and 6.5. Color b-value of extracts was increased according to lower ethanol concentration and longer extraction time. Color a-value and b-value was increased more in stem than in root Extracts were increased rapidly within $2{\sim}3\;hr$. The extract in $30{\sim}70%$ ethanol was $0.84{\sim}1.34%(w/v)$ with root Main free sugar of extracts was sucrose in root. The eleutherosides were extracted rapidly within 3 hr, moreover were increased in water or $30{\sim}70%$ ethanol more than 95% ethanol concentration. Extraction of acanthoic acid from root was more affected on ethanol concentration than extracted time, moreover it was detected only trace by extracting with water. Furthermore, acanthoic acid was extracted rapidly within 2 hr in $50{\sim}70%$ ethanol, and was extracted 3 times higher with 70% ethanol than with 30% ethanol. The content of acanthoic acid in residue after extraction was affected largely by extraction solvents. The extraction efficiency in 70, 50 and 35% of ethanol concentration was about 95, 90 and 35% respectively. The eleutherosides were extracted to 95% with water or nature of water and ethanol. Therefore, the reflux extraction in $40{\sim}70%$ ethanol concentration for $3{\sim}5\;hr$ was adequate for extraction of functional materials from Acanthopanax koreanum.

Effect of Collection Seasons on the Oocyte Quality and Developmental Competence of Oocytes Derived from Korean Native Cows (Hanwoo) by Ovum Pick-Up (OPU 채란계절이 한우의 난자 품질 및 발달 능력에 미치는 영향)

  • Implementation of smart embryo technologies in cattle e.g. ovum pick-up followed by in vitro embryo production (OPU-IVP). Seasonal variation is important factor for follicular growth, oocytes quality, quantity and developmental competence. Therefore the aim of present study was carried out to investigated whether the seasons (hot and cool) effect on follicular development, oocyte recovery and subsequent embryo development. Follicular oocytes were aspirated from Korean native cows (Hanwoo) by the ovum pick-up (OPU) method, which was performed 24 times during two different seasons, the hot (July to September) and cool (October to December), from OPU donors. The recovered oocytes were classified according to morphological categories and used for in vitro embryo production (IVEP). The mean number of total follicles was significantly higher (p<0.05) during the hot season ($18.32{\pm}2.26$) compared to cool season ($15.41{\pm}3.34$). Furthermore, seasons did not significantly effect on the number of oocytes recovered (hot season: 41.16% vs. cool season: 46.14%). However, the average number of Grade A oocytes was significantly greater during hot ($1.75{\pm}1.86$) season compared to the cool season ($1.00{\pm}1.46$), but there was no significant difference of other grades oocytes. The cleavage rate (hot: 66.67% vs. cool: 63.3%) and embryo development (hot: 58.95% vs. cool: 56.97%) did not differ significantly between the seasons. In conclusion, the results of present study suggest that the season (hot and cool) does not have effects on the oocyte recovery and embryo developmental competence of in vitro cultured embryos.
