새싹채소의 생리활성 특성 및 그 가공품의 제조 (Physiological Characteristics and Manufacturing of the Processing Products of Sprout Vegetables)

  • 무, 적양배추, 청경채, 메밀 및 브로콜리 씨앗을 이용하여 새싹채소 및 성장이 완료된 일반채소를 재배하였다. 이들에 대한 비타민 함량을 측정한 결과 각 시료에 따라 큰 편차를 나타내었으며, 비타민 $B_1$은 싹이 트면서 그 함량이 감소하거나 검출되지 않았다. 이는 비타민 $B_1$이 성장에 사용되는 영양소이기 때문인 것으로 판단된다. 새싹채소의 무기물 함량은 일반채소 보다 2~10배 높게 나타났다. 새싹의 성장에 다량 이용되는 무기물은 칼륨이었고, 나트륨은 가장 적게 소비되었다. 일반채소에 비하여 새싹채소가 그 $\alpha$-amylase 활성 및 종합당화력이 높은 것으로 나타났으며, 이러한 경향은 이들을 이용하여 가공한 동결건조분말에서도 동일한 경향을 나타내어 전체적으로 싹 틔움에 따라 효소활성도가 비례적으로 증가함을 확인할 수 있었다. 새싹채소의 장점은 각종 영양소가 풍부하고, 가정에서 직접 재배가 가능하여 농약이나 화학처리를 하지 않기 때문에 안심하고 먹을 수 있다는 점인 것으로 판단된다.

Changes in the Total Lipid, Neutral Lipid, Phospholipid and Fatty Acid Composition of Phospholipid Fractions during Pastirma Processing, a Dry-Cured Meat Product

  • Pastirma is a dry-cured meat product, produced from whole beef or water buffalo muscles. This study was carried out to investigate the effect of production stages (raw meat, after curing, after $2^{nd}$ drying and pastirma) on the total lipid, neutral lipid, phospholipid and fatty acid composition of phospholipid fraction of pastirma produced from beef M. Longissimus dorsi muscles. The pH and colour ($L^*$, $a^*$ and $b^*$) analyses were also performed in raw meat and pastirma. It was found that pastirma production stages had significant effects (p<0.01) on the total amounts of lipid, neutral lipid and phospholipid, and the highest amounts of lipid, neutral lipid and phospholipid were detected in pastirma. In pastirma, neutral lipid ratio was determined as $79.33{\pm}2.06%$ and phospholipid ratio as $20.67{\pm}2.06%$. Phospholipids was proportionately lower in pastirma than raw meat. Pastirma production stages affected pentadecanoic acid (15:1) (p<0.01), linoleic acid (18:2n-6) (p<0.05), ${\gamma}-linoleic$ acid (18:3n-6) (p<0.05), erucic acid (22:1n-9) (p<0.05), docosapentaenoic acid (22:5n-6) (p<0.05), total unsaturated fatty acid (${\Sigma}USFA$) (p<0.05) and total saturated fatty acid (${\Sigma}SFA$) (p<0.05) ratios of phospholipid fraction and also the moisture content (p<0.01). Pastirma process also affected pH and colour ($L^*$, $a^*$ and $b^*$) values (p<0.01), and these values were higher in pastirma than raw meat.

Valencia 오렌지에서 내열성 및 비내열성 Pectinesterase 분리 정제 (A Study on Separation of Thermolabile and Thermostable Pectinesterase from Valencia Orange)

  • Valencia 오렌지로부터 추출한 펙틴에스테라제(pectinesterase, PE) 중에서 가열처리하지 않은 PE 조효소를 CM-Sephadex C-50, Phenyl Sepharose CL-4B와 CM-Biogel A의 chromatography를 차례로 수행하여 내열성이 약한 TLPE(thermolabile pectinesterase)와 내열성이 강한 TSPE(thermostable pectinesterase)를 분리 정제하였다. PE 중에서 가열처리한 PE 조효소에서는 위와 같은 정제과정으로 TSPE 만을 분리하였다. TLPE는 비활성 1,005 units/mg이고 35배로 정제된 것을 13.6% 회수할 수 있었으며, 반면에 가열하지 않고 분리한 TSPE는 비활성 3,115 units/mg이고 100.5배로 정제된 것을 1.5% 얻을 수 있었으나 가열하여 분리한 TSPE는 비활성 1,803 units/mg이고 가열처리한 조효소가 140배로 정제된 것을 15.4% 얻을 수 있었다.

  • 쌀보리의 합리적인 제분 방법을 규명하고저 tempering 최적조건 부분 도정후의 제분성, 밀과의 혼합 제분성 등을 test mill 을 이용, stock stream 별로 검토해 본 결과 다음과 같은 결과를 얻었다. 1. 쌀보리의 최적 tempering 조건으로서는 수분 13. 5%에서 48시간 일차 tempering 을 한 후 이어 제분 30분전 0. 5% 가수 처리하는 것이 가장 유리하였다. 2. tempering 전후 부분 도정을 실시하므로써, 회분 함량은 저하시킬 수 있었으나 수율이 크게 저하되고, 부분도정 후 제분이란 두 단계의 가공 과정이 필요하므로 비효율적이었다. 3. 쌀보리를 밀과 혼합하여 제분할 때, 쌀보리의 혼합비율은 $10{\sim}20%$가 합리적이며, 이때 사용되는 원료 밀은 연질보다 경질의 밀이 더욱 유리하였다.

Characterization and Comparative Evaluation of Milk Protein Variants from Pakistani Dairy Breeds

  • The aim of study was to scrutinize the physicochemical and protein profile of milk obtained from local Pakistani breeds of milch animals such as Nilli-Ravi buffalo, Sahiwal cow, Kajli sheep, Beetal goat and Brela camel. Physicochemical analysis unveiled maximum number of total solids and protein found in sheep and minimum in camel. Buffalo milk contains the highest level of fat (7.45%) while camel milk contains minimum (1.94%). Ash was found maximum in buffalo (0.81%) and sheep (0.80%) while minimum in cow's milk (0.71%). Casein and whey proteins were separated by subjecting milk to isoelectric pH and then analyzed through sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). The results showed heterogeneity among these species. Different fractions including αS1, αS2, κ-casein, β-casein and β-lactoglobulen (β-Lg) were identified and quantitatively compared in all milk samples. Additionally, this electrophoretic method after examining the number and strength of different protein bands (αS1, αS2, β-CN, α-LAC, BSA, and β-Lg, etc.), was helpful to understand the properties of milk for different processing purposes and could be successfully applied in dairy industry. Results revealed that camel milk was best suitable for producing allergen free milk protein products. Furthermore, based on the variability of milk proteins, it is suggested to clarify the phylogenetic relationships between different cattle breeds and to gather the necessary data to preserve the genetic fund and biodiversity of the local breeds. Thus, the study of milk protein from different breed and species has a wide range of scope in producing diverse protein based dairy products like cheese.

Milk Conjugated Linoleic Acid (CLA) Profile and Metabolic Responses of Dairy Cows Fed with High-temperature-micro-time (HTMT) Treated Diets Containing High Quantity Extruded Soybean (ESB)

  • A feeding trial was conducted to examine the effect of high-temperature-micro-time (HTMT) processing of diets containing extruded soybean (ESB) in high quantity on milk fat production, metabolic responses, and the formation of conjugated linoleic acid (CLA) and trans-vaccenic acid (TVA). Twenty-one multiparous Holstein cows in mid-lactation were blocked according to milk yield in the previous lactation. Cows within each block were randomly assigned to either normal concentrate or HTMT treated diets containing ESB (7.5% HTMT-ESB and 15% HTMT-ESB). It was hypothesized that the HTMT-ESB would affect the undegradable fatty acids in the rumen and, thus, would modify the fatty acid profile of milk fat. Both 7.5% and 15% HTMT-ESB did not affect milk yield, fat, protein, lactose and solid-not-fat (SNF), but the proportion of cis-9, trans-11 CLA in milk fat was significantly increased by these treatments. Content of TVA in milk fat was not affected by HTMT-ESB. The HTMT-ESB influenced the fatty acid profile in milk fat, but there was little difference between 7.5% and 15% of supplementation. HTMT-ESB feeding significantly decreased the concentration of plasma insulin and glucose, while plasma growth hormone (GH), triglyceride (TG), non-esterified fatty acid (NEFA) and HDLcholesterol were increased by 7.5% and 15% ESB-HTMT supplementation in comparison to the control group (p<0.05). However, no significant difference was observed in plasma LDL-cholesterol, insulin like growth factor (IGF)-1, T3, T4, and leptin concentrations among treatments (p>0.05). The present results showed that cis-9, trans-11 CLA production was increased by HTMT treatment of dietary ESB without reduction of milk fat, and the unchanged milk fat and yield was assumed to be associated with the constant level of thyroid hormones, leptin, and IGF-1.

금감 가공식품(加工食品)의 제조(製造)와 품질특성(品質特性) (Manufacture of Processed Foods and its Quality Characteristics from Kumquats, a Citrus Variety Produced in Cheju)

  • 제주산 금감을 소재로 원료특성, 냉동금감의 기호성, 금감잼 등의 가공식품의 제조와 특성 등을 검토하였다. 시설재배한 영파금감은 과실의 청경이 29.5 mm, 과중이 16.10g, 껍질의 두께가 4.63 mm, 단산비가 20으로서 생식용 및 가공용 금감으로서 우수하였다. 유기산 함량은 citric acid 58.27%, malic acid 32.49%, tartaric acid 9.14%, maleic acid 0.08%로서 malic acid 함량이 높아 온주밀감쥬스와는 매우 다른 특성을 나타내었다. $-18^{\circ}C$에서 6개월간 저장한 냉동금감에 대한 기호성에서 외관, 맛, 물성 모두 보통 이상으로 평가되어 좋은 것으로 판단되었으며, 냉동금감의 비타민 C 함량은 74.94 mg/100g으로 법동기간 중에 초기에 감소하다가 후기에는 큰 변화를 보이지 않았다. 오렌지쥬스만으로 제조한 잼의 경도는 1,294 g-force이며, 금감쥬스의 혼합비율이 높을수록 경도는 비례적으로 감소하였고 다른 texture parameter는 큰 차이를 보이지 않았다. 관능검사 결과, 금감쥬스를 $40{\sim}60%$, 함유한 금감잼은 오렌지쥬스 만으로 제조한 잼에 비하여 기호성이 우수하였다.

음파를 이용한 비파괴 수박 내부품질 판정 시스템 개발 (Development of Nondestructive Evaluation System for Internal Quality of Watermelon using Acoustic Wave)

  • 음파를 이용한 수박의 내부품질을 비파괴적으로 판정하는 시스템을 개발하였다. 개발된 수박품질판정기는 크기가 $200{\times}200\;mm$, 무게가 290 g로 휴대가 가능하여 수박 재배포장, 집하장, 슈퍼마켓 등 현장에서 쉽게 사용가능하도록 개발하였다. 수박의 음파수집한 후 음파데이터에서 영교차율, 피크(peak)점간 거리 표준편차, 시간파형에서 진폭 적분값, 주파수대역 에너지, 영점빈도수와 같은 특징인자를 추출하여 내부품질을 판정할 수 있는 주성분회귀 모델개발에 이용하였다. 개발된 수박의 주성분회귀 모델의 상관계수는 0.79, SEC는 0.28로 판정정확도는 87% 이었으며, 모델 검증한 결과 상관계수가 0.78, SEP가 0.785로 판정정확도가 83%이었다. 생산자 내부품질판정 정확도를 조사한 결과, 생산자 판정 정확도는 75% 이었다.

Prevalence of pale, soft, and exudative (PSE) condition in chicken meat used for commercial meat processing and its effect on roasted chicken breast

  • Background: Studies on prevalence of pale, soft, exudative (PSE) condition in Sri Lankan poultry industry is minimal. Hence, the objective of present study was to determine the incidence of PSE chicken meat in a commercial meat processing plant and to find out its consequences on meat quality traits of roasted chicken breast. Method: A total of 60 breast fillets were randomly selected, evaluated based on color L* value, and placed into 1 of 2 categories; PSE (L* > 58) or normal meat ($L*{\leq}58$). A total of 20 breast fillets (10 PSE and 10 normal) were then analyzed for color, pH, and water holding capacity (WHC). After processing those into roasted chicken breast, cooking loss, color, pH, WHC, and texture values were evaluated. A sensory evaluation was conducted using 30 untrained panelists. Results: The incidence of PSE meat was 70 % in the present experiment. PSE fillets were significantly lighter and had lower pH values compared with normal fillets. Correlation between the lightness and pH was negative (P < 0.05). Although there was no significant difference in color, texture, and WHC values between the 2 groups after processing into roasted chicken breast (P > 0.05), an approximately 3 % higher cooking loss was observed in PSE group compared to its counterpart (P < 0.05). Moreover, cooking loss and lightness values showed a significant positive correlation. Nevertheless, there were no significant differences in sensory parameters between the 2 products (P > 0.05). Conclusions: These results indicated that an economical loss can be expected due to the significantly higher cooking loss observed in roasted breast processed from PSE meat.

Quantitative Analysis of Milk-Derived microRNAs and Microbiota during the Manufacturing and Ripening of Soft Cheese

  • MicroRNAs (miRNAs) are abundant in bovine milk and milk derived from other livestock, and they have functional roles in infants and in the secretion process of mammary glands. However, few studies have evaluated miRNAs in dairy processes, such as during cheese making and ripening. Thus, we investigated the characteristics of milk-derived miRNAs during the manufacturing and ripening of Camembert cheese as well as the microbiota present using the quantitative reverse transcription polymer chain reaction (RT-qPCR) and 16S rRNA pyrosequencing, respectively. Pyrosequencing showed that the cheese microbiota changed dramatically during cheese processing, including during the pasteurization, starter culture, and ripening stages. Our results indicated that the RNA contents per $200mg/200{\mu}l$ of the sample increased significantly during cheese-making and ripening. The inner cheese fractions had higher RNA contents than the surfaces after 12 and 22 days of ripening in a time-dependent manner (21.9 and 13.2 times higher in the inner and surface fractions than raw milk, respectively). We performed a comparative analysis of the miRNAs in each fraction by RT-qPCR. Large amounts of miRNAs (miR-93, miR-106a, miR-130, miR-155, miR-181a, and miR-223) correlated with immune responses and mammary glands were present in aged cheese, with the exception of miR-223, which was not present on the surface. Considerable amounts of miRNAs were also detected in whey, which is usually disposed of during the cheese-making process. Unexpectedly, there were no significant correlations between immune-related miRNAs and the microbial populations during cheese processing. Taken together, these results show that various functional miRNAs are present in cheese during its manufacture and that they are dramatically increased in amount in ripened Camembert cheese, with differences according to depth.
