Seahorse-derived peptide suppresses invasive migration of HT1080 fibrosarcoma cells by competing with intracellular α-enolase for plasminogen binding and inhibiting uPA-mediated activation of plasminogen

  • ${\alpha}$-Enolase is a glycolytic enzyme and a surface receptor for plasminogen. ${\alpha}$-Enolase-bound plasminogen promotes tumor cell invasion and cancer metastasis by activating plasmin and consequently degrading the extracellular matrix degradation. Therefore, ${\alpha}$-enolase and plasminogen are novel targets for cancer therapy. We found that the amino acid sequence of a peptide purified from enzymatic hydrolysates of seahorse has striking similarities to that of ${\alpha}$-enolase. In this study, we report that this peptide competes with cellular ${\alpha}$-enolase for plasminogen binding and suppresses urokinase plasminogen activator (uPA)-mediated activation of plasminogen, which results in decreased invasive migration of HT1080 fibrosarcoma cells. In addition, the peptide treatment decreased the expression levels of uPA compared to that of untreated controls. These results provide new insight into the mechanism by which the seahorse-derived peptide suppresses invasive properties of human cancer cells. Our findings suggest that this peptide could emerge as a potential therapeutic agent for cancer.

  • The have been many reports that phenoloxidase are correlated with development in many fungi. C. congregatus, one of nushroom-forming basidiomycetes, which requires light for its development also has phenoloxidases. In C. congragatus, there are two sets of membrane-associated phenoloxidase (PHO I and PHO II) which are differentiated by their isozyme patterns, and each enzyme set consists of two different subtrate specific enzyme protein; o-tolidine reacting enzyme, and DOPA reacting enzyme. PHO I which is localized by a protoplast-concanavalin A technique by using a new solidifying agent, Pluronic Polyol F 127, instead of agar appears in the vegetative hyphae, and PHO II appears at the early primordial stage on agar and at the sclerotial stage of liquid shake cultures. Inhibition of PHO I with the enzyme inhibitors inhibits mushroom formation as well as melanization of the vegetative hyphae at concentrations which do not inhibit the vegetative growth. PHO I deficient mutants do not form mushrooms or melanins, and the mutants show abnormal nuclear migration patterns. PHO II has roles; possibly cementing the adjacent hyphae during the actual three dimensonal structure formation, and melanizing mushrooms and sclerotia. The possible roles of PHO I in the light reception complex and in melanin formation, the function of malanin, and possible roles of postulated post translational modifying enzymes which regulate the phenoloxidases, nuclear migration pattern, and self-nonself recognition mechanism are discussed.

고등학교 세계지리 교과서의 사막포도 형성에 대한 고찰 (A Review on the Formation of Desert Pavements in High School Textbooks of World Geography)

  • High school textbooks of world geography show geomorphic featuresin arid environments such as sand dune, yardang and ventifact which are largely created by aeolian processes. Desert pavements, ubiquitous armored surfaces composed of a mosaic of clasts in hot and arid regions, are introduced as a major landform which can be attributed to wind erosion. However, they are formed by a variety of processes including deflation, surface runoff, upward clast migration and dust accretion that cause coarse particles concentration at the surface. The deflation by wind leaving a lag of coarse clasts has been solely regarded at home, even though the classical mechanism of deflation has been evaluated as a relatively unimportant process of pavement formation abroad through empiricalstudies. The accretionary model is gaining wider acceptance, thus implying that desert pavements could be formed through deposition of aeolian material. In addition, sheetflood and upward migration of clasts, irrelevant to the aeolian processes, could also create stone pavements. As a consequence, the deflation process in high school textbooks has to be urgently modified into a range of processes including aeolian mantling. By stressing that desert pavements are an exceptional geomorphic feature in deserts where wind is a predominant geomorphic agent, they can be used as a good example to demonstrate that a landform is not monogenetic.

Methyl Linderone Suppresses TPA-Stimulated IL-8 and MMP-9 Expression Via the ERK/STAT3 Pathway in MCF-7 Breast Cancer Cells

  • Methyl linderone (ML), a cyclo-pentenedione, was isolated from the fruit of Lindera erythrocarpa Makino (family Lauraceae). This plant has well-known anti-inflammatory effects; however, the anti-cancer effects of ML have not yet been reported. Thus, in the present study we investigated the effects of ML on the metastasis of human breast cancer cells. We used 12-O-tetradecanoyl phorbol-13-acetate (TPA)-stimulated MCF-7 cells as the cell model to study the effects of ML on invasion and migration. ML was found to reduce the invasion and migration rate of TPA-stimulated MCF-7 cells. Moreover, it inhibited two metastasis-related factors, matrix metalloproteinase-9 (MMP-9) and interleukin-8 (IL-8), at the mRNA and protein expression levels, in TPA-treated MCF-7 cells. The mechanism by which ML exerted these effects was through the inhibition of translocation of activator protein-1 (AP-1) and signal transducer and activator of transcription-3 (STAT3), mediated via phosphorylation of extracellular signal-regulated kinase (ERK). Taken together, our findings indicated that ML attenuated the TPA-stimulated invasion and migration of MCF-7 cells by suppressing the phosphorylation of ERK and its downstream factors, AP-1 and STAT3. Therefore, ML is a potential agent for the treatment of breast cancer metastasis.

캐티온염료를 이용한 Silk/Cationic Dyeable Polyester 혼합사의 염색거동 (Dyeing Behavior of Silk/CDP Mixed Filament with a Cationic Dye)

  • Silk/CDP 혼합사의 염색거동을 살펴보기 위하여 pH, 염색온도, 조제 등 염색조건을 변화시켜가면서 고찰한 결과 다음과 같은 결론을 얻었다. 1. 견섬유는 염욕의 pH가 증가함에 따라 일계 pH까지 염착량이 증가하는 경향을 나타냈으나 CDP 섬유는 pH에 따른 염착량의 변화가 거의 나타나지 않았다. 염색온도의 염착거동에 대한 영향은 염색온도가 증가하면 초기염착속도는 증가하는 반면 최종 염착량은 줄어드는 일반적인 경향을 나타내었다. 특히 CDP섬유의 경우 유리전이온도 이하에서도 염착속도가 상당히 느린 결과를 보였다. 2. Carrier를 사용하여 silk/CDP 혼합사를 염색하는 경우 염욕의 pH 및 염색온도에 따라 각 component에의 염료 흡착 및 탈착거동, 염착량 변화 등이 차이가 나는 것으로 나타났다. 또한 carrier가 없는 경우에는 CDP component에는 거의 염착이 이루어지지 않았다. 3. Silk/CDP 혼합사의 염색시에 나타나는 이염현상은 carrier의 존재하에서 CDP component의 구조 이완에 따라 염착능력이 증대되고 염욕의 평형이 깨어지면서 silk component의 탈착과 동시에 친화력이 큰 CDP component,쪽으로 염료이동이 일어나는 것으로 나타났다.

Resveratrol blunts tumor necrosis factor-${\alpha}$-induced monocyte adhesion and transmigration

  • The leukocyte recruitment and transmigration across the endothelial barrier into the vessel wall are crucial steps in atherosclerosis. Leukocyte trafficking on the endothelium is elicited by induction of endothelial adhesion molecules, and its transmigration is mediated by degradation of basement membrane proteins through enzymatic activity of matrix metalloproteinases (MMP). The current study investigated whether resveratrol, a polyphenol present in grapes and red wine, was capable of inhibiting leukocyte adhesion to tumor necrosis factor (TNF)-${\alpha}$-activated endothelium. It was found that resveratrol inhibited the TNF-${\alpha}$-activated endothelial expression of vascular cell adhesion molecule-1 in a dose-dependent manner. In addition, resveratrol hampered THP-1 monocyte adhesion to activated endothelial cells. This study further examined whether resveratrol interfered with transendothelial migration of leukocytes. The MMP-2 gelatinolytic activity of endothelial cells was enhanced by TNF-${\alpha}$, which was attenuated by an addition of ${\geq}25{\mu}M$ resveratrol. In addition, 25 ${\mu}M$ resveratrol mitigated the MMP-9 activity of THP-1 cells, followed by a marked inhibition of transendothelial migration. These results demonstrated that resveratrol suppressed monocyte adhesion and migration induced by TNF-${\alpha}$ through modulating expression of adhesion molecules and gelatinolytic activity of MMP. These findings suggest that dietary resveratrol may be therapeutic agent for inhibiting leukocyte recruitment into the subendothelium during inflammatory atherosclerosis.

Rhus verniciflua Stokes extract suppresses migration and invasion in human gastric adenocarcinoma AGS cells

  • BACKGROUND/OBJECTIVES: Many studies have suggested that Rhus verniciflua Stokes (RVS) and its extract are anticancer agents. However, RVS had limited use because it contains urushiol, an allergenic toxin. By improving an existing allergen-removal extraction method, we developed a new allergen-free Rhus verniciflua Stokes extract (RVSE) with higher flavonoid content. In this study, we examined whether RVSE inhibits the ability of AGS gastric cancer cells to migrate and invade. MATERIALS/METHODS: The flavonoids content of RVSE was analyzed by HPLC. The effects of RVSE on migration and invasion in AGS cells were analyzed by each assay kit. Matrix metalloproteinase (MMP)-9, plasminogen activator inhibitor-1 (PAI-1) and urokinase-type plasminogen activator (uPA) protein expression was analyzed by protein antibody array. The Phosphorylation of signal transducer and activator of transcription (STAT) 3 were assayed by Western blot analysis. RESULTS: RVSE treatment with 0-100 ㎍/mL dose-dependently reduced the ability of AGS cells to migrate and invade. Notably, treatment with RVSE strongly inhibited the expression of MMP-9 and uPA and the phosphorylation of STAT3. In contrast, RVSE treatment dramatically increased the expression of PAI-1. These results indicate that the inhibition of MMP-9 and uPA expression and STAT3 phosphorylation and the stimulation of PAI-1 expression contributed to the decreased migration and invasion of AGS cells treated with RVSE. CONCLUSIONS: These results suggest that RVSE may be used as a natural herbal agent to reduce gastric cancer metastasis.

Tas13D Inhibits Growth of SMMC-7721 Cell via Suppression VEGF and EGF Expression

  • Objective: Taspine, isolated from Radix et Rhizoma Leonticis has demosntrated potential proctiective effects against cancer. Tas13D, a novel taspine derivative synthetized by structure-based drug design, have been shown to possess interesting biological and pharmacological activities. The current study was designed to evaluate its antiproliferative activity and underlying mechanisms. Methods: Antiproliferative activity of tas13D was evaluated by xenograft in athymic mice in vivo, and by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) and cell migration assays with human liver cancer (SMMC-7721) cell lines in vitro. Docking between tas13D and VEGFR and EGFR was studied by with a Sybyl/Surflex module. VEGF and EGF and their receptor expression was determined by ELISA and real-time PCR methods, respectively. Results: Our present study showed that tas13D inhibited SMMC-7721 xenograft tumor growth, bound tightly with the active site of kinase domains of EGFR and VEGFR, and reduced SMMC-7721 cell proliferation (IC=34.7 ${\mu}mol/L$) and migration compared to negative controls. VEGF and EGF mRNAs were significantly reduced by tas13D treatment in a dose-dependent manner, along with VEGF and EGF production. Conclusion: The obtained results suggest that tas13D inhibits tumor growth and cell proliferation by inhibiting cell migration, downregulating mRNA expression of VEGF and EGF, and decreasing angiogenic factor production. Tas13D deserves further consideration as a chemotherapeutic agent.

흑삼의 신생혈관 억제활성에 대한 연구 (Study on Antiangiogenic Effect of Black Ginseng Radix)

  • Objectives : This study was performed to investigate the influence of black ginseng radix extracts (BG) and ginsenoside Rg3, Rg5 on basic fibroblast growth factor (bFGF) induced proliferation, migration and capillary tubule-like formation of human umbilical vein endothelial cells (HUVECs). Methods : HUVECs were cultured with BG and ginsenoside Rg3, Rg5 at different concentrations (60, 125, 250, 500, $1,000{\mu}g/m\ell$) for 2 day In the presence of bFGF, respectively. XTT was used to detect the proliferation. Migration and tube formations were examined to detect the antiangiogenesis. Also, the chick embryo chorioallantoic membrane (CAM) assay was performed to detect the antiangiogenesis. Results : BG and ginsenoside Rg3, Rg5 significantly inhibited bFGF-induced endothelial cell proliferation and migration in a dose-dependent manner. Tube formation in bFGF-induced HUVECs were suppressed by BG and ginsenoside Rg3, Rg5. Moreover, BG and ginsenoside Rg3, Rg5 (30-$50{\mu}g$/egg) inhibited new blood vessel formation on the growing CAM. Conclusions:Based on the present results, it can be suggested that BG has a potential chemopreventive agent via antiangiogenesis.

Radially patterned polycaprolactone nanofibers as an active wound dressing agent

  • Background The objectives of this study were to design polycaprolactone nanofibers with a radial pattern using a modified electrospinning method and to evaluate the effect of radial nanofiber deposition on mechanical and biological properties compared to non-patterned samples. Methods Radially patterned polycaprolactone nanofibers were prepared with a modified electrospinning method and compared with randomly deposited nanofibers. The surface morphology of samples was observed under scanning electron microscopy (SEM). The tensile properties of nanofibrous mats were measured using a tabletop uniaxial testing machine. Fluorescence-stained human bone marrow stem cells were placed along the perimeter of the radially patterned and randomly deposited. Their migration toward the center was observed on days 1, 4, and 7, and quantitatively measured using ImageJ software. Results Overall, there were no statistically significant differences in mechanical properties between the two types of polycaprolactone nanofibrous mats. SEM images of the obtained samples suggested that the directionality of the nanofibers was toward the central area, regardless of where the nanofibers were located throughout the entire sample. Florescence images showed stronger fluorescence inside the circle in radially aligned nanofibers, with significant differences on days 4 and 7, indicating that migration was quicker along radially aligned nanofibers than along randomly deposited nanofibers. Conclusions In this study, we successfully used modified electrospinning to fabricate radially aligned nanofibers with similar mechanical properties to those of conventional randomly aligned nanofibers. In addition, we observed faster migration along radially aligned nanofibers than along randomly deposited nanofibers. Collectively, the radially aligned nanofibers may have the potential for tissue regeneration in combination with stem cells.
