Effect of pH of Casing Soil and Calcium on Mycelial Growth and Yield of Agaricus bisporus (Lge.) Sing (복토의 pH 및 칼슘이 양송이의 균사생장 및 수량에 미치는 영향)

  • The highest fresh weight of Agaricus bisporus was obtained at pH 7.5 of casing soil. Above or below this pH level, the yields were significantly reduced. Stimulatory effect of calcium on the mycelial growth depended upon pH of casing soil, In neutral or slightly alkaline soil, the stimulatory effect was increased. The effects of supplementing calcium carbonate or hydrated lime to casing soil were assumed not only the influence of pH of casing soil, but also stimulatory effect of calcium ion on mycelial growth of A. bisporus.

  • Quantitative analysis was achieved by gas-liquid chromatographic method (GLC) with a single column system of OV-17 for 16 of protein amino acids in mushroom (agaricus bisporus).   • For the purpose of isolation and screening of tyrosinase inhibitory activity from edible mushrooms, Pleurotus ostreatus, Auricularia auricula-Judae, Umbilicaria esculenta, Agaricus bisporus, Flammuline velutipes, Lentinus edodes, Ganoderma lucidum, and Coriouls versicolor were examined by tracing inhibitory activities against tyrosinase, utilizing L-3,4-dihydroxyphenylalanine (L-DOPA) as a substrate. Among the eight edible mushrooms tested, Umbilicaria esculenta showed potent enzyme inhibitory activities above 7804% against tyrosinase in ethylacetate (EtOAc) extracts. Ganoderma lucidum and Agaricus bisporus showed inhibitory activities of 67.3% and 51.5% in water extracts. EtOAc extracts of Umbilicaria esculenta was fractionated from silicagel column chromatography and one fraction showed the most inhibitory activity of 60.9%. The three bands (Rf=0.38, 0.27, 0.19) were isolated from preparative TLC of the fraction for purification and identified as mixtures of orsellinate, methyl orsellinate, methyl lecanorate, and methyl gyrophorate by high pressure liquid chromatography (HPLC), ultravisible spectrophotometer (UV), mass spectrophotometer (Mass), nuclear magnetic resonance spectrometer (NMR).

Optimization on Organoleptic Properties of Mushroom (Agaricus bisporus) Pickles using Response Surface Methodology (반응표면법을 이용한 양송이버섯 피클의 관능적 특성 최적화)

  • The purpose of this study was to determine the organoleptic properties of mushroom pickles made at various compounding ratios according to central composite design for optimum organoleptic properties. In this study, various kinds of mushroom pickle were made at different compounding ratios of vinegar, sugar and salt-critical ingredients of the pickle recipe and the products were presented to an expert panel, who graded the subjects in 7 degrees for 5 items: color, flavor, hardness, taste and overall quality. As a result of sensory quality, mushroom pickles with 300g of vinegar, 150g of sugar and 60g of salt achieved the highest grade. Meanwhile, the results of Response Surface Methodology were different from the sensory quality results, showing that the optimum mixing conditions for overall organoleptic properties of mushroom pickle were 279.58g of vinegar, 179.34g of sugar and 59.09g of salt.   • Attempts were made to investigate on the fatty acids of Agaricus bisporus, a cultivated edible mushroom. Fats were extracted from it and saponified with alcoholic potassium hydroxide. Isolated fatty acids were methylated and were subjected to column chromatography and G.L.C. Eleven saturated fatty acids, i.e., dodecanoic, tridecanoic, tetradecanoic, pentadecanoic, hexadecanoic, heptadecanoic, octadecanoic, eicosenoic, uncosenoic, docosenoic, and tricosenoic acids, were identified. Especially palmitic and stearic acids were abundant. Nine unsaturated fatty acids appear to exist in the carpophore.

  • Inhibitory effects of the mushrooms on the growth and urease of Helicobacter pylori (HP), which is associated with human gastroduodenal diseases such as gastritis, peptic ulcer and gastric carcinoma, were investigated. Most of the mushroom extracts did not show inhibitory effect on HP urease except Coriolus versicolar, Auricularia auricular Sarcodon aspratus and Flammulina velutipes. The extract of Ganoderma lucidum, Coriolus versicolar, Gyropora esculenta and Agaricus bisporus var. albidus inhibited the growth of HP. When their extracts were fractionated, the ether fraction of Ganoderma lucidum and Agaricus bisporus var. albidus were the most effective. Among seven components separated from the ether fraction of G. lucidum extract by silica gel column chromatography, P3 was the most potent: MIC was $200{\mu}g/ml$. However, P3 did not inhibit the urease.

  • A. bisporus is the fifth most cultivated mushroom in Korea, and approximately 10,757 tons were cultivated in 2015. The genetic diversity of collected strains in Korea and commercial cultivars was analyzed using inter-simple sequence repeat (ISSR) markers. ISSR markers known to be comparable among A. bisporus spp. were selected from various markers. Totally, 16 markers, namely the ISSR markers 807, 808, 810, 811, 834, 835, 836, 841, 842, P3, P8, P17, P22, P30, P38, and P39, were evaluated to discriminate between ASI 1110, 1114, 1115, 1238, 1246, 1365, 1366, and 1369 for selecting suitable markers in 16 markers. The ISSR markers P31, P38 and P39 exhibited various fingerprints that could help classify the strains in species. Using the three markers, genetic relationships among 39 strains, including commercial cultivars, such as SaeA and SaeYeon, were analyzed using the UPGMA method. The results of the analysis of the genetic relationships between commercial cultivars and collected strains in Korea confirmed that the commercial cultivars were different from the collected strains in Korea. These results suggested that the ISSR markers P31, P38, and P30 could be used for selecting the commercial cultivars of A. bisporus.

  • Molecular spslemaucs of Agaricus species was investigated on the base of the sequences of the internal transcribed spaceriITS) regions in ribosomal DNA (rDNA). The sequences of the ITS region in 5 species and two group of Agaricus genus were resolved. In the phylogenetic trees. the species generally divided inlo two subclusters, refered to here as the group I and group 11. The group I consisted of Agaricus blazei ATCC 76739, Agarictrs blazei species cultivated in Korean hmings. Ago/-icus anmensis IMSNU 32049 and Agaricus can~pestris VPI-OKM 25665. Between Agaricus blazei NCC 76739 and the Agaricus blazei species cultivated in Korean farmings had the variation in lhe 5 nucleotide on the ITS regions. These varieties were presumed the variation by the geographic and cultivated conditions. In addition the subgroup of group I was formed by Agaricus arvensis LMSNU 32049 and Agaricus carnpests VPI-OKM 25665. The group IT included Agnrictrs bispoms CH 3004 and Agaricus pocillotor DUKE-J 173.

  • Agaricus bitorquis is an edible white mushroom of the genus that is cultivated at high temperature($25{\pm}1^{\circ}C$) unlike A. bisporus is cultivate at $16{\pm}2^{\circ}C$. Unlike Agaricus bisporus, an edible white A. bitorquis mushroom is cultivated at high temperature ($25{\pm}1^{\circ}C$). Most farmers cultivate this mushroom for a long cultivation period in Korea. For this reason, we made heterokayons to develop a new cultivar that generate fruitbodies for short cultivation period. Over one hundred SSIs(single spores isolates) were collected from selected A. bitorquis ASI1151 and ASI1349 strains. Seventy-three SSIs were germinated on CDA(compost dextrose agar) media after 20 days (minimum) or 83 days (maximum) incubation under different media condition. The mycelial growth rate of germinated SSIs was different. 9 homokaryons in ASI 1151 and 11 homokaryons in ASI 1349 from SSIs were selected by OPN-02 primer in RAPD analaysis. Also this primer was used to select heterokaryon that cross among each homokaryon with compatible locus. Therefore 44 compatible matings were confirmed of 99 crossed lines.

Biochemical Characterization of Agaricus bisporus Dikaryon Strains (양송이 이핵균주의 생화학적 특성 검정)

  • Button mushroom (Agaricus bisporus) strains from diverse origins were compared in this study to obtain basic information on their growth and biochemical properties that are helpful for breeding. Among 31 dikaryotic strains tested, most strains showed better mycelial growth on oatmeal agar than on MEA and PDA. Mycelia of the mushroom strains revealed ${\beta}$-glucosidase activity the most clearly among the seven extracellular enzymes tested. All the strains showed protease activity, but ${\beta}$-glucosidase activity was found in 27 strains and xylanase activity was found in 30 strains. The activity of avicelase, CM-cellulase, amylase, and pectinase was detected in less than 20 strains. These results implied that enzymatic characteristics could be used as a criterion of strain selection for breeding study.
