Quantifiable Downregulation of Endogenous Genes in Agaricus bisporus Mediated by Expression of RNA Hairpins

  • Functional gene studies in the cultivated white button mushroom Agaricus bisporus have been constrained by the absence of effective gene-silencing tools. Using two endogenous genes from A. bisporus, we have tested the utility of dsRNA hairpin constructs to mediate downregulation of specific genes. Hairpin constructs for genes encoding orotidine 5'-monophosphate decarboxylase (URA3) and carboxin resistance (CBX) were introduced into A. bisporus using Agrobacteriummediated transfection. Although predicted changes in phenotype were not observed in vitro, quantitative-PCR analyses indicated unambiguously that transcripts in several transformants were substantially reduced compared with the non-transformed controls. Interestingly, some hairpin transformants exhibited increased transcription of target genes. Our observations show that hairpin transgenic sequences can mediate downregulation of A. bisporus endogenous genes and that the technology has the potential to expedite functional genomics of the mushroom.

Development of a Segregating Population with Biological Efficiency in Agaricus bisporus

  • In this study, we made a population with high biological efficiency (BE) to investigate the complex genetic architecture of yield-related traits in Agaricus bisporus. MB-013 crossed between bisp 015-p2 and bisp 034-p2, had high BE. Additionally MB-013 was an intervarietal hybrid that intercrosses with A. bisporus var. burnettii, bisp 015, and A. bisporus var. bisporus, bisp 034. One hundred and seventy homokaryons were selected using the cleaved amplified polymorphic sequence (CAPS) markers (PIN primer/HaeIII) from 300 single spore isolates (SSIs). One hundred $BC_1F_1$ hybrids were obtained by crossing the homokaryons of MB-013 with bisp15-p1. The population of 100 BC1F1 hybrids is suitable for analyses of BE.

  • Agaricus bisporus is a popular edible mushroom that is cultivated worldwide. Due to its secondary homothallic nature, cultivated A. bisporus strains have low genetic diversity, and breeding novel strains is challenging. The aim of this study was to investigate the genetic diversity and population structure of globally collected A. bisporus strains using simple sequence repeat (SSR) markers. Agaricus bisporus strains were divided based on genetic distance-based groups and model-based subpopulations. The major allele frequency (MAF), number of genotypes (NG), number of alleles (NA), observed heterozygosity (HO), expected heterozygosity (HE), and polymorphic information content (PIC) were calculated, and genetic distance, population structure, genetic differentiation, and Hardy-Weinberg equilibrium (HWE) were assessed. Strains were divided into two groups by distance-based analysis and into three subpopulations by model-based analysis. Strains in subpopulations POP A and POP B were included in Group I, and strains in subpopulation POP C were included in Group II. Genetic differentiation between strains was 99%. Marker AB-gSSR-1057 in Group II and subpopulation POP C was confirmed to be in HWE. These results will enhance A. bisporus breeding programs and support the protection of genetic resources.

Yield, Nutrient Characteristics, Ruminal Solubility and Degradability of Spent Mushroom (Agaricus bisporus) Substrates for Ruminants

  • This study was conducted to evaluate the yield, nutrient characteristics, ruminal solubility, degradability and disappearance of spent mushroom (Agaricus bisporus) substrates for ruminants. The annual yield of spent Agaricus bisporus substrates was measured to be about 210,000 tons (M/T) in South Korea. The surface soil-removed spent substrates had nutritional characteristics of high crude ash (375 g/kg) and Ca (32 g/kg), medium protein (134 g/kg CP), and high fiber (384 g/kg NDF on a DM basis). Compared with initial mushroom substrates, spent mushroom substrates had twice higher (p<0.0001) CP content and 22.0% lower (p<0.0001) NDF content on an organic matter basis. Compared with raw rice straw, spent rice straw had much higher (p<0.05) predicted ruminal degradabilities and disappearances of DM and CP and a little lower (p<0.05) predicted degradability and disappearance of NDF. In conclusion, the general feed-nutritional value of spent mushroom (Agaricus bisporus) substrates appeared to improve after cultivation of mushrooms.

Development of Air-jet Washer for the Agaricus Bisporus (공기분사에 의한 양송이 버섯 세척기 개발)

  • This study was conducted to develop an agaricus bisporus washing machine which uses compressed air to remove foreign materials attached on the surface of agaricus bisporus. A prototype of the washing machine was constructed, and performance of removing foreign materials was tested. Research results are as follows: 1. Several transferring methods including PE roller rotation, brush roller rotation, PE screw rotation, vibration plate, and belt conveyor were evaluated. Roller, screw, and vibration methods caused damages on the surface of the products, but belt conveyor method caused the least damages. 2. For air jet, a stationary nozzle type and a rotational type were evaluated. The best air jet nozzle was the jet-type nozzle, and the rotational type was more effective than stationary type nozzle. 3. With the conveyer belt, box type moving method and the rotational air jet nozzle, the washing machine showed the best performance when higher than 5.4${\times}$105 Pa of air jet pressure and lower than 0.047 m/s of moving speed was used. Working performance of the system was 650 kg/h, and the damaging rate was 1.2 %.

The Preparation of Mushroom Pieties and Change in Quality during Storage (버섯 피클(Fickle) 제조 및 저장 중의 품질변화)

  • The mushroom pickles were prepared as a processed mushroom product. The changes in pH, total acidity, brix. color and texture of the mushroom pickles (Lentinus edodes, Pleurotus ostreatus, Agaricus bisporus and Pleurotus eryngii) were measured during storage at 4$^{\circ}C$. In addition, sensory evaluations on the mushroom pickles were performed. While the pH of Lentinus edodes and Pleurotus ostreatus pickles declined slowly, the pH of Agaricus bisporus and Pleurotus eryngii pickles declined rapidly during storage. The total acidity of Lentinus edodes pickles rose slowly, while the acidity Pleurotus ostreatus, Agaricus bisporus and Pleurotus eryngii pickles rose rapidly. As for the color of mushroom pickles, "L" , "a" and "b" values changed slightly. The changes in texture were determined by a rheometer Hardness, cohesiveness, springiness, gumminess and brittleness of Lentinus edodes pickles were lower than those of the other mushroom pickles. In the sensory evaluation of mushroom pickles, Lentinus edodes showed higher acceptability ratings than the other mushrooms in taste, color, clearness, texture and overall acceptability. This study showed that among the mushrooms tested, Lentinus edodes was the most acceptable mushroom pickle for storage.

Inhibition of Inducible Nitric Oxide Synthase by Agaricus bisporus Extract in RAW 264.7 Macrophages

  • Agaricus bisporus, also known as white button mushroom, is one of the most popular mushrooms consumed in Korea. This mushroom contains high concentrations of flavanoids and exhibits antioxidant activity. In this study, we examined the effects of Agaricus bisporus ethanol extract (ABE) on lipopolysaccharide (LPS)-induced inflammation in RAW 264.7 cells. Nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) protein levels were assessed in cells treated with $100\;{\mu}M$ LPS in the presence or absence of ABE. 0.5 mg/mL of ABE suppressed NO production significantly. Moreover, ABE inhibited levels of iNOS protein. Taken together, these results suggest that ABE exerts anti-inflammatory activity in LPS-induced inflammation in RAW 264.7 cells.

  • In order to study the protease from Agaricus bisporus (Lange), the crude protease preparation was separated by fractionation of mushroom extracts with ammonium sulfate. It was found that extracts from Agaricus bisporus (Lange) Sing. contained protease. The optimum pH of the enzyme was 6.0, and the pH range at which the enzyme was stable was 4.0 to 7.0. The optimum temperature at which the enzyme showed the highest proteolytic activity was $50^{\circ}C$, while the enzyme was instantly inactivated at about $60^{\circ}C$. The enzyme activity was inhibited by $Ag^+$, $Hg^{2+}$, $Cu^{2+}$, $Ba^{2+}$, $Fe^{3+}$, $Co^{2+}$, $Ca^{2+}$, $Pb^{2+}$, $Mg^{2+}$ and $Mn^{2+}$. The $K_m$ value was 0.32 mM with Hammarsten casein.

Study on the Genetic Diversity and Biological Characteristics of Wild Agaricus bisporus Strains from China

  • 90 wild Agaricus strains from China, including 44 Agaricus bisporus strains identified preliminarily by isozyme electrophoresis, were studied by the techniques of SRAP and ISSR. 18 special SRAP bands and 12 special ISSR bands were analyzed, the strains were clustered and a demdrogram was obtained. The results showed that the strains were divided into 2 groups, wild A. bisporus group and the other Agaricus group. It is similar to the result of isozyme electrophoresis. 41 wild A. bisporus strains from Sichuan and Tibet were divided into 4 groups based on their growing places, suggesting the regionally difference of the strains to be quite obvious. Some white wild A. bisporus strains from Xinjiang and Tibet had special patterns, resulting in lower coefficient values with other wild A. bisporus strains. The biological characteristics of three wild A. bisporus strains were analyzed, and the results showed: 1. The wild strains grew slowly on PDA medium with weak appressed mycelia, and grew normally in kernel or fermented cottonseed shell substrate. 2. They grew faster than control strain As2796 under lower temperature of $16^{\circ}C$, and higher temperature of $32^{\circ}C$, with optimum growing temperature of $20-24^{\circ}C$, which was $4^{\circ}C$ lower than that of control strain. 3. In the cultivation with manure compost via twice fermentation, the mycelia grew normally in compost and quite slowly in casing soil, and the fruitbodies occurred less and late with easily opening and low production. 4. The fruitbody was off-white with flat and scaled cap, long stipe and dark gill. The bisporus basidia occupied 70-80% and trisporus basidia 20-30% of the total basidia. 5. Heterokaryotic monospore isolates could fruit in cultivation, and the homokaryotic isolates could cross with those derived from overseas wild A.bisporus strains. 6. The electrophoresis phenotype of isozymes such as esterase etc. belonged to high production type (H type). 7. The RAPD patterns made much difference from those of high production, good quality or hybrid strains, which indicated that the wild strains produce a new kind of RAPD type.

Physical Properties of fish Paste Containing Agaricus bisporus (양송이 함유 어묵의 물성 특성)

  • Fish paste containing mushroom, Agaricus bisporus, was prepared for the supplementary value of the product, and it was tested for the color, textural properties, and sensory attributes. With increasing amounts of the mushroom in fish paste, L value decreased and a value increased in the color changes while the b value was not significantly different. As the result of textural properties, folding test in all test samples showed AA that mean good flexibility. The hardness, elasticity, gumminess and brittleness of fish paste with the mushroom were increased but the strength was not significantly different. In overall acceptance of sensory evaluation, fish paste containing 10% of mushroom received the highest score. These results suggest that Agaricus bisporus can be applied to fish paste for the purpose of the varieties of product.
