Survey of Airborne Fungi Levels in 24 Seasonal Divisions and Correlation Analysis with Meteorological Elements (24절기 야외 부유곰팡이 농도 조사 및 기상요소와의 상관성 분석)

  • To identify daily and annual changes in outdoor airborne fungi, it is necessary to shorten the collection cycle and increase the number of measurements. In this study, measurements were performed by employing an air sampler and potato dextrose agar media on the rooftop of National Research Institute of Cultural Heritage during a period of one year (August 2018 to July 2019). The collection cycle spanned the twenty-four seasonal divisions and the collection time was 2 p.m. and 11 p.m.. Meteorological elements were collected at intervals of one hour. Furthermore, the concentration of airborne fungi was monitored and correlation analysis with meteorological elements was subsequently conducted. Obtained results indicate that the concentration of airborne fungi is found to be highest in November, autumn, night, followed by autumn, summer, winter, and spring. The concentration, type, and dominant species of airborne fungi can vary depending on factors such as rainfall, typhoons, and yellow dust (fine dust). The concentration of airborne fungi indicates a strong positive linear relationship between precipitation, number of precipitation days, and relative humidity. The concentration of airborne fungi was related to the period of increase of dead plants in terms of nutrition source, and to the high relative humidity conditions including rainfall in terms of meteorological elements.

Effects of Embryo Development Stage and Gelrite Concentration on Plant Regeneration in Seed Culture of Rice (벼의 종자배양에서 배의 성숙정도와 Celrie 농도가 캘러스 형성 및 식물체 재분화에 미치는 영향)

  • To identify the effects embryo developmental stage and gelrite concentration on plant regeneration in seed culture of rice, mature and immature seeds of rice were cultured on the $N_{6}$ medium supplemented with2 mg/$\ell$ 2.4-D and different levels of gelrite(0.2~1.0%). The calli formed immature embryos were produced more plants than those from mature embryos. The maximum frequency of plant regeneration was achieved in the culture of the calli of immature embryos which was harvested at the 21$^{th}$ day after pollination. The plant regeneration on the medium with gelrite was more accelerate than that on the medium with agar. The highest frequency(55%) of plant regeneration was obtained from the calli transferred to the medium with 6g/$\ell$ gelrite.

Distribution of Airborne Fungi in the Public Facilities (다중이용시설 내 공기 중의 진균 분포)

  • Recently, there is increasing interest in indoor air pollution. Fungal spores are one of the pollutants that is the causes of hypersensitivity reactions, bronchial asthma, allergic rhinitis. This study was performed to assess the levels of fungi concentration in public facilities and airborne fungi were collected with the air sampler using DRBC(Dichloran rose bengal chloramphenicol) agar. It was found that airborne fungi concentration were related to the humidity and the people movement. 10 genera of fungi was isolated and identified and the most common culturable fungi were Penicillium, Cladosporium, and Aspergillus.

  • Microorganisms capale of utilizing linear alkylbenzene sulfonates(LAS) as sole carbon source were isolated from industrial effluent by using LAS agar plates. The isolated strains were identified as Salmonella sp(BC-2) and Escherichia sp.(BC-3) from the results of morphological, cultural and biochemical tests. The optimal condition for the growth and biodegradation of LAS was the initial pH 7.0 and LAS concentration 0.1%. The isolated BC-2 and BC-3 strains harbored plasmid and LAS-degrading activity was lost when the plasmids were cured by mitomycin C. The plasmids were transformed into E. coli and transformants have the LAS-degrading activity. Isolated strains were examined for primary biodegradation rate of LAS in the medium by methylene blueactive substance(MBAS) method. Of these isolates, BC-2 and BC-3 strains degradated LAS upto 60% and high resistant to CdCl$_{2}$ and HgCl$_{2}$. Isolated strains were sensitive to chloramphenicol, kanamycin, rifampicin, streptomycin and tetracycline but resistant to ampicillin and lincomycin.] Its minimal inhibitory concentration(MIC) for ampicillin was more than 1500 $\mu $g/ml.

  • Tissue culture systems to optimize regeneration plant species of Ocimum spp were evaluated as a method to micropropagate individual plants and to better study their biology in vitro. Ocimum species were also evaluated for the production of natural plant products during and following the regeneration process. The primary goal of this project was to enhance the regeneration efficiency of basil. Several factors were examined using different Ocimum species and commercial varieties. The effect of cytokinin combination, activated charcoal, gelling agents, and different carbon sources were investigated. Anthocyanin callus spots were produced only in four varieties among six tested. 'Sweet Dani' showed the best results on anthocyanin accumulation, while 'African beauty', 'Tree basil' and 'Methylcinnamate' produced only a few spots. Shoot regeneration was only achieved from 'Sweet Dani' explants. As the activated charcoal concentration increased, callus formation rate decreased respectively compare to the controls for all varieties. There was a decrease in callus growth with increasing concentration of agar and phytagel.

Characteristics of Cadmium-Resistant Bacillus subtilis DT134 (Bacillus subtilis DT134의 카드뮴 저항성)

  • Bacillus subtilis DT134 was resistant to 50-fold higher concentration of cadmium ions (Cd2+) than cadmium-sensitive B. subtilis BD224 in Luria Broth (LB) medium. Minimal inhibition concentration test in LB agar plates also showed similar results. The elevated cadmium resistance of B. subtilis DT134 strongly suggested a possible existence of cadmium resistance gene in it. Southern blot with Staphylococcus aureus cadA gene fragment (757 bp NlaIV-XmnI cadA DNA fragment) as probe was carried out to test the existence and similarity of the gene. In high stringency condition, there was no detectable signal, but in low stringency, a strong signal specific to the cadA probe could be detected. These results strongly suggested that there was some similarity between total DNA of B. subtilis DT134 and S. aureus pl258 in terms of cadmium resistance gene and the resistance mechanism might be an efflux mechanism. The subsequent efflux experiment showed that the cadmium resistance mechanism of B. subtilis DT134 was also due to the efflux of cadmium.

  • The purpose of this study is to determine the antibacterial effect of Dilute Formocresol and Eugenol and Propylene glycol. The experimental drugs are Formocresol in Propylene glycol (5, 10, 20%) and Eugenol in Propylene glycol (50, 75, 100%) and Propylene glycol. The organisms selected for study were Staphylococcus aureus, Pseudomonas aeruginosa, and Bacillus subtilis which are found in infected root canals and are highly resistant to antiseptics. Isolated bacteria were inoculated on blood agar plate and the plates were incubated at $37^{\circ}C$ for 18 hours and the zones of inhibition then measured. The results were as follows ; 1. The antibacterial action of Formocresol was effective even at 5-10 percent and the action increased when higher concentration was used. 2. The antibacterial action of Eugenol was not effective and the action decreased when higher concentration was used. 3. Propylene glycol itself possessed some antibacterial properties and showed that the antibacterial action of Propylene glycol might be almost the same as that of Eugenol. 4. Among the experimental organisms, Pseudomonas aeruginosa was found to be the most resistant to all the experimental drugs.

Optimization of Genetic Transformation Conditions for Korean Soybean Cultivars (국내 콩(Glycine max) 품종 형질전환 초기조건 확립)

  • In order to establish highly efficient gene transfer condition at early stage of soybean transformation, various experiments were performed and compared their efficiencies by transient GUS analysis; those conditions are genotype determination of Korean soybean cultivars for amenability to Agro-infection, appropriate agar and selective agent concentration, orientation of explant placement, hormone pre-culture, and liquid selection condition. In the genotype screen of Korean soybean varieties, 14 amenable genotypes were selected. For efficient Agrobacterium washing, cefotaxime was chosen and hygromycin at the concentration of 10 and 15 ppm was used as selection agent in the media. Agar concentration was slightly better in 0.6% and 0.8% for both shoot and callus formation, and explant placement with adaxial side down showed high frequency of GUS expression. For wounding treatment, oriental needle was efficient than scalpel for shoot formation and gene transfer. To increase the frequency of gene transfer, hormone pre-treatment was applied. BA at the concentration of 5 and 10 ppm resulted in better survival at the late stage of selection in shoot elongation media. Selection in liquid media after hormone pre-treatment seemed to be effective to remove the escaped non-transformants at early stage of procedure. Considering the results obtained, Eunhakong could be the first choice as a material for soybean transformation among Korean soybean genotypes.

Physical Characteristics and Antagonistic Effect of Ampelomyces (Ampelomyces의 생리적 특성 및 길항능력)

  • During the period of June, 2005 to May, 2008, 44 host plants infected with powdery mildew were collected in the Jeon-ju and Jang-su districts of Jeonbuk province and in the Jang-sung district of Jeonnam province. The hyperparasites, Ampelomyces were confirmed in 12 plant species. Most of the pycnidium shapes of Ampelomyces were circular or oval shaped, and the sizes were different even within the same host plant, and also the color of pycnidium was ranged from light brown to dark brown. Ampelomyces species were isolated from 4 hosts including Impatiens balsamina L., Cucurbita pepo, Rudbeckia laciniata var. elatier and Youngia sonchifolia, and thus the most appropriate 12 Ampelomyces strains for the current experiment were selected. When analyzing the selected 12 strains' incubational and nutritional characteristics, the malt extract agar was the most appropriate media. When investigating the effect of osmotic pressure on the spore germination, 0.15M NaCl concentration was the optimum germination concentration. When the isolated Ampelomyces sp. was tested in-vitro, it was found to be effective to control in other plant pathogens, isolated Ampelomyces showed no pathogenicity to the plant. strains isolated . studied on rDNA ITS sequence analysis. The rDNA ITS sequence data of Ampelomyces sp. isolate BSLAH16 from Impatiens balsamina L. were analyzed and identified.

In vitro Antimicrobial Activity of a New Isolate Streptomyces sp. BCNU 1030 (신규 분리균주 Streptomyces sp. BCNU 1030의 in vitro 항균활성)

  • This work focused on screening and characterizing antibiotic-producing actinomycetes to develop new antibiotics that can overcome the growing resistance of disease-causing microbes. One-hundred actinomycetes strains were isolated from soil samples from Chungcheongbuk-do, Korea using various kinds of actinomycetes isolation media, including a starch casein agar medium and potato dextrose agar (PDA). Among them, strain BCNU 1030 was determined to show strong antimicrobial activity against methicillin-resistant Staphylococcus aureus (MRSA). Biochemical, physiological, and 16S rRNA sequence analyses indicated that strain BCNU 1030 belonged to the genus Streptomyces. Strain BCNU 1030 exhibited antibiotic activity against a wide range of bacteria, especially methicillin-resistant Staphylococcus aureus (MRSA). The minimum inhibitory concentration (MIC) of BCNU 1030 dichloromethane extract was determined to be $0.78\;{\mu}g/ml$ for MRSA CCARM 3090. Therefore, Streptomyces sp. BCNU 1030 has potential for anti-MRSA drug development.
