Vitamin C가 방사선과 Aflatoxin B1을 투여한 흰쥐의 혈청과 간장의 지질성분 및 지방산 조성에 미치는 영향 (The Effects of Vitamin C on Lipid Contents and Fatty Acid Compositions of Serum and Liver in Rats Treated with Radiation or Aflatoxin B1)

  • Aflatoxin은 Aspergillus속 곰팡이로부터 생성되며 사람에게 있어서 간독성 및 간암을 유발하는 잠재력을 가진 곰팡이 독소이며, 지질과산화 반응은 aflatoxin $B_1$에 의한 세포 산화적 손상 시 발생하는 주요 현상 중의 하나이다. 방사선은 수술, 항암약물요법과 더불어 임상 시 중요한 치료방법이나, 정상세포에 방사선을 조사하였을 때 반응성이 높은 활성산소와 과산화라디칼$(OH{\cdot})$을 생성하여 세포막의 불포화지방산을 지질과산화물로 변성시켜 세포 산화적 손상을 일으킨다. 따라서 본 연구의 목적은 aflatoxin $B_1$과 방사선조사 후 유도된 간과 혈청의 지질성분 및 지방산 조성에 미치는 vitamin C의 영향을 조사하기 위해 수행되었다. X-ray 조사는 실험기간 내 단 1회로 실험사육기간 첫 일에 조사하였고 X-ray 조사 후 vitamin C를 투여하였으며 vitamin C 투여 1시간 후 $AFB_1$을 투여하였다. Vitamin C와 $AFB_1$은 모두 복강투여로 실험 사육 첫 일부터 1회 시작하여 3일에 한번씩, 5회 반복 투였으며 실험동물 사육기간은 총 15일로 하였다. 혈청 총콜레스테롤 농도는 $1\sim5$군에서 유의적인 차이를 볼 수 없었는데 방사선과 $AFB_1$을 함께 처리한 6군에서는 $130.37{\pm}9.14mg/dL$로서 전군에서 가장 높은 수치를 나타낸 반면 6군에 vitamin C를 혼합 투여한 7군에서는 $90.23{\pm}6.15mg/dL$로서 가장 낮았으며 이는 중성지질과 유사한 경향이었다. 또한 간장의 총콜레스테롤과 유리 콜레스테롤도 이와 유사한 경향이었다. HDL 농도는 방사선과 $AFB_1$을 함께 처리한 것에 vitamin C를 혼합 투여한 7군이 $59.57{\pm}9.56mg/dL$로서 p<0.01 수준에서 현저히 증가하였다. 혈청의 인지질과 콜레스테롤 에스테르의 지방산 조성에서 vitamin C를 혼합 투여한 3,5,7군에서 PUFA 함량이 증가한 반면, SFA 함량은 감소하여 P/S 비율, n-3P/n-6P 비율은 증가하는 경향이었으며 이는 간장의 인지질, 콜레스테롤 에스테르, 총 지질의 지방산조성에서도 같은 경향을 볼 수 있었다.

Quality Evaluation of Five Commercial Enzyme Linked Immunosorbent Assay Kits for Detecting Aflatoxin B1 in Feedstuffs

  • The objective of this study was to evaluate the quality of five commercial enzyme linked immunosorbent assay (ELISA) kits (A, B, C, D, and E) from different suppliers for detecting aflatoxin $B_1$ ($AFB_1$). $AFB_1$-free corn samples supplemented with different levels of $AFB_1$ (5, 10, and $20{\mu}g/kg$) were used as positive controls and 6 replicates of each control sample were tested to evaluate the accuracy and precision of these kits. In addition, we also evaluated the performance of these ELISA kits for $AFB_1$ in 30 feed samples, including corn, distillers dried grains with soluble, wheat samples, soybean meal, and poultry feed, which were verified by high performance liquid chromatography. Results showed that the coefficients of variation ranged from 1.18% to 16.22% in intra-plate and 2.85% to 18.04% in inter-plate for the determination of $AFB_1$. The half maximal inhibitory concentration for five kits ranged from 3.72 to $7.22{\mu}g/kg$. The quantitation limits of $AFB_1$ were all under the legal limit in China but somewhat inconsistent with kit instructions. Although the recovery rate of four of the five kits were either less than 90% or more than 110%, all these values were acceptable in practice. Two kits had high false positive rates (C and E). In conclusion, our results revealed that the qualities of five tested ELISA kits were significantly different.

암모니아 및 가성소다 처리가 Mycotoxin 오염 사료용 볏짚의 사일레지 저장 및 반추위 미생물의 섬유소 분해에 미치는 영향 (The Effect of Ammonia and Sodium Hydroxide Treatment on the Storage and Rumen Microbial Fiber Degradation in Silage of Rice Straw Contaminated Mycotoxin)

  • 본 연구는 볏짚 사일리지의 곰팡이 독소 저감을 위한 효율적 방안으로 화학적 처리 방법에 따른 효능에 관한 진보적 연구 자료를 제공하고자 실시하였다. 화학적 처리 방법으로 암모니아 및 가성소다를 이용 곰팡이독소에 오염된 볏짚의 4% 수준으로 각각 처리하고 사일리지 저장에 따른 곰팡이독소 저감, 발효 품질 및 섬유소 소화에 미치는 영향을 평가 하였다. 모든 실험구에서 Aflatoxin B1, B2, G1, G2 그리고 Fomonisin B1, B2 뿐만 아니라 Dexynivalenol은 검출되지 않았으며, Ochratoxin A와 Zearalenone이 검출되었다. 그리고 Ochratoxin A은 대조구(41.23g/kg)보다 화학적 처리구에서 낮게 검출되었다(p<0.05). Zearalenone은 대조구(600.33㎍/kg) 및 암모니아 처리구(376.00㎍/kg)에 비하여 가성소다 처리구(297.44㎍/kg)가 더 낮은 결과를 나타내었다(p<0.05). 볏짚 사일리지의 pH는 암모니아 처리구(4.63)가 가장 낮았고, 가성소다 처리구가 가장(8.19) 높았다(p<0.05). lactic acid 함량은 대조구와 암모니아 처리구(10.85와 10.98mM)는 유사한 수준이었으나 가성소다 처리구(8.80mM)가 가장 낮았다(p,0.05). Propionic acid는 대조구(0.55mM)가 화학처리구에 비하여 높았으며(p<0.05), 암모니아와 가성소다 처리구간(0.09와 0.07mM)에는 유사한 함량을 나타내었다. 화학처리에 따른 볏짚 사일리지의 반추위 미생물의 NDF 및 ADF의 분해율 모두 가성소다 처리구가 가장 높은 분해율을 보였고 다음으로 암모니아 처리구이었으며, 대조구가 가장 낮은 수준을 나타내었다(p<0.05). 따라서 본 연구결과는 볏짚 사일리지 제조공정에서 암모니아 및 가성 소다 처리가 곰팡이 독소 저감 및 볏짚 사일리지의 반추위 미생물 분해율 증진에 좋은 영향을 주는 것으로 사료되며, 가성소다 처리는 암모니아 처리에 비하여 곰팡이 독소 저감 및 섬유소 분해율 증진에 더 효율적이었으나 사일리지 발효에는 비효율적 영향을 준 것으로 사료된다.

Determination of Aflatoxin M1 and Heavy Metals in Infant Formula Milk Brands Available in Pakistani Markets

  • Aflatoxin $M_1$ ($AFM_1$) after its bioconversion from aflatoxin $B_1$ in animal liver becomes the part of milk while heavy metals get entry into milk and milk products during handling in the supply chain. Aflatoxin $M_1$ and heavy metals being toxic compounds are needed to be monitored continuously to avoid any ailments among consumers of foods contaminated with such toxicants. Thirteen commercially available infant formula milk (IFM) brands available in Pakistani markets were analyzed for the quantitative determination of $AFM_1$ and heavy metals through ELISA and atomic absorption spectrophotometer, respectively. $AFM_1$ was found positive in 53.84% samples while 30.76% samples were found exceeding the maximum EU limit i.e. $0.025 {\mu}g/kg$ for $AFM_1$ in IFM. Heavy metals lead (Pb) and cadmium (Cd) were found below the detection limits in any of the sample, whereas the concentrations of iron (Fe), zinc (Zn) and nickel (Ni) ranged between 45.40-97.10, 29.72-113.50 and <$0.001-50.90 {\mu}g/kg$, respectively. The concentration of Fe in all the tested brands was found in normal ranges while the concentrations of Zn and Ni were found exceeding the standard norms. Elevated levels of $AFM_1$, Zn and Ni in some of the tested IFM brands indicated that a diet completely based on these IFM brands might pose sever health implications in the most vulnerable community i.e., infants.

Effect of Aflatoxin B1 on the Function of Peritoneal Macrophage from Mule Duck

  • This study was conducted to investigate the effect of aflatoxin $B_1$ ($AFB_1$) alone or mixed function oxidase (MFO)-activated $AFB_1$ on various functions of mule duck peritoneal macrophages. Duck peritoneal macrophages were incubated with $AFB_1$ 0, 5, 10, 20, 50 and $100 {\mu}g/ml$ for 12 h. The cell viability significantly declined as the concentration of $AFB_1$ increased and more obviously detrimental effects was noticed in MFO-metabolized $AFB_1$ treatments. Either in opsonized or unopsonized Candida albicans, phagocytotic ability of macrophages was decreased with the elevation of the concentration of $AFB_1$. Significantly higher levels of macrophages were damaged in MFO-metabolized $AFB_1$ than $AFB_1$ alone in concentrations above $20{\mu}g/ml$. The cytotoxicity activity was in the range of 41 to 33% after exposure to $AFB_1$ 5 to $100{\mu}g/ml$, and a significant higher TNF-like substance secretion by lipopolysaccharide (LPS) stimulation was obtained. When LPS was present in the medium, the percentage of cytotoxicity was higher than all treatments of $AFB_1$ both with and without MFO-activation in the absence of LPS. The results suggest that MFO-metabolized $AFB_1$ can alter cell viability and morphology of duck macrophages more than $AFB_1$ administered alone. Both with and without MFOactivation, $AFB_1$ has detrimental effects on phagocytotic ability and TNF-like substance secretion, increasing with level of $AFB_1$.

Aflatoxin B1 Detoxification by Aspergillus oryzae from Meju, a Traditional Korean Fermented Soybean Starter

  • Aflatoxins are classified as Group 1 (carcinogenic to humans) by the International Agency for Research on Cancer. In this study, a total of 134 fungal strains were isolated from 65 meju samples, and two fungal isolates were selected as potential aflatoxin $B_1$ ($AFB_1$)-biodetoxification fungi. These fungi were identified as Aspergillus oryzae MAO103 and A. oryzae MAO104 by sequencing the beta-tubulin gene. The two A. oryzae strains were able to degrade more than 90% of $AFB_1$ (initial concentration: $40{\mu}g/l$) in a culture broth in 14 days. The mutagenic effects of $AFB_1$ treated with A. oryzae MAO103 and MAO104 significantly decreased to 5.7% and 6.4%, respectively, in the frame-shift mutation of Ames tests using Salmonella typhimurium TA98. The base-substituting mutagenicity of $AFB_1$ was also decreased by the two fungi. Moreover, $AFB_1$ production by Aspergillus flavus was significantly decreased by the two A. oryzae strains on soybean-based agar plates. Our data suggest that the two $AFB_1$-detoxifying A. oryzae strains have potential application to control $AFB_1$ in foods and feeds.

Protective Effect of Korean Red Ginseng against Aflatoxin B1-Induced Hepatotoxicity in Rat

  • Korean red ginseng (KRG), the steamed root of Panax ginseng Meyer, has a variety of biological properties, including anti-inflammatory, antioxidant and anticancer effects. Aflatoxin $B_1$ ($AFB_1$) produced by the Aspergillus spp. causes acute hepatotoxicity by lipid peroxidation and oxidative DNA damage, and induces liver carcinoma in humans and laboratory animals. This study was performed to examine the protective effects of KRG against hepatotoxicity induced by $AFB_1$ using liver-specific serum marker analysis, histopathology, and terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling. In addition, to elucidate the possible mechanism of hepatoprotective effects, superoxide dismutase, catalase, glutathione peroxidase, and malondialdehyde were analyzed. Rats were treated with 250 mg/kg of KRG (KRG group) or saline ($AFB_1$ group) for 4 weeks and then received 150 ${\mu}g/kg$ of $AFB_1$ intraperitoneally for 3 days. Rats were sacrificed at 12 h, 24 h, 48 h, 72 h, or 1 wk after $AFB_1$ treatment. In the KRG pre-treatment group, serum alanine aminotransferase, aspartate aminotransferase, and malondialdehyde levels were low, but superoxide dismutase, catalase, and glutathione peroxidase activities were high as compared to the $AFB_1$ alone group. Histopathologically, $AFB_1$ treatment induced necrosis and apoptosis in hepatocytes, and led to inflammatory cells infiltration in the liver. KRG pre-treatment ameliorated these changes. These results indicate that KRG may have protective effects against hepatotoxicity induced by $AFB_1$ that involve the antioxidant properties of KRG.

Induction of Changes in Morphology, Reactive Nitrogen/Oxygen Intermediates and Apoptosis of Duck Macrophages by Aflatoxin B1

  • The purpose of this study was to investigate the effects of aflatoxin $B_1$ ($AFB_1$) on the ultracellular morphology alteration, apoptosis induction and reactive nitrogen and oxygen intermediates production of peritoneal macrophages (DPM) from mule ducks. The ducklings were purchased from a commercial hatchery, and were fed a corn-soybean based diet. As the ducklings were grown up to 3 wk of age, the Sephadex-elicited peritoneal exudative cells (PEC) were used as the source for duck peritoneal macrophages. The ultracellular morphology study showed that significant number of cells shifted from category I (normal cell with ruffled membrane) and II (cell membrane blebbing) to category III (cell membrane blebbing and even rupture) after DPM were incubated with $AFB_1$ ($20{\mu}g/ml$) for 12 to 48 h. When DPM were exposed to $AFB_1$ in vitro, the production of NO, $H_2O_2$ and $O_2{^-}$ in macrophages was reduced after 12-48 h incubation with previous LPS stimulation. There was a DNA laddering pattern observed in DPM incubated with $AFB_1$ 5, 10, 20, 50 or $100{\mu}g/ml$ for 12 h. Evidence also revealed that the percentage of apoptotic cells was increased along with the elevation of $AFB_1$ concentration. The results suggest that $AFB_1$ exposure causes duck macrophages going on apoptotic pathway through evidence of ultracellular morphology alteration and DNA laddering in agarose electrophoresis. The production of reactive nitrogen and oxygen intermediates of duck macrophages also depressed after $AFB_1$ exposure, and this implied that $AFB_1$ could cause deteriorated functions of bacteriocidal and tumoricidal activity in duck macrophages.

LC-MS/MS를 이용한 반려동물 사료에서 곰팡이독소 동시분석법 유효성 확인 (Validation of simultaneous mycotoxin analysis method in pet food using LC-MS/MS)

  • The simultaneous analysis of mycotoxins using LC-MS/MS, a food official analysis method, was applied with compound feed for pets with high consumer preferences. In this study, the linearity of all calibration curves showed good linearity of 0.99 or more. and both the accuracy (recovery rate) and precision (repeatability) criteria of the concentration range for each mycotoxin in the National Agricultural Products Quality Management Service's Validation and Verification Guidelines were met. And as a result of analyzing FAPAS QCM in the same way, it was assesed that the z-scores of Aflatoxin B1, Ochratoxin A, Zearalenone, and Fumonisin B1, were within ±2 range. This study showed that the application of the food official analysis method to compound feed for pets is suitable.

식품중 총 아플라톡신의 노출량 평가 (Exposure Assessment of Total Aflatoxin in Foods)

  • HPLC/FLD를 이용하여 곡류, 견과류 등 총 565건을 대상으로 총 아플라톡신 함량을 조사하여 노출량을 평가하였다. B와 G 그룹의 검출한계는 각각 0.05, 0.07 ng/g이였고, 쌀, 땅콩버터 및 고춧가루에 대한 아플라톡신($B_{1}$, $B_{2}$, $G_{1}$$G_{2}$)의 회수율은 양호한 수준이었다. 총 아플라톡신은 전체 시료 565건 중 27건(4.77%)이 검출되었으며, 검출빈도는 곡류 0.17%, 견과류 0.35%, 가공식품 3.01%, 기타식품 1.24%을 보여주었고, 두류 및 건조과일류는 검출되지 않았다. 식이섭취량을 고려한 총 아플라톡신의 1일 1인당 평균섭취량은 0.04 ng/kg bw/day이였다.
