Aflatoxin에 노출된 닭에서 활성탄과 어성초의 독성완화 효과 (Alleviative effects of activated charcoal and Houttuynia cordata Thunb. in broiler chickens during aflatoxicosis)

  • This study was conducted to evaluate the alleviative effects of activated charcoal (AC) and Houttuynia cordata (HC) singly or in combination in broiler chickens during aflatoxicosis. Activated charcoal (1% or 0.5%) and H. cordata (1% or 0.5%) were mixed into the diets for the ability to reduced the deleterious effects of 2.4mg total aflatoxin $(AFB_1)kg^{-1}$ diet on growing broiler chickens from 1 to 21 days of age. A total of 160 1-day-old (Hyline Variety Brown) broiler chicks were housed in eight treatment groups [Control, $AFB_1$, AC 1%, HC 1%, $AFB_1$ plus AC 1% plus HC 1%, $AFB_1$ plus AC 1% plus HC 0.5%, $AFB_1$ plus AC 0.5% plus HC 1%, $AFB_1$ plus AC 0.5% plus HC 0.5%] each consisting of 20 chicks. Compared to control, 2.4mg $AFB_1$ alone treatment group significantly decreased body weight gains of chickens. The addition of mixed AC 1% and HC 1% including 6, 7 groups to the 2.4mg $AFB_1$-containing diet moderately reduced the adverse effects of $AFB_1$ on performances of chickens. The chickens consuming 2.4mg $AFB_1$ plus AC 0.5% and HC 0.5%-containing diet showed very slightly reduced the adverse effects on investigated parameters compared to the $AFB_1$ only treated group. Also, the single addition of AC or HC to the $AFB_1$-free diet had no adverse effects in chickens. These results suggest that AC and HC mixed can reduced the aflatoxicosis in broilers and may be contribute to a solution of the aflatoxicosis problem in poultry production.

Effect of Polyherbal Feed Supplement "Growell" during Induced Aflatoxicosis, Ochratoxicosis and Combined Mycotoxicoses in Broilers

  • An experiment was conducted to study the protective role of polyherbal feed supplement (Growell) during induced mycotoxicosis in broilers. A total of 240 Vencobb broilers were divided at day old stage into eight equal groups. Group A served as control and was given plain feed, group B, D, F and H were given Growell at 0.35 g/kg of feed. Group C, D, G and H were given dietary aflatoxin $B_1$ at 0.2 ppm and groups E, F, G and H were given ochratoxin A at 0.2 ppm in feed to study effect of Growell on individual aflatoxicosis, ochratoxicosis and combined mycotoxicosis of broilers. The chicks were given their respective feeds from 1st day to 6th week of age and were vaccinated at 7th and 28th day of age with Lasota strain of Newcastle disease virus. There was no statistically significant effect of mycotoxins individually or in combination on body weight of broilers. However, body weights were highest in group B and lowest in co-mycotoxicated group G. Feed conversion ratio was best in group B followed by A, D, F, E, H and G. Significant improvement in haemoglobin values was observed in broilers due to feeding of Growell in ochratoxin and co-mycotoxicated groups. There was no significant effect of mycotoxin treatment on PCV, TEC and TLC of broilers. Due to single and combined mycotoxicosis, there was reduction in serum total protein, albumin, cholesterol and triglyceride and rise in alkaline phosphatase, creatinine and uric acid levels. Supplementation of diets with Growell reduced the alterations induced due to mycotoxins. There was a significant rise in per cent organ weight of liver and reduction of that of spleen, bursa of Fabricius and thymus of broilers fed mycotoxins. Protection from alteration in per cent organ weight of these organ by supplementation of Growell was recorded. The observed impaired immune response and histopathological changes in liver, kidney, spleen, bursa of Fabricius and thymus of broilers given mycotoxins were protected by supplementation of Growell.

Biochemical and Histopathological Study of Aflatoxicosis on Ross 308 Broiler Chicks

  • Totally, one hundred and sixty 1-day-old Ross 308 broiler chicks were fed with a diet containing 0, 0.5, 1.0, and 2.0 mg of aflatoxin $B_1(AFB_1)/kg$ of feed for 21 days. Body weight was lower for the $AFB_1$-treated broilers than for the control group. At 14 and 21 DPF, the broilers fed with 2.0 mg of $AFB_1/kg$ of feed weighed significantly lower than those of the other groups (p<0.05). Relative liver weights increased significantly in a dose-dependent manner, and relative spleen weights were significantly high in the chicks fed with 2.0 mg of $AFB_1/kg$ of feed at 21 DPF (p<0.001). Biochemical analyses showed that total protein and albumin levels decreased significantly at 7 and 14 DPF for the chicks of the group fed with 2.0 of mg $AFB_1/kg$ of feed, compared with those fed with 0.5 and 1.0 mg of $AFB_1/kg$ of feed (p<0.05). AST and ALT levels increased significantly at 14 and 21 DPF (p<0.05), and the AST levels, particularly, increased dose-dependently (p<0.05). Histopathological analyses showed that the liver tissues of the $AFB_1$-treated chicks showed significant lesions, including hemorrhage, hepatocyte necrosis, inflammatory cell infiltration, and fatty degeneration. The severity of both hepatocyte necrosis and inflammatory cell infiltration appeared to increase dose- and time-dependently. Similarly, hepatic fibrosis increased dose-dependently (p<0.05). The results of this study could improve our understanding of parameters used for evaluating aflatoxicosis in poultry.

Study on Applying Artichoke Extract to Lessen The Toxicity of Aflatoxin to Chicken

  • The Artichoke extract at 10% was used to add in drinking water to understand its effect on Aflatoxicosis of chickens. The Artichoke extract at the dose of 6 ml per liter of drinking water was given (experiment group) or not (control group) and to Hybro chickens (150 birds), during the first 49 days of life. Also, the chickens were fed with foodstuff containing 200 ppb or 500 ppb Aflatoxin $B_1$. Results showed that, the chickens having Artichoke extract: (1) Had overcome the growth retardation caused by the toxin at concentration of 200 ppb and 500 ppb of Aflatoxin $B_1$ (an addittonal weight gain of about 200-400 g/bird). (2) The feed conversion was improved (a reduction of 200-400 g of feed per kg of bird living weight). (3) Aflatoxicosis lesions were mild in the chickens, which fed 500 ppb of Aflatoxin $B_1$ or not found in those having the toxin 200 ppb. The blood examinations at 28th and 49th days of the trial gave the following results: (1) The Artichoke extract had an effect of suppressing the changes of blood cell numbers, hemoglobin amount. packed cell volume. leukocyte formula that were caused by Aflatoxin $B_1$. (2) The Artichoke extract had an effect of suppressing the diminution oj sugar, protein levels and the increase of the levels of GOT, GPT, alkaline phosphatase and bilirubin in the blood of intoxicated chickens. There was not or very Jew residue of Aflatoxin $B_1$ contained in the liver and muscle of chickens intoxicated by Aflatoxin $B_1$ having Artichoke, that was much lower than the allowed level in animal products.

Effects of Modified Montmorillonite Nanocomposite on Growing/Finishing Pigs during Aflatoxicosis

  • Experiments were conducted to determine the efficacy of modified montmorillonite nanocomposite (MMN) to reduce the toxic effects of aflatoxin (AF) in growing/finishing pigs. 96 weaned pigs were assigned to four dietary treatment groups (0 g of MMN and 0 mg of AF/kg feed, 3 g of MMN/kg feed, 0.1 mg of AF/kg feed, and 3 g of MMN plus 0.1 mg of AF/kg feed). Body weight gain (BW gain), feed/gain ratio, serum biochemical values and enzyme activities were evaluated. Compared with the control, AF alone markedly reduced BW gain and resulted in a significantly higher feed/gain ratio. There were no differences in BW gain and feed/gain ratio between 0.3% MMN or 0.3% MMN plus AF and the control. These results suggested that the deleterious effects of AF were ameliorated by MMN addition. AF intake markedly increased relative organ weights of liver, kidney, spleen and pancreas, and resulted in significant alterations of serum parameters. However, these parameters for pigs fed diets containing MMN and AF returned to normal values, indicating that MMN had the ability to recover the AF-decreased performance, organ damage and to correct aberrations in serum parameters. These findings in our study suggested that MMN can effectively modulate the toxicity of AF in growing/finishing pigs and may offer a novel approach to the preventive management of aflatoxicosis in animals.

In-field evaluation of clinoptilolite feeding efficacy on the reduction of milk aflatoxin M1 concentration in dairy cattle

  • Background: Clinoptilolite is a natural zeolite with high adsorption capacity for polar mycotoxins such as aflatoxins. The efficacy of clinoptilolite in ameliorating the toxic effects of aflatoxicosis has been proven in monogastric animals, but there is no such evidence for ruminants. The aim of this study was to evaluate, under field conditions, whether the dietary administration of clinoptilolite in dairy cows could reduce the concentration of aflatoxin M1 ($AFM_1$) in bulk-tank milk, in farms with higher than or close to $0.05{\mu}g/kg$ of milk (European maximum allowed residual level). An objective of the present study was also to investigate the effect of particle size of clinoptilolite on aflatoxin binding. Methods: Fifteen commercial Greek dairy herds with AFM1 concentrations in bulk tank milk ${\geq}0.05{\mu}g/kg$ were selected. Bulk tank milk AFM1 was determined prior to the onset and on day 7 of the experiment. Clinoptilolite was added in the total mixed rations of all farms at the rate of 200 g per animal per day, throughout this period. Two different particle sizes of clinoptilolite were used; less than 0.15 mm in 9 farms (LC group) and less than 0.8 mm in 6 farms (HC group). Results: Clinoptilolite administration significantly reduced $AFM_1$ concentrations in milk in all farms tested at an average rate of 56.2 % (SD: 15.11). The mean milk $AFM_1$ concentration recorded on Day 7 was significantly (P < 0.001) lower compared to that of Day 0 ($0.036{\pm}0.0061$ vs. $0.078{\pm}0.0074{\mu}g/kg$). In LC group farms the reduction of milk $AFM_1$ concentration was significantly higher than HC group farms ($0.046{\pm}0.0074$ vs. $0.036{\pm}0.0061{\mu}g/kg$, P = 0.002). As indicated by the Pearson correlation, there was a significant and strong linear correlation among the milk $AFM_1$ concentrations on Days 0 and 7 (R = 0.95, P < 0.001). Conclusions: Dietary administration of clinoptilolite, especially of smallest particle size, at the rate of 200 g per cow per day can effectively reduce milk $AFM_1$ concentration in dairy cattle and can be used as a preventive measure for the amelioration of the risks associated with the presence of aflatoxins in the milk of dairy cows.

항산화 비타민과 Aflatoxin$B_1$의 혼합 투여가 마우스간의 지질 함량 및 지방산 조성에 미치는 영향 (The Effects of Aflatoxin $B_1$Co-administrated with Antioxidant Vitamins on Lipid Contents and Fatty Acids Composition of Liver in Mice)

  • Aflatoxin은 자연속에 존재하며 높은 독성을 가진 곰팡이 독소이다. 이들 곰팡이 독소는 Aspergillus flavus와 Aspergillus parasiticus로부터 생성되는 대사산물로서 식품에서 다양한 오염 경로를 가지고 발생한다. 특히 Aflatoxin B$_1$(AFB$_1$)은 사람에게 있어서 간독성 및 간암을 유발하는 잠재력을 가진 곰팡이 독소이며, 지질과산화 반응은 aflatoxin B$_1$에 의한 세포 산화적 손상시 발생하는 주요 현상중의 하나이다. 따라서 본 연구의 목적은 aflatoxin B$_1$과 항산화 비타민 혼합 투여 후 유도된 간과 혈청의 지질 구성 성분 및 지방산 조성에 미치는 영향을 조사하기 위하여 수행되었다. 특히 비타민 C와 비타민 E는 중요한 항산화제에 속하는 것으로 이들을 복강내 주사(i.p, intraperitoneal injection)를 통하여 각각 10 mg/kg, 63.8 mg/kg씩 male ICR mice에 투여하였으며 1시간후 0.4mg/kg의 AFB$_1$을 동일한 방법으로 투여하였다. AFB$_1$에 의하여 유도된 지질구성 성분의 변화에 있어서 AFB$_1$, 단독 투여군과 항산화 비타민 혼합 투여군을 비교하였을 때 혈청에서의 유리콜레스테롤, 중성지질, 총 콜레스테롤등은 AFB$_1$, 단독 투여군에서 2배 이상 증가되었다. 간에서의 지방산 조성에서도 AFB$_1$, 단독 투여군에서 항산화비타민 혼합투여군보다 콜레스테롤 에스테르, 중성지질, 총콜레스테롤 및 인지질의 P/S ratio가 감소하였다. 이상의 결과로 미루어 볼 때 AFB$_1$에 의한 세포손상이 항산화 비타민에 의해 감소되는 효과가 있는 것을 확인할 수 있었다.
