Application of Electron-Beam Irradiation Combined with Aging for Improvement of Microbiological and Physicochemical Quality of Beef Loin

  • The combined effects of irradiation and aging temperature on the microbial and chemical quality of beef loin were investigated. The samples were vacuum-packaged, irradiated at 0 or 2 kGy using electron-beam (EB), and stored for 10 d at different aging temperatures (2, 14, or 25℃). The microbial growth, shear values, meat color, and nucleotide-related flavor compounds of the samples were analyzed. The irradiation effect on inactivation of foodborne pathogens was also investigated. The population of Listeria monocytogenes and Escherhia coli O157:H7 inoculated in beef samples decreased in proportion to the irradiation dose. Irradiation reduced the total aerobic bacteria (TAB) over the storage, but higher aging temperature increased the TBA. Thus TAB increased sharply in non-irradiated and high temperature-aged (14, 25℃) beef samples after 5 d. With increasing aging temperature and aging time, shear force values decreased. Lipid oxidation could be reduced by short aging time at low aging temperature. The color a* values of the irradiated beef were lower than those of the non-irradiated throughout the aging period. As aging period and temperature increased, IMP decreased and hypoxanthine increased. Considering microbial and physicochemical properties, irradiation can be used for raw beef to be aged at relatively high temperature to shorten aging time and cost.

  • DO, ORP and pH values measured during SBR operation can provide information about removal reaction of organic contaminants and nutrient materials in the reactor. It is already generalized control strategy to control reaction phase time using their special patterns indicating the end of the removal reactions. However, those informations are limited to point out the end time of oxidative reaction in the aerobic phase or reductive reaction in the anoxic phase without giving quantitative value of influent loading level. In this research, a diagnosis algorithm which can estimate the loading level of carbon and ammonia as high, medium and low was developed using the basic measurements like DO, ORP, and pH. It will be possible to know the level of influent loading rate from those online measurements without experimental analysis.

Effect of Palm oil and Soybean oil on the Quality and Shelf-life of Yugwa Base (팜유와 대두유가 유과 바탕의 품질과 저장성에 미치는 영향)

  • The shelf-life of Yugwa(Busuge) is very short because of the low oxidative stability of soybean oil which is conventionally used as the frying oil of Yugwa base with high porous structures. To identify the possibility of replacing soybean oil with palm oil whose oxidative stability is high, POV(peroxide value), AV(acid value), colorness (L. a and b), hardness and sensory characteristics during storage of the Yugwa base fried with the soybean oil and palm oil stored in the anaerobic packaging or the aerobic packaging at 10 and $30^{\circ}C$ for 75 days were measured. POV and AV of the Yugwa base fried with palm oil were significantly lower than those of Yugwa fried with the soybean oil. Hardness and colorness were not different each other and, sensory evaluation was slightly higher than that fried with soybean oil. Lipid absorption amount was not significantly different between the Yugwa base fried with palm oil and the one fried with soybean oil.

Changes in the Ammonia-Oxidizing Bacteria Community in Response to Operational Parameters During the Treatment of Anaerobic Sludge Digester Supernatant

  • The understanding of the relationship between ammonia-oxidizing bacteria (AOB) communities in activated sludge and the operational treatment parameters supports the control of the treatment of ammonia-rich wastewater. The modifications of treatment parameters by alteration of the number and length of aerobic and anaerobic stages in the sequencing batch reactor (SBR) working cycle may influence the efficiency of ammonium oxidation and induce changes in the AOB community. Therefore, in the research, the impact of an SBR cycle mode with alternating aeration/mixing conditions (7 h/1 h vs. 4 h/5.5 h) and volumetric exchange rate (n) on AOB abundance and diversity in activated sludge during the treatment of anaerobic sludge digester supernatant at limited oxygen concentration in the aeration stage (0.7 mg $O_2/l$) was assessed. AOB diversity expressed by the Shannon-Wiener index (H') was determined by the cycle mode. At aeration/mixing stage lengths of 7 h/1 h, H' averaged $2.48{\pm}0.17$, while at 4 h/5.5 h it was $2.35{\pm}0.16$. At the given mode, AOB diversity decreased with increasing n. The cycle mode did not affect AOB abundance; however, a higher AOB abundance in activated sludge was promoted by decreasing the volumetric exchange rate. The sequences clustering with Nitrosospira sp. NpAV revealed the uniqueness of the AOB community and the simultaneously lower ability of adaptation of Nitrosospira sp. to the operational parameters applied in comparison with Nitrosomonas sp.

A Role and Properties of $C_{1}$ Enriched Cellulase Fraction from Anaerobic Clostridium thermocellum in Cellulose Degradation (섬유소 분해시 혐기성 Clostridium thermocellum이 생산하는 Cellulase의 $C_{1}$ 성분의 역할과 성질)

  • A $C_{1}$ enriched cellulase fraction was separated from culture filtrate of anaerobic Clostridium thermocellum by hydroxyapatite column chromatography. The separated fraction showed strong synergistic action with $C_{x}$ component (endo-$\beta$-1, 4-glucanase) in digestion of crystalline cellulose, similar to the other aerobic cellulolytic microorganisms. Unlike the $C_{x}$ component the $C_{1}$ enriched fraction was rapidly inactivated by oxidation at the atmospheric condition. The enzyme activity was significantly enhanced by the addition of reducing agents, especially $\beta$-mercaptoethanol, which indicates that a $C_{1}$ component has a lot of sulfhydryl groups essential for the enzyme activity. The effect of metal ions on $C_{1}$ activity was also investigated. The $C_{1}$ fraction was found to be thermally stable compare to endo-$\beta$-1,4-glucanase. Optimal temperature and pH were found to be $60^{\circ}C$ and 6.0, respectively.

Microbiological and Physicochemical Quality of Irradiated Ground Beef as Affected by Added Garlic or Onion

  • The effects of garlic and onion on the microbiological and physicochemical properties of irradiated ground beef patties were evaluated. Ground beef was mixed with 0.5%(w/w) minced garlic or 2.5%(w/w) minced onion, vacuum-packed in oxygen-impermeable nylon/PE bags and then electron beam-irradiated at 2.5 kGy. All samples were kept at $4^{\circ}C$ for 8 d. Irradiation resulted in a 2-log CFU/g reduction in both aerobic and coliform bacteria. The microbial counts gradually increased during storage, but those in beef that contained garlic or onion were 1-log CFU/g lower than those of the control samples after 8 d of storage. The pH value decreased during storage in all meat samples, but this decrease was greater in non-irradiated beef than in irradiated beef. The lipid oxidation, volatile basic nitrogen content and Hunter color values of raw patties were generally not affected by irradiation or the addition of garlic and onion. Sensory evaluation of cooked patties showed that the off-odor was less pronounced in samples that contained added garlic or onion than in control samples, and that the overall acceptability of beef formulated with garlic was the highest. Overall, the results of this study indicate that the addition of garlic or onion in combination with irradiation treatment enhanced the microbial quality and improved the sensory quality of irradiated ground beef.

Changes in Phosphorus and Sediment Oxygen Demand in Coastal Sediments Promoted by Functionalized Oyster Shell Powder as an Oxygen Release Compound

  • In this study, we performed a sediment elution experiment to evaluate water quality in terms of phosphorus, as influenced by the dissolved oxygen consumed by sediments. Three separate model column treatments, namely, raw, calcined, and sonicated oyster shell powders, were used in this experiment. Essential phosphorus fractions were examined to verify their roles in nutrient release from sediment based on correlation analyses. When treated with calcined or sonicated oyster shell powder, the sediment-water interface became "less anaerobic," thereby producing conditions conducive to partial oxidation and activities of aerobic bacteria. Sediment Oxygen Demand (SOD) was found to be closely correlated with the growth of algae, which confirmed an intermittent input of organic biomass at the sediment surface. SOD was positively correlated with exchangeable and loosely adsorbed phosphorus and organic phosphorus, owing to the accumulation of unbound algal biomass-derived phosphates in sediment, whereas it was negatively correlated with ferric iron-bound phosphorus or calcium fluorapatite-bound phosphorus, which were present in the form of "insoluble" complexes, thereby facilitating the free migration of sulfate-reducing bacteria or limiting the release from complexes, depending on applied local conditions. PCR-denaturing gradient gel electrophoresis revealed that iron-reducing bacteria were the dominant species in control and non-calcined oyster shell columns, whereas certain sulfur-oxidizing bacteria were identified in the column treated with calcined oyster powder.

Assessment of chicken thigh meat quality of Ross 308 broiler of animal welfare certified farm

  • Objective: This study aimed to evaluate the difference in the thigh meat quality of Ross 308 broiler from conventional and welfare farms. Methods: Thigh meat samples of Ross 308 broilers (age, 35 d; carcass weight, 1.1 kg) from conventional farm (RCF, n = 60) and animal welfare farms (RAWF, n = 60) were analyzed. Proximate composition, pH, color (lightness, redness, and yellowness), water-holding capacity (WHC), shear force, total aerobic bacteria (TAB), and volatile basic nitrogen (VBN) were measured and the levels of bioactive compounds such as dipeptides (anserine and carnosine), creatine, creatinine, and their anti-oxidation activity were determined. Results: The RCF and RAWF did not differ significantly in their proximate composition, WHC, color, and creatine and carnosine levels. The pH value was significantly lower in RAWF than in RCF on day 7. The shear force value was significantly higher in RAWF than in RCF throughout the storage duration. TAB in RCF on day 9 were significantly higher than those in RAWF. The VBN content of RAWF was significantly lower than that of RCF after 5 days of storage. Creatinine content was significantly higher in RAWF (3.50 mg/100 g) than in RCF (3.08 mg/100 g) on day 1. Along with higher carnosine and anserine contents of RAWF, it had significantly higher 2,2-diphenyl-1-picrylhydrazyl and 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activities than those of RCF. Conclusion: These results imply that the animal welfare farming system beneficially affects the overall oxidative stability of Ross 308 thigh meat.

The Effect of Washing of Carcasses with Sodium Hypochlorite Solution and Vacuum Packaging on the Microbiological and Physiochemical Quality of the Breast Meat from Old Hen during Storage at 4℃ (차아염소산나트륨 세척 및 진공 포장이 노계 가슴육의 냉장 저장 중 미생물학적 및 이화학적 품질에 미치는 영향)

  • This study was conducted to examine the effect of spray-washing old hens (old laying hens, old molting hens and old breeder hens) carcasses with sodium hypochlorite (50 ppm) solution and vacuum packaging on the microbiological and physiochemical quality of breast meat during storage at $4^{\circ}C$. There were no significant differences of cooking loss and texture among breast meats from different birds with spray-washing by water or sodium hypochlorite solution before storage. The numbers of total aerobic bacteria of aerobically packaged and only water-washed breast meat were higher than 7 Log CFU/g (the limitation population for spoilage of meat) after 7 days of storage. However, the numbers of total aerobic bacteria of vacuum packaged breast meat washed by sodium hypochlorite solution were lower than 7 Log CFU/g even after 14 days of storage. The volatile basic nitrogen content of vacuum packaged breast meat were lower than 20 mg% (the limitation value for spoilage of meat) regardless of the washing method, while those of aerobically packaged breast meat washed by tap water and sodium hypochlorite solution were 41.1 and 20.1 mg%, respectively. In addition, lipid oxidation and change of pH in breast meat was inhibited by vacuum packaging when compared with that of aerobic packaged breast meat during storage. Therefore, the results indicated that the washing old hen's carcasses with sodium hypochlorite (50 ppm) solution and vacuum packaging could improve the shelf-life of meat from old birds. This result can provide the basic information for industry, which are seeking for export market.

Analysis of Nitrogen and Phosphorus Benthic Diffusive Fluxes from Sediments with Different Levels of Salinity (염분농도에 따른 호소 퇴적물 내 질소 및 인 용출 특성 분석)

  • The study involved the categorization of domestic lakes located in South Korea into three groups based on their salinity levels: upstream reservoirs with salinity less than 0.3 psu, estuarine reservoirs with salinity ranging from 0.3 to 2 psu, and brackish lagoons with salinity exceeding 2 psu. Subsequently, the research assessed variations in the concentrations of total nitrogen (T-N) and total phosphorus (T-P) in the sediment of these lakes using statistical analysis, specifically one-way analysis of variance (ANOVA). Additionally, a laboratory core incubation test was conducted to investigate the benthic nutrient fluxes in Songji lagoon (salinity: 11.80 psu), Ganwol reservoir (salinity: 0.73 psu), and Janggun reservoir (salinity: 0.08 psu) under both aerobic and anoxic conditions. The findings revealed statistically significant differences in the concentrations of T-N and T-P among sediments in the lakes with varying salinity levels (p<0.05). Further post-hoc analysis confirmed significant distinctions in T-N between upstream reservoirs and estuarine reservoirs (p<0.001), as well as between upstream reservoirs and brackish lagoons (p<0.01). For T-P, a significant difference was observed between upstream reservoirs and brackish lagoons (p<0.01). Regarding benthic nutrient fluxes, Ganwol Lake exhibited the highest diffusive flux of NH4+-N, primarily due to its physical characteristics and the inhibition of nitrification resulting from its relatively high salinity. The flux of NO3--N was lower at higher salinity levels under aerobic conditions but increased under anoxic conditions, attributed to the impact of salinity on nitrification and denitrification. Additionally, the flux of PO43--P was highest in Songji Lake, followed by Ganwol Lake and Janggun Reservoir, indicating that salinity promotes the diffusive flux of phosphate through anion adsorption competition. It's important to consider the influence of salinity on microbial communities, growth rates, oxidation-reduction processes, and nutrient binding forms when studying benthic diffusive nutrient fluxes from lake sediments.
