The Effects of Salt and NaNO2 on Fatty Acid Composition, Free Amino Acids, Microbial Counts and Sensory Characteristics of Dry-cured Ham Processed under Korean Environment (소금과 아질산염 처리수준이 자연숙성 건염햄의 지방산 조성, 유리아미노산, 미생물수 및 관능적 특성에 미치는 영향)

  • The study analyzed the effects of salt concentration [high salt (HS) and low salt (LS)] and sodium nitrite ($NaNO_2$), which are typically utilized in Korean processing facilities, on fatty acid composition, free amino acids, microbial counts and sensory characteristics of processed dry-cured ham. Four different treatments were considered: three hams (11.30 kg) salted with 92 g/kg salt (w/w) (HS), three hams (10.65 kg) treated with HS and 100 ppm $NaNO_2$ (HS+$NaNO_2$), three hams (11.42 kg) salted with 62 g/kg salt (w/w) (LS), and three hams (10.62 kg) treated with LS and 100 ppm $NaNO_2$ (LS+$NaNO_2$). Fatty acid composition analysis revealed significantly (p<0.05) higher saturated fatty acid and lower (p<0.05) unsaturated fatty acid in the HS+$NaNO_2$ group compared with the other groups. Glutamate, alanine and lysine free amino acids were higher than the other free amino acids. The processing conditions did not significantly affect the free amino acids of biceps femoris muscles, except for the proline content (p>0.05). In sensory evaluation, the fermentation aroma of the LS group was higher than that of the HS group. The aerobic counts consistently ranged from from $2.3{\times}10^2$ to $1.11{\times}10^4$ CFU/g. Escherichia coli including strain O157:H7, Staphylococcus aureus, and Salmonella spp. were not detected.

Chemical Changes of Meju made with Barly Bran Using Fermentation (보리등겨로 제조한 메주의 발효기간에 따른 각종 성분 변화)

  • For investigation of new utilization as jang-products, Meju was prepared using barely bran. As barley meju was fermented, change of pH was $5.2{\sim}5.6$, it was indistinguishable change. L-value of color was changed from 46.9 to 60.3, that meant it was getting moe dark. The counts of aerobic bacteria were $4.8{\times}10^7{\sim}5.6{\times}10^9$ CFU/g, it was extraordinarily increased during fermentation. Counts of Yeast, molds, and bacteria were $9.1{\times}10^6{\sim}5.0{\times}10^8$ CFU/g, $8.3{\times}10^5{\sim}6.9{\times}10^7$, and $2.0{\times}10^2{\sim}4.5{\times}10^6$ CFU/g, respectively. Crude ash content was $3146.0{\sim}7147.4$ mg%. The level of K was the highest in quantity among the crude ash in barely meju. 7 free sugars(i.e., raffnose, stachyose, inositol, fructose, glucose, arabinose, and maltose), 3 volatile organic acid(i.e., acetic acid, propionic acid, and butyric acid) and 4 non-volatile organic acid(i.e., fumaric acid, ${\alpha}-ketoglutaric$ acid, malic acid, and citric acid) were detected. The content of free amino acid was $596.3{\sim}1580.8$ mg%. Glutamic acid was most abundant component among the amino acids, 2nd abundant component was alanine, it's content was $79.9{\sim}165.3$ mg%, 3rd abundant component was leucine, it's count was $41.7{\sim}161.6$ mg%. Finally, essential amino acid content was revealed $33.2{\sim}40.38%$.

Quality Characteristics of Cupcakes Added with Opuntia ficus-indica var. saboten Powder (백년초열매 분말 첨가 컵케이크의 품질특성)

  • Antioxidative activity and functional properties of the powder obtained from ground Opuntia ficus-indica var. saboten fruit were studied. Cupcakes containing 1,3, and 5% (w/w) of O. ficus-indica vu. saboten powders were prepared and evaluated for their sensory property, textural quality, and shelf-life. The antioxidative activity, measured by DPPH radical scavenging activity of O. ficus-indica var. saboten powder, increased as the concentrations of O. ficus-indica var. saboten powder increased. No coliform bacteria, yeasts, and molds were detected in either the O. ficus-indica var. saboten powder or in the cupcakes. However, total aerobic bacteria counts were 4.41 log CFU/g in the O. ficus-indica var. saboten powder and were a negligible level (<$10^2$ CFU/g) in the cupcakes. Moisture content of the cupcakes was not significantly different in all samples. Lightness (L) and yellowness (b) of cupcake color decreased as the concentration of O. ficus-indicar var. saboten powder increased, whereas the redness (a) increased. Increasing the concentration of O. ficus-indica var. saboten powder, the mechanical characteristics of the cupcakes, such as hardness, gumminess, and chewiness, while decreasing cohesiveness and springiness. The sensory properties, such as color, flavor, taste, texture, and overall acceptability, of the cupcakes containing the 1% 0. ficus-indica var. saboten powder were superior to the control sample. The results exhibited that the adding the O. ficus-indica var. saboten powder into the cupcakes increased antioxidant activity and showed no effect on shelf-life of the cupcakes. The highest quality improvement was obtained by incorporating the 1% (w/w) of 0. ficus-indica var. saboten powder into the cupcake formula.

Microbiological Hazard Analysis of Hot Pepper Farms for the Application of Good Agricultural Practices (GAP) System (농산물우수관리제도 (GAP) 적용을 위한 고추농가의 미생물학적 위해도 평가)

  • The objective of this study was to determine microbiological risk factors in hot pepper farms for the application of good agricultural practices (GAP). Samples were collected from cultivation environments and utensils, plants, workers, and air at 3 hot pepper farms located in Cheongsong, Korea and were tested to detect sanitary indications [aerobic plate bacteria (APC), coliform, and Escherichia coli], foodborne pathogens, and fungi. APC, coliform, and fungi were detected at the levels of 0.7~6.2, 0.2~4.7, and 0.4~4.3 log CFU, respectively, in the three farms. Four (4.4%; l leaf, l irrigation water, and 2 soil) of 90 samples collected were revealed to be E. coli positives. For foodborne pathogens, Staphylococcus aureus was only detected at $1.0log\;CFU/100cm^2$ in the worker's cloth of B farm, and Bacillus cereus was detected at the levels 1.0~2.5 log CFU in the cultivation environments and utensils and worker of B and C farms. However, other pathogens were not detected. The results demonstrated potential microbiological risks for hot pepper cultivated in the farms. Therefore, a management system to minimize the microbial risk such as GAP is required to ensure the safety of hot pepper.

Characteristics of Seepage Water and Groundwater in a Coastal LPG Storage Cavern of Jeonnam (전남 해안 LPG 저장공동 유출수와 주변 지하수의 수질특성)

  • Water curtain of an underground LPG storage cavern is a facility to prevent leakage of high pressure gases, for which groundwater should flow freely towards the cavern and groundwater level also must be stably maintained. In this study, in order to evaluate qualities of seepage water and surrounding groundwater of an underground LPG storage cavern in Yeosu, 4 rounds of samplings, field measurements and laboratory analyses (February, May, August, October of 2007) were conducted. According to field measurements, pH was weak acidic to neutral but it gradually increased with time. Electrical conductivity (EC) of groundwater near a salt stack showed very high values between 10.47 and 38.50 mS/cm. Dissolved oxygen (DO) showed a very wide range of 0.20~8.74 mg/L and a mean of oxidation-reduction potential (ORP) was 159 mV, which indicated an oxidized condition. Levels of $Fe^{2+}$ and $Mn^{2+}$ were mostly less than 3 mg/L. All of seepage waters showed a Na-Cl type while only groundwater near the salt stack showed a Na-Cl type with a high total dissolved solid. The other groundwaters exhibited typical $Ca-HCO_3$ types. Levels of aerobic bacteria were mostly very high (573-39,520 CFU/mL). Based on the analyses of these hydrochemistry and biological characteristics, it is concluded that there are no particular problems in groundwater and seepage water, which not causing a trouble in the cavern operation. However, both for control of bio-clogging and for sustainable operation of the water curtain system, a regular hydrochemical and microbiological monitoring is required for the seepage water and surrounding groundwater.

  • Biogas production from agricultural wastes were summarized as follows: 1. Biogas Generation Characteristics of Various Manures and Residues a. Gas yield from crop residues like rice straw, rice hull, corn stalk and coconut husk can be improved by addition of animal manures. b. Gas yield from coconut husk can be improved through aerobic fermentation for at least one week before loading in the digester. c. Gas yield from fresh rice straw is better than from pre-fermented one, whether alone or in combination with animal manures. d. Initial study has shown that fresh azolla can be substituted for animal manures in manurerice straw combinations and gas yield derived based on unit volatile solids loaded is actually better than for manure-residue combinations. e. Gas production is highly sensitive to substrate pH and becomes almost nil at a pH of below 6. 2. Effect of ambient conditions and other factors on biogas production in a house hold-size digester. a. Results showed that compaction of rice straw in straw-manure combination can reduce gas yield compared with loosely mixed straw. b. The effective gas production period extended to 70 days using freshly threshed rice straw and fresh cattle manure as feed material. c. Underground and above ground digesters with shade have relatively more stable substrate temperature than aboveground exposed digesters. This relative temperature instability may likely be the reason for lower gas yield for the exposed aboveground digester loaded with loose straw-cattle manure substrate, compared with the underground digester with the same substrate. 3. Economic Analysis a. Based on prevailing costs of fuel, materials, and labor in the Philippines, biogas produced from the household size system is cheaper than either LPG or kerosene. b. If other benefits like organic fertilizer, pollution control and convenience are considered, biogas will surely be the best alternative fuel source.

Impact of Compost Application on Improvement of Rice Productivity and Quality in Reclaimed Soil (간척지 토양에서 퇴비처리가 벼의 생산 및 품질개선에 미치는 영향)

  • This study was conducted to reduce the dependability of farmers on chemical fertilizers for rice cultivation. Soil chemical and biological properties were monitored before experiment and at the time of harvesting. The results showed that EC, available $SiO_2$, and exchangeable $Ca^{2+}$ were decreased at the time of harvesting while pH, OM, and exchangeable $K^+$ and $Mg^{2+}$ were remain unchanged, compared with soil before experiment. Population of aerobic bacteria, Bacillus sp., and fungi were also increased at the time of harvesting in the paddy field, compared with before fertilization, in the treatment of 50% soil-testing fertilizer+ 50% compost. Concentrations of N, P, and K in rice leaves increased with the fertilizers application, maximum increase was recorded in 50% soil-testing fertilizer+ 50% compost. Non-significant difference was observed in the morphological parameters of rice among the treatments. The chlorophyll contents of rice leaf increased in a similar fashion up to 60 days, thereafter, sharp decrease was observed in all the treatments. Maximum yield (per 10a) was recorded in the field treated with 50% soil-testing fertilizer+ 50% compost followed by standard applied fertilizer, 70% soil-testing fertilizer+ 30% compost, soil-testing fertilizer and unfertilized plot. Amylose content showed non-significant difference within the treatments. Protein content increased with the use of fertilizers and best protein content was recorded in the treatment of 50% soil-testing fertilizer+ 50% compost. It was concluded that the amount of the chemical fertilizer used was directly proportional to the protein content of rice grain. However, the palatability of rice grown in unfertilized field was better than the treatments but minimum yield was obtained. Hence, the treatment of 50% soil-testing fertilizer+ 50% compost, was the best among the fertilizer combinations for rice cultivation as supported by the yield, protein and palatability index.

Comparison of Liquefying Efficiency of Mixed Organic Fertilizer as Affected by Aeration Time and the Ratio of Organic Fertilizer to Water (폭기시간과 유기질비료 농도에 따른 혼합유기질비료의 액비화 특성비교)

  • This study was conducted to evaluate the liquefying efficiency of mixed organic fertilizer in different conditions. The organic fertilizer was composed of sesame oil cake, rice bran, fish meal, ground bone meal etc, and made by fermenting process. It included $23g\;kg^{-1}$, $17.0g\;kg^{-1}$, $23.9g\;kg^{-1}$, $290g\;kg^{-1}$ of N, $P_2O_5$, $K_2O$, organic matter, respectively. In one test, the mixed organic fertilizer was added in the proportion of 10% to water 90% and aerated continuously, for 2, 8 hours per day, and not aerated as control. In the other test, ratios of organic fertilizer to water were 5%, 10%, 20% and aerated for 2 hours a day. With the increase of liquefying time, pH, EC and $NH_4-N$ increased without relation to aeration time. After 10 days, liquid organic fertilizer aerated for 2 hours a day contained $634mg\;N\;kg^{-1}$, $68.1mg\;P_2O_5\;kg^{-1}$, $453mg\;K_2O\;kg^{-1}$, which was not significantly different from 8 hours a day or continuous aeration. Then extraction ratios of inorganic contents were 27.6%, 4.0% and 18.9%, respectively. Continuous aeration resulted in increasing the viable number of aerobic bacteria, spore forming bacteria and fungi in liquefied solution. Higher ratio of organic fertilizer to water increased EC, $NH_4-N$ and other inorganic matter contents, but decreased extraction ratio of nutrients in liquid fertilizer. The liquid organic fertilizer of 20% contained $1,140mg\;N\;kg^{-1}$, $35.4mg\;P_2O_5\;kg^{-1}$, $544mg\;K_2O\;kg^{-1}$ after 10 days. Then extraction ratios were 24.8%, 2.4% and 13.6%, respectively. The ratio of organic fertilizer to water was positively correlated with only spore forming bacteria, Pseudomonas spp. among microorganisms.

Effect of 10 Weeks Smart Machine Circulation Exercise on Body Composition, Lung Function, Blood Lipids and Insulin Resistance in Obesity Middle-aged Women (10주간 스마트머신 순환운동이 비만 중년여성의 체조성, 폐기능, 혈중지질 및 인슐린 저항성에 미치는 영향)

  • The purpose of this study was to analyze the effects of smart machine circulation exercise on body composition, lung function, blood lipid and insulin resistance in obesity middle-aged women among 40-60 years by dividing them into a smart machine circulation exercise group(n=8), and control group(n=6). The smart machine circulation exercise program included 55-minutes sessions thrice each week at the following intensities: The strength of aerobic exercise is applied to smart machines by linking the smart machine with the POLAR T31; the 1-4 week is 40-50%HRR, 5-8 week is 50-60%HRR, and 9-10 week is 60-70%HRR. The strength of the resistance exercise was tested using a smart machine based on the constant velocity motion, and then, using the 1-RM data value, applied 40% 1-RM for 1-4 weeks, 60% 1-RM for 5-8 weeks, and 80% 1-RM for 9-10 week. As a results, body composition indicated that weight, BMI, %BF, WHR had a significant interaction effect. Lung function indicated that FVC levels significantly changes in the exercise group and the between groups difference in changes at 10week was significant. Also, FVC and FEV1 significantly showed interaction effect. TC, TG and HDL-C levels significantly changes in smart machine circulation exercise group and the between-group difference in changes after 10 weeks was significant. TC, TG and HDL-C significantly showed interaction effect. Insulin resistance demonstrated that Insulin, Glucose and HOMA-IR levels significantly showed difference over 10 weeks between group. Therefore, the 10 weeks smart machine circulation exercise positively effects on the body composition, lung function, blood lipids, and insulin resistance in obesity middle-aged women and this smart machine circulation exercise can improve their obesity and prevent obesity.

Distribution of foodborne pathogens in red pepper and environment (고추와 재배환경의 식품매개 병원균 분포)

  • This study was performed to investigate the extent of microbial contamination, the presence of enterotoxin genes, and the antibiotic susceptibility of Bacillus cereus in 58 red pepper plants and 43 environmental samples (soil, irrigation water, and gloves) associated with the plant cultivation. The detected counts of total aerobic bacteria, coliform bacteria, Escherichia coli, Bacillus cereus, and Staphylococcus aureus were lower in these samples, as compared to the regulations of standards for foods; moreover, pathogens, such as E. coli, E. coli O157:H7, Listeria monocytogenes, and Salmonella spp., were not detected. Genes encoding hemolysin BL enterotoxins (hblA, hblC, and hblD) as well as non-hemolytic enterotoxins (nheA, nheB, and nheC) were detected in 23 B. cereus specimens that were isolated from the test samples and had β-hemolytic activity. Interestingly, B. cereus is resistant to β-lactam and susceptible to non-β-lactam antibiotics. However, in this case, the isolated B. cereus specimens exhibited a shift from resistant to intermediate in response to cefotaxime and from susceptible to intermediate in case of rifampin, trimethoprim-sulfamethoxazole, vancomycin, clindamycin, and erythromycin. Therefore, the levels of B. cereus should be monitored to detect changes in antibiotic susceptibility and guarantee their safety.
