The aeration of an overtopping wave on a vertical structure generated by a plunging wave was investigated through laboratory measurements of void fraction. The overtopping wave occurring after wave breaking becomes multi-phased and turbulent with significant aeration, so that the void fraction of the flow is of importance. In this study, fiber optic reflectometer and bubble image velocimetry were employed to measure the void fraction, velocity, and layer thickness of the overtopping flow. Mean properties were obtained by ensembleand time-averaging the repeated instantaneous void fractions and velocities. The mean void fractions show that the overtopping wave is very high-aerated near the overtopping wave front and relatively low-aerated near the deck surface and rear free surface of the wave. The flow rate and momentum of the overtopping flow estimated using the measured data show that the void ratio is an important parameter to consider in the multiphase flow. From the similarity profiles of the depth-averaged void fraction, velocity, and layer thickness, one-dimensional empirical equations were obtained and used to estimate the flow rate and momentum of the overtopping flow.
A scaling up study for the exopolysaccharide (EPS) production by submerged culture of Ganoderma lucidum was carried out in jar fermenter systems (2.6, 20 and 75 L) under bi-staged pH process. Profiles of dissolved oxygen (DO) and volumetric coefficient of oxygen transfer ($k_La$) as a function of operating variables (agitation speed and aeration rate) was investigated, and a correlation between $k_La$ and operating variables was analysed statistically. Under bi-staged pH process, no limitation of DO was observed at agitation speeds tested in the range of 200 and 600 rpm, and the highest EPS production was obtained at the level of DO of $40{\sim}80%$. From the regression analysis, the relation between $k_La$, gas velocity (Vs), stirrer speed (N) and impeller diameter (Di) could be expressed as : $$k_La=0.555{\times}Vs^{0.42}{\times}(N^3{\times}Di^2)^{0.33}\;(R^2=0.925,\;p<0.05)$$ It was found that under 2.6 L jar fermenter, the optimum agitation speed and aeration rate was 400 rpm and 1 vvm, respectively, obtaining the EPS production of 15.43 g/L. Under the submerged cultivation of G. lucidum in jar fermenters of $2.6{\sim}75\;L$, the similar EPS yields at each fermenter were achieved during scaling up based on $k_La$, and $k_La$ value for maximum EPS production was $85.4{\pm}26.70\;h^{-1}$.
A simple method to determine viable cell numbers of each species in mixed culture was developed. The oxygen uptake rate (OUR) equals to the product of the specific OUR and the size of the microbial population. In a mixed culture, the OUR is a result of the respiration activities of each sub-population. The OUR was determined from the slope of the linear relationship between time and the decrease of dissolved oxygen concentration when aeration was stopped. The specific OUR was calculated from the slope of the viable cell number versus OUR curve. These values for C. lusitaniae at 20 and $30^{\circ}C$ were $1.36{\times}10^{-9}$ and $3.90{\times}10^{-9}$ and those for P tannoPhilus at 20 and $30^{\circ}C$ were $0.59{\times}10^{-9}$ and $1.86{\times}10^{-9}$ [(%/s)/(cells/ml)J. respectively. Using these values, viable cell numbers were calculated after the OURs of mixed culture at two temperatures were measured. A good agreement between the viable cell numbers determined by this method and by plate count was obtained.
Effect of various fermentation parameter was investigated on the production of prednisolone by microbial $\Delta$'-dehydrogenation of hydrocortisone. The microbial conversion process was conducted by using pseudo-crystallo-fermentation techniques. The optimum temperature for the bioconversion process was found to be 35$^{\circ}C$. It was noted that the production rate of prednisolone was little affected within the initial pH range of 6.5-7.8, and also by the use of surfactant, Tween 80. Production rate of prednisolone was significantly reduced by the use of the antifoam agent, neolin. In a fermentor operation, however, large amount of antifoam agent should be used to remove foams generated by the high aeration rate, which resulted in n lower production rate of prednisolone than that from the shake flask experiment.

Batch cultures were carried out to optimize the exo-polysaccharide production by liquid cultures of Pleurotus ferulae. Among the various carbon sources, when $5\%$ of glucose was used, the maximum mycelial growth and exo-polysaccharide concentration reached were 8.78 g/l and 3.59 g/l, respectively. Yeast extract and polypeptone were identified as the most suitable nitrogen sources. In particular, when a mixture of $1\%$ of polypeptone and $0.8\%$ of yeast extract was used, 9.52 g/l of mycelial growth and 4.09 g/l of exo-polysaccharide were obtained. In the case of mineral sources, K$_2$HPO$_4$ and MgSO$_4$$\codt$7H$_2$0 were found to be the best mineral sources for mycelial growth and exo-polysaccharide production. Under the optimized culture conditions, the agitation speed and aeration were investigated for mycelial growth and exopolysaccharide production in a jar fermentor. The maximum mycelial growth and exo-polysaccharide concentration at 1.5 vvm and 200 rpm obtained were 13.2 g/l and 4.95 g/l, respectively, after 10 days of culture, which were $76\%$ and $79\%$ higher than those of the basal medium. The specific growth rate was decreased with the increase of mycelial growth. However, the specific production rate of the exo-polysaccharide was proportionally increased with the specific growth rate. The proposed model profiles showed good agreement with the experimental results for the mycelial growth and exo-polysaccharide production. The specific production rate using the optimized medium was higher than that of basal medium.
A membrane module including grid was designed and introduced to MBR (membrane bio-reactor) for the purpose of better control of membrane fouling. It could be anticipated that the grid enhances the shear force of fluid-air mixture into the membrane surface by even-distributing the fluid-air to the membrane module. As MLSS concentration, packing density which is expressed in the ratio of the housing and the cross-sectional area of membrane fibers ($A_m/A_t$) and air-flow rate were changed, membrane foulings were checked by monitoring fouling resistances. The total fouling resistance ($R_c+R_f$) without grid installation (i.e., control) was $2.13{\times}10^{12}m^{-1}$, whereas it was reduced to $1.69{\times}10^{12}m^{-1}$ after the grid was installed. Regardless of the grid installation, the $R_c+R_f$ increased as the packing density increased from 0.09 to 0.28, however, the increment of resistance for the grid installation was less than that of the control. Increase in the air flow rate did not always guarantee the reduction of fouling resistance, indicating that the higher air flow rate can partially de-flocculate the activated sludge flocs, which led to severer membrane fouling. Consequently, installation of grids inside the housing have brought a beneficial effect on membrane fouling and optimum air flow rate is important to keep the membrane lowering fouling.
Haematococcus pluvialis was cultured autotrophically and mixotrophically for astaxanthin production in bubble column photobioreactors with $CO_2$ and/or acetate supplements. Four different schemes were tested using 5% $CO_2$ balanced with 95% $N_2$ at the aeration rate of 0.1 vvm and supplementation of acetate (l g/L) to the basal medium, MBBM. Average specific growth rates based on cell total volume of each case were 0.138 $day^{-1}$ in control (without $CO_2$ and acetate supplements), 0.125 $day^{-1}$ with only acetate supplementation, 0.315 $day^{-1}$ with only $CO_2$ supplement, 0.304 $day^{-1}$with both $CO_2$ and acetate. The growth rate under $CO_2$ supply was much higher than that without $CO_2$ supply. DCO and DO seemed to be a possible limitation factor. Another interesting observation was the growth rate under $CO_2$ supplement (without acetate) was higher than that under both $CO_2$ and acetate supply. This result is the opposite of the previously reported works. From the result, the existence of acetate may suppress the dark reaction of $CO_2$ fixation or photosynthesis. Conclusively, the growth of H. pluvialis under photoautotrophic condition found to be higher than that under mixotrophic condition.
In the process of producing chitosan from crustacean shell, the use of excessive acid and alkli is causing the problems of environmental pollution and of production cost. In this study, one way to solve these problems is to cultivate fungi, then, to extract chitosan from the cell wall. By means of flask incubation and batch cultivation, the optimum cultivation conditions for mass production of continuous cultivation was found. Four strains used for the production of fungal chitosan were Gongronella butleri IF08080, Absidia coerulea IF05301, Rhizopus delemar IF04775, Mucor tuberculisporus IF09256. In flask incubation to select strain of producing much chitosan by means of experiment of the effect of initial pH, Absidia coerulea IFO 5301 had highest yield in FCs, 258.1 $\pm$ 47.3 mg/200 $m\ell$l at pH 6.5. In flask incubation under the optimum cultivation condition, temperature 27$^{\circ}C$, culture time 6days, glucose 2%, peptone 1%, (NH$_4$)$_2$ SO$_4$ 0.5%, $K_2$HPO$_4$ 0.1 %, Nacl 0.1 %, MgSO$_4$ㆍ7$H_2O$ 0.05%, CaCl$_2$ㆍ2$H_2O$ 0.01 %, the yield of DCW brought the highest yields. In batch bioreactor, the optimum cultivation condition was that cell suspended solution was 70 $m\ell$, aeration rate 0.5 l/min, agitation rate 800 rpm, culture time 36 hr. In continuous bioreactor, the optimum substrate flow rate was 4 ι/day.
Effects of soil disinfection, fungicide application, and narrow ridge cultivation on ginger rhizome rot development were examined in two naturally-infested fields at Seosan, Choongnam province. Soil disinfection treatments were assigned to main plots, and fungicide and ridge treatments to sub-plots in a split plot design with three replications. The rhizome rot started in late July, and progressed rapidly until late September with the peak incidence in mid-august to early September. Soil disinfection by dazomet application showed the most prominent inhibition effects in both fields, where the disease was reduced by the treatment from 17.5% to 4.8% in one field, and from 51.0% to 2.2% in the other field. Three to five applications of fungicide metalaxyl-copper during the growing season inhibited the disease by 89.7% in one field, but less effectively in the other field. Narrow ridge cultivation reduced the disease effectively by 78.1% and 63.9%, compared to the unridged control plots in each field, respectively. Germination rate of seed-rhizomes and growth of ginger plants were similar between treatments, except when the plots received improper aeration after applying dazomet, and then the germination rate was significantly reduced. The greatest yields were obtained in the disinfected plots, regardless of rhizome rot incidence, except one control plot with very little disease. Ginger yield was negatively correlated with disease severity. However, the yield of ridge plots averaged 58∼59% compared to those of the unridged plots, due mainly to the half planting rate of the ridge plots. In spatial progress, the disease in the disinfected plots started from a single focus of the inoculum, and spread into the adjacent areas only, whereas in the untreated plots, the disease started from many foci that were distributed over the plot, and rapidly progressed to make an epidemic during the season. The soil density of P. myriotylum in the disinfected plots was not changed or, if not, increased slightly during the season. However, in the untreated plots it increased rapidly to reach the density 3 to 5 times greater by the end of the season.
In this study, a process combined biofiltration with sulfur-utilizing autotrophic denitrification and membrane separation was proposed to examine the efficiency of nitrogen removal. As an experimental device, hollow-fiber module was installed in the center of reactor to generate the flux forward sulfur layer in the cylinder packed with granular sulfur. In addition, a simple module was installed in activated sludge aeration tank which inside and outside of sulfur-using denitrification module was covered with microfilter and the module was considered as an alternative of clarifier. The experiment for developing new MBR process was carried out for three years totally. As the results of first two-year experiment, successful nitrogen removal performance was revealed with lab-scale test and pliot scale plant using artificial wastewater and actual plating wastewater. In this year, pilot scale test using actual domestic wastewater was performed to prove field applicability. As the results, high-rate nitrogen removal performance was confirmed with about 0.19 kg ${NO_3}^--N/m^3$ day of rate. Also significant fouling and pressure increase were not found during the experiment. And, the production ratio of sulfate and the consumption ratio of alkalinity showed a slightly higher value about 311 mg ${SO_4}^{2-}/L$ and 369 mg $CaCO_3$/L, respectively. In conclusion, the developed MBR process can be utilized as an alternative for retrofiting existing wastewater plants as well as new construction of advanced sewage wastewater treatment plants, with cost-effective merit.
